Abstract
A quantitative understanding of the constraints on protein evolution is important for many aspects of biology. I describe the use of high-throughput selections followed by deep sequencing to map the constraints at all sites in key proteins of influenza virus. I then explain how the resulting experimental information can be used to define how biophysical constraints and biomedically relevant selection pressures shape the evolution of this virus in nature.
