Abstract

The present study was undertaken to determine whether progesterone and/or its early metabolite(s) associate with macromolecules in the decidual (traumatized) and pseudopregnant rat and rabbit uterus. Under both in vivo and in vivo conditions progesterone-7α- 3H-derived radioactivity binds to 4-5S protein(s) in the uterine cytosol fraction of both species. In addition, it was shown that the decidual rat uterus rapidly converts progesterone to 3α-OH-5α:-pregnan-20-one and that this is the major early metabolite of progesterone in the cytoplasm of this tissue. Both progesterone and 3α-OH-5α-pregnan-20-one bind to 4-5S cytoplasmic protein(s), suggesting that the early metabolite, in addition to progesterone, may play some role in regulating the progestational response of the uterus.
