Abstract

This chapter discusses the locomotor activity of leukemic blasts from patients with acute myeloid leukemia and its relationship with receptors for IgG Fc. In an experiment described in the chapter, receptors for IgG Fc on leukemic blasts were determined using ox RBC coated with IgG anti-ox RBC antibody. Surface immunoglobulins were assayed by using fluorescein isothiocyanate-conjugated F(ab′) 2 antisera monospecific for heavy chain of immunoglobulins. Chemotaxis assay was performed using modified Boyden's chambers. The results of the chemotaxis in acute myeloblastic, acute myelomonocytic, and acute promyelocytic leukemia are discussed in the chapter. Patients 1–6 had few leukemic blasts with IgG receptors. Leukemic blasts from these patients either did not move or moved poorly toward both casein and EAS. No surface immunoglobulin (SIg) was found on the leukemic blasts. Patients 7 and 8 had 78% and 44% blasts that bore IgG receptors but lacked surface SIg. The leukemic blasts from both of these patients moved very well toward chemoattractants. Patient 9 had 28% leukemic blasts with IgG Fc receptors and 15% with SIgG. Patients 10 and 11 with acute myelomonocytic leukemia had few leukemic blasts with IgG Fc receptors, and they moved very poorly toward casein endotoxin activated serum.
