Abstract

Polydeoxythymidylate [poly(dT)] strongly inhibits the translation of reticulocyte globin mRNA in a system derived from Krebs ascites cells. The polynucleotide has little effect on chain elongation by pre-existing reticulocyte or ascites cell polyribosomes, suggesting that it blocks translation primarily at the level of peptide chain initiation. The inhibition appears to be non-competitive with 9-S reticulocyte RNA but can be partially reversed by ascites cell ribosomes or 100 000 × g supernatant fraction.
