Abstract
目的 探讨Ⅰ、Ⅲ型前胶原末端肽在评价试验性高血压大鼠(SHR)颈动脉血管间质胶原重构及逆转中的意义.方法选择16周龄雄性SHR各12只分别为SHR组和厄贝沙坦组(30 mg·kg-1·d-1),另选同龄雄性Wistar大鼠12只为正常对照组;颈总动脉切片采用HE染色、苦味酸--天狼星红(PSR)染色分别检测颈动脉壁厚/腔径比值、胶原面积百分比;采用放免法测定三组血清中PCⅠ、PCⅢ的浓度.结果 SHR组颈动脉壁厚/腔径比值较正常对照组高,厄贝沙坦组治疗20周后,较SHR组下降;SHR组胶原面积百分比较正常对照组高,厄贝沙坦组治疗20周后,较SHR组也有下降;SHR组血清PCⅠ、PCⅢ和PCⅠ/Ⅲ均较正常对照组明显增高,应用厄贝沙坦后PCⅢ下降明显,与正常对照组的差异亦无意义;PCⅠ和PCⅠ/Ⅲ虽有下降,但与正常对照组的差距仍然明显.结论 SHR颈动脉血管存在间质胶原重构,应用厄贝沙坦可以得到一定程度的逆转,血清PCⅠ能同步反映血管胶原面积百分比的变化,能够用于评价试验性高血压大鼠颈动脉血管胶原重构及逆转的变化。
