Abstract
Calf, mouse and guinea pig DNA's have been eluted on methylated albumin kieselguhr and hydroxyapatite columns in order to study the elution patterns of nuclear satellite DNA's and to set up new techniques for their isolation and purification. Fractionation of DNA on these columns depends on the base composition; this is at least partially true in the case of hydroxyapatite if flat and continuous gradients are used. Some satellite DNA's behave abnormally: particularly mouse satellite DNA is eluted before the main DNA on the methylated albumin kieselguhr column and after on the hydroxyapatite column. Using two subsequent methylated albumin kieselguhr columns the two calf satellite DNA's (densities, 1.713 g/ml and 1.721 g/ml) have been obtained in pure form. Also guinea pig satellite DNA's can be isolated from the main DNA, but they were not obtained separated one from the other.
