Abstract

Objective To investigate the effects of rosiglitazone,a ligand of peroxisome proliferators activated receptor gamma (PPAR-γ),on the expression of matrix metalloproteinase (MMP)-2 and MMP-9 in hepatic stellate cells (HSCs).Methods HSCs were cultured in medium containing different concentrations of rosiglitazone (5,10,15,20 μmol/L respectively),and harvested at 24 h.The expression of MMP-2 and MMP-9 was detected by using reverse transcription polymerase chain reaction.Results The MMP-2 expression in HSCs cultured with different concentrations of rosiglitazone (5,10,15,20 μmol/L)was 0.5708 ±0.0609,0.8900 ±0.0823,1.1348 ±0.1205 and 1.4490 ±0.0832 respectively,and it in control group was 0.3237 ± 0.0796.The MMP-9 expression in HSCs cultured with different concentrations of rosiglitazone (5,10,15,20 μmol/L) was 0.5487 ±0.0770,0.7554 ±0.0510,0.9497 ±0.0451 and 1.1088 ±0.0777 respectively,and it in control group was 0.3220 ±0.0592.Conclusion PPAR-γ ligand rosiglitazone may up-regulate the expression of MMP-2 and MMP-9 in HSCs dose-dependently to some extent.
