Abstract

Objective To observe the effects of photodynamic therapy (PDT) on proliferation and apoptosis of human breast carcinoma cell line MCF-7 in vitro.Methods MCF-7 cells were treated with PSD-007 for 2 h under the influence of low-level laser (635 nm) therapy at different doses.Then the optical density (OD) values and survival rates of MCF-7 cells were measured by MTT assay at different time (0.5,1,3,6,12,24 h) after PDT.The nucleus morphological changes of the MCF-7 cells stained by DAPI were observed under the reversal fluorescence microscope.The apoptosis was analyzed using flow cytometry through Annexin/PI double-labeled staining.Results PSD-007 mediated PDT inhibited the proliferation of MCF-7 cells,which depended on laser power and time after PDT.The morphological study proved that the multinucleate giant cells existed in the apoptotic cells in PSD-007 PDT.Flow cytometry of Annexin/PI double-labeled staining verified the results as well.Conclusion PDT inhibit the proliferation of MCF-7 cells in vitro by inducing apoptosis,and the results show an effect of time and dose dependence.
