Abstract

Objective: This study aims to investigate the effect of Twist1 on human breast carcinoma E-cadherin. Methods: pcDNA-Twist1 was transfected into human breast carcinoma cell line MCF-7. The expression of E-cadherin was detected via real-time polymerase chain reaction and immunofluorescence staining. Wound-healing assay was performed to examine the migration ability of cells, and the 3D culture was used to observe vasculogenic mimicry (VM) formation. Results: MCF-7 cells overexpressing Twist1 resulted in the suppression of E-cadherin expression and abnormal subcellular location. Migration ability and VM formation were also enhanced. Conclusion: Twist1 prevented E-cadherin expression as well as promoted migration ability and VM formation in human breast carcinoma cell line.
