Abstract
In this brief overview, I summarize my career in NMR from 1980 to the present. I describe the following developments that were made in my laboratory: the transferred NOE for probing the conformation of ligands bound to large proteins, assignment and structure determination of DNA and RNA, restrained molecular dynamics and simulated annealing for macromolecular structure determination, protein structure determination of small proteins, 3-D and 4-D heteronuclear NMR, structure determination of large proteins and complexes including both protein–DNA and protein–protein complexes up to ∼150 kDa, and finally methods based on paramagnetic NMR to probe NMR invisible, sparsely populated states of macromolecules.
