Abstract

This chapter provides an overview of cleaning of glassware and apparatus. All glassware containing bacterial cultures or otherwise contaminated should be sterilized by autoclaving for 20 min at 121°C, which will also liquefy any solid media and allow easy removal. Plastic Petri dishes and disposable pipettes and bottles should be sterilized by autoclaving for 20 min at 121°C before discarding. Microscope slides and coverslips having been discarded into jars of disinfectant on the work bench can be disposed of without further sterilization. The sterilization of culture media, containers and instruments is essential in bacteriological work for the isolation and maintenance of pure cultures. Bacteriological tests for sterility may be used to confirm the efficiency of the procedures used.
