Abstract

Summary Monoclonal antibodies (McAbs) have been produced which discriminate between the two major protein components of the PSI reaction centre designated the P 700 chl a-protein (PSI-AI and PSI-A2). Rats were immunised with PSI enriched preparations from lettuce (Lactuca sativa L.) leaves. Antibody-secreting cultures were selected which bind to both «native» and SDS-denatured PSI preparations in an enzyme-linked immunosorbent assay (ELISA) and were subsequently screened using electroblotted PSI preparations after SDS-PAGE electrophoresis. McAbs were selected which bind to polypeptides with apparent m wt 60-66 kD. These McAbs react specifically with 60-66 kD polypeptides in crude membrane preparations from lettuce leaves. MAC 305 binds to the slower migrating polypeptide, molecular weight approximately 66 kD, while MAC 304, MAC 306 and MAC 307 bind to a polypeptide, molecular weight approximately 60 kD. The McAbs MAC 305 and MAC 304 also cross react with polypeptides from wheat and pea.
