Abstract

This chapter covers the First-Generation and Next-Generation (NextGen) methods for sequencing DNA. It includes discussions of dideoxy and “shotgun” sequencing strategies. This is followed by a section on the analysis of the large amount of data produced by DNA sequencing projects, and the data banks that exist to store the information. There is an analysis of the history and methods of the Drosophila Genome Project and its importance for the Human Genome Project. This is followed by a discussion of NextGen sequencing methods, including third-generation sequencing. The problems associated with defining genes, and finding them within a sequenced genome are discussed. The chapter concludes with a discussion of the “Post-Genomic Era,” including the Earth Bio-Genome Project.
