Abstract
Since the lens is under anaerobic conditions, the pentose phosphate cycle is important for energy production from glucose. Glucose-6-phosphate dehydrogenase (G6PDH) is the rate-limiting enzyme and determines the production of reduced nicotinamide adenine dinucleotide phosphate (NADPH) and ribose-5-phosphate (R5P) from the pentose phosphate cycle. NADPH is a regulator for antioxidation and R5P is essential for nucleic acid synthesis. 18S ribosomal RNA (18S rRNA) is the active center of protein synthesis in the 40S ribosomal subunit. The messenger RNA (mRNA) levels of G6PDH and 18S rRNA are good indicators for evaluating oxidative status and the effects of antioxidants. In this investigation, the mRNA levels of G6PDH and 18S rRNA in the anterior capsule of cataractous lens increased after intake of an antioxidant supplement that included lutein, β-carotene, multivitamins, and trace elements. The increases were more significant among female patients. These data indicate that intake of an antioxidant supplement can upregulate the pentose phosphate cycle and may further regulate antioxidation in the lens.
