Publisher Summary This chapter discusses various in vivo methods. Rabbit intraductal method is inexact but convenient method because 6–8 determinations can be performed on the same rabbit. Rabbit intracerebral test is not quantitative, but it is convenient for the localization of prolactin-inhibiting or releasing areas and for the testing of the lactogenic activity of unknown substance. Milk yield test in post-partum rats test is not specific to exogenous prolactin because of surplus of endogenous prolactin. Systemic pigeon crop gland weight method is of no practical use. A major problem in the local assay methods is the occurrence of non-specific inflammatory or other reactions, resulting from the injection of tissue extracts or body fluids. The experienced investigator can, in most cases, distinguish between a true prolactin response and a purely inflammatory reaction.
