Abstract

Pichia pastoris are single-celled fungi and have long been used for heterologous protein production that can grow to a high cellular density at an industrial level. Despite the prevailing circumstances of its elevated protein productivity, the streamlining of P. pastoris refinement is still imperative due to strain and product-specific tasks such as promoter strength, expression system, methanol utilization type, beta-galactosidase super secretion (BGS secretion), bioreactor cultivation, systems biology, genomics, transcriptomics, proteomics, metabolomics, and metabolic modeling. In this book chapter, we summarized all these parameters along with integrated omics analysis approaches collaborating with multiple studies of P. pastoris. P. pastoris is becoming an indispensable platform with these combined engineering strategies. Understanding the parameter of the P. pastoris promoter and the development of novel cultivation strategies that include the substrate and product and metabolic engineering are appraised here.
