Abstract
ABSTRACT Aim: To evaluate the IL-35 Gene Polymorphism in Periodontitis subjects with and without type 2 diabetes mellitus. Methodology: 96 participants were categorized into three groups, Group 1: Systemically and Periodontally healthy individuals, Group 2: Systemically healthy patients with Generalized chronic periodontitis, Group 3: Generalized chronic periodontitis with type 2 diabetes Mellitus. Genomic DNA was extracted from the whole venous blood samples collected from the participants using The PureLink Genomic Kit. The region spanning rs1580809257 and rs1580801731 were amplified using appropriate primer and reaction conditions. Determination of genotypes for each of the polymorphisms assessed using Amplification Refractory Mutation System (ARMS) - based PCR and Restriction Fragment Length Polymorphism (RFLP). The distribution of genotype and allele frequencies for Case and control groups were compared using the Chi-square test. Hardy Weinberg equilibrium is calculated. Results: The gene and allele frequency were found to be similar in all three groups. Conclusion: The study reveals that polymorphism of rs1580809257 and rs1580801731 in the IL-35 gene is neither associated with Periodontitis patients nor Periodontitis with Diabetes patients.
