Abstract

This study aims to investigate the effect of celery leaf infusa on decreasing level of blood glucose in rat (Rattus norvegicus). Twenty five rats were divided into 5 groups namely first treatment group (P1) as negative control. Second treatment group (P2) as positive control, rats were induced with alloxan. Third treatment group (P3) rats were induced with alloxan and given 5% celery leaf infusa. Fourth treatment group (P4) rats were induced with alloxan and given 10% celery leaf infusa. Fifth treatment group (P5) rats were induced alloxan and given 15% celery leaf infusa for 14 days. The level of blood glucose of the rat was determined before treatment, after given aloksan and after given celery leaf infusa. Data were analyzed using Analysis of Varians (ANOVA). Results showed that administration of celery leaf infusa for 14 days, show significant effect to decreasing level of glucose in rat (P0.01). It can be concluded that the administration of celery leaf infusa concentrated 5%, 10% and 15% for 14 days show significant effect to decreased level of glucose in rats.

MATERI DAN METODE

Penelitian ini merupakan penelitian eksperimental dengan menggunakan Rancangan Acak Lengkap Faktorial (RAL Faktorial) yang dibagi atas 5 kelompok dengan 5 kali ulangan. Dua puluh lima ekor tikus putih jantan diadaptasi selama 1 minggu dengan memberikan pakan standar dan air minum secara ad libitum. Seledri dicuci bersih dan dipotong kasar, dan direbus dengan 100 ml air dalam panci infusa selama 15 menit terhitung mulai suhu mencapai 90oC, sambil sekali-sekali diaduk. Setelah dingin, disaring menggunakan kain flannel dan ditambahkan air secukupnya melalui ampas sampai diperoleh volume 100 ml, maka akan diperoleh infusa daun seledri sebanyak 100 ml dengan konsentrasi 10%. Untuk konsentrasi 5% dan 15%, 5 gram dan 15 gram daun seledri masing-masing direbus dengan 100 ml air (Depkes, 2009). Kadar glukosa darah diukur dengan menggunakan Gluco-Dr setelah masa adaptasi, setelah pemberian aloksan dan setelah pemberian infusa daun seledri. Analisis Data Data penurunan kadar glukosa darah dianalisis dengan menggunakan Analysis of Variance (ANOVA) Rancangan Acak Lengkap Faktorial (RAL Faktorial). Bila terdapat pengaruh perlakuan terhadap kadar glukosa darah maka dilanjutkan dengan Uji Jarak Ganda Duncan (Gaspersz, 1989)
