Abstract

Publisher Summary This chapter discusses the preparation and determination of gluconic, 2-ketogluconic, and 5-ketogluconic acid. To D-glucose dissolved in Na 2 CO 3 , a solution of KI is added, which is incubated at 30 ° for 30 minutes. Further, CO 2 -free nitrogen gas is bubbled through the mixture for 1 hour at 30 ° to remove free I 2 . Iodide and iodate ions are removed by passing the solution through an anion exchange resin such as Dowex 50-H + . Finally, gluconic acid is eluted from the column with 0.05 N H 2 SO 4 . D-Gluconic acid may be determined by measuring the change in rotation because of formation of the molybdate complex, or enzymatically by measuring the extent of DPN reduction coupled with the conversion of the gluconic acid to 6-phosphogluconic acid. Although both methods are described here, the enzymatic method is recommended. In addition, manometric methods have also been described.
