Abstract

The problem of developing conditions most efficient cloning primary mesenchymal stem cells is extremely important. The important part of culture media containing growth factors, cell populations, is a blood serum of animals. It continues to be an indispensable component of the substratum for most cell cultures. The value of serum for cell growth, detection components that stimulate proliferation is the subject of interest of many researchers. We know that the most commonly used for this purpose fetal bovine serum, but its widespread use is limited, primarily, the lack and high cost. So, search replace fetal bovine serum cheaper sera are promising. Our studies show that the composition of the substrate has a significant impact on the speed of colony forming ability of bone marrow stem cells of rabbit 0 and 1 passages and their proliferative activity. Thus, in the experiment with partial replacement of fetal bovine serum to horse serum, the first cell colonies began to appear already on 5 - 6th day of cultivation, while in control Petri dishes - only on 6 -7 th day. Note that the colonies in culture dishes cells with partial replacement of fetal bovine serum to horse serum were much more numerous and evenly placed throughout the area of the bottom of the cups, in comparison with culture dishes cells with fetal bovine serum. The increase clonogenic and proliferative activity of undifferentiated mesenchymal stem cells in bone marrow of the rabbit, obviously, it occurs due to the presence in horse serum cytokines, that is removed from the dormant state of the cultured cells at 0 passage. Also activate markers of proliferation of these cells, which in turn proves the possibility of partial replacement of fetal bovine serum to horse serum.
