Investigation of the Occurrence of Zoonotic Intestinal Parasites along the Karmanasa River Bank in Lalitpur, Nepal.

Increasing urbanization has particularly affected rivers and their outer edges in cities, including Kathmandu Valley, which encompasses Lalitpur, the nation's third-largest city. This study aims to conduct a parasitological survey to investigate the occurrence of zoonotic intestinal protozoa and helminths along the Karmanasa River bank in central Nepal. Faecal samples from openly defaecating animals were collected via non-invasive techniques, and coproscopy was carried out using direct wet mount, concentration and acid-fast staining methods to ensure reliable findings. The findings showed that all the faecal samples were positive for intestinal parasites as follows: buffaloes (15/15), cats (5/5), cattle (30/30), chickens (7/7), dogs (15/15), goats (15/15), pigs (20/20) and rats (12/12). A total of 28 intestinal parasites were reported, out of which 21 species possess zoonotic potentialities, and each host was reported to harbour at least one zoonotic parasite. Entamoeba sp., Cryptosporidium sp., hookworm, Trichuris sp., Trichostrongylus and Balantidium coli were among the predominant zoonotic parasites. The use of the landscape for livestock grazing and the presence of free-ranging animals could have all added to the zoonotic risks. Moreover, the excretion of a moderate to large number of zoonotic parasite eggs in the faecal samples indicates a transmission risk. The study detected 21 species of intestinal zoonotic parasites circulating along the landscape in the study area, indicating a higher risk of cross-transmission. Therefore, strategic treatment of livestock and free-ranging animals and periodic public health awareness programs for the local inhabitants are highly recommended.

Prevalence and genotypic characterization of zoonotic intestinal protozoan parasites in transboundary migratory Mongolian Gazelles (Procapra gutturosa): the first survey.

The migration of Mongolian gazelles (Procapra gutturosa) poses a potential risk of outbreak for zoonotic intestinal protozoan parasite infections. This study aims to investigate the infection status of zoonotic intestinal protozoan parasites in these migratory Mongolian gazelles. We collected 120 fecal samples from Mongolian gazelles during their migration from Mongolia to China in December 2023. These samples were analysed using amplification and sequencing of partial SSU rRNA genes to detect the 4 presence of zoonotic intestinal protozoan parasites and characterize their genotypes. Our analysis revealed the presence of several zoonotic intestinal protozoan parasites in the sampled Mongolian gazelles. Cryptosporidium spp. was detected in 14.17% (17/120) of the samples, followed by Cystoisospora belli in 13.33% (16/120), Blastocystis sp. in 16.67% (20/120) and Cyclospora cayetanensis in 30.00% (36/120). Moreover, we identified novel host-adapted genotypes of Cryptosporidium spp. and C. belli, as well as the presence of ST2 and ST13 Blastocystis sp. subtypes, while distinct genotypes were found in Blastocystis sp. and C. cayetanensis. This study revealed the status of 4 prevalent zoonotic intestinal protozoan parasite infections in Mongolian gazelles and provided crucial insights into their characteristics. The prevalence of these parasites in the population highlights the potential risk of cross-border transmission of infectious diseases associated with long-distance migration. Furthermore, the identification of novel genotypes contributes to our understanding of the genetic diversity and adaptation of these parasites. These findings can inform the development of protective measures to mitigate the impact of these infections on the health and survival of Mongolian gazelles.

Chromosome-level genome assembly of Cryptosporidium parvum by long-read sequencing of ten oocysts

Cryptosporidium parvum is a zoonotic parasite of the intestine and poses a threat to human and animal health. However, it is difficult to obtain a large number of oocysts for genome sequencing using in vitro culture. To address this challenge, we employed the strategy of whole-genome amplification of 10 oocysts followed by long-read sequencing and obtained a high-quality genome assembly of C. parvum IIdA19G1 subtype isolated from a pre-weaning calf with diarrhea. The assembled genome was 9.13 Mb long and encompassed eight chromosomes with six capped by telomeric sequences at one or both ends. In total, 3,915 protein-coding genes were predicted, exhibiting a high completeness with 98.2% single-copy BUSCO genes. To our current knowledge, this represents the first chromosome-level genome assembly of C. parvum achieved through the combined use of whole-genome amplification of 10 oocysts and long-read sequencing. This achievement not only advances our understanding of the genomic landscape of this zoonotic intestinal parasite, but also provides valuable resources for comparative genomics and evolutionary analyses within the Cryptosporidium clade.

Prevalence and Genetic Characterization of Giardia duodenalis and Blastocystis spp. in Black Goats in Shanxi Province, North China: From a Public Health Perspective.

Blastocystis spp. and Giardia duodenalis are two prevalent zoonotic intestinal parasites that can cause severe diarrhea and intestinal diseases in humans and many animals. Black goat (Capra hircus) farming is increasingly important in China due to the remarkable adaptability, high reproductive performance, rapid growth rate, and significant economic value of black goats. A number of studies have indicated that black goats are the potential reservoir of multiple zoonotic protozoans in China; however, the prevalence and zoonotic status of G. duodenalis and Blastocystis spp. in black goats in Shanxi Province is still unknown. Thus, a total of 1200 fecal samples of black goats were collected from several representative regions at different altitudes in Shanxi Province and were examined for the presence and genotypes of G. duodenallis and Blastocystis spp. by amplifying the beta-giardin (bg), glutamate dehydrogenase (gdh), and triosephosphate isomerase (tpi) loci of G. duodenalis and SSU rRNA of Blastocystis spp. using PCR and sequence analysis methods, respectively. The overall prevalence of G. duodenalis and Blastocystis spp. in black goats in Shanxi Province were 7.5% and 3.5%, respectively. Two assemblages (B and E) of G. duodenalis and four subtypes (ST5, ST10, ST14, and ST30) of Blastocystis spp. were identified, with assemblage E and ST10 as the prevalent genotype and subtype in black goats, respectively. One novel multilocus genotype (MLG) was identified in MLG-E and was designated as MLG-E12. For both G. duodenalis and Blastocystis spp., the prevalence was significantly related to the region and age groups (p < 0.05). This is the first report on the prevalence of G. duodenalis and Blastocystis spp. in black goats in Shanxi Province. These results not only provide baseline data for the prevention and control of both parasites in black goats in Shanxi Province, but also enhance our understanding of the genetic composition and zoonotic potential of these two parasites.

Cryptosporidium spp. in large-scale sheep farms in China: prevalence and genetic diversity

Cryptosporidium spp. are significant zoonotic intestinal parasites that induce diarrhea and even death across most vertebrates, including humans. Previous studies showed that sheep are important hosts for Cryptosporidium and that its distribution in sheep is influenced by geography, feeding patterns, age, and season. Environmental factors also influence the transmission of Cryptosporidium. Molecular studies of Cryptosporidium in sheep have been conducted in only a few regions of China, and studies into the effect of sheep-housing environments on Cryptosporidium transmission are even rarer. To detect the prevalence of Cryptosporidium in large-scale sheep-housing farms, a total of 1241 fecal samples were collected from sheep, 727 environmental samples were taken from sheep housing, and 30 water samples were collected in six regions of China. To ascertain the existence of the parasite and identify the species of Cryptosporidium spp., we conducted nested PCR amplification of DNA extracted from all samples using the small-subunit (SSU) rRNA gene as a target. For a more in-depth analysis of Cryptosporidium spp. subtypes, C. xiaoi-and C. ubiquitum-positive samples underwent separate nested PCR amplification targeting the 60 kDa glycoprotein (gp60) gene. The amplification of the Cryptosporidium spp. SSU rRNA gene locus from the whole genomic DNA of all samples yielded a positive rate of 1.2% (20/1241) in fecal samples, 0.1% (1/727) in environmental samples, and no positive samples were found in water samples. The prevalence of Cryptosporidium spp. infection in large-scale housed sheep was 1.7%, which was higher than that in free-ranging sheep (0.0%). The highest prevalence of infection was found in weaning lambs (6.8%). Among the different seasons, the peaks were found in the fall and winter. The most prevalent species were C. xiaoi and C. ubiquitum, with the former accounting for the majority of infections. The distribution of C. xiaoi subtypes was diverse, with XXIIIc (n = 1), XXIIId (n = 2), XXIIIe (n = 2), and XXIIIl (n = 4) identified. In contrast, only one subtype, XIIa (n = 9), was found in C. ubiquitum. In this study, C. xiaoi and C. ubiquitum were found to be the predominant species, and Cryptosporidium was found to be present in the environment. These findings provide an important foundation for the comprehensive prevention and management of Cryptosporidium in intensively reared sheep. Furthermore, by elucidating the prevalence of Cryptosporidium in sheep and its potential role in environmental transmission, this study deepens our understanding of the intricate interactions between animal health, environmental contamination, and public health dynamics.

Occurrence and genotypic identification of Blastocystis sp., Enterocytozoon bieneusi, and Giardia duodenalis in dairy cattle in Heilongjiang Province, China

Blastocystis sp., Enterocytozoon bieneusi, and Giardia duodenalis are three common zoonotic intestinal parasites, and cattle are important hosts of these three intestinal protozoa. In this study, 1632 fecal samples were collected from dairy farms in Heilongjiang Province, China, and screened for Blastocystis sp., E. bieneusi, and G. duodenalis using polymerase chain reaction. Of these, 149 (9.13%) were positive for three zoonotic pathogens, including 104 (6.40%), 22 (1.35%), and 23 (1.41%) for Blastocystis sp., E. bieneusi, and G. duodenalis, respectively. Based on partial SSU rRNA gene sequencing analysis, 104 positive samples of Blastocystis sp. were found, and a total of nine known subtypes were identified, including ST10 (61), ST3 (18), ST14 (6), ST26 (7), ST24 (3), ST25 (2), ST1 (2), ST5 (2), and ST21 (1). Among these, three subtypes (ST1, ST3, and ST5) were recognized as zoonotic subtypes, and two subtypes (ST10 and ST14) were specific to animals. All 23 Giardia duodenalis-positive samples belonged to assemblage E (n = 23) based on sequenced beta-giardin (bg) and triosephosphate isomerase (tpi) genes. Three known genotypes of E. bieneusi, namely J (n = 9), I (n = 6), and BEB4 (n = 7), were identified by sequence analysis of the internal transcriptional spacer region gene. Our study provides basic data for prevention and control in Heilongjiang Province; however, further research is required to better understand the prevalence and public health significance of these pathogens in the Heilongjiang region.

Prevalence of Zoonotic Intestinal Parasites in Meat Goats in Southern Thailand.

Background: Intestinal parasites not only lead to poor goat health and productivity but can also affect human health. We investigated the distribution of zoonotic intestinal parasites among goats in southern Thailand to assess the human health risks. Materials and Methods: Overall, 561 fecal samples were collected randomly from goats in farms (55) in Songkhla, Satun, Pattalung, Pattani, Yala, and Narathiwat provinces between January 2020 to January 2022. Parasite eggs were detected via the standard protocol formalin ether concentration technique (FECT). Results: Samples containing one or more intestinal parasites accounted for 97.33% (543/561) of the total samples, and the percentage of zoonotic parasite was 80.04% (449/561). Among the positive samples, two types of zoonotic helminths were identified, Strongylid egg type (79.32%) and Fasciola spp. (2.14%). Strongylid egg type was highly abundant in samples from Satun (84.62%), Songkhla (82.74%), Pattalung (79.31%), and Yala (77.27%). Fasciola spp. was mostly found in samples form Pattalung (13.79%) and Songkhla (1.12%). Eimeria spp. was the mostly identified protozoa 25.67%. It was mostly identified in samples from Narathiwat (75%), Pattani (41.9%), and Songkhla (31.41%). Mixed infections accounted for 40.82% of the total with 33 patterns. It was found that Strongylid egg types occurred in 28 patterns (84.85%). Strongylid egg types-Eimeria spp.-pattern was the most commonly found. Conclusion: Based on the results, zoonotic helminth infection was found together with widespread and high diversity of intestinal parasites infection in meat goats, which affect animal health and may pose a risk to human health. Mass deworming programs with a proper anthelmintic drugs and good agricultural practices need to be encouraged by the government so that people who contact with goats should practice good hygiene behavior. Farm management should be applied by intensive goat raising with pasture circulating to reduce parasite egg contamination. Moreover, fresh fertilizers from goat faces should be avoided.

Intestinal Helminth Infections Among Domesticated Cats in Malate, Manila, Philippines

Intestinal helminthiasis is a public health issue in developing nations particularly those which have insufficient access to clean water, sanitary infrastructures, and lacks public health education. Due to the zoonotic potential of some intestinal helminths, cats, and other mammalian species with direct contact to humans may act as reservoir hosts and provide a medium for the transmission of zoonotic infections. This study aimed to determine the prevalence of intestinal helminths among domesticated cats in Malate, Manila, Philippines. A total of 25 cat stools were collected, processed using Formol-Ether Concentration Technique (FECT) and were microscopically examined. The overall prevalence of intestinal helminths was 76% (19/25). The intestinal helminths identified include roundworms of phylum Nematoda: Toxocara cati (44%) and Ascaris spp. (20%), whipworm: Trichuris spp. (24%), and hookworm (12%). All of the identified intestinal parasites have zoonotic potential and domestic cats are significant reservoirs of zoonotic intestinal parasites that can facilitate the transmission of infection to humans. Therefore, an innovative one-health approach strategy which includes constant monitoring and control of stray and feral cats, access to potable water, public health education, and more sanitary infrastructures, can help resolve intestinal helminthiasis crisis in the Philippines.

Occurrence and Genotypic Identification of Blastocystis spp., Enterocytozoon bieneusi, and Giardia duodenalis in Leizhou Black Goats in Zhanjiang City, Guangdong Province, China.

Blastocystis spp., Enterocytozoon bieneusi, and Giardia duodenalis are three common zoonotic intestinal parasites that cause severe diarrhea and enteric diseases. Leizhou black goats are characterized by a high reproductive rate, fast growth, and good meat quality, making them one of the pre-eminent goat breeds in China. Goats are reportedly common reservoirs of these three intestinal pathogens, but no information on their prevalence or genotypic distributions in black goats in Guangdong Province, China, is available. A total of 226 fecal samples were collected from goats in Zhanjiang city and genomic DNA was extracted from them. The presence of the three pathogens was detected using nested PCR targeting the sequences encoding SSU rRNA (Blastocystis spp.), the internal transcribed spacer of rRNA (ITS; E. bieneusi), as well as beta-giardin, glutamate dehydrogenase, and triosephosphate isomerase (G. duodenalis). All PCR products were sequenced to determine the species and genotypes of the organisms. The total prevalence rates of Blastocystis spp., E. bieneusi, and G. duodenalis were 33.63% (76/226), 17.70% (40/226), and 24.78% (56/226), respectively. Four subtypes of Blastocystis spp. were detected: ST5 (n = 6), ST10 (n = 50), ST14 (n = 14), and ST21 (n = 6). Among them, ST10 was the dominant genotype, accounting for 65.79% of strains, followed by the genotypes ST14 (18.42%), zoonotic ST5 (7.89%), and ST21 (7.89%). Four genotypes of E. bieneusi were detected: CHG3 (n = 32), CM21 (n = 4), CHG1 (n = 2), and ET-L2 (n = 2). Among these, CHG3 was the dominant genotype. Assemblage E (n = 54) and concurrent assemblages A and E (n = 2) were identified in the G. duodenalis-positive goats using multilocus genotyping. Blastocystis spp., E. bieneusi, and G. duodenalis infections were common in Leizhou black goats, all of which have zoonotic genotypes, indicating the potential risk of zoonotic transmission. Our results provide basic data for the prevention and control of these three intestinal pathogens. Further studies are required to better understand their genetic characteristics and zoonotic potential in Guangdong Province.

The Intensity of Infection and Public Health Perception of Potentially Zoonotic Intestinal Parasites of Dogs in Kwara Central, Nigeria

Background: The close relationship between dogs and humans and the possibility of intestinal parasite transmission from dogs to humans calls for frequent assessment of these potential zoonotic intestinal parasites in dogs and the possibility of their transmission to humans. Objectives: This study aimed to determine the presence, intensity of infection, and public health perception of potentially zoonotic intestinal parasites in dogs of Kwara Central, Nigeria. Methods: The study was conducted in 28 locations in Kwara Central Senatorial District of Kwara State, Nigeria. Three hundred and five healthy dogs were sampled. Two hundred and thirty respondents (dog owners or handlers) were questioned using a well-structured questionnaire containing open-ended and closed-ended questions. Fecal samples from the sampled dogs were subjected to the direct fecal smear technique, simple fecal centrifugation flotation technique, formalin-ethyl acetate concentration technique, and the modified Ziehl-Neelsen staining technique. Oocysts or eggs per gram of feces were counted using the modified McMaster technique. Results: Seven different intestinal parasites (Ancylostoma spp., Cryptosporidium spp., Dipylidium caninum, Isospora spp., Strongyloides stercoralis, Toxocara spp., and Uncinaria stenocephala) were detected, with a prevalence ranging from 2.30% to 25.25%. Of these parasites, 6 were zoonotic. The mean intensities of infection were 91.43 eggs per gram (EPG) for D. caninum¸ 96.52 EPG for S. stercoralis, 129.36 ±28.12 oocysts per gram (OPG) for Isospora spp., 165.17±19.88 for Toxocara spp., 240.00±44.42 for EPG for U. stenocephala, and 303.64±31.83 EPG for Ancylostoma spp. Some dog owners and handlers were not cautious about possible zoonotic parasite transmission from dogs. Conclusion: Zoonotic intestinal parasites of dogs are present and prevalent in Kwara Central, Nigeria. There is a need to educate the public on the possibility of zoonotic parasite transmission to humans.

Giardia duodenalis-induced G0/G1 intestinal epithelial cell cycle arrest and apoptosis involve activation of endoplasmic reticulum stress in vitro.

Giardia duodenalis is a zoonotic intestinal protozoan parasite that may cause host diarrhea and chronic gastroenteritis, resulting in great economic losses annually and representing a significant public health burden across the world. However, thus far, our knowledge on the pathogenesis of Giardia and the related host cell responses is still extensively limited. The aim of this study is to assess the role of endoplasmic reticulum (ER) stress in regulating G0/G1 cell cycle arrest and apoptosis during in vitro infection of intestinal epithelial cells (IECs) with Giardia. The results showed that the mRNA levels of ER chaperone proteins and ER-associated degradation genes were increased and the expression levels of the main unfolded protein response (UPR)-related proteins (GRP78, p-PERK, ATF4, CHOP, p-IRE1, XBP1s and ATF6) were increased upon Giardia exposure. In addition, cell cycle arrest was determined to be induced by UPR signaling pathways (IRE1, PERK and ATF6) through upregulation of p21 and p27 levels and promotion of E2F1-RB complex formation. Upregulation of p21 and p27 expression was shown to be related to Ufd1-Skp2 signaling. Therefore, the cell cycle arrest was induced by ER stress when infected with Giardia. Furthermore, the apoptosis of the host cell was also assessed after exposure to Giardia. The results indicated that apoptosis would be promoted by UPR signaling (PERK and ATF6), but would be suppressed by the hyperphosphorylation of AKT and hypophosphorylation of JNK that were modulated by IRE1 pathway. Taken together, both of the cell cycle arrest and apoptosis of IECs induced by Giardia exposure involved the activation of the UPR signaling. The findings of this study will deepen our understanding of the pathogenesis of Giardia and the associated regulatory network.

The prevalence of potentially zoonotic intestinal parasites in dogs and cats in Moscow, Russia.

This study was aimed to determine the prevalence of Toxocara canis/cati, Strongyloides stercoralis, Giardia spp., and Cryptosporidium spp., which occur and are potentially zoonotic to humans in domestic dogs and cats in Moscow (Russia). The fecal flotation method and larvae detection by microscopy of a direct feces smear were performed to detect Toxocara, Giardia spp., and Cryptosporidium spp. The total parasitic prevalence in dogs was as follows: Giardia spp.: 10.2 % (226/2208), Cryptosporidium spp.: 2.7 % (60/2208), T. canis: 2 % (45/2208), S. stercoralis larvae: 1.1 % (25/2208). The younger animals under were infected more than those over 12 months of age (p<0.001). The preva lence rates were along these lines: Giardia spp. (18.2 %), Cryptosporidium spp. (5.7 %), T.canis (3 %), S. stercoralis larvae (2.3 %). The overall prevalence in cats was as follows: Giardia spp. - 5.2 % (71/1350), Cryptosporidium spp. - 4.8 % (65/1350), T. cati - 4.1 % (56/1350). Similarly to dogs, the infection rates were higher in cats under 12 months of age Giardia spp. (8.2 %), Cryptosporidium spp. (8.6 %), T. cati (7.5 %. Analysis of combined infections in dogs revealed the following combinations: Giardia spp. and Cryptosporidium spp. (35.5 %) larvae of S. stercoralis sp. and Giardia spp. (32.3 %), T.canis and Giardia spp. (22.6 %), T.canis and Cryptosporidium spp. (6.6 %), T.canis and S.stercoralis and (3.2 %), respectively. In cats, only two coinfections by Giardia spp. and Cryptosporidium spp. (58.3 %), and T.cati with Giardia spp. (41.7 %) were noticed. Further research is needed to study the spread of parasitic diseases in pet animals. The data will improve countermeasures to prevent these diseases' spread among animals and humans.

Infection with Cryptosporidium parvum Affects Secondary Sexual Characteristics of Male Mice by Altering the Pheromone Content in Preputial Gland

The olfactory acuity of female mice allows them to discriminate the urinary odors of males. Parasitic infection can reduce the odor attractiveness of male mice to females and result in female aversion or avoidance responses in odor selection. However, the chemical signaling changes in the pheromone contents produced by the foreskin gland were not fully revealed after parasitic infection. Cryptosporidium parvum (C. parvum) is a common zoonotic intestinal parasite and has a wide range of hosts, including human, domestic animals, and wild animals. In this study, we immunosuppressed ICR/CD-1 male mice by dexamethasone sodium phosphate treatment. After C. parvum infection, physiological indexes such as body weight and organ weight were significantly decreased. Furthermore, the gene expression level of MUP (major urinary protein) in liver and urine were significantly down-regulated, which could be the reason for the decrease in urine attractiveness to females. GC-MS was performed to analyze the changes in the pheromone produced by the preputial gland before and after parasitic infection, and the results indicated that the levels of different pheromones were significantly reduced after parasitic infection. In summary, this study reveals that C. parvum infection damages the secondary sexual characteristics of male ICR/CD-1 male mice and decreases the pheromone content produced by the foreskin gland.

Molecular characterization and prevalence of Cryptosporidium spp. in sheep and goats in western Inner Mongolia, China

Cryptosporidium spp. are zoonotic intestinal parasites that infect fish, birds, reptiles and mammals. Cryptosporidium spp. are common cause of diarrhea. In this study, a total of 1032 fecal samples were collected from the rectums of sheep and goats. The samples were analyzed using nested polymerase chain reaction (nested PCR) based on the small subunit ribosomal RNA (SSU rRNA) gene of Cryptosporidium spp. The average infection rate of Cryptosporidium spp. was 2.23% (n = 23), and three Cryptosporidium species were identified, namely Cryptosporidium ubiquitum (8/23), Cryptosporidium andersoni (5/23) and Cryptosporidium xiaoi (10/23). Subtyping of C. ubiquitum and C. xiaoi was carried out by DNA sequence analysis of the 60-kDa glycoprotein (gp60) gene. Eight C. ubiquitum isolates were identified as zoonotic subtype XIIa. Nine C. xiaoi isolates were identified as subtypes XXIIIc (n = 1), XXIIIf (n = 3) and XXIIIg (n = 5). Subtype XXIIIg was first found in Chinese sheep. C. ubiquitum subtype XIIa was found in both sheep and goats, suggesting that sheep and goats are important sources of C. ubiquitum infections.

Prevalence, risk factors and zoonotic potential of intestinal parasites in dogs from four locations in Morocco.

Dogs can harbor various intestinal parasites that have serious clinical, economic, and zoonotic impact. In Morocco, the epidemiological status of those parasites is largely unknown. This study aimed to obtain data on the prevalence of intestinal parasites in various Moroccan dog populations, to identify associated risk factors, to evaluate people's knowledge regarding zoonotic parasites and to estimate the risk of human infection. A total of 291 fecal samples were analyzed using a 33% Zinc Sulphate (ZnSO4) centrifugal flotation and a sheather's sugar simple flotation techniques. In addition, 100 dog owners were asked to fill out a questionnaire about their knowledge regarding canine zoonotic intestinal parasites. Overall, 58% of sampled dogs were positive for at least one parasite species. Ancylostoma/Uncinaria spp. (31.9%), Toxascaris leonina (27.4%), Toxocara canis (27.1%), Cystoisospora spp. (13.4%) and Giardia spp. (7.2%) were the most frequently isolated parasites. The overall prevalence was associated with the dogs' activity, and feces consistency. More specifically, the prevalence of Ancylostoma/Uncinaria spp., Toxascaris leonina and Cystoisospora spp. was associated with dogs' activity and feces consistency and, additionally with age for Cystoisosporaspp.Toxocara canis prevalence was associated with age and feces consistency, while Giardia spp. prevalence was associated with dogs' activity. A weak awareness among dog owners regarding the zoonotic potential of canine intestinal parasites was noticed (33%) especially when their knowledge was compared to another endemic zoonotic disease, i.e. rabies (85%). Furthermore, the rate of respecting the vaccination protocol (82%) was significantly higher than the rate of respecting the deworming protocol (47%). We conclude that intestinal parasites, including the zoonotic ones, were highly prevalent in the dogs sampled in this study whereas people's knowledge about those parasites was very limited. These findings suggest that the risk of human infection is very likely, which highlights the need for effective control programs and health education.

Prevalence and genetic diversity of Giardia duodenalis in pet dogs from Zhengzhou, central China and the association between gut microbiota and fecal characteristics during infection

As a common zoonotic intestinal parasite, Giardia duodenalis could infect humans and various mammals worldwide, including pet dogs, leading to giardiasis. This study detected the infection of G. duodenalis in asymptomatic pet dogs in Zhengzhou, and evaluated the possibility of zoonosis and the relationship between gut microbiota and fecal characteristics. We randomly collected 448 fresh fecal samples from Zhengzhou, and G. duodenalis was screened based on the beta-giardin (bg), glutamate dehydrogenase (gdh), and triose phosphate isomerase (tpi) genes. The difference of gut microbiota between five G. duodenalis-positive and five G. duodenalis-negative samples was investigated by 16S rRNA gene sequencing. The overall prevalence of G. duodenalis was 7.1% (32/448) based on bg, gdh, and tpi locus, two G. duodenalis assemblages (C = 13, D = 14) and five (15.6%) mixed infection (C + D) were identified. Moreover, compared with the G. duodenalis-negative group, the diversity of gut microbiota increased in G. duodenalis-positive group. The decrease of Lactobacillus spp. and considerable increase of Prevotella spp. were associated with the fecal characteristics. These results show that the transmission of zoonotic giardiasis between humans and pet dogs is rare in Zhengzhou, central China, and support the use of Lactobacillus spp. as a potential probiotic agent to improve intestinal health in dogs, or even humans, by treating G. duodenalis. Therefore, the public health significance of G. duodenalis to humans, companion animals, and the environment should be further evaluated from One Health perspective.

Detection of zoonotic-borne parasites in Rattus spp. in Klang Valley, Malaysia

Background and Aim:Rattus spp. are the most common animals capable of adapting to their environment. They can be reservoirs or vectors of diseases that facilitate the transmission of zoonotic-borne parasites to humans. Hence, a study on the detection of parasites in rat populations in urban areas is crucial to prepare for emerging zoonosis. Therefore, this study aims to identify blood parasites, ectoparasites, and helminths in Rattus spp. from wet markets located in Klang Valley, an urban area with a high-density human population.Materials and Methods:A total of 32 rats were trapped in several wet markets in Klang Valley, Malaysia. They were anesthetized for morphometric examination followed by exsanguination. Various parasitological techniques such as perianal tape test, simple flotation, direct examination of the intestine, and fecal smear were performed for intestinal parasite detection; hair plucking, skin scraping, and full body combing for ectoparasite identification; and blood smear, microhematocrit centrifugation, and buffy coat techniques for blood parasite detection.Results:The rats were identified as Rattus rattus (71.9%) and Rattus norvegicus (28.1%). The only blood protozoan found was Trypanosoma lewisi. The ectoparasites identified belonged to two broad groups, mites (Laelaps spp. and Ornithonyssus spp.) and fleas (Xenopsylla cheopis), known to be parasitic zoonotic disease vectors. The zoonotic intestinal parasites were cestodes (Hymenolepis nana), nematodes (Nippostrongylus brasiliensis, Strongyloides spp., Trichuris spp., Capillaria spp., and Syphacia spp.), and intestinal protozoa (coccidian oocysts and Giardia spp.). Microscopic images showing Giardia spp. are the first report of this organism in rats in Malaysia.Conclusion:Rats caught in this urban area of the Klang Valley harbor parasites can pose a potential zoonotic threat to humans, raising public health concerns because of their proximity to densely populated urban areas.

Zoonotic intestinal parasites in free-ranging dogs (Canis lupus familiaris): a risk to public health in a Mexican Protected Area

Domestic dogs (Canis lupus familiaris) have been in contact with humans for thousands of years, playing an important role in societies. Nonetheless, the lack of responsible ownership has contributed to the transition from companion dogs to free-ranging or feral dogs that can be reservoirs of zoonotic parasites. Our goal was to identify zoonotic intestinal parasites in free-ranging dogs in a Mexican Protected Area. A total of 132 scat samples from free-ranging dogs were collected and examined using the Faust flotation technique. We identified a total of nine parasite species, four platyhelminthes, and five nematodes. Eight of nine identified parasite are zoonotic. The most frequent zoonotic parasites are Ancylostoma caninum and Ascaris spp. (19.7% each) followed by Toxascaris leonina (17.4%) and Uncinaria stenocephala (7.6%). The least frequent are Dipylidium caninum (2.2%), Capillaria spp., Hymenolepis diminuta, and Hymenolepis nana (0.75% each). This study provides the first description of intestinal zoonotic parasites richness in free-ranging dogs within a Mexican Protected Area. The presence of zoonotic parasites in canine scats represents a high risk to public health, mainly for the transmission of some species through cutaneous and visceral migrans larvae, especially in infants and kids. We recommend specific measures to prevent, control and mitigate the presence of free-ranging dogs in Protected Areas.

Cryptosporidium and Giardia Infection in Pet Animals: A review Study in Iraq

Cryptosporidium parvum and Giardia lamblia are the most common zoonotic intestinal protozoan parasites causing diarrheal diseases Cryptosporidiosis and Giardiasis are significant health concerns in both humans and animals; for this reason, these parasites acquired public and veterinary importance. Dogs and cats are susceptible to infection and play an important role in transmitting these protozoans to human. Recently in Iraq, there is an interest in keeping pet animals and an increase of stray cats and dogs which accompany livestock. Some people started to attract stray cats through feeding and became like house animals. This literature review focused on the results of Iraqi researchers to overview the importance prevalence of these parasites on both human and animals. This literature review recommended to educate pet owners especially children and follow veterinarians’ instructions, to control and prevent transmission of these zoonotic diseases and future researches should focus and investigate from these finding and highlight the role of domestic animals in transmission of these diseases.

Prevalence of Helminths Parasites among Dogs and Risk Factors of Zoonotic Infections by Dog Owners in Bende Local Government Area, Abia State, Nigeria

Dogs, as the most common pet animal worldwide, habours a wide range of intestinal parasites with zoonotic potential, thus raising serious concern over public health. This study presents the intestinal helminthes in dogs and risk factors associated with dog owners in Bende Local Government Area of Abia State, Nigeria. The dogs’ faecal samples were examined using formal-ether sedimentation techniques. Questionnaire was administered to elicit responses from the dog owners. A total of 110 faecal samples collected randomly from dogs were processed and examined for helminthes eggs. The overall prevalence of gastrointestinal helminthes was 39.1%. Prevalence was higher in Bende-Ovu 39.7% than Umuoshie 38.1%. There was no statistical relationship found in both communities (P&gt;0.05). The four species of parasites identified were Ancylostoma caninum (44.2%), Toxocara canis (30.2%), Diphylidium caninum (23.3%) and Trichuris vulpis (2.3%) with Ancylostoma caninum occurring most 59.2% in Bende-Ovu and Toxocara canis (56.2%) in Umuoshie. This was statistically significant (P&lt;0.05). Basenji breeds had the highest infection (51.1%) while Greyhound breed had the least prevalence (2.3%). Female (47.8%) were more infected in Bende-Ovu than male (35.6%) while in Umusohie, male (39.1%) were more infected than the female (36.8%). Prevalence pattern was age dependent in the two communities but this was statistically insignificant (P&gt;0.05). There is greater statistical significance (P&lt;0.05) in the prevalence of breed in relation to sex of the dogs in Bende-Ovu (39.7%) as compared with dogs in Umuoshie (38.1%; P&gt;0.05). Age related prevalence in the breed of dogs’ in Bende-Ovu and Umuoshie were not statistically significant (P&gt;0.05). Questionnaire analyses revealed that majority (64.6%) of the dog owners are not aware of canine zoonosis. Most (58.4%) of them don’t take diseases transmitted by dogs seriously. The trend of the breed and species prevalence of zoonotic intestinal parasites of dogs observed in this study constitutes public health problems in the study area. Therefore, intervention measures such as health education should be provided to dog owners to reduce the risk of transmission of parasites from dogs to humans.
