Or the caveman who discovered that a stone hammer could crack a skull as well as a nut. Saade et al. recognized an opportunity in the variable difficulty of releasing proteins from “His-tag” complexes. The affinity of His-tagged proteins for Ni-agarose is very high and displacing the tags requires imidazole. If the pH is in the right range, tagged proteins are not removed by organic solvents, high ionic strength, or denaturants. Thus, if only one member of a complex is His-tagged, other members can be eluted with common methods until the last ones are removed by a pH shift or imidazole. (Is this a case of a silk purse from a sow's ear?). Csk: The>150 x Go-To guy pp. 4944–4961 C-terminal Src kinase, Csk, must be an extremely busy protein – handling tasks in signal transduction and cell communication and cell growth and maintenance (previously recognized) to the regulation of nucleic acid and nucleobase metabolism, protein metabolism, energy pathways, transport, and biogenesis and organization of cells (newly discovered by Yang et al.). These researchers turned up over 150 unique partners of Csk using MALDI-TOF, shotgun LC-MS/MS, immunofluorescence, and other proteomic techniques. Some were direct, some indirect as judged by the presence of one or more Csk-SH2-binding domains. Src Family Kinases, SFKs are normally negatively controlled by Csk. Over-expression and over-activation of SFKs have been associated with cancerous tumors of the colon, liver, pancreas, stomach, breast, ovaries and lung. Separate the wheat from the chaff before feeding to analyst pp. 4978–4984 Looking in a haystack for a needle is the proverbial impossible task. If you know that you are looking for a steel needle, however, it suddenly becomes much easier – you can use a large magnet to preprocess the haystack. Collect all the ferrous material from the haystack, then search through that for the needle. Renard et al. applied the principle to peptide identification in MS/MS data sets from human (2 types), yeast, and mouse cell lysates. It takes some work to make it work, however. Various relatively simple spectrum preprocessing routines are built into the most popular peptide identification software packages and may not function well following the stand-alone preprocessing package unless both are optimized to work together.
