Variability in CitXET expression and XET activity in Citrus cultivar Huangguogan seedlings with differed degrees of etiolation.

Considering the known effects of xyloglucan endotransglycosylase (XET) on plant growth and development, we aimed to determine whether XETs help to regulate the growth and elongation of Huangguogan shoots and roots. We confirmed a possible role for XET during seedling etiolation. Our results revealed that the roots of etiolated seedlings (H-E) were longer than those of green seedlings (H-G). However, shoot length exhibited the opposite pattern. We also observed positive and negative effects on the xyloglucan-degrading activity of XET in the root sub-apical region and shoots of etiolated Huangguogan seedling, respectively. There was a significant down-regulation in CitXET expression in the etiolated shoots at 15 days after seed germination. On the contrary, it was significantly increased in the root sub-apical region of etiolated and multicolored seedlings at 15 days after seed germination. The XET coding sequence (i.e., CitXET) was cloned from Huangguogan seedlings using gene-specific primers. The encoded amino acid sequence was predicted by using bioinformatics-based methods. The 990-bp CitXET gene was highly homologous to other XET genes. The CitXET protein was predicted to contain 319 amino acids, with a molecular mass of 37.45 kDa and an isoelectric point of 9.05. The predicted molecular formula was C1724H2548N448O466S14, and the resulting protein included only one transmembrane structure. The CitXET secondary structure consisted of four main structures (i.e., 21% α-helix, 30.72% extended strand, 9.09% β-turn, and 39.18% random coil). Analyses involving the NCBI Conserved Domains Database (NCBI-CDD), InterPro, and ScanProsite revealed that CitXET was a member of the glycosyl hydrolase family 16 (GH16), and included the DEIDFEFLG motif. Our results indicate that the differed degrees of etiolation influenced the CitXET expression pattern and XET activity in Huangguogan seedlings. The differential changes in XET activity and CitXET expression levels in Huangguogan seedlings may influence the regulation of root and shoot development, and may be important for seedling etiolation.

DkXTH8, a novel xyloglucan endotransglucosylase/hydrolase in persimmon, alters cell wall structure and promotes leaf senescence and fruit postharvest softening.

Fruit softening is mainly associated with cell wall structural modifications, and members of the xyloglucan endotransglucosylase/hydrolase (XTH) family are key enzymes involved in cleaving and re-joining xyloglucan in the cell wall. In this work, we isolated a new XTH gene, DkXTH8, from persimmon fruit. Transcriptional profiling revealed that DkXTH8 peaked during dramatic fruit softening, and expression of DkXTH8 was stimulated by propylene and abscisic acid but suppressed by gibberellic acid and 1-MCP. Transient expression assays in onion epidermal cells indicated direct localization of DkXTH8 to the cell wall via its signal peptide. When expressed in vitro, the recombinant DkXTH8 protein exhibited strict xyloglucan endotransglycosylase activity, whereas no xyloglucan endohydrolase activity was observed. Furthermore, overexpression of DkXTH8 resulted in increased leaf senescence coupled with higher electrolyte leakage in Arabidopsis and faster fruit ripening and softening rates in tomato. Most importantly, transgenic plants overexpressing DkXTH8 displayed more irregular and twisted cells due to cell wall restructuring, resulting in wider interstitial spaces with less compact cells. We suggest that DkXTH8 expression causes cells to be easily destroyed, increases membrane permeability and cell peroxidation, and accelerates leaf senescence and fruit softening in transgenic plants.

Analysis of xyloglucan endotransglycosylase/hydrolase (XTH) genes and diverse roles of isoenzymes during persimmon fruit development and postharvest softening.

Xyloglucan endotransglycosylase/hydrolase (XTH) enzymes have played a role in the remodeling of cell wall hemicelluloses. To investigate the function of XTHs in persimmon (Diospyros kaki L.) fruit development and postharvest softening, five cDNAs (DkXTH1 to DkXTH5), whose putative proteins contained the conserved DEIDFEFLG motif of XTH, were cloned. Real time quantitative PCR analysis revealed that DkXTH1, DkXTH4, and DkXTH5 peaked in immature expanding fruit, and their higher expression was observed along with higher fruit firmness in cold-treated fruit or firmer cultivar fruit during storage. The opposite gene expression patterns were observed in DkXTH2 and DkXTH3, which reached maxima concomitance with pronounced fruit softening. Meanwhile, the xyloglucan endotransglycosylase (XET) enzymes play important roles in both the rapid growth and ripening of persimmon fruit. Furthermore, the recombined DkXTH1 and DkXTH2 proteins showed significant XET activity without any detected XEH activity. However, the XET activity of recombined DkXTH2 protein had a higher affinity for small acceptor molecules than that of recombined DkXTH1 protein. The former might prefer to participate in cell wall restructuring, and the latter is more inclined to participate in cell wall assembly. Besides, DKXTH proteins could function by targeting to the cell wall under regulation of a signal peptide. The data suggested that individual DKXTHs could exhibit different patterns of expression, and the encoded products possessed specific enzymatic properties conferring on their respective functions in growth and postharvest softening of persimmon fruit.

Group III-A XTH genes of Arabidopsis encode predominant xyloglucan endohydrolases that are dispensable for normal growth.

The molecular basis of primary wall extension endures as one of the central enigmas in plant cell morphogenesis. Classical cell wall models suggest that xyloglucan endo-transglycosylase activity is the primary catalyst (together with expansins) of controlled cell wall loosening through the transient cleavage and religation of xyloglucan-cellulose cross links. The genome of Arabidopsis (Arabidopsis thaliana) contains 33 phylogenetically diverse XYLOGLUCAN ENDO-TRANSGLYCOSYLASE/HYDROLASE (XTH) gene products, two of which were predicted to be predominant xyloglucan endohydrolases due to clustering into group III-A. Enzyme kinetic analysis of recombinant AtXTH31 confirmed this prediction and indicated that this enzyme had similar catalytic properties to the nasturtium (Tropaeolum majus) xyloglucanase1 responsible for storage xyloglucan hydrolysis during germination. Global analysis of Genevestigator data indicated that AtXTH31 and the paralogous AtXTH32 were abundantly expressed in expanding tissues. Microscopy analysis, utilizing the resorufin β-glycoside of the xyloglucan oligosaccharide XXXG as an in situ probe, indicated significant xyloglucan endohydrolase activity in specific regions of both roots and hypocotyls, in good correlation with transcriptomic data. Moreover, this hydrolytic activity was essentially completely eliminated in AtXTH31/AtXTH32 double knockout lines. However, single and double knockout lines, as well as individual overexpressing lines, of AtXTH31 and AtXTH32 did not demonstrate significant growth or developmental phenotypes. These results suggest that although xyloglucan polysaccharide hydrolysis occurs in parallel with primary wall expansion, morphological effects are subtle or may be compensated by other mechanisms. We hypothesize that there is likely to be an interplay between these xyloglucan endohydrolases and recently discovered apoplastic exo-glycosidases in the hydrolytic modification of matrix xyloglucans.

Structural basis for the action of xyloglucan endotransglycosylases/hydrolases: Insights from homology modeling

Xyloglucan endotransglycosylase/hydrolases (XTH) are believed to play an important role in modifying the cell wall structure through two different, but related actions: the cleavage of a cross-linking xyloglucan polymer (xyloglucan endohydrolase (XEH) activity) and transfer of a newly generated end to another sugar polymer (xyloglucan endotransglycosylase (XET) activity). These enzymes normally show predominantly either XET or XEH. Question remains concerning the origin of the XET or XEH activity of poplar (Populus trichocarpa) XTH proteins as well as the 3-dimensional structural features that might contribute to the different activities for different phylogenetic groups. Previous investigations have demonstrated that the key structural features controlling the activity may involve the loop 1 and 2 regions of the XTH enzymes. Here we report homology models for 40 poplar (Populus trichocarpa) XTH proteins and analyze their loop 1 and 2 regions. These analyses allow us to generate some hypotheses concerning the possible activities of these enzymes. The results show that the protein models for subfamilies (SFs) I/II/IV match well with that of the known XET enzyme (Ptt-XET16-34 from a Populus hybrid), suggesting that they might mainly function as the XET. It is found that the SF III-A models match well with that of the known XEH TmXTH1. Therefore, they may function as XEH as well. Although the SF III-B members are found to have a truncated loop 2, comparison of the homology models with the existing TmXTH1 structure seems to suggest that they might have relatively high possibility to function as XEH instead of XET.

Xyloglucan Endo-transglycosylase (XET) Functions in Gelatinous Layers of Tension Wood Fibers in Poplar—A Glimpse into the Mechanism of the Balancing Act of Trees

Tension wood is a specialized tissue of deciduous trees that functions in bending woody stems to optimize their position in space. Tension wood fibers that develop on one side of the stem have an increased potency to shrink compared with fibers on the opposite side, thus creating a bending moment. It is believed that the gelatinous (G) cell wall layer containing almost pure cellulose of tension wood fibers is pivotal to their shrinking. By analyzing saccharide composition and linkage in isolated G-layers of poplar, we found that they contain some matrix components in addition to cellulose, of which xyloglucan is the most abundant. Xyloglucan, xyloglucan endo-transglycosylase (XET) activity and xyloglucan endo-transglycosylase/hydrolase (XTH) gene products were detected in developing G-layers by labeling using CCRC-M1 monoclonal antibody, in situ incorporation of XXXG-SR and the polyclonal antibody to poplar PttXET16-34, respectively, indicating that xyloglucan is incorporated into the G-layer during its development. Moreover, several XTH transcripts were altered and were generally up-regulated in developing tension wood compared with normal wood. In mature G-fibers, XTH gene products were detected in the G-layers while the XET activity was evident in the adjacent S(2) wall layer. We propose that XET activity is essential for G-fiber shrinking by repairing xyloglucan cross-links between G- and S(2)-layers and thus maintaining their contact. Surprisingly, XTH gene products and XET activity persisted in mature G-fibers for several years, suggesting that the enzyme functions after cell death repairing the cross-links as they are being broken during the shrinking process.

Patterns of enzymatic activity of cell wall-modifying enzymes during growth and ripening of apples

Fruit softening is thought to result from extensive cell wall modifications that occur during ripening. These modifications are the result, at least in part, of the activity of members of cell wall-modifying enzymes from the same families involved in the cell wall loosening which promote tissue extension and growth. In this work, the activities of a set of pectolytic and non-pectolytic cell wall-modifying enzymes, namely polygalacturonase (PG; endo-and exo-acting), pectin methylesterase (PME), pectate lyase (PL), β-galactosidase (β-Gal), α- l-arabinofuranosidase (AFase), endo-1,4-β-glucanase (EGase), xyloglucan endotransglycosylase (XET) and expansin, were monitored during growth and ripening of ‘Mondial Gala’ apple ( Malus × domestica Borkh.) fruit. After optimisation of protein extraction protocols and standard activity assays, activity could be detected in all the assays, except for endo-PG. The overall results suggest that fruit growth and ripening are possibly coordinated by members of the same families of cell wall-modifying enzymes, although different isoforms may be involved in distinct developmental processes. Based on the trend of total activity measured in vitro using equal amounts of protein per developmental stage, the role of EGase seems to be more prominent during growth than during ripening, and XET activity is most important only after the fruit stopped growing and is maintained throughout ripening. β-Gal and AFase activities increased after harvest as the fruit became over-ripe. On the other hand, exo-PG, PL and expansin activities increase from that in unripe fruit to fruit at harvest but are maintained at similar levels thereafter, throughout the over-ripe stages. The patterns of activity observed are discussed in relation to published information about ripening of apples and to results reported using other species.

Analysis of a xyloglucan endotransglycosylase/hydrolase (XTH) from the lycopodiophyte Selaginella kraussiana suggests that XTH sequence characteristics and function are highly conserved during the evolution of vascular plants

A tissue print followed by a xyloglucan endotransglycosylase assay revealed that XET activity is present at sites of cell elongation in both roots and shoots of the lycopodiophyte Selaginella kraussiana. This paper provides the first report and analysis of a xyloglucan endotransglycosylase/hydrolase (XTH) cDNA sequence, isolated from a club moss. In silico analysis of the deduced amino acid sequence revealed a strong conservation of the XET-domain described in higher plants. The catalytic site (DEIDLEFLG) varies in only one amino acid compared with the consensus sequence and was shown to be functional after recombinant expression of Sk-XTH1 in Pichia pastoris. Sk-XTH1 displays xyloglucan endotransglycosylase activity over a broad pH (4.5-7.5) and temperature range (4-30 degrees C), but it shows no hydrolase activity. The catalytic site is followed by a consensus sequence for N-linked glycosylation. Four terminal cysteines were shown to stabilize a putative XET-C terminal extension region, which includes conserved amino acids, involved in the recognition and binding of the substrates. The N-linked sugar interactions as well as the disulphide bridges were shown to be necessary to perform XET activity. The presence of a highly conserved XTH sequence and function in a microphyllophyte suggests that XTHs were present before the divergence of lycopodiophytes and euphyllophytes. It also points to a possible key role for XTHs in the production of a cell wall that allowed the further evolution of land plants.

Production of poplar xyloglucan endotransglycosylase using the methylotrophic yeast Pichia pastoris

The gene XET16A encoding the enzyme xyloglucan endotransglycosylase (XET) from hybrid aspen (Populus tremula x tremuloides Mich) was transformed into Pichia pastoris GS115 and the enzyme was secreted to the medium. The influence of process conditions on the XET production, activity, and proteolytic degradation were examined. Inactivation of XET occurred in the foam, but could be decreased significantly by using an efficient antifoam. Rich medium (yeast extract plus peptone) was needed for product accumulation, but not for growth. The proteolytic degradation of the enzyme in the medium was substantially decreased by also adding yeast extract and peptone to the glycerol medium before induction with methanol. Decreasing the fermentation pH from 5.0 to 4.0 further reduced the proteolysis. The specific activity was further improved by production at 15 degrees C instead of 22 degrees C. In this way a XET production of 54 mg/L active enzyme could be achieved in the process with a specific activity of 18 Unit/mg protein after a downstream process including centrifugation, micro- and ultrafiltration, and ion exchange chromatography.

Organization of cell walls in Sandersonia aurantiaca floral tissue.

Visual symptoms of the onset of senescence in Sandersonia aurantiaca flowers begin with fading of flower colour and wilting of the tissue. When fully senescent, the flowers form a papery shell that remains attached to the plant. The cell walls of these flowers have been examined to determine whether there are wall modifications associated with the late stages of expansion and subsequent senescence-related wilting. Changes in the average molecular size of pectin were limited through flower opening and senescence, although there was a loss of neutral sugar-containing side-branches from pectins in opening flowers, and the total amounts of pectin and cellulose continued to rise in cell walls of fully senescent sandersonia flowers. Xyloglucan endotransglycosylase activity increased in opening and mature flowers, but declined sharply as flowers wilted. Concomitantly, the proportion of hemicellulose polymers of increasing molecular weight increased from flower expansion up to the point at which wilting occurred. Approximately 50% of the non-cellulosic neutral sugar in mature flower cell walls was galactose, primarily located in an insoluble cell wall fraction. Total galactose in this fraction increased per flower with maturity, then declined at the onset of wilting. Beta-galactosidase activity was low in expanding tepals, but increased as flowers matured and wilted.

Release of complexed xyloglucan endotransglycosylase (XET) from plant cell walls by a transglycosylation reaction with xyloglucan-derived oligosaccharides

Incubation of isolated NaCl-washed cell walls from epicotyls of pea ( Pisum sativum) and nasturtium ( Tropaeolum majus) with solutions of various oligosaccharides released among others the cell wall marker enzyme xyloglucan endotransglycosylase (XET, EC 2.4.1.207). The greatest release of XET occurred upon incubation of the cell walls with xyloglucan-derived oligosaccharides (XGOS, DP 7-9). Concomitantly, reduced radioactive nonasaccharide 〚 3H〛-XLLGol (Gal 2.Xyl 3.Glc 3.〚1- 3H〛-glucitol) was incorporated into the cell walls. Subsequent hydrolysis of the radioactively labelled cell walls with Trichoderma cellulase liberated XGOS-alditols, DP 7-9 as the sole radioactive products indicating that 〚 3H〛-XLLGol was incorporated into the cell wall xyloglucan by transglycosylation, as an entity. Oligosaccharides of cello-, chito- and/or oligoglucurono-series were much less effective than XGOS but a substantial liberation of XET and other proteins from plant cell walls could be achieved by the nucleophile 0.1 M imidazole. The specific release of the cell wall-associated XET activity by incubation with xyloglucan-derived oligosaccharides and the simultaneous incorporation of the tritiated xyloglucan nonasaccharide en bloc into the cell walls indicates that XET is present in the cell walls in form of a competent glycosyl-enzyme complex which decomposes by transglycosylation of its glycan moiety to added xyloglucan-oligosaccharide acceptors. This finding suggests a new concept for the regulation of activity of cell wall-associated glycanases/transglycosylases: they exist in plant cell walls in a transiently latent state as covalent glycosyl-enzyme complexes and are active only when suitable glycosyl acceptors become available.

Characterization of a tomato xyloglucan endotransglycosylase gene that is down-regulated by auxin in etiolated hypocotyls.

The reorganization of the cellulose-xyloglucan matrix is proposed to serve as an important mechanism in the control of strength and extensibility of the plant primary cell wall. One of the key enzymes associated with xyloglucan metabolism is xyloglucan endotransglycosylase (XET), which catalyzes the endocleavage and religation of xyloglucan molecules. As with other plant species, XETs are encoded by a gene family in tomato (Lycopersicon esculentum cv T5). In a previous study, we demonstrated that the tomato XET gene LeEXT was abundantly expressed in the rapidly expanding region of the etiolated hypocotyl and was induced to higher levels by auxin. Here, we report the identification of a new tomato XET gene, LeXET2, that shows a different spatial expression and diametrically opposite pattern of auxin regulation from LeEXT. LeXET2 was expressed more abundantly in the mature nonelongating regions of the hypocotyl, and its mRNA abundance decreased dramatically following auxin treatment of etiolated hypocotyl segments. Analysis of the effect of several plant hormones on LeXET2 expression revealed that the inhibition of LeXET2 mRNA accumulation also occurred with cytokinin treatment. LeXET2 mRNA levels increased significantly in hypocotyl segments treated with gibberellin, but this increase could be prevented by adding auxin or cytokinin to the incubation media. Recombinant LeXET2 protein obtained by heterologous expression in Pichia pastoris exhibited greater XET activity against xyloglucan from tomato than that from three other species. The opposite patterns of expression and differential auxin regulation of LeXET2 and LeEXT suggest that they encode XETs with distinct roles during plant growth and development.

Leaf area is stimulated in Populus by free air CO2 enrichment (POPFACE), through increased cell expansion and production

ABSTRACTThe effects of free‐air CO2 enrichment (FACE) on leaf growth in Populus, was studied. For the first time in field conditions, both the production and expansion of leaf cells were shown to be sensitive to atmospheric carbon dioxide. Leaf area expansion rate and final leaf size were stimulated under FACE for three species (Populus x euramericana (I‐214), P. nigra (Jean Pourtet) and P. alba (2AS‐11), with the largest effect observed for P. x euramericana (61%). In this species and in P. nigra, both epidermal cell size and cell number were increased, whereas for P. alba, only cell production was increased in FACE. Two findings suggest that changes in the cell wall may be important in explaining larger leaf cells in FACE: (i) Leaf cell wall extensibility of rapidly growing leaves increased in all species in FACE; and (ii) an increase in xyloglucan endotransglycosylase activity, a cell wall‐loosening enzyme, was increased in FACE and associated with leaf growth rate. The results suggest that the mechanisms by which FACE promotes leaf growth differ, depending on species. Despite this, increases in final leaf size provide an important component driving increased biomass accumulation in POPFACE, during this first year of rapid growth, prior to canopy closure. The question as to whether these effects are the result of a direct response to CO2, or are driven indirectly through substrate availability remains unresolved, although evidence from the literature suggests that the latter mechanism is most likely.

Functional characterisation of Nicotiana tabacum xyloglucan endotransglycosylase (NtXET-1): generation of transgenic tobacco plants and changes in cell wall xyloglucan.

To study the function of xyloglucan endotransglycosylase (XET) in vivo we isolated, a tomato (Lycopersicon esculentum Mill.) XET cDNA (GenBank AA824986) from the homologous tobacco (Nicotiana tabacum L.) clone named NtXET-1 (Accession no. D86730). The expression pattern revealed highest levels of NtXET-1 mRNA in organs highly enriched in vascular tissue. The levels of NtXET-1 mRNA decreased in midribs with increasing age of leaves. Increasing leaf age was correlated with an increase in the average molecular weight (MW) of xyloglucan (XG) and a decrease in the relative growth rates of leaves. Transgenic tobacco plants with reduced levels of XET activity were created to further study the biochemical consequences of reduced levels of NtXET-1 expression. In two independent lines, total XET activity could be reduced by 56% and 37%, respectively, in midribs of tobacco plants transformed with an antisense construct. The decreased activity led to an increase in the average MW of XG by at least 20%. These two lines of evidence argue for NtXET-1 being involved in the incorporation of small XG molecules into the cell wall by transglycosylation. Reducing the incorporation of small XG molecules will result in a shift towards a higher average MW. The observed reduction in NtXET-1 expression and increase in the MW of XG in older leaves might be associated with strengthening of cell walls by reduced turnover and hydrolysis of XG.

Xyloglucan endotransglycosylase: a role after growth cessation in harvested asparagus

Little is known about the mode of xyloglucan endotransglycosylase (XET) activity in cell walls once the turgor, which drives expansion, is reduced. Such a situation exists when growing shoots are excised from the parent plant, and is the case for many commercially valuable vegetable crops, e.g. asparagus, Asparagus officinalis L. XET activity was present in all zones of rapidly growing, immature asparagus spears, but with highest levels at the spear base where elongation growth had ceased. Activity increased in all parts of the spear for up to 72 h after harvest. Two members of the XET-related gene family in asparagus (AoXET1 and AoXET2) were isolated and mRNA corresponding to these clones accumulated at low levels, particularly in the basal zone during spear growth. Transcript levels increased in all parts of the asparagus spear after harvest, but this increase did not coincide with the increase in XET activity. The harvest-related changes to xyloglucan molecular weight were restricted to slight, segment-specific, up- or down-shifts. However, this may hide strategic alterations to linkages leading to a more rigid wall without major changes in overall molecular weight. The initial postharvest surge in XET activity could be related to harvest stresses such as water deficit, but we propose that the later induction of AoXET1 and AoXET2 is linked to the development of lignified secondary cell walls.

Cell expansion in the epidermis: microtubules, cellulose orientation and wall loosening enzymes

Two models of isolated epidermis were used to demonstrate that the net orientation of cellulose microfibrils in the cell wall is related to mechanical properties of the tissue, and can be used as an indicator for wall anisotropy. In the developing plant epidermis, cells expand in one or two directions in the plane of the plant surface. In epidermis cells actively expanding in one direction (elongation), the orientation of cortical microtubules closely matches the net cellulose orientation. In epidermis cells expanding in two directions, the orientation of the parallel microtubules does not coincide with the net cellulose orientation in the adjacent cell wall. The orientation of cortical microtubules is thus not always a reliable indicator of wall characteristics. In both types of epidermis, a high rate of expansion correlates with a high activity of xyloglucan endotransglycosylase (XET), as determined in situ . This high activity alone cannot explain unidirectional wall expansion.

Cell wall metabolism in fruit softening and quality and its manipulation in transgenic plants.

Excessive softening is the main factor limiting fruit shelf life and storage. Transgenic plants modified in the expression of cell wall modifying proteins have been used to investigate the role of particular activities in fruit softening during ripening, and in the manufacture of processed fruit products. Transgenic experiments show that polygalacturonase (PG) activity is largely responsible for pectin depolymerization and solubilization, but that PG-mediated pectin depolymerization requires pectin to be de-methyl-esterified by pectin methylesterase (PME), and that the PG beta-subunit protein plays a role in limiting pectin solubilization. Suppression of PG activity only slightly reduces fruit softening (but extends fruit shelf life), suppression of PME activity does not affect firmness during normal ripening, and suppression of beta-subunit protein accumulation increases softening. All these pectin-modifying proteins affect the integrity of the middle lamella, which controls cell-to-cell adhesion and thus influences fruit texture. Diminished accumulation of either PG or PME activity considerably increases the viscosity of tomato juice or paste, which is correlated with reduced polyuronide depolymerization during processing. In contrast, suppression of beta-galactosidase activity early in ripening significantly reduces fruit softening, suggesting that the removal of pectic galactan side-chains is an important factor in the cell wall changes leading to ripening-related firmness loss. Suppression or overexpression of endo-(1-->4)beta-D-glucanase activity has no detectable effect on fruit softening or the depolymerization of matrix glycans, and neither the substrate nor the function for this enzyme has been determined. The role of xyloglucan endotransglycosylase activity in softening is also obscure, and the activity responsible for xyloglucan depolymerization during ripening, a major contributor to softening, has not yet been identified. However, ripening-related expansin protein abundance is directly correlated with fruit softening and has additional indirect effects on pectin depolymerization, showing that this protein is intimately involved in the softening process. Transgenic work has shown that the cell wall changes leading to fruit softening and textural changes are complex, and involve the coordinated and interdependent activities of a range of cell wall-modifying proteins. It is suggested that the cell wall changes caused early in ripening by the activities of some enzymes, notably beta-galactosidase and ripening-related expansin, may restrict or control the activities of other ripening-related enzymes necessary for the fruit softening process.

In vivo colocalization of xyloglucan endotransglycosylase activity and its donor substrate in the elongation zone of Arabidopsis roots.

We have developed a method for the colocalization of xyloglucan endotransglycosylase (XET) activity and the donor substrates to which it has access in situ and in vivo. Sulforhodamine conjugates of xyloglucan oligosaccharides (XGO-SRs), infiltrated into the tissue, act as acceptor substrate for the enzyme; endogenous xyloglucan acts as donor substrate. Incorporation of the XGO-SRs into polymeric products in the cell wall yields an orange fluorescence indicative of the simultaneous colocalization, in the same compartment, of active XET and donor xyloglucan chains. The method is specific for XET, as shown by competition experiments with nonfluorescent acceptor oligosaccharides, by negligible reaction with cello-oligosaccharide-SR conjugates that are not XET acceptor substrates, by heat lability, and by pH optimum. Thin-layer chromatographic analysis of remaining unincorporated XGO-SRs showed that these substrates are not extensively hydrolyzed during the assays. A characteristic distribution pattern was found in Arabidopsis and tobacco roots: in both species, fluorescence was most prominent in the cell elongation zone of the root. Proposed roles of XET that include cell wall loosening and integration of newly synthesized xyloglucans could thus be supported.

Purification of xyloglucan endotransglycosylase based on affinity sorption of the active glycosyl-enzyme intermediate complex to cellulose.

Xyloglucan endotransglycosylase (XET) catalyzes the cleavage of xyloglucan (XG) molecules by a transglycosylation mechanism involving two steps: (a) endocleavage of the beta-(1,4)-linked polyglucosyl backbone of the xyloglucan molecule with formation of a glycosyl-enzyme intermediate; (b) transfer of the glycosyl residue from the intermediate to the C-4 position of the nonreducing end glucosyl unit of another molecule of XG or an XG-derived oligosaccharide with liberation of the enzyme (Z. Sulová et al., 1998, Biochem. J. 330, 1475-1480). The formation of a relatively stable active complex of XET with XG and the tendency of xyloglucan to bind tightly via hydrogen bonds to cellulose were exploited in the present method of purification of XET. Crude extracts from nasturtium (Tropaeolum majus) cotyledons and other plant sources containing the enzyme were mixed with XG in order to form the XET:XG complex, which was applied onto cellulose. Unadsorbed proteins were removed by washing and the XET was released from the adsorbed XET:XG complex by transglycosylation of its glycosyl moiety to added XG-derived oligosaccharides. The described procedure resulted in an over 100-fold increase in specific activity of XET in a single step. Further purification of the enzyme to homogeneity was achieved by gel-permeation chromatography on Bio-Gel P30. Similar procedure could be used for purification of XET from other plant sources, such as lentil (Lens culinaris) seeds, pea (Pisum sativum) epicotyls, and supernatant of suspension-cultured Catharanthus roseus cells.
