The relationship between working memory and anxiety in individuals with early treated phenylketonuria (PKU).

Although early diagnosis and treatment prevent the severe impairments associated with untreated phenylketonuria (PKU), individuals with early treated PKU (ETPKU) nonetheless experience significant neurocognitive and psychological sequelae, including difficulties in working memory (WM) and increased risk of anxiety. The primary objective of the present study was to examine the extent to which anxiety may moderate the relationship between ETPKU and WM performance. A sample of 40 adults with ETPKU and a demographically comparable sample of 40 healthy adults without PKU completed a comprehensive assessment of WM performance and anxiety symptomatology. Data were collected using a variety of remote assessment methods (e.g., web-based neurocognitive tests, semistructured interview, report-based measures). The ETPKU group demonstrated significantly poorer WM performance as compared to the non-PKU group. The groups did not differ significantly in anxiety; however, high anxiety was more common in the ETPKU group (53% of sample) than the non-PKU group (33%). A significant interaction between anxiety, metabolic control (as reflected by Phe levels), and WM performance was observed for the ETPKU group. Individuals with high anxiety and/or high Phe levels (> 360 μmol/L) performed poorer than the non-PKU group. Individuals with low anxiety and relatively low Phe levels (< 360 μmol/L) performed comparably to the non-PKU group. Anxiety was found to moderate the relationship between Phe levels and WM performance in individuals with ETPKU. This finding underscores the importance of accounting for anxiety when evaluating neurocognitive performance in individuals with ETPKU whether for research or clinical purposes. White matter lesion load determines exercise-induced dopaminergic plasticity and working memory gains in aging

Age-related dopamine reductions have been suggested to contribute to maladaptive working memory (WM) function in older ages. One promising intervention approach is to increase physical activity, as this has been associated with plasticity of the striatal dopamine system and WM improvements, however with individual differences in efficacy. The present work focused on the impact of individual differences in white-matter lesion burden upon dopamine D2-like receptor (DRD2) availability and WM changes in response to a 6 months physical activity intervention. While the intervention altered striatal DRD2 availability and WM performance in individuals with no or only mild lesions (p < 0.05), no such effects were found in individuals with moderate-to-severe lesion severity (p > 0.05). Follow-up analyses revealed a similar pattern for processing speed, but not for episodic memory performance. Linear analyses further revealed that lesion volume (ml) at baseline was associated with reduced DRD2 availability (r = −0.41, p < 0.05), and level of DRD2 change (r = 0.40, p < 0.05). Taken together, this study underlines the necessity to consider cerebrovascular health in interventions with neurocognitive targets. Future work should assess whether these findings extend beyond measures of DRD2 availability and WM.

Effects of hypotension and hypertension on source memory and working memory

Objectives The effects of chronic low and high blood pressure on memory are unclear due to divergent results, originating in part due to participant misclassifications. The aim of this study was to compare source memory and working memory performance in individuals diagnosed with hypotension or hypertension with the performance of normotensive participants. Hypertensive and hypotensive individuals were receiving medical treatment. Method From a sample of 1656 participants, 219 were identified as hypertensive, and 37 were identified as hypotensive. Each of these two groups was compared with normotensive individuals matched by age, education and sex. Source memory performance and working memory performance were assessed through computerized tasks. Results Source memory accuracy was poorer in hypotensive and hypertensive individuals than in normotensive individuals, and spatial working memory discrimination was inferior in hypertensive participants compared to normotensive individuals. Conclusion Blood pressure impairment should be considered a major concern because it has been linked to severe cardiovascular and cerebrovascular diseases. Furthermore, here we show that it has negative effects on the two types of memory that are most essential for preserving a self-sufficient lifestyle.

Frontal Underactivation During Working Memory Processing in Adults With Acute Partial Sleep Deprivation: A Near-Infrared Spectroscopy Study.

Individuals with partial sleep deprivation may have working memory (WM) impairment, but the underlying neural mechanism of this phenomenon is relatively unknown. The present study examined neural processing during WM performance in individuals with and without partial sleep deprivation using near-infrared spectroscopy (NIRS). Forty college students (10 males) were equally split into Sufficient Sleep (SS) and Insufficient Sleep (IS) groups based on self-reports of previous night's sleep duration. Participants in the SS group obtained the recommended amounts of sleep according to various sleep organizations (i.e., >7.0 h), whereas those in the IS group obtained amounts of sleep no greater than the lower limit of the recommendation (i.e., ≤7.0 h). All participants underwent an n-back paradigm with a WM load (i.e., 3-back) and a control condition (i.e., 0-back) while their prefrontal hemodynamics were recorded by NIRS. The IS and SS groups performed the tasks comparably well. However, unlike the SS group, which exhibited bilateral frontal activation indicated by increased oxyhemoglobin concentration and decreased deoxyhemoglobin concentration during WM processing (i.e., 3-back > 0-back), the IS group did not exhibit such activation. In addition, levels of WM-related frontal activation, especially those on the left side, correlated with sleep duration the night before, even when habitual sleep duration was controlled for. The findings suggest the presence of frontal lobe dysfunction in the absence of evident WM difficulties in individuals with acute partial sleep deprivation. They also highlight the importance of a good night's sleep to brain health.

Reduced Frontal Activations at High Working Memory Load in Mild Cognitive Impairment: Near-Infrared Spectroscopy

Background: Some functional magnetic resonance imaging studies have reported altered activations in the frontal cortex during working memory (WM) performance in individuals with mild cognitive impairment (MCI), but the findings have been mixed. The objective of the present study was to utilize near-infrared spectroscopy (NIRS), an alternative imaging technique, to examine neural processing during WM performance in individuals with MCI. Methods: Twenty-six older adults with MCI (7 males; mean age 69.15 years) were compared with 26 age-, gender-, handedness-, and education-matched older adults with normal cognition (NC; 7 males; mean age 68.87 years). All of the participants undertook an n-back task with a low (i.e., 0-back) and a high (i.e., 2-back) WM load condition while their prefrontal dynamics were recorded by a 16-channel NIRS system. Results: Although behavioral results showed that the two groups had comparable task performance, neuroimaging results showed that the MCI group, unlike the NC group, did not exhibit significantly increased frontal activations bilaterally when WM load increased. Compared to the NC group, the MCI group had similar frontal activations at low load (p > 0.05 on all channels) but reduced activations at high load (p < 0.05 on 4 channels), thus failing to demonstrate WM-related frontal activations (p < 0.05 on 9 channels). In addition, we found a positive correlation between the left WM-related frontal activations and WM ability primarily in the NC group (r_s = 0.42, p = 0.035), suggesting a relationship between frontal hypoactivation and WM difficulties. Conclusion: The present findings suggest the presence of frontal dysfunction that is dependent on WM load in individuals with MCI.

Exploring spatial working memory performance in individuals with Williams syndrome: The effect of presentation format and configuration

Williams syndrome (WS) is a neurodevelopmental disorder associated with an impaired capacity for visuospatial representation. Individuals with WS have a specific weakness in spatial processing, while visual components are relatively well preserved. This dissociation is apparent in working memory function too. The present study aimed to further investigate spatial working memory performance in individuals with WS, analyzing whether their impaired WM performance regards both simultaneous and sequential spatial formats, and whether presenting configurations differently might reduce their difficulties. These issues were examined by administering simultaneous and sequential spatial tasks, in which the information to be recalled was presented in random or arranged configurations. Our results showed that individuals with WS performed less well than typically developing (TD) children in the spatial-simultaneous task, but not in the spatial-sequential one. The presence of a pattern enhanced the performance of both groups, but the difference between the two groups' performance in the spatial simultaneous task remained, albeit to a lesser degree.

E23 Functional Brain Correlates Of Psychiatric Function In Presymptomatic Huntington's Disease: The Image-hd Study

<h3>Background</h3> Changes in prefrontal brain responses as well as psychiatric deficits are detected in Huntington’s disease (HD) prior to diagnosis. <h3>Aims</h3> This study investigated the relationship between psychiatric symptoms and functional brain activity during working memory performance in individuals with pre-symptomatic hd (pre-hd) using the image-hd data set. <h3>Methods/techniques</h3> Pre-HD far from onset (pre-hd far, n = 17), pre-hd close to onset (pre-hd close, n = 18), and control (n = 32) groups completed an n-back task (0, 1 and 2-back) during functional magnetic resonance imaging (fMRI). Brain regions of interest included the bilateral dorsolateral prefrontal cortex (dlpfc) and anterior cingulate cortex (acc). Correlations were performed between blood-oxygen-level-dependent (bold) activity in each brain region and scores on a battery of neuropsychiatric inventories, while controlling for age, gender, and accuracy on the n-back task. <h3>Results/outcome</h3> For both 1- and 2-back conditions, significant negative correlations were found in the pre-hd close group between neuropsychiatric disturbance and bold activation in both the right dlpfc and acc. During 2-back, a significant negative correlation was found in the pre-hd far group between depression symptoms and right dlpfc bold. For 1-back, no significant correlations were found in the pre-hd far group. <h3>Conclusions</h3> These findings suggest that as pre-hd individuals approach onset, increased psychiatric disturbance is associated with decreased functional brain activity, which becomes more wide-spread during higher working memory loads.

Dynamic working memory performance in individuals with single-domain amnestic mild cognitive impairment

Previous studies have observed poorer working memory performance in individuals with amnestic mild cognitive impairment than in healthy older adults. It is unclear, however, whether these difficulties are true only of the multiple-domain clinical subtype in whom poorer executive functioning is common. The current study examined working memory, as measured by the self-ordered pointing task (SOPT) and an n-back task, in healthy older adults and adults with single-domain amnestic mild cognitive impairment (aMCI). Individuals with single-domain aMCI committed more errors and required longer to develop an organizational strategy on the SOPT. The single-domain aMCI group did not differ from healthy older adults on the 1-back or 2-back, but had poorer discrimination on the 3-back task. This is, to our knowledge, the first characterization of dynamic working memory performance in a single-domain aMCI group. These results lend support for the idea that clinical amnestic MCI subtypes may reflect different stages on a continuum of progression to dementia and question whether standardized measures of working memory (span tasks) are sensitive enough to capture subtle changes in performance.
