Effect of Dietary Xylanase Inclusion on Growth Performance, Nutrient Digestibility, and Digesta Viscosity of Weaned Pigs Fed Wheat-Soybean Meal-Based Diets.

(1) Background: This study aimed to evaluate the effects of dietary xylanase addition on growth performance, nutrient digestibility, volatile fatty acids, and digesta viscosity at different digestive sites in weaned pigs fed wheat-soybean meal-based diets with reduced metabolizable energy. (2) Methods: A total of 312 weaned pigs (5.1 ± 0.9 kg, 20 ± 2 days of age) were assigned to one of six dietary treatments. The experimental diets were formulated in a three-phase nursery feeding program: phase 1 (d0-d7), phase 2 (d8-d21), and phase 3 (d22-d42). The experimental diets consisted of a wheat-soybean meal-based diet formulated to meet pig requirements (positive control, PC); the PC diet with a reduction of 100 kcal of metabolizable energy (ME) (negative control, NC); and the NC diet with either 900, 1800, 3600, or 7200 units of xylanase. Feed disappearance and body weight were measured at d7, 14, 21, and 42 in the nursery phase. The pen fecal score was assessed daily from d0 to d14 and three times a week from d15 to d28. On d21-d24 of the experiment (12 pigs per day), one pig per pen was selected for sample collection: ileal, cecal, and mid-colon digesta for viscosity and ileal digesta, feces for nutrient digestibility, and feces and cecal digesta for the measurement of volatile fatty acid. (3) Results: The addition of xylanase to the NC diets did not improve pig growth performance (body weight, feed conversion ratio, and average daily gain; p > 0.10) during the entire nursery phase. In Week 2 and Week 3, pigs fed xylanase had a lower (χ2 < 0.05) incidence of fecal scores 3 and 4 (diarrhea) than the PC and NC diets. In addition, the apparent total tract digestibility of neutral detergent fiber and acid detergent fiber increased linearly (p < 0.1) in response to xylanase addition. Xylanase addition (900 to 7200 U) decreased digesta viscosity in the colon compared to the PC and NC diets. Furthermore, xylanase addition resulted in a lower (p < 0.05) concentration of acetic, propionic, butyric, valeric, and total volatile fatty acid in cecal samples compared to PC. The addition of xylanase resulted in greater acetic and valeric acid concentrations in cecal samples compared to the NC group (p < 0.10). (4) Conclusions: Xylanase addition can improve nutrient digestibility, particularly at the total tract level, and reduce viscosity in the hindgut, which could be related to decreasing the occurrence of looseness. However, its impact on growth performance was minimal in wheat-soybean meal-based diets with a reduction of 100 kcal of ME.

PSI-14 Effect of dietary xylanase supplementation on growth performance, nutrient digestibility, and digesta viscosity of weaned pigs fed wheat-soybean meal-based diets

Abstract Xylanase supplementation can improve nutrient digestibility in pig diets by degrading cell wall non-starch polysaccharides and reducing digesta viscosity as well as promoting intestinal health of pigs after weaning. This study determined the effects of dietary xylanase supplementation on growth performance, apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of nutrients, and digesta viscosity in different digestive sites of weaned pigs. A total of 312 weaned pigs [body weight (BW) = 5.1 ± 0.9 kg] were weaned into 48 pens and assigned to one of six dietary treatments, each treatment with 8 pens, 6 to 7 pigs per pen, and sex ratios maintained within BW blocks. Experimental diets were formulated in a 3-phase nursery feeding program: Phase 1 (d 0 - d 7), Phase 2 (d 8 - d 21), and Phase 3 (d 22 – d 42). Experimental diets consisted of a wheat/soybean meal-based diet formulated to meet pig requirements (positive control, PC), the PC diet with reduction of 100 kcal of ME (negative control, NC), and the NC diet supplemented with either 900, 1,800, 3,600, or 7,200 units of xylanase. Feed disappearance and body weight were measured at d 7, 14, 21, and 42. Pen fecal score was assessed daily during d 0 to 14 and three times a week during d 15 to 28. On d 21 to 24 of the experiment (12 pigs per day), one pig per pen was selected for sample collection: ileal, cecal, and mid-colon digesta for viscosity and ileal digesta and feces for nutrient digestibility. Orthogonal polynomial contrasts were used to determine the linear and quadratic effects of increasing levels of xylanase in NC diets. Contrast analysis was used to compare PC and NC groups with xylanase-supplemented groups. Fecal score data were analyzed using the PROC FREQ procedure. Supplementing xylanase to NC diets did not improve pig growth performance (P &amp;gt; 0.05) through the nursery period. In wk 2 and wk 3, pigs fed the PC and NC diets had a greater (χ2 &amp;lt; 0.05) incidence of fecal scores 3 &amp; 4 (diarrhea) than the xylanase-supplemented pigs. The AID of DM and ATTD of crude protein, gross energy, and NDF were greater (P &amp;lt; 0.05) in the PC and 900-xylanase supplemented diets than in the other dietary treatments. In addition, the ATTD of NDF and ADF increased linearly (P &amp;lt; 0.1) in response to xylanase supplementation. Xylanase supplementation (900 to 7,200 U) decreased digesta viscosity in colon when compared with PC and NC diets. Overall, xylanase supplementation can improve nutrient digestibility, particularly at total tract level, and reduce viscosity in the hindgut, which could be related with decreasing the occurrence of looseness. However, the effects on growth performance were not expressive when xylanase was supplemented to wheat-soybean meal-based diets with reduction of 100 kcal of ME.

Effects of enzyme supplementation on growth performance, digestibility of phosphorus, femur parameters and fecal microbiota in growing pigs fed different types of diets.

A 42-days study was conducted to evaluate the effects of different dietary types (corn-or wheat-soybean meal-based diet) and phytase (Phy) or a multi-carbohydrase and phytase complex (MCPC) supplementation on growth performance, digestibility of phosphorus (P), intestinal transporter gene expression, plasma indexes, bone parameters, and fecal microbiota in growing pigs. Seventy-two barrows (average initial body weight of 24.70 ± 0.09 kg) with a 2 × 3 factorial arrangement of treatments and main effects of diet type (corn-or wheat-soybean meal-based-diets) and enzyme supplementation (without, with Phy or with MCPC). Each group was designed with 6 replicate pens. The MCPC increased (p < 0.05) average daily gain (ADG) and final body weight (BW). A significant interaction (p = 0.01) was observed between diet type and enzyme supplementation on apparent total tract digestibility (ATTD) of P. The ATTD of P was higher (p < 0.05) in wheat soybean meal-based diets compared to corn-soybean meal-based diets. Compared with the corn-soybean meal-based diet, the relative expression of SLC34A2 and VDR genes in the ileum and SLC34A3 in jejunum of growing pigs fed the wheat-soybean meal based diet was lower (p < 0.05). The MCPC significantly reduced (p < 0.05) the relative expression of TRPV5 and CALB1 genes in the ileum and increased the expression of CALB1 in the duodenum compared to control diet. The phytase increased (p < 0.05) the relative expression of SLC34A1 gene in the duodenum in comparison to control diet and MCPC-supplemented diet. The Ca and P contents in plasma from pigs fed corn-soybean meal-based diet were higher (p < 0.05) than those from pigs fed wheat-soybean meal-based diet, and the parathyroid hormone (PTH) and calcitonin (CT) concentrations were lower (p < 0.05) than those fed wheat-soybean meal-based diet. The content of Ca and P in the femur and the bone strength of pigs in the corn-soybean meal group were significantly higher (p < 0.05) than those in the wheat-soybean meal groups. The phytase increased (p < 0.05) the Ca and P content and bone strength of the femur. Additionally, diet type and both enzymes significantly improved fecal microbial diversity and composition. Taken together, diet type and exogenous enzymes supplementation could differently influence the growth performance, utilization of phosphorus, intestinal transporter gene expression, bone mineralization and microbial diversity and composition in growing pigs.

Effect of Cyberlindnera jadinii yeast on growth performance, nutrient digestibility, and gut health of broiler chickens from 1 to 34 d of age

The effect of dietary graded levels of Cyberlindnera jadinii yeast (C. jadinii) on growth performance, nutrient digestibility, and gut health of broilers was evaluated from 1 to 34 d of age. A total of 360 male broiler chicks were randomly allocated to 1 of 4 dietary treatments (6 replicate pens each) consisting of a wheat-soybean meal-based pelleted diet (Control or CJ0), and 3 diets in which 10% (CJ10), 20% (CJ20), and 30% (CJ30) of the crude protein were supplied by C. jadinii, by gradually replacing protein-rich ingredients. Body weight and feed intake were measured at d 1, 11, 22, and 32. Pellet temperature, durability, and hardness increased linearly (P < 0.05) with C. jadinii inclusion, with highest (P < 0.05) values for CJ30. Up until d 22, feed conversion ratio (FCR) was similar between treatments (P=0.169). Overall, increasing C. jadinii inclusion linearly increased (P=0.047) feed intake but had no effect on weight gain or mortality. FCR increased (P < 0.05) linearly with increasing C. jadinii inclusion but only birds fed CJ30 had a significantly poorer FCR compared to the Control. Ileal digestibility was not affected by C. jadinii inclusion, however, there was a significant linear decrease in crude protein and phosphorus, and a tendency for a decrease in fat digestibility. Apparent metabolizable energy (AME) decreased (P < 0.001) quadratically with increasing C. jadinii and was significantly lower in CJ30 compared to the Control. Ileal concentrations of volatile fatty acids (VFAs) were not affected by C. jadinii inclusion, but butyric acid and total VFAs were linearly and quadratically increased and were significantly higher in cecal digesta of birds fed CJ20 and CJ30. Increasing C. jadinii inclusion was associated with an increase (P < 0.05) in the relative abundance of lactobacillus in the ileum and cecum. In conclusion, C. jadinii yeast can supply up to 20% of the total dietary protein without negatively affecting performance, digestibility, or gut health of broilers. The potential confounding role of feed processing and C. jadinii cell wall components on broiler performance is discussed.

Xylanase supplementation of pelleted wheat-based diets increases growth efficiency and apparent metabolizable energy and decreases viscosity of intestinal contents in broilers

This study was designed to test graded supplementation of a thermostable xylanase in pelleted, wheat-based diets fed to broiler chickens over a 28-d period. A total of 600 Ross 708 male broilers were allotted to 1 of 5 dietary treatments: positive control (PC), negative control (NC; 125 kcal of AME/kg diet reduction relative to PC), and NC supplemented with 10, 15, or 30 g/ton of xylanase. Wheat-soybean meal-based diets were pelleted and fed in 2 feeding phases (14-d each). Study outcomes included growth performance, AME, and ileal digesta viscosity with 20 battery cages of 6 birds per treatment. Data were analyzed by 1-way ANOVA along with estimation of Pearson correlation coefficients. Whereas no difference between NC and PC was observed for BW gain, NC birds exhibited increased (P < 0.05) feed intake during each feeding phase and overall, which caused improvements (P < 0.05) in feed conversion ratio (FCR) for PC vs. NC birds. The analyzed AME of PC birds was 112 kcal/kg of diet greater (P < 0.05) than for NC birds, though no differences in digesta viscosity were observed. Xylanase supplementation of the NC diet at 15 or 30 g/ton elicited overall improvements (P < 0.05) in BW gain beyond the PC, while the 30 g/ton level equalized feed intake with the PC. Regardless of level, xylanase supplementation improved (P < 0.05) the FCR relative to the NC, thereby equalizing the response with the PC. Similarly, supplementation with any xylanase level increased (P < 0.05) AME over the NC, making all treatments synonymous with the PC. Digesta viscosity of all xylanase-supplemented treatments was decreased relative to both the NC and PC treatments. Overall, this study provided clear evidence that addition of a thermostable xylanase to pelleted wheat-based diets elicited improvements in growth performance of broilers concomitant with a reduction in digesta viscosity and elevation of analyzed dietary AME content.

Effects of xylanase on growth performance, nutrient digestibility, serum metabolites, and fecal microbiota in growing pigs fed wheat-soybean meal-based diets.

This experiment investigated the effects of xylanase on growth performance, nutrient digestibility, serum metabolites, and fecal microbiota in growing pigs fed wheat-soybean meal-based diets. Seventy-two crossbred pigs (Duroc × [Landrace × Large White]) pigs (body weight of 23.30 ± 1.51 kg) were allotted two treatments with six pens per treatment and six pigs per pen. The diets were a wheat-soybean meal-based diet (Control group) and a wheat-soybean meal-based diet supplemented with 500 U/kg xylanases (XYL group). The experiment was divided into two periods (phase 1: days 1 to 35 and phase 2: days 36 to 70). Xylanase improved G:F during phase 1 and the entire experiment (P < 0.05) and tended to improve G:F during phase 2 (P = 0.09). Compared with the control group, pigs in the XYL group had greater apparent total tract digestibility of dry matter, organic matter, and gross energy on days 35 and 70 (P < 0.05) and had greater apparent ileal digestibility of amino acids (histidine, lysine, methionine, and serine) on day 70 (P < 0.05). The fecal microbiota in the XYL group contained greater abundances of g_Terrisporobacter, g_Lactobacillus, g_Clostridium_sensu_stricto_1, and g_Romboutsia than the Control group on day 70. Xylanase increased the fecal Lactobacillus populations on day 35 (P < 0.05). On days 35 and 70, xylanase reduced the fecal E. coli populations (P < 0.05). Supplementing xylanase to wheat-soybean meal-based diets collectively improved fecal microbiota, and nutrient digestibility, thereby improving growth performance in growing pigs.

Supplementation of multi-enzymes alone or combined with inactivated Lactobacillus benefits growth performance and gut microbiota in broilers fed wheat diets.

The effects of multi-enzymes mixture supplementation or combination with inactivated Lactobacillus on growth performance, intestinal barrier, and cecal microbiota were investigated in broilers at the age of 15–42 days fed a wheat-based diet. A total of 576 broilers (12 broilers/cage; n = 12) were used and divided into four groups and randomly allotted to four experimental diets throughout grower (15–28 days of age) and finisher (29–42 days of age) phases. Diets consisted of a corn-soybean meal-based diet (BD), a wheat-soybean meal-based diet (WD), and WD supplemented multi-enzymes (WED) or combined with inactivated Lactobacillus (WEPD). The results showed that the average daily gain (ADG) and body weight (BW) were reduced in broilers fed WD diet compared with those fed BD diet during the grower period (P < 0.05). Broilers in the WED or WEPD group had higher ADG and BW during the grower period (P < 0.05) and had a lower feed-to-gain ratio (F/G) compared to broilers in the WD group during the grower and overall periods (P < 0.05). Improved expression of intestinal barrier genes (claudin-1, ZO-1, and mucin-2) was observed in WEPD compared to the BD or WD group (P < 0.05). Compared to the BD group, the WD group decreased the abundance of Oscillospira, norank_f__Erysipelotrichaceae, and Peptococcus, which are related to anti-inflammatory function and BW gain. The WD also increased Bifidobacterium and some short-chain fatty acid (SCFA)-producing bacteria (Anaerotruncus, Blautia, and Oscillibacter), and Barnesiella, which were presumed as “harmful microbes” [false discovery rate (FDR) < 0.05]. WED and WEPD groups, respectively, improved Bilophila and Eubacterium_hallii_group compared with those in the WD group (FDR < 0.05). In addition, the Enterococcus abundance was reduced in the WEPD group compared to the WD group (FDR < 0.05). Higher acetate and total SCFA concentrations were observed (P < 0.05) among broilers who received a WD diet. Compared with the WD group, the WED or WEPD group further increased cecal propionate content (P < 0.05) and tended to improve butyrate concentration. These results suggested that supplemental multi-enzymes alone and combined with inactivated Lactobacillus could improve the growth performance based on the wheat-based diet and offer additional protective effects on the intestinal barrier function of broilers.

Evaluation of early feed access and algal extract on growth performance, organ development, gut microbiota and vaccine-induced antibody responses in broiler chickens

Hatching concepts such as on-farm hatching provide an opportunity to supply newly hatched chickens with optimal nutrition that support growth and development of a healthy gut. Brown algae contain bioactive compounds, especially laminarin and fucoidan that may improve intestinal health and immune responses. This study aimed to examine the effects of early access to feed and water posthatch and feed supplementation with algal extract rich in laminarin from Laminaria digitata, on growth performance, organ and microbiota development and antibody production. A total of 432 Ross 308 chicks were allotted to 36 rearing pens in a 2 × 3 factorial design with two hatching treatments and three dietary treatments. During chick placement, half of the pens were directly provided access to feed and water (Early) while half of the pens were deprived of feed and water for 38 h (Late). The chicks were fed three different starter diets until day 6; a wheat-soybean meal-based control diet, a diet with low inclusion of algal extract (0.057%) and a diet with high inclusion of algal extract (0.114%). Feed intake and BW were registered on pen basis at placement, days 1, 6, 12, 19, 26, 33 and 40. To induce antibody responses, all chicks were vaccinated against avian pneumovirus on day 10. Three chicks per pen were selected as focal animals and used for blood sampling on days 10 and 39. On days 6, 19, and 40, two birds per pen were killed and used for organ measurement and caecal digesta sampling for gut microbiota analysis using the Illumina Miseq PE 250 sequencing platform. Results showed that algal extract did not influence gut microbiota, gut development or vaccine-induced antibody responses. However, during the first 38 h, early-fed chicks consumed on average 19.6 g of feed and gained 27% in BW, while late-fed chicks lost 9.1% in BW which lowered BW and feed intake throughout the study (P < 0.05). Late chicks also had longer relative intestine, higher relative (g/kg BW) weight of gizzard and proventriculus but lower relative bursa weight on day 6 (P < 0.05). No effects of hatching treatment on microbiota or antibody response were detected. The microbiota was affected by age, where alpha diversity increased with age. In conclusion, this study showed that early access to feed but not algal extract improved the growth performance throughout the 40-day growing period, and stimulated early bursa development.

Net energy, energy utilization, and nitrogen and energy balance affected by dietary pea supplementation in broilers.

Pea starch consists predominantly of C-type of amylopectin chain which is more resistant to digestive enzymes than A-type of starch thus slowly digested in poultry. It was hypothesized that the presence of slowly digested pea starch in broiler diets will increase net energy and the efficiency of energy utilization in broilers. Two experiments were performed to investigate starch digestibility of pea at different incubation times (in vitro study) and the effect of dietary pea on heat increment and net energy in broilers using an open-circuit respiratory calorimetry system (in vivo study). One-day-old Ross 308 male broilers were fed a common starter crumble from d 1 to 10 and standard grower diets thereafter. At d 21, birds were transferred to the chambers each housing 2 birds. Each treatment was replicated 6 times with 2 identical runs of 3 replicates per treatment. A wheat-soybean meal-based diet was used as a control and the treatment diet contained 500 g of pea/kg pea. In vitro study showed that pellet processing increased (P < 0.001) starch digestibility, particularly at shorter times for wheat and a much larger response for pea. Birds offered the pea-based diet had lower (P = 0.002) feed intake, lower (P = 0.020) body weight gain, but a similar (P > 0.05) FCR compared to those offered the wheat-based diet. Net energy (NE) and apparent metabolizable energy (AME) values were higher in the pea-based diet than in the wheat-based diet (P = 0.037 for NE and P = 0.018 for AME). Heat production, respiratory quotient, heat increment of feed, efficiency of utilization of gross energy for AME, and efficiency of utilization of AME for NE did not differ (P > 0.05) between the 2 treatments. There was no effect (P > 0.05) of pea on the total tract digestibilities of dry matter, crude protein and ash, but the total tract digestibility of starch was higher (P = 0.022) in the pea-based diet compared to the wheat-based diet. This study provides insight into the energy metabolism of broilers offered a pea-based diet and indicates that dietary pea supplementation increases dietary AME and NE but has no effect on heat increment of feed and the efficiency of energy utilization in broilers.

116 Meta-analysis of 3 trials: the effect a novel consensus bacterial 6-phytase variant on phosphorus digestibility and retention in piglets fed inorganic phosphate free diets

Abstract A meta-analysis was conducted to investigate the effect of a novel consensus bacterial 6-phytase variant on apparent total tract digestibility (ATTD) of phosphorus (P) and P retention in piglets (initial body weight of 9.6 ± 1.8 kg). Database was constructed using 3 studies (unpublished), contributing 189 data points. All studies had a positive control (PC) diet with adequate levels of calcium (Ca; 0.57–0.85%) and digestible P (0.25–0.29%), a negative control (NC) diet with low Ca and digestible P levels (total removal of inorganic P). Piglets were fed a corn or wheat soybean meal-based diet with rice bran or sunflower meal for 7–10 d, followed by 4 d fecal and urine collection. Total analyzed Ca, total P, calculated digestible P and phytate P levels in NC varied from 0.48 to 0.79%, 0.37 to 0.40%, 0.16 to 0.18%, and 0.24 to 0.28%, respectively, among studies. The NC diet was supplemented with a novel consensus bacterial 6-phytase variant (PhyG) or a commercial Buttiauxella sp. phytase (PhyB), at 500 and 1,000 FTU/kg. Data were analyzed using Mixed Model by JMP 14.0. Addition of both phytases improved (P &amp;lt; 0.05) ATTD P compared to NC diets. The NC reduced digestible P (g/kg) vs PC, both phytases showed greater (P &amp;lt; 0.05) digestible P (g/kg) when compared to NC and recovered to the level of PC. Digestible P improvement (g/kg) over NC showed a significant phytase effect, where PhyG supplementation had 17% greater (P &amp;lt; 0.05) digestible P improvement when compared to PhyB. Phosphorus retention improved (P &amp;lt; 0.05) with phytase supplementation, with 1,000 FTU/kg showing greater (P &amp;lt; 0.05) P retention when compared to 500 FTU/kg. It was concluded that both PhyG and PhyB are efficient in improving ATTD P and P retention, PhyG showed greater digestible P improvement when compared to PhyB, and 1,000 FTU/kg showed greater effect than 500 FTU/kg.

Effects of Optimal Carbohydrase Mixtures on Nutrient Digestibility and Digestible Energy of Corn- and Wheat-Based Diets in Growing Pigs

This experiment investigated the effects of two optimal carbohydrase mixtures (OCMs) on nutrient digestibility and digestible energy in growing pigs fed corn-based and wheat-based diets. These OCMs were screened based on the corn-based diet and wheat-based diet, respectively, using an in vitro method developed by our research group in previous studies. The results show that the respective OCM improved the total tract digestibility of most macronutrient and digestible energy for pigs fed both corn-based diet and wheat-based diet. These findings are useful for the rational utilization of carbohydrases in the swine industry.This study aimed to evaluate the effects of optimal carbohydrase mixture (OCM) on macronutrients and amino acid digestibility and the digestible energy (DE) in growing pigs fed the corn-soybean meal-based diet (CSM) and the wheat-soybean meal-based diet (WSM). A total of 36 ileal-cannulated pigs (50.9 ± 4.9 kg initial body weight) were allotted to four dietary treatments randomly, which included CSM and WSM diets, and two diets supplied with corresponding OCM. These OCMs were screened using an in vitro method from our previous study. After the five day adaptation period, fecal samples were collected from d six to seven, and ileal digesta samples were collected on d 8 and 10. Chromic oxide was added as an indigestible marker. The results show that the addition of OCM improved the apparent ileal digestibility (AID) of dry matter (DM), ash, carbohydrate (CHO), neutral detergent fiber, and gross energy (GE) and the apparent total tract digestibility (ATTD) of DM, CHO, and GE in CSM diet (p < 0.05), but reduced the apparent hindgut disappearance (AHD) of DM in CSM diet (p < 0.05). The ATTD of DM, crude protein (CP), ether extract (EE), ash, and GE and the AHD of DM, CP, EE, ash, CHO, and GE in WSM diet were improved by the OCM addition (p < 0.05), whereas the AID of DM, CP, ash, CHO, and GE were decreased (p < 0.05). The respective DE contents in CSM and WSM diets were increased from 15.45 to 15.74 MJ/kg and 15.03 to 15.49 MJ/kg under the effects of OCM (p < 0.05). Similar to the trend of AID of CP, the OCM addition increased the AID and standardized ileal digestibility (SID) of Ile, Thr, and Cys in CSM diet, but decreased the AID and SID of Ile, Phe, Thr, Val, Ala, Pro, Ser, and Tyr in WSM diet. In conclusion, the OCMs screened by an in vitro method could improve the total tract nutrient digestibility and DE for pigs fed corn-based diet or wheat-based diet but had inconsistent effects on the ileal digestibility of nutrients and energy.

Effect of dietary supplementation of xylanase in a wheat-based diet containing canola meal on growth performance, nutrient digestibility, organ weight, and short-chain fatty acid concentration in digesta when fed to weaned pigs.

This study was designed to determine the effect of dietary supplementation of xylanase on growth performance, nutrient digestibility, organ weight, digesta pH, and concentration of short-chain fatty acids (SCFA) of weaned pigs fed wheat-canola meal (CM) diets over a 35-d period. A total of 144 piglets (72 barrows and 72 gilts) weaned at 18 ± 2 d of age, with initial body weight (BW) of 6.2 ± 0.7 kg, received one of eight dietary treatments based on randomized complete block design. BW and feed intake were recorded weekly to calculate average daily gain (ADG), average daily feed intake (ADFI), and feed efficiency (G:F). Treatments consisted of a control wheat-soybean meal-based diet and wheat-regular (RCM), dehulled (DCM), or coarse CM (CCM) without and with 16,000 BXU/kg xylanase (Econase XT). All diets contained 500 FTU/kg of phytase (Quantum Blue 5G) and titanium dioxide (0.3%). Apparent total tract digestibility (ATTD) of neutral detergent fiber (NDF), crude protein (CP), phosphorous (P), calcium (Ca), dry matter, and diet were determined. On day 35, one pig per pen was euthanized to evaluate the main factors of protein, xylanase supplementation, and sex on organ weight, ileal and colon digesta pH, and colon digesta concentrations of SCFA. The main factors did not affect growth performance. Xylanase supplementation improved nutrient digestibilities in all diets and increased ileal and colonic digesta pH without affecting the growth performance of weaned pigs fed wheat and CM-based diets. A protein-xylanase effect (P < 0.05) resulted in increasing the ATTD of NDF from 28% to 32% and from 29% to 37% for RCM and DCM, respectively. The ATTD of CP was greater (P < 0.05) with xylanase supplementation (75% vs. 70%). Xylanase supplementation increased ATTD of P and Ca. A three-way interaction (P < 0.05) for protein-xylanase-gender for colon pH, acetic, and propionic acid in the colon digesta of pigs indicated that, in addition to the protein source, piglet sex could have influenced how xylanase works. Xylanase supplementation increased (P < 0.05) the weight of the liver and spleen and tended (P < 0.10) to increase the size of the kidney. In conclusion, dietary supplementation of xylanase increased nutrient digestibility and digesta pH but did not influence the growth performance of weaned pigs fed wheat and CM-based diets over a 35-d period.

Evaluation of broiler chickens’ digestibility of Neurospora intermedia biomass

A total of 70 broiler chickens were used to evaluate the apparent ileal digestibility coefficient (AIDC) of protein and amino acids, and apparent digestibility coefficient (ADC) of energy in the protein rich Fungal Biomass of Neurospora intermedia (FBN) obtained from bioethanol production. The chickens were housed in 10 pens with seven chickens per pen and fed one of two experimental diets between day 28 and 35 of age. The experimental diets were wheat-soybean meal-based control diet and a diet composed of 70% control diet and 30% of FBN. There were no difference (P > 0.05) between the control and FBN diet on chick feed intake and body weight at day 35. However, the AIDCs of crude protein and amino acids were significantly (P < 0.01) higher in control diet than in the FBN diet except for proline and phenylalanine. The AIDC of CP (0.74), cysteine (0.68), methionine (0.70), AME (15.6), and threonine (0.69) in FBN were comparable to the corresponding values in other protein-rich feedstuffs such as soybean meal and fish meal. The results from this study show that FBN may be an alternative protein source for poultry.

Effect of wheat distillers dried grains with solubles and exogenous xylanase on laying hen performance and egg quality.

Despite the rapid increase in the use of wheat distillers dried grains with solubles (DDGS) within the poultry sector, little information is available on how the long-term feeding of this by-product will affect the performance of laying hens. This experiment examined the effect of wheat DDGS, with and without exogenous xylanase on dietary apparent metabolizable energy (AME), total tract dry matter retention (DMR), nitrogen retention (NR), fat digestibility (FD) coefficients, feed intake (FI), bodyweight gain (BWG), eggs laid, mean egg weight, egg mass output, and egg quality characteristics including albumin height (AH), Haugh units (HU), yolk color, eggshell strength, and thickness. A total of 320 Hy-Line brown laying hens were randomly allocated to 80 enriched layer colonies (groups of 4). A total of 2 control wheat-soybean meal-based diets were formulated to contain 11.60MJ/kg. One of the diets contained 300g/kg wheat DDGS, while the other was DDGS free, with a respective crude protein content of 171.1g/kg and 166.5g/kg. Both diets were divided by 2 and half of them were supplemented with 2,500 U/kg of xylanase, resulting in 4 diets in total. Data was analyzed as a 2 × 2 factorial arrangement of treatments with analysis of variance (ANOVA). Diets were fed ad libitum from 17 to 43 wk of age and data was collected from 23 to 43 wk. The inclusion of wheat DDGS reduced (P < 0.001) dry matter retention (DMR), FI, BWG, eggs laid, mean egg weight, and egg mass. However, xylanase supplementation improved AME and NR in diets containing wheat DDGS and FD in diets without DDGS (DDGS × xylanase, P < 0.05) and tended to improve (P < 0.10) BWG and egg mass output. For egg quality measurements, the inclusion of DDGS improved (P = 0.046) HU values, eggshell strength (P < 0.001), and increased (P < 0.001) yolk color intensity. This experiment showed xylanase can be used to improve the AME and NR of DDGS-based diets. However, the long-term feeding of 300g/kg wheat DDGS negatively impacts the productive performance of hens.

Effects of pea extrusion and enzyme and probiotic supplementation on performance, microbiota activity and biofilm formation in the broiler gastrointestinal tract

ABSTRACT1. The effects of supplementation of broiler chicken diets with pea meal, carbohydrase enzymes and a probiotic were investigated for potential performance improvement.2. Raw or extruded pea meal (cv Model, grown in Poland) was included in a wheat-soybean meal-based diet at 250 g/kg. The diets were unsupplemented (control) or supplemented with either carbohydrase enzymes (200 U/kg xylanase and 10 U/kg β-glucanase in feed) or a probiotic (Bacillus subtilis), or both. The diets were fed to Ross 308 broilers aged 9–28 days.3. After two additional days, chick gastrointestinal tracts were excised and analysed for the presence of Bacillus subtilis biofilm; and the ileal and caecal digesta were analysed for bacterial enzyme activities and to determine the concentration of short-chain fatty acids (SCFAs).4. Feeding the pea-based diet supplemented with the probiotic compromised feed utilisation, due to higher feed intake. The addition of enzymes to the raw, but not the extruded, pea containing diet partially ameliorated this effect (pea form × additives; P < 0.002).5. In the ileal digesta, interactions between the dietary treatments were observed for the activities of all bacterial glycolytic enzymes and for SCFA concentrations. β-glucosidase, α-galactosidase and β-glucuronidase were highest in birds fed the diet containing extruded pea supplemented with the probiotic and enzymes (pea form x additives; P = 0.018 to P < 0.006). In the caecal digesta, interactions were observed for bacterial enzyme activities, but not for total SCFA concentration. Biofilm formation in the caecum indicated that the probiotic strain was metabolically active in the broiler gut.6. In conclusion, supplementation of diets containing raw or extruded pea meal with enzymes and a Bacillus subtilis spore-based probiotic modulated microbiota activity but had no clear effects on broiler performance. Probiotic administration did not cause excessive fermentation in the ileum and caecum but enhanced Bacillus subtilis spp. biofilm formation in the caecum, which may be indicative of a beneficial effect on gut health.

The Effect of Different Dietary Levels and Sources of Methionine on the Growth Performance of Turkeys, Carcass and Meat Quality

Abstract The aim of this study was to determine the effect of different dietary levels and sources of methionine (Met) on the growth performance of turkeys, carcass and meat quality. A total of 816 Hybrid Converter turkeys in 6 groups and 8 replications were fed wheat-soybean meal-based diets supplemented with three sources of Met: DL-isomer, L-isomer and DL-hydroxy analog (DLM, LM and MHA, respectively). In four 4-week periods (from 1 to 16 weeks of age), the Met content of turkey diets corresponded to the level recommended by NRC (1994) or was increased by approximately 50% to match the intake recommended by some breeding companies. Increased dietary Met content resulted in a higher final body weight (BW) of turkeys (P=0.002) and a lower feed conversion ratio (FCR) (P=0.049), but had no effect on carcass dressing percentage and most parameters of carcass quality. The higher dietary Met level contributed to a decrease in meat pH, a lower contribution of redness and a smaller muscle fiber diameter (P=0.028, P=0.040 and P=0.004, respectively). The higher dietary Met level had no influence on the redox status of meat, but it reduced the incidence of lymphoid cell infiltration between muscle fibers threefold (P=0.003). Throughout the experiment, no significant differences were noted in the growth performance parameters of turkeys, irrespective of Met source. MHA contributed to higher abdominal fat content, lower dry matter (DM) content and lower catalase (CAT) activity in breast meat, compared with DLM and LM. Increased dietary Met content, approximately 50% higher than that recommended by NRC (1994), regardless of Met source, led to higher final BW of turkeys, but had no effect on carcass dressing percentage and most parameters of carcass quality.

Effects of dietary inclusion of high- and low-tannin faba bean (Vicia faba L.) seeds on microbiota, histology and fermentation processes of the gastrointestinal tract in finisher turkeys

This study evaluated the effects of dietary replacement of soybean meal (SBM) with graded levels of faba bean (FB) seeds with high or low tannin content (HT or LT) on the gastrointestinal function and growth performance of turkeys at 13–18 weeks of age. Hybrid Converter turkeys were distributed into 7 treatments corresponding to 7 different finisher diets: a control wheat-soybean meal-based (FB0) diet and experimental diets where SBM was partially replaced with HT or LT seeds at 100, 200 and 300 g/kg. Each treatment comprised 210 turkeys, with seven replicate pens and 30 birds per pen. The LT treatment decreased jejunal crypt depth (vs. FB0; P = 0.049) and the experimental factors had no significant effect on the analysed caecal histological parameters. In comparison with the FB0 diet, diets containing HT and LT FB contributed to an increase in the total bacterial counts (P = 0.001 and P = 0.033) and Bacteria domain (P = 0.001 and P = 0.060), and a decrease in the counts of Bacteroides (P = 0.002 and P = 0.013). Diets containing LT FB reduced the abundance of Salmonella bacteria, relative to the FB0 diet (P = 0.011) and diets with HT FB (P = 0.023). The LT treatment decreased the counts of total bacteria and Bacteria domain (P = 0.005), in comparison with the HT treatment. The highest ileal short-chain fatty acid (SCFA) concentrations were observed in response to the LT200 diet. LT diets stimulated increased SCFA production in the caeca, relative to the FB0 diet (P = 0.022), and the opposite effect was noted when HT and SBM dietary treatments were compared. In comparison with HT diets, LT diets led to a desirable increase in the concentrations of all major fatty acids (acetic, propionic and butyric) in the caecal contents. It can be concluded that FB seeds enhanced fermentation processes in the gastrointestinal tract of turkeys. In comparison with HT FB, LT seeds improved selected parameters of intestinal function, including a decrease in the counts of Salmonella bacteria (P = 0.023), increased SCFA production (including butyrate; P = 0.001), and a decrease in the pH of intestinal digesta (P = 0.105). In conclusion, both LT and HT FB seeds, the latter containing up to 7.1 g/kg tannins, can be included in finisher turkey diets at up to 300 g/kg as a safe and effective substitute for SBM.

Investigations of the nutritive value of meals of double-low rapeseed and its influence on growth performance of broiler chickens

Four experiments were carried out to study the possible differences in metabolizable energy (ME) of meals (RSM) or expeller meals (RSE) from double-low rapeseed (Expt. 1), the influence of processing on ME (Expt. 2) and on relative phosphorus (P) bioavailability (Expt. 3) in RSM, and effect of RSM inclusion on growth performance of broilers (Expt. 4). For Expt. 1, diets with 300 g/kg RSM from 11 RSM and 4 RSE varieties were fed to broilers from d 14 to 21, with excreta collection on d 19 to 21. Each treatment had 8 replicates and 3 birds per replicate. Energy metabolizability of RSM of a specialized high glucosinolate variety (V275OL) was greater (P < 0.05) than all the other varieties. In Expt. 2, two RSM varieties were processed with mild or conventional processing condition. There were no variety effects on ME, but ME and MEn were greater (P < 0.01) for RSM processed by mild processing condition. In Expt. 3, P bioavailability of RSM was determined, relative to MSP, using growth performance and tibia ash as responses. Phosphorus relative bioavailability values were greater (P < 0.05) in RSM of DK Cabernet variety processed using the mild processing condition. In Expt. 4, two RSM varieties were added to wheat-soybean meal-based diet at the rates of 50, 100, 150, or 200 g/kg and fed to broilers from d 0 to 42. Inclusion of 150 and 200 g/kg of RSM resulted in reduced weight gain and increased feed conversion ratio (FCR) compared (P < 0.01) with the lower inclusion levels during the starter phase. For the entire trial (d 0 to 42), weight gain was greater (P < 0.01) for birds receiving diets with RSM from PR46W21 variety. It was concluded from the experiments that apart from the residual ether extract content, variety differences had no impact on ME of RSM, conventional processing reduced ME and relative bio-availability of P; and that the maximum level of RSM inclusion depends on maximum growth performance level desired.

The effect of DL-, L-isomers and DL-hydroxy analog administered at 2 levels as dietary sources of methionine on the metabolic and antioxidant parameters and growth performance of turkeys

A hypothesis was verified that dietary methionine (Met) improves the growth and antioxidant status of turkeys, and that its effects depend on dietary inclusion levels and sources. A total of 816 female Hybrid Converter turkeys was fed wheat-soybean meal-based diets supplemented with 3 sources of Met: DL-, L-isomers and DL-hydroxy analog (DLM, LM, and MHA, respectively). In 4 4-week periods (from one to 16 wk of age), dietary Met content corresponded to NRC (1994) recommendations or was increased by approximately 50% (in one to 8 wk by 44 to 46% and in 9 to 16 wk by 55 to 56% vs. the NRC guidelines) to match the recommendations of some breeding companies. Increased Met content resulted in higher final body weights of turkeys (P = 0.002), an improved feed conversion ratio (P = 0.049), increased total glutathione concentration and ferric reducing ability of plasma (FRAP) values, and decreased malondialdehyde (MDA) concentration (all P < 0.001) in the blood plasma of turkeys. In comparison with DLM, LM and MHA contributed to an increase in plasma glutathione concentration (P = 0.001), a decrease in plasma triacylglycerol (P = 0.003) and uric acid (P = 0.001) concentrations, and a decrease in liver MDA (P = 0.001) levels. A decrease in plasma MDA (vs. DLM) and lipid peroxides (LOOH) (vs. DLM and LM) concentrations as well as a decrease in plasma superoxide dismutase (SOD) activity (vs. DLM and LM) also were noted in the MHA treatment (P = 0.016, P = 0.001 and P = 0.011, respectively). In conclusion, the results of the study indicate that the antioxidant status of turkeys could be affected by dietary Met levels and sources. The dietary Met content increased by 50% relative to NRC recommendations, improved the growth performance of turkeys, and strengthened their antioxidant defense system. In comparison with DLM, LM and MHA could be considered positive nutritional factors as manifested by a beneficial decrease in plasma and hepatic MDA concentrations as well as an increase in plasma glutathione levels, and the effect of MHA was more pronounced.

Effect of different dietary methionine levels on the growth performance and tissue redox parameters of turkeys

A total of 630 8-week-old female Hybrid Converter turkeys were divided (based on their body weights) into 6 groups, with 7 replicates per group and 15 birds per replicate. All birds were fed identical isocaloric and isonitrogenous wheat-soybean meal-based diets without (group 1) or with (groups 2 to 6) increasing levels of supplemental methionine (Met). The total content of Met in diets 1 to 6 was as follows (%): 0.29, 0.32, 0.40, 0.47, 0.56, and 0.61 at 9 to 12 wk of age and 0.24, 0.28, 0.34, 0.42, 0.47, and 0.55 at 13 to 16 wk of age. In both feeding phases, dietary Met levels in group 3 corresponded to those recommended by the National Research Council (NRC) (1994). Different dietary Met concentrations had no influence on feed intake, the final body weights of turkeys or carcass dressing percentage. Only in the first experimental feeding period (9 to 12 wk), the lowest dietary Met content significantly deteriorated the feed conversion ratio (FCR), whereas the highest Met content led to a significant improvement in FCR. After 8 wk of experimental feeding, dietary treatment 1 contributed to a significant increase in the activity of catalase (CAT) (blood and breast muscles) and superoxide dismutase (liver), an increase in lipid peroxides concentrations (blood, breast muscle) and a decrease in total glutathione (GSH+GSSG) content (breast muscles), in comparison to treatment 3 which is comparable to NRC recommendations. The highest level of dietary Met significantly increased blood total antioxidant potential (FRAP) values and glutathione content in the liver. To sum up, in the final feeding period between 9 and 16 wk of age, the growth performance of female turkeys was not deteriorated by dietary Met deficiency or excess (–30% and up to +50% relative to NRC recommendations, respectively). The total antioxidant potential can be effectively increased by dietary Met supplementation, but the highest Met level may lead to unbalanced oxidative changes in the body as indicated by lower FRAP values and a lower GSH/GSSG ratio in the liver.
