Red-Light-Photosensitized NO Release and Its Monitoring in Cancer Cells with Biodegradable Polymeric Nanoparticles.

The role of nitric oxide (NO) as an "unconventional" therapeutic and the strict dependence of biological effects on its concentration require the generation of NO with precise spatiotemporal control. The development of precursors and strategies to activate NO release by excitation in the so-called "therapeutic window" with highly biocompatible and tissue-penetrating red light is desirable and challenging. Herein, we demonstrate that one-photon red-light excitation of Verteporfin, a clinically approved photosensitizer (PS) for photodynamic therapy, activates NO release, in a catalytic fashion, from an otherwise blue-light activatable NO photodonor (NOPD) with an improvement of about 300 nm toward longer and more biocompatible wavelengths. Steady-state and time-resolved spectroscopic and photochemical studies combined with theoretical calculations account for an NO photorelease photosensitized by the lowest triplet state of the PS. In view of biological applications, the water-insoluble PS and NOPD have been co-entrapped within water-dispersible, biodegradable polymeric nanoparticles (NPs) of mPEG-b-PCL (about 84 nm in diameter), where the red-light activation of NO release takes place even more effectively than in an organic solvent solution and almost independently by the presence of oxygen. Moreover, the ideal spectroscopic prerequisites and the restricted environment of the NPs permit the green-fluorescent co-product formed concomitantly to NO photorelease to communicate with the PS via Förster resonance energy transfer. This leads to an enhancement of the typical red emission of the PS offering the possibility of a double color optical reporter useful for the real-time monitoring of the NO release through fluorescence techniques. The suitability of this strategy applied to the polymeric NPs as potential nanotherapeutics was evaluated through biological tests performed by using HepG2 hepatocarcinoma and A375 melanoma cancer cell lines. Fluorescence investigation in cells and cell viability experiments demonstrates the occurrence of the NO release under one-photon red-light illumination also in the biological environment. This confirms that the adopted strategy provides a valuable tool for generating NO from an already available NOPD, otherwise activatable with the poorly biocompatible blue light, without requiring any chemical modification and the use of sophisticated irradiation sources.

Cryodesiccation-driven crystallization preparation approach for zinc(II)-phthalocyanine nanodots in cancer photodynamic therapy and photoacoustic imaging.

Multifunctional nanodots represent an emerging platform for overcoming the delivery challenges of poorly water-soluble drugs for use in the diagnosis and treatment of cancer. The authors describe the preparation of nanocrystallites composed of the water-insoluble photosensitizer zinc(II)-phthalocyanine in the form of nanodots by applying a cryodesiccation-driven crystallization approach. Modification of the surface of the nanodots with Pluronic F127 and folic acid endows them with excellent water solubility and stealth properties in blood. Under near-infrared (NIR) photoexcitation at 808nm, the nanodots are shown to produce singlet oxygen, which is widely used in photodynamic therapy of cancer. The nanodots exhibit strong NIR absorbance at 808nm and can be used as a non-toxic contrast agent for photoacoustic imaging of tissue. Disassembly of Hydrophobic Photosensitizer by Biodegradable Zeolitic Imidazolate Framework-8 for Photodynamic Cancer Therapy.

Photodynamic therapy (PDT), an alternative to conventional cancer therapeutics, has gained increasing attention for its noninvasive advantage and simultaneous fluorescence imaging property. PDT is a tripartite process that functions in the simultaneous presence of a photosensitizer (PS), light, and available oxygen molecules. However, many highly efficient PSs are hydrophobic and highly tend to self-aggregate in aqueous solution, leading to quick quenching of the PDT effect. Here we construct zeolitic imidazolate framework-8 (ZIF-8) containing water-insoluble photosensitizer zinc(II) phthalocyanine (ZnPc), a typical hydrophobic PS, by one-step coprecipitation process, named as ZnPc@ZIF-8. The micropores of ZIF-8 act as molecular cages to separate and maintain hydrophobic ZnPc in the monomeric state and protect it against self-aggregation, which enables the encapsulated ZnPc to generate cytotoxic singlet oxygen (1O2) under light irradiation (650 nm) in aqueous condition. The formed nanosystem of ZnPc@ZIF-8 can be endocytosed by cancer cells and exhibits red fluorescent emission with excellent photodynamic activity for cancer treatment in vitro. In addition, ZnPc@ZIF-8 is acid-sensitive and would completely degrade after PDT, which can be monitored by the self-quenching of fluorescence emission of ZnPc. This work paves a facile way for resolving the problem of solubility and bioavailability of hydrophobic PS by utilizing metal-organic frameworks as nanocarriers.

Water-Insoluble Photosensitizer Nanocolloids Stabilized by Supramolecular Interfacial Assembly towards Photodynamic Therapy

Nanoengineering of hydrophobic photosensitizers (PSs) is a promising approach for improved tumor delivery and enhanced photodynamic therapy (PDT) efficiency. A variety of delivery carriers have been developed for tumor delivery of PSs through the enhanced permeation and retention (EPR) effect. However, a high-performance PS delivery system with minimum use of carrier materials with excellent biocompatibility is highly appreciated. In this work, we utilized the spatiotemporal interfacial adhesion and assembly of supramolecular coordination to achieve the nanoengineering of water-insoluble photosensitizer Chlorin e6 (Ce6). The hydrophobic Ce6 nanoparticles are well stabilized in a aqueous medium by the interfacially-assembled film due to the coordination polymerization of tannic acid (TA) and ferric iron (Fe(III)). The resulting Ce6@TA-Fe(III) complex nanoparticles (referenced as Ce6@TA-Fe(III) NPs) significantly improves the drug loading content (~65%) and have an average size of 60 nm. The Ce6@TA-Fe(III) NPs are almost non-emissive as the aggregated states, but they can light up after intracellular internalization, which thus realizes low dark toxicity and excellent phototoxicity under laser irradiation. The Ce6@TA-Fe(III) NPs prolong blood circulation, promote tumor-selective accumulation of PSs, and enhanced antitumor efficacy in comparison to the free-carrier Ce6 in vivo evaluation.

Size-engineered biocompatible polymeric nanophotosensitizer for locoregional photodynamic therapy of cancer

Current approaches in use of water-insoluble photosensitizers for photodynamic therapy (PDT) of cancer often demand a nano-delivery system. Here, we report a photosensitizer-loaded biocompatible nano-delivery formulation (PPaN-20) whose size was engineered to ca. 20nm to offer improved cell/tissue penetration and efficient generation of cytotoxic singlet oxygen. PPaN-20 was fabricated through the physical assembly of all biocompatible constituents: pyropheophorbide-a (PPa, water-insoluble photosensitizer), polycaprolactone (PCL, hydrophobic/biodegradable polymer), and Pluronic F-68 (clinically approved polymeric surfactant). Repeated microemulsification/evaporation method resulted in a fine colloidal dispersion of PPaN-20 in water, where the particulate PCL matrix containing well-dispersed PPa molecules inside was stabilized by the Pluronic corona. Compared to a control sample of large-sized nanoparticles (PPaN-200) prepared by a conventional solvent displacement method, PPaN-20 revealed optimal singlet oxygen generation and efficient cellular uptake by virtue of the suitably engineered size and constitution, leading to high in vitro phototoxicity against cancer cells. Upon administration to tumor-bearing mice by peritumoral route, PPaN-20 showed efficient tumor accumulation by the enhanced cell/tissue penetration evidenced by in vivo near-infrared fluorescence imaging. The in vivo PDT treatment with peritumorally administrated PPaN-20 showed significantly enhanced suppression of tumor growth compared to the control group, demonstrating great potential as a biocompatible photosensitizing agent for locoregional PDT treatment of cancer.

Amphiphilic chitosan modified upconversion nanoparticles for in vivo photodynamic therapy induced by near-infrared light

Upconversion nanoparticles (UCNPs) have attracted much attention as potential photosensitizer carriers for photodynamic therapy (PDT) in deep tissues. In this study, hydrophilic UCNPs were prepared by coating amphiphilic chitosan (N-succinyl-N′-octyl chitosan, SOC) on the surface of hydrophobic oleic acid-capped NaYF4 UCNPs (OA-UCNPs). Water-insoluble photosensitizer zinc(II) phthalocyanine (ZnPc) was loaded into the SOC-coated UCNPs (SOC-UCNPs) via hydrophobic interactions to form a novel drug delivery system for in vivo deep tissue PDT triggered by near-infrared (NIR) light. The ZnPc-loaded SOC-UCNPs exhibited good dispersibility, excellent optical properties and good photostability. Under NIR light irradiation, the measurement of singlet oxygen production indicated that ZnPc effectively generated singlet oxygen induced by the emission from the UCNPs. Cell viability assays showed the low cytotoxicity of the UCNPs after surface modification. In vitro and in vivo therapeutic investigation evidenced the prominent PDT effects of ZnPc-loaded SOC-UCNPs upon NIR light irradiation. Furthermore, NIR imaging of ICG derivative-loaded SOC-UCNPs after intra-tumoral injection indicated the high retention of SOC-UCNPs only in the tumor site. Histological examination confirmed the negligible toxicity of ZnPc-loaded SOC-UCNPs on other organs. All the results demonstrate the promising potential of using SOC-UCNPs as new photosensitizer carriers for PDT of cancer and other diseases in deep tissues.

Tumor specificity and therapeutic efficacy of photosensitizer-encapsulated glycol chitosan-based nanoparticles in tumor-bearing mice

We reported the development of new nanoscale drug carriers, chitosan-based nanoparticles (CNPs) that can be used for photodynamic therapy. These carriers could encapsulate a photosensitizer, protophorphyrin IX (PpIX), and deliver it to tumor tissue. We already reported that CNPs presented the enhanced tumor target specificity in cancer therapy and imbibed various water insoluble anticancer agents into the hydrophobic multicores of nanoscale particles. In this study, we prepared photosensitizer-encapsulated CNPs by self-assembling amphiphilic glycol chitosan–5β-cholanic acid conjugates in an aqueous environment and then encapsulating the water-insoluble photosensitizer (PpIX), with high drug-loading efficiency (>90%) by using a dialysis method. Freshly prepared PpIX-encapsulated CNPs (PpIX-CNPs) had an average diameter of 290 nm and were stable in aqueous solutions for 1 month. As nanoscale drug carriers, PpIX-CNPs exhibited a sustained release profile in vitro and were non-toxic to tumor cells in the dark. In a cell culture system, we observed rapid cellular uptake of the PpIX-CNPs and the released PpIX from CNPs became highly phototoxic upon visible irradiation. In SCC7 tumor-bearing mice, PpIX-CNPs exhibited enhanced tumor specificity and increased therapeutic efficacy compared to free PpIX. Taken together, our results indicate that PpIX-CNPs have potential as an effective drug delivery system for clinical photodynamic therapy.

Photodynamic inactivation of enveloped viruses by buckminsterfullerene

Photodynamic reactions induced by singlet oxygen-generating agents are known to inactivate enveloped viruses. In this report we demonstrate that the water-insoluble photosensitizer buckminsterfullerene (C 60) can be used to mediate the inactivation of enveloped viruses. Viruses from two different families, Semliki Forest virus (SFV, Togaviridae) and vesicular stomatitis virus (VSV, Rhabdoviridae) were used as model systems. Buffered solutions containing C 60 plus either of these viruses were illuminated with visible light for up to 5 h, resulting in a loss of infectivity of more than 7 log 10/ml (TCID 50). Furthermore, it was demonstrated that this viral inactivation was oxygen-dependent and equally efficient in solutions containing protein. C 60 yields singlet oxygen in very high amounts and is completely inert to photo-oxidative destruction. In addition, it can be easily removed and recycled from aqueous solutions. For these reasons, it may prove useful in the inactivation of viruses in biological systems.
