A critical note on the new english title for Walras Éléments

Donald Walker and Jan van Daal’s recently translated the 3rd edition of Léon Walras’s Éléments d'économie politique pure under the title Elements of Theoretical Economics, in place of the conventional translation Elements of Pure Economics. This critical note points out that this new English title for Walras’s Éléments is inappropriate because it ignores Walras’s position as founder of the ‘Lausanne school’, with Vilfredo Pareto, as Walras’s successor, embracing Walras’s term ‘pure’ economics and, in his Cours d’économie politique, creating a distinction between pure and applied economics that is inconsistent with the translation of économie politique pure as ‘theoretical economics’.

Comment on Kokshoorn, B, and Blankers, BJ 'Response to Grisedale, KS and van Daal, A: comparison of STR profiling from low template DNA extracts with and without the consensus profiling method'

Kokshoorn and Blankers responded to our recent article by saying that replicate analysis and consensus profiling of low template samples was best in terms of reliability and objectivity. We agree that the consensus approach has benefits, particularly in eliminating non-repeating spurious alleles from the final profile. However, with the development of statistical models that can accommodate stochastic effects and allele drop in, it may be beneficial to perform a single amplification with three times the amount of template, since much information is lost from the profile using the consensus approach.

Response to Grisedale and Van Daal: comparison of STR profiling from low template DNA extracts with and without the consensus profiling method

In a recent contribution to this journal Grisedale and Van Daal concluded that a single STR analysis of all available template DNA is to be preferred over replicate analyses and a consensus approach when analyzing low template DNA samples. A single STR analysis approach does not allow for an assessment of the validity of the resulting DNA profile. We argue that the use of replicate amplifications is the best way to objectively quantify the extent of the stochastic variation in the data. By applying consensus methodology and/or a probabilistic model, the interpretation of the data will therefore be more objective and reliable.

Comment on “Low copy number typing has yet to achieve “general acceptance”” by Budowle, B., et al., 2009. Forensic Sci. Int. Genetics: Supplement Series 2, 551–552

Bruce Budowle, Arthur Eisenberg, and Angela van Daal recently authored an article entitled “Low Copy Number typing has yet to achieve ‘general acceptance”’ [1]. We have concerns about both the title and the content of this article.

Sequencing of Part of the Goat agouti Gene and SNP Identification

The agouti gene plays an important role in the synthesis and distribution of coat color in domestic animals. Most attention has been focused on the study of sequencing, identification of single nucleotide polymorphisms (SNPs), and association analysis with color variation in a number of animals as well as human beings (Voisey and van Daal 2002; Voisey et al. 2006; Rieder et al. 2001; Vage et al. 1997; Eizirik et al. 2003). Altogether, 10 color patterns have been observed and were postulated to be caused by 10 alleles at the agouti locus, with the allele for white or tan color dominant and the nine others codominant (Adalsteinsson et al. 1994). No more molecular data for the goat agouti gene have been obtained, but it has been shown to be located at 13q22 (Schibler et al. 1998). Earlier we sequenced intron 2 (AY291442) and exon 4 (DQ058664) of the goat agouti gene and found that allele T of 423G[T in DQ058664 might be responsible for the black phenotype or be linked with the causative site in goats (Tang et al. 2008). In this paper, other parts of the goat agouti gene have been sequenced and assembled (EF587236) and some SNPs have been identified in order to provide more useful information for further study of the association between variations and coat colors.

Conservation of intron position indicates separation of major and variant H2As is an early event in the evolution of eukaryotes

Genomic clones of Drosophila and Tetrahymena histone H2A variants were isolated using the corresponding cDNA clones (van Daal et al. 1988; White et al. 1988). The site corresponding to the initiation of transcription was defined by primer extension for both Drosophila and Tetrahymena genomic sequences. The sequences of the genomic clones revealed the presence of introns in each of the genes. The Drosophila gene has three introns: one immediately following the initiation codon, one between amino acids 26 and 27 (gln and phe), and one between amino acids 64 and 65 (glu and val). The Tetrahymena gene has two introns, the positions of which are identical to the first two introns of the Drosophila gene. The chicken H2A.F variant gene has been recently sequenced and it contains four introns (Dalton et al. 1989). The first three of these are in the same positions as the introns in the Drosophila gene. The fourth intron interrupts amino acid 108 (gly). In all cases the sizes and the sequences of the introns are divergent. However, the fact that they are in conserved positions suggests that at least two of the introns were present in the ancestral gene. A phylogenetic tree constructed from the sequences of the variant and major cell cycle-regulated histone H2A proteins from several species indicates that the H2A variant proteins are evolutionarily separate and distinct from the major cell cycle-regulated histone H2A proteins. The ancestral H2A gene must have duplicated and diverged before fungi and ciliates diverged from the rest of the eukaryote lineage. In addition, it appears that the variant histone H2A proteins analyzed here are more conserved than the major histone H2A proteins.
