Reducing conflict between the common vampire bat Desmodus rotundus and cattle ranching in Neotropical landscapes

AbstractIncreased cattle ranching in Neotropical landscapes has led to human–wildlife conflicts that complicate the relationship between agricultural production and biodiversity conservation. We review the literature related to conflicts between livestock production and the common vampire bat Desmodus rotundus, specifically, the factors that influence the incidence of problems caused by bites to cattle. We summarise the strategies proposed to reduce these problems and propose actions from the perspectives of cattle management, landscape ecology and the biology of the common vampire bat. The literature shows that free‐range extensive management systems, where cattle graze in areas with forest cover, near riparian zones, caves, secondary vegetation (generally shrublands and immature trees), and vegetation fragments and edges, increase the implications and the severity of the conflict. As a result of different selection forces, the wing morphology and echolocation characteristics of the common vampire bat facilitate its movement under these landscape conditions, thus favouring a scenario of greater interaction with cattle. We propose the establishment of a ‘buffer zone’ to separate the cattle as far as possible (at least 1 km) from the elements of the landscape that are key for the movement patterns of common vampire bats in cattle ranching areas. The feeding events of this species are positively associated with host availability and landscape elements that provide habitat and connectivity, which shows that the cause of this conflict originates from the process of cattle production and the invasion of the habitat of the common vampire bat. Anti‐rabies vaccination programmes in cattle and forest–cattle separation are the most important strategies to minimise conflict and prevent rabies outbreaks.

How to make a vampire.

Herein, we compared the developmental maturity of the cranium, limbs, and feeding apparatus in a perinatal common vampire bat relative to its mother. In addition, we introduce a method for combining two computed tomographic imaging techniques to three-dimensionally reconstruct endocasts in poorly ossified crania. The Desmodus specimens were scanned using microcomputed tomography (microCT) and diffusible iodine-based contrast-enhanced CT to image bone and soft tissues. Muscles of the jaw and limbs, and the endocranial cavity were segmented using imaging software. Endocranial volume (ECV) of the perinatal Desmodus is 74% of adult ECV. The facial skeletal is less developed (e.g., palatal length 60% of adult length), but volumes for alveolar crypts/sockets of permanent teeth are nearly identical. The forelimb skeleton is uniformly less ossified than the distal hind limb, with no secondary centers ossified and an entirely cartilaginous carpus. All epiphyseal growth zones are active in the brachium and antebrachium, with the distal radius exhibiting the greatest number of proliferating chondrocytes arranged in columns. The hind limb skeleton is precociously ossified from the knee distally. The musculature of the fore limb, temporalis, and masseter muscles appear weakly developed (6-11% of the adult volume). In contrast, the leg and foot musculature is better developed (23-25% of adult volume), possibly enhancing the newborn's capability to grip the mother's fur. Desmodus is born relatively large, and our results suggest they are born neurally and dentally precocious, with generally underdeveloped limbs, especially the fore limb.

AFFINITY OF BRAZILIAN WILD MAMMAL IMMUNOGLOBULINS TO BACTERIAL PROTEINS A AND G.

Staphylococcal A and streptococcal G proteins are widely used in immunoassays when specific immunological reagents are unavailable, such as for wild animals. The affinity of bacterial proteins A and G to the immunoglobulins of seven Brazilian mammals were tested, including black-tufted marmoset (Callithrix penicillata, n = 5), golden-bellied capuchin (Sapajus xanthosternos, n = 13), woolly mouse opossum (Micoureus demerarae, n = 6), long-nosed armadillo (Dasypus novemcinctus, n = 5), collared anteater (Tamandua tetradactyla, n = 5), ocelot (Leopardus pardalis, n = 6), and vampire bat (Desmodus rotundus, n = 5). Blood samples were collected from animals that were rescued in peri-urban rainforest fragments. Sera pools of each species were tested by ELISA to determine the intensity of each bacterial protein affinity to the immunoglobulins. When comparing the affinity to both proteins, immunoglobulins from D. rotundus, S. xanthosternos, and T. tetradactyla presented a higher affinity to protein G, whereas a higher affinity to protein A was found for immunoglobulins of C. penicillata and L. pardalis. The only species that presented a very low affinity to both bacterial proteins was M. demerarae. This study can be used as a reference for further studies on the development of sensitive and specific immunodiagnostic assays to be used for the monitoring of the health of these wild mammals.

Reduced injury risk links sociality to survival in a group-living primate

Sociality has been linked to a longer lifespan in many mammals, including humans. Yet, how sociality results in survival benefits remains unclear. Using 10 years of data and over 1,000 recorded injuries in rhesus macaques (Macaca mulatta), we tested two injury-related mechanisms by which social status and affiliative partners might influence survival. Injuries increased individual risk of death by 3-fold in this dataset. We found that sociality can affect individuals' survival by reducing their risk of injury but had no effect on the probability of injured individuals dying. Both males and females of high social status (measured as female matrilineal rank and male group tenure) and females with more affiliative partners (estimated using the number of female relatives) experienced fewer injuries and thus were less likely to die. Collectively, our results offer rare insights into one mechanism that can mediate the well-known benefits of sociality on an individual's fitness.

La Rabia, una enfermedad ancestral muy vigente

Rabies is an acute and fatal infectious disease. It mainly affects the central nervous system and the virus usually enters the body through the bite of a rabid animal. It is a zoonosis that still kills between 40,000 and 70,000 people each year and 40% of them are children. This disease has been known and referred to since the time of the Greeks. In several countries, urban rabies transmitted by dogs continues: however, in 2019, Mexico was recognized as “free of canine rabies by the World Health Organization. After the transmission of canine rabies to humans was controlled, bat rabies has become the leading cause of rabies in humans and animals in several Latin American countries. Vampire bat rabies is a problem that challenges public and animal health authorities, and solutions must be implemented in a coordinated manner, using and improving detection of the virus and antibodies in vaccinated animals, carrying out active surveillance. It is urgent to implement the “One Health” vision to combat this and other common diseases.

Drivers of Spatial Expansions of Vampire Bat Rabies in Colombia.

Spatial expansions of vampire bat-transmitted rabies (VBR) are increasing the risk of lethal infections in livestock and humans in Latin America. Identifying the drivers of these expansions could improve current approaches to surveillance and prevention. We aimed to identify if VBR spatial expansions are occurring in Colombia and test factors associated with these expansions. We analyzed 2336 VBR outbreaks in livestock reported to the National Animal Health Agency (Instituto Colombiano Agropecuario-ICA) affecting 297 municipalities from 2000-2019. The area affected by VBR changed through time and was correlated to the reported number of outbreaks each year. Consistent with spatial expansions, some municipalities reported VBR outbreaks for the first time each year and nearly half of the estimated infected area in 2010-2019 did not report outbreaks in the previous decade. However, the number of newly infected municipalities decreased between 2000-2019, suggesting decelerating spatial expansions. Municipalities infected later had lower cattle populations and were located further from the local reporting offices of the ICA. Reducing the VBR burden in Colombia requires improving vaccination coverage in both endemic and newly infected areas while improving surveillance capacity in increasingly remote areas with lower cattle populations where rabies is emerging.

Novel Borrelia Genotypes from Brazil Indicate a New Group of Borrelia spp. Associated with South American Bats.

The bacterial genus Borrelia comprises vector-borne spirochetes that have been classified into three major groups: the relapsing fever group (RFG), the Borrelia burgdorferi Johnson, Schmid, Hyde, Steigerwalt & Brenner sensu lato group (Bbsl), and the reptile-monotreme group (RMG). All three groups have been associated mainly with ticks and wild animals, especially rodents, birds, and reptiles. Here, we searched for Borrelia infection among 99 vampire bats [Desmodus rotundus (É. Geoffroy)] (Chiroptera: Phyllostomidae) from the Brazilian semiarid region. Through molecular investigation of bat internal organs, haplotypes of a potentially novel Borrelia organism were detected in 5% (5/99) of the bats. Borrelia DNA was detected in the liver, blood, spleen, kidney and brain, suggesting a systemic infection. Phylogenetic analyses inferred from partial sequences of the borrelial rrs and flaB genes indicated that the vampire bat-associated Borrelia sp. of this study form a monophyletic group with a newly reported Borrelia associated with a Colombia bat, distinct from the three main currently recognized groups of Borrelia spp., Bbsl, RFG, and RMG. These novel bat-associated Borrelia spp. from South America might have arisen through an independent event along the borrelial evolutionary history, since previous molecular reports of Borrelia organisms in bats or bat-associated ticks from Africa, Europe, and North America were all classified in the RFG.

Identification of viral RNA sequences in vampire bats (Desmodus rotundus) from central Mexico

Bats are important reservoirs of viral entities that cause diverse economical and health problems in several sectors. The purpose of this study was to determine the prevalence of the rabies virus in vampire bats. We also tested for 3 other zoonotic viruses in the collected tissue. We captured 45 vampire bats (Desmodus rotundus), which were measured and sexed under standard parameters. We followed an RT-PCR reaction to amplify the viral products of four virus types: chikungunya (CHIKV), dengue (DENV), rabies (RABV) and Zika virus (ZIKV) from cerebral tissue. We obtained an amplicon of 100 bp for ZIKV in 2 samples, representing a prevalence of 4.4%. An amplicon of 581 bp for RABV was observed in 9 individuals, which is equivalent to 20% of our sample. We obtained 4 different haplotypes for RABV and a single haplotype for ZIKV. We did not record the presence of DENV and CHIKV. We corroborated the presence of the family Rhabdoviridae in the vampire bat and recorded the presence of ZIKV for the first time in this bat species.

Bucking and charging defense of Baird’s tapir (Tapirella bairdii) from common vampire bats (Desmodus rotundus)

Los estudios con cámaras trampa han capturado gran diversidad de comportamientos de la fauna silvestre, resaltando la importancia de la ecología del comportamiento para cumplir con los objetivos de gestión y conservación de la vida silvestre. Reportamos intentos de depredación de vampiros comunes (Desmodus rotundus) hacia el tapir centroamericano (Tapirella bairdii) y describimos la respuesta defensiva del tapir. En septiembre de 2020, capturamos registros de esta interacción interespecífica en una estación de cámara trampa ubicada en la zona de amortiguamiento de la Reserva Rincón Rainforest en la zona noroeste de Costa Rica. Registramos vampiros intentando alimentarse de al menos 2 ejemplares de tapir centroamericano. Al detectar a los vampiros, los tapires reaccionaban para repelerlos sacudiéndose, corriendo, girando y persiguiendo a los murciélagos en vuelo. Estos registros generan nuevo conocimiento sobre el comportamiento defensivo de los tapires y son relevantes para preguntas relacionadas a la evolución del sigilo en el ataque de los murciélagos vampiro. Además, enfatizan el papel del monitoreo de estas interacciones interespecíficas en relación con la zoonosis en paisajes modificados por humanos.

Ecological determinants of rabies virus dynamics in vampire bats and spillover to livestock.

The pathogen transmission dynamics in bat reservoirs underpin efforts to reduce risks to human health and enhance bat conservation, but are notoriously challenging to resolve. For vampire bat rabies, the geographical scale of enzootic cycles, whether environmental factors modulate baseline risk, and how within-host processes affect population-level dynamics remain unresolved. We studied patterns of rabies exposure using an 11-year, spatially replicated sero-survey of 3709 Peruvian vampire bats and co-occurring outbreaks in livestock. Seroprevalence was correlated among nearby sites but fluctuated asynchronously at larger distances. A generalized additive mixed model confirmed spatially compartmentalized transmission cycles, but no effects of bat demography or environmental context on seroprevalence. Among 427 recaptured bats, we observed long-term survival following rabies exposure and antibody waning, supporting hypotheses that immunological mechanisms influence viral maintenance. Finally, seroprevalence in bats was only weakly correlated with outbreaks in livestock, reinforcing the challenge of spillover prediction even with extensive data. Together our results suggest that rabies maintenance requires transmission among multiple, nearby bat colonies which may be facilitated by waning of protective immunity. However, the likelihood of incursions and dynamics of transmission within bat colonies appear largely independent of bat ecology. The implications of these results for spillover anticipation and controlling transmission at the source are discussed.

Social effects of rabies infection in male vampire bats (Desmodus rotundus).

Rabies virus (RABV) transmitted by the common vampire bat (Desmodus rotundus) poses a threat to agricultural development and public health throughout the Neotropics. The ecology and evolution of rabies host-pathogen dynamics are influenced by two infection-induced behavioural changes. RABV-infected hosts often exhibit increased aggression which facilitates transmission, and rabies also leads to reduced activity and paralysis prior to death. Although several studies document rabies-induced behavioural changes in rodents and other dead-end hosts, surprisingly few studies have measured these changes in vampire bats, the key natural reservoir throughout Latin America. Taking advantage of an experiment designed to test an oral rabies vaccine in captive male vampire bats, we quantify for the first time, to our knowledge, how rabies affects allogrooming and aggressive behaviours in this species. Compared to non-rabid vampire bats, rabid individuals reduced their allogrooming prior to death, but we did not detect increases in aggression among bats. Yellow fever virus investigation in tissues of vampire bats Desmodus rotundus during a wild yellow fever outbreak in Brazilian Atlantic Forest

In the last decade a large outbreak of Yellow Fever (YF) has been observed in Brazilian Atlantic Forest region, traditionally a non-endemic area. In this scenario, the role of wild mammal species as YF reservoirs can be questioned, especially the hematophagous bat, Desmodus rotundus. So, the objective of this study was to analyze molecularly the presence of the YF virus (YFV) in hematophagous bats during a YF outbreak in Brazil. Twenty-one samples were collected from seven adult male hematophagous bats D. rotundus. As YFV is considered a viscerotropic and neurotropic virus, samples of liver, kidney and brain were collected and molecularly analyzed using the RT qPCR technique. The animals were captured according to ethical protocols during a YF outbreak in Brazil in 2017, from a region of the Brazilian Atlantic Forest. The results revealed that the analyzed tissue samples were not infected with the YFV. The negative results for this bat species allow us to infer that other animals may be reservoirs of this virus in this ecosystem and they probably have not been identified yet. Therefore, health surveillance actions are essential to monitor the role of wild animals in the YF dissemination in Brazilian Atlantic Forest and alert to the possibility of new geographic amplification of areas where YF occurs. This research encourages the new search about the role of wild animals on YFV transmission and reinforces the importance of epidemiological surveillance in the transmission of human infectious diseases.

A recombinant rabies vaccine that prevents viral shedding in rabid common vampire bats (Desmodus rotundus).

Vampire bat transmitted rabies (VBR) is a continuing burden to public health and agricultural sectors in Latin America, despite decades-long efforts to control the disease by culling bat populations. Culling has been shown to disperse bats, leading to an increased spread of rabies. Thus, non-lethal strategies to control VBR, such as vaccination, are desired. Here, we evaluated the safety and efficacy of a viral-vectored recombinant mosaic glycoprotein rabies vaccine candidate (RCN-MoG) in vampire bats (Desmodus rotundus) of unknown history of rabies exposure captured in México and transported to the United States. Vaccination with RCN-MoG was demonstrated to be safe, even in pregnant females, as no evidence of lesions or adverse effects were observed. We detected rabies neutralizing antibodies in 28% (8/29) of seronegative bats post-vaccination. Survival proportions of adult bats after rabies virus (RABV) challenge ranged from 55–100% and were not significantly different among treatments, pre- or post-vaccination serostatus, and route of vaccination, while eight pups (1–2.5 months of age) used as naïve controls all succumbed to challenge (P<0.0001). Importantly, we found that vaccination with RCN-MoG appeared to block viral shedding, even when infection proved lethal. Using real-time PCR, we did not detect RABV nucleic acid in the saliva samples of 9/10 vaccinated bats that succumbed to rabies after challenge (one was inconclusive). In contrast, RABV nucleic acid was detected in saliva samples from 71% of unvaccinated bats (10/14 sampled, plus one inconclusive) that died of the disease, including pups. Low seroconversion rates post-vaccination and high survival of non-vaccinated bats, perhaps due to earlier natural exposure, limited our conclusions regarding vaccine efficacy. However, our findings suggest a potential transmission-blocking effect of vaccination with RCN-MoG that could provide a promising strategy for controlling VBR in Latin America beyond longstanding culling programs.

Geographic Distribution of Common Vampire Bat Desmodus rotundus (Chiroptera: Phyllostomidae) Shelters: Implications for the Spread of Rabies Virus to Cattle in Southeastern Brazil.

Desmodus rotundus bats show a complex social structure and developed adaptive characteristics, considered key features of a pathogen disseminator, such as the rabies virus, among bats and other mammals, including cattle and humans. Our aim was to understand the correlation between the environment and the ecological features of these bats in bovine rabies outbreaks. Geostatistical analyses were performed, covering 104 cattle positives for rabies, between 2016 and 2018, in 25 municipalities, in addition to the characteristics of D. rotundus colonies mapped during this period in the state of São Paulo, Brazil. Data from the shelters showed that 86.15% were artificial, mainly abandoned houses (36.10%) and manholes (23.87%), in addition to demonstrating a correlation between these shelters and a higher concentration of bovine rabies cases. Due to their adaptive capacity, these bats choose shelters close to the food source, such as livestock. In Brazil, D. rotundus is the main transmitter of rabies and the cause of outbreaks in cattle and deaths in humans, considering the advance of humans in previously preserved ecosystems. There seems to be a correlation between the impact of anthropic changes on the environment, mainly for the expansion of pasture for cattle and the outbreaks of bovine rabies in this area.

ZOONOSIS CONTROL POLICY IN THE STATE OF RIO GRANDE DO SUL (PART 2)

Background: This is the transcription of the interview with the scientist, graduated in veterinary medicine, coordinator of the Herbivorous Rabies Control Program, Wilson Hoffmeister Júnior, was interviewed. The Inspector of the Secretariat of Agriculture, Livestock, and Rural Development (SEAPDR) of the State of Rio Grande do Sul, which develops one of the work fronts of sanitary defense. Objective: to analyze the work of prevention and control of rabies in the state of Rio Grande do Sul (Brazil). Methods: the interview was formulated using the Herbivorous Rabies Control Program (PNCRH-RS) as an information base. Results and Discussion: The PNCRH-RS is a public policy program that has operated for decades in the state of Rio Grande do Sul, and it has contributed to the elimination of certain types of rabies in the state. In addition to keeping rabies transmitted by vampire bats under control, preventing or reducing economic losses, and ensuring the health and quality of the herd in the state of Rio Grande do Sul. Conclusions: the uninterrupted continuity of the PNCRH-RS guaranteed the economic viability of rural producers, increased their profitability, and ensured animal health and public health in the state of Rio Grande do Sul.

How to Make a Vampire: Part II. Getting Ready to Feed

The mammalian feeding system is made up of multiple components that influence facial variation, including the masticatory muscles, bony jaw apparatus, and teeth. However, it is not fully understood how the development of these components are coordinated and are impacted by life history and diet.Bats are the only mammals that continue to nurse until they are almost adult size, and often after they become volant. Common vampire bats (Desmodus rotundus) have a relatively prolonged juvenile period among bats, can nurse up to nine months, and later solely subsist off of blood. Feeding system maturity may therefore be delayed in bats, and particularly delayed in Desmodus, as a result of their lengthy juvenile period and liquid diet. In this study, we examine the developmental maturity of the cranium and feeding apparatus in the late fetus of a common vampire bat (Desmodus rotundus) relative to an adult (its mother).The heads of each specimen were microCT scanned twice. Conventional microCT was used to image mineralized tissues. Diffusible iodine‐based contrast‐enhanced CT (diceCT) imaging was used to image soft tissues. Cranial length and palate length measurements were acquired from unstained scans to evaluate size and jaw apparatus development. Masticatory muscles (e.g., medial pterygoid, zygomaticomandibularis) and permanent tooth crypt volumes were acquired from stained tissue scans to evaluate masticatory muscle and tooth development. Developmental maturity of these structures was assessed by calculating a percentage of the growth completed (i.e., [fetal muscle volume/adult muscle volume]*100).In the fetus, permanent tooth crypt volume is already comparable to the adult condition despite cranial length and palate length being &lt;75% of adult size. Masticatory muscle volume maturity is highest in the lateral pterygoid muscle (~35%), whereas other masticatory muscles are far less developed (e.g., medial pterygoid volume maturity, ~20%; temporalis profundus volume maturity, ~20%; temporalis medius volume maturity, ~6%).These results reveal highly precocious permanent tooth development in Desmodus. Although this may facilitate transition to hematophagy, Desmodus do not fly until eight to ten weeks, so the precocious incisors may primarily aid the infant to grip its mother’s skin. Differential development of the masticatory muscles may also be influenced by the need for young bats to use their mouths to help anchor themselves to their mother early in life. This may explain the advanced maturity of the lateral pterygoid muscle, which would protrude the jaw and help to facilitate a more secure grip.

One step closer to a transmissible vaccine for rabies virus.

A recent study in PLOS Biology shows that a betaherpesvirus circulating with the vampire bat, Desmodus rotundus, could serve as an effective vector for a transmissible vaccine capable of reducing the risk of rabies virus spillover in Peru.

Longitudinal deep sequencing informs vector selection and future deployment strategies for transmissible vaccines.

Vaccination is a powerful tool in combating infectious diseases of humans and companion animals. In most wildlife, including reservoirs of emerging human diseases, achieving sufficient vaccine coverage to mitigate disease burdens remains logistically unattainable. Virally vectored "transmissible" vaccines that deliberately spread among hosts are a potentially transformative, but still theoretical, solution to the challenge of immunising inaccessible wildlife. Progress towards real-world application is frustrated by the absence of frameworks to guide vector selection and vaccine deployment prior to major in vitro and in vivo investments in vaccine engineering and testing. Here, we performed deep sequencing on field-collected samples of Desmodus rotundus betaherpesvirus (DrBHV), a candidate vector for a transmissible vaccine targeting vampire bat-transmitted rabies. We discovered 11 strains of DrBHV that varied in prevalence and geographic distribution across Peru. The phylogeographic structure of DrBHV strains was predictable from both host genetics and landscape topology, informing long-term DrBHV-vectored vaccine deployment strategies and identifying geographic areas for field trials where vaccine spread would be naturally contained. Multistrain infections were observed in 79% of infected bats. Resampling of marked individuals over 4 years showed within-host persistence kinetics characteristic of latency and reactivation, properties that might boost individual immunity and lead to sporadic vaccine transmission over the lifetime of the host. Further, strain acquisitions by already infected individuals implied that preexisting immunity and strain competition are unlikely to inhibit vaccine spread. Our results support the development of a transmissible vaccine targeting a major source of human and animal rabies in Latin America and show how genomics can enlighten vector selection and deployment strategies for transmissible vaccines.
