Development and Validation of Stability Indicating UV Spectrophotometric Method for Estimation of Resveratrol in Bulk and Tablet Dosage Form

A simple, sensitive, accurate, trustworthy, and stable UV spectrophotometric approach has been devised to quantify resveratrol in pharmaceutical formulations and bulk materials. After scanning the UV spectrum from 200 to 400 nm, the highest wavelength for absorption was determined to be 306 nm. For the medication, Beer’s rule has been adhered to within a concentration range of 1 to 5 μg/mL. While the developed method was able to recover a good amount of drug (%Recovery), the precision study’s percentage RSD values were ≥2%. The method demonstrated effective functionality for a pharmaceutical dosage form containing resveratrol, free from interference from the excipients. Correspondingly, the limit of detection (LoD) for resveratrol were 0.13 and 0.40 μg/mL. The results of this investigation have been confirmed in accordance with ICH standards. Simultaneous UV Spectrophotometric Method for Estimation of Berberine and Withanolide in Polyherbal Immunity Booster Dosage Form

The majority of commercial herbal preparations used to improve immunity lack standardization with regard to the active ingredients marker molecules. The objective of present study is the simultaneous estimation of berberine and withanolide in herbal immunity booster tablet using UV spectrophotometric method. The method was validated according to ICH guidelines. The results indicated that berberine and withanolide have maximal absorption wavelength i.e. λmax at 348 nm and 231 nm, respectively. Withanolide and berberine were found to behave linearly over the concentration range of 5–25μg/mL with R2 value of 0.9998 and 0.9999, respectively. Withanolide and berberine have shown % relative standard deviation (RSD) values of 0.07 and 0.09, respectively, in accuracy study. The intraday and interday precision was deemed adequate with a % RSD fewer than 2. Further, the limit of detection (LOD) and limit of quantization (LOQ) value for berberine and withanolide were found to be 0.12 mg/mL and 0.35mg/mL and 0.07, 0.20 mg/mL, respectively. In conclusion, the proposed method was validated in accordance with the International Conference on Harmonization (ICH) guidelines. The developed method can be employed to determine how much berberine and withanolide are present in pharmaceutical preparations.

Development and Validation of Simple UV Spectrophoto-metric Method for Estimation of Azithromycin in Dosage Form using Hydrotropic Solubilization Technique

Objective: A new UV method was developed for the estimation of azithromycin in tablets using the hydrotropic solubilization method and it was validated as per ICH guidelines. Methods: The solubility of azithromycin was studied in various hydrotropic agents and the one with the best solubility was considered as solvent for developing and validating the UV spectrophotometric method. Results: The best solubility of azithromycin was witnessed in a 2M sodium acetate solution in water. The absorption maximum of azithromycin in 2M sodium acetate was found to be 221 nm. The method was found to be linear in the range of 10-50µg/ml. The precision of the method was good and the recovery of drugs was well within the acceptance limits of 80-120%. Conclusion: The proposed UV method was found suitable for the estimation of azithromycin in formulations and is simple, selective, reproducible, and accurate with good precision, and can be successfully applied to routine analytical purposes.

Development and validation of UV spectrophotometric method for estimation of naringenin in phytosomal formulation: interlaboratory comparison, capability, and statistical analysis.

The goal of the current work was to establish a simple, reproducible, and reliable UV spectrophotometric method for determining naringenin in phytosomal formulations. The solvent methanol was optimized and spectrophotometric analysis was carried out at 289nm. The proposed method was also validated for linearity, specificity, accuracy, precision, ruggedness, and robustness in accordance with the International Council for Harmonisation (ICH). With a correlation coefficient of 0.9982, the concentration of naringenin in the range of 2-14μg/ml follows Lambert-Beer rule. The accuracy ranged between 99.33 and 99.75%. The predicted percent recovery was found to be 99.48 ± 0.41. It was found that the limits of detection (LOD) and quantification were 0.54μg/ml and 1.66μg/ml, respectively. The normality of the data is accepted by the Shapiro-Wilk test (P = 0.9065) and Shapiro-Francia test (W = 0.9866). On the Bland-Altman plot, an acceptable repeatability coefficient was found. The remarkable inter-laboratory repeatability was proved by the Youden plot, which was also utilized to spot random and total errors. Levey-Jennings charts and other control diagrams have demonstrated that the method is statistically controlled. Greater values of Cp (1.03) and Cpk (0.83) were seen in the capability analysis, indicating that the method could analyze the samples reliably and consistently with minimum variation. The validation report demonstrated that the proposed method was appropriate for naringenin detection and analysis in phytosomal formulations, indicating that it can be employed for routine laboratory analysis as well.

Two simple, precise and economical UV methods have been developed for the simultaneous estimation of Sitagliptinphosphate and Metformin hydrochloride in bulk and pharmaceutical dosage form. Method A is Absorbance maximamethod,whichisbasedonmeasurementofabsorptionatmaximumwavelengthof266nmand232nmforSitagliptinphosphateandMetforminhydrochloriderespectively.MethodBisareaundercurve(AUC),inthewavelength range of244-279 nm for Sitagliptin phosphateand222-240nm for Metformin hydrochloride. Linearityfordetector response wasobserved in the concentration range of25-225µg/ml for Sitagliptin phosphate and 2-12µg/ml for Metformin hydrochloride. The accuracy of the methods was assessed by recovery studies and was found tobe99.64%and98.98%forSitagliptinphosphateandMetforminhydrochloride.Thedevelopedmethodwasvalidatedwithrespecttolinearity,accuracy(recovery),precisionandspecificity.Theresultswerevalidatedstatistically as per ICH Q2 R1 guideline and were found to be satisfactory. The proposed methods were successfullyappliedforthedeterminationofforSitagliptinphosphateandMetforminhydrochlorideincommercialpharmaceuticaldosage form.

Development of UV spectrophotometric method for estimation of curcumin in bulk drug and nanogel formulation: A hydrolytic degradation studies

The main purpose of this research was to develop and validate a cost effective UV spectrophotometric method for determination of curcumin in bulk drug and nanogel formulation. The method was validated according to the guideline of International Conference on Harmonisation (ICH). The curcumin was found to be soluble in methanol with λmax of 425 nm. Curcumin over the concentration level of (1-8 µg mL) depicted linear relation in the plot of concentration vs. absorbance. The regression equation of graph was y = 0.1137x - 0.0138 with R value of 0.999. The limits of detection and quantification were 0.23 μg mLand 0.75 μg mL, respectively. The method was accurate, precise, reproducible and robust since all the samples analyzed had relative standard deviation less than 2%. No statistically significant difference between theoretical and measured concentrations was detected. The degradation kinetics of free curcumin (k = 0.042 µg mL.h, 0.53 µg mL.h and 0.068 µg mL.h) and nanogel formulation (k = 0.178 h, 0.003 h and 0.005 h) were zero and first order, respectively. The proposed method was found to be specific while estimating curcumin loaded nanogel formulation without interference of excipients. The developed method was cost effective and can be used for routine quality control analysis of curcumin.

The method was developed by using 0.1 N HCl as a solvent and absorbance was measured at 312 nm. The drug exhibited the linearity in the concentration range of 2-12 μg/ml with a correlation coefficient of 0.9978. The % recovery of the drug was found to be 98.62 % - 100.5 %. The method was validated as per ICH guidelines. The proposed method is economical and sensitive for the estimation of Famciclovir in bulk and tablet dosage form.

The main purpose of this study was to develop a simple precise, rapid and accurate UV-visible spectrophotometric method for determination of Bumetanide in spiked human urine by extracting the Bumetanide from spiked human urine using ethyl acetate after extraction it was scanned between 200-400nm by using UV detector and its absorbance maxima was found to be 222nm.The calibration curve was linear in the range of 1-17 µg/ml. .the recovery and assay studies of bumetanide were within 93-94.85% indicating that the proposed method can be estimation of bumetanide.

Abstract: Background: Tea tree oil, also known as Melaleuca alternifolia essential oil is widely used in skin care cosmeceuticals for its antibacterial, antifungal, analgesic and anti-inflammatory effect. The complex and variable composition of the oil poses many challenges to the analytical chemist and a recent survey of analytical methods indicated only a few simple and validated methods for estimation of the oil. Aim: A quality by design approach has therefore been adopted to develop a simple and novel UV spectrophotometric method for estimation of tea tree oil in bulk and cosmeceutical creams. Materials and Methods: UV spectrophotometric method was developed for estimation of Tea tree oil using dichloromethane- methanol (54 % v/v) solvent system. The method was then validated under optimized conditions for accuracy, precision and ruggedness, limit of detection and limit of quantification as per ICH: Q2 (R1) guidelines. Results: A characteristic spectral peak was observed at 267 nm and linearity was established over a concentration range of 10-160 mcg/ml with R2 value of 0.9961. Accuracy based on recovery studies, 100.5-113.6 %, precision and ruggedness based on % RSD values less than 2 %, LOD, 0.1277mcg/ml and LOQ and 4.195 mcg/ml indicated that the method is sensitive enough to be used for routine estimation. Conclusion: The method is robust and has been used to determine the essential oil content of two cosmeceutical creams. Tramadol could be a painkiller just like an opioid. It acts within the central system (CNS) to alleviate pain. Tramadol is employed to treat moderate to severe pain in adults widely used for clinical purposes. During this respect, we aimed to develop an easy and economic UV spectrophotometric method for estimation of Tramadol Hydrochloride in bulk, injection, and tablet dosage form and validate as per ICH guidelines. Analytical research and development may be a process that assures quality attributes and internal control of the products. Tramadol shows the most absorbance at wavelength 270nm. Tramadol showed the linearity range 20-160µg per ml for this procedure with correlation (R2) was 0.9998. The current methods were found to be simple, linear, precise, accurate, and sensitive and may be used for routine internal control analysis for the estimation of Tramadol Hydrochloride bulk and tablet dosage form.
