Sensory-Guided Identification and Characterization of Kokumi-Tasting Compounds in Green Tea (Camellia sinensis L.).

The chemical substances responsible for the kokumi taste of green tea infusion are still unclear. Here, we isolated the kokumi compound-containing fractions from green tea infusion through ultrafiltration, and the major kokumi compounds were characterized as γ-Glu-Gln and γ-Glu-Cys-Gly (GSH) through ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS). The results indicated that peptides and amino acids were essential compounds in the kokumi-enriched fractions for conducting the sense of kokumi. L-theanine had an enhancing effect on the kokumi taste of green tea infusion, which was confirmed in the sensory reconstitution study. Thus, peptides, especially γ-Glu-Gln and GSH, are the major kokumi compounds in green tea infusion, which has the potential of improving the flavor of tea beverages.

Residue, dissipation, and dietary intake evaluation of fenpyroximate acaricide in/on guava, orange, and eggplant under open field condition.

Fenpyroximate is a widely used acaricide applicable in many crops. In this study, the residue behavior of fenpyroximate on eggplant, orange, and guava was investigated. The chronic and acute dietary intake was calculated at several sampling points, and preharvest intervals (PHI) were proposed to ensure compliance with the existing maximum residue levels. A simple extraction protocol combined with ultrahigh-performance liquid chromatography–tandem mass spectrometry (UHPLC-MS/MS) was employed to quantify residue levels. The method was successfully validated according to the European Union (EU) guidelines, and a limit of quantification of 0.01 mg/kg was set. The dissipation patterns in all crops could be described by the first-order kinetics model with half-lives of 1.7, 2.2, and 1.9 days for eggplants, guavas, and oranges, respectively. The dietary risk assessment at the authorized or more critical application patterns was acceptable for the consumers. For oranges and eggplant, a PHI of 3 and 7 days, respectively, can be proposed; however, a proposal was not possible for guava due to the absence of maximum residue limits (MRLs) and quantitative residue findings at all sampling points tested. The current work not only contributes to the practical application of fenpyroximate related to residue management in dryland areas, such as Egypt, but can also be used to estimate the appropriate PHIs and support the authorization of plant protection products as supplementary information.

ATP-Responsive Strand Displacement Coupling with DNA Origami/AuNPs Strategy for the Determination of Microcystin-LR Using Surface-Enhanced Raman Spectroscopy.

The DNA origami-mediated self-assembly strategy has emerged as a powerful tool in surface-enhanced Raman spectroscopy (SERS). However, these self-assembly approaches typically do not possess high detection specificity. Herein, a novel strategy based on adenosine triphosphate (ATP)-responsive strand displacement (ARSD) coupling with DNA origami/AuNPs for SERS analysis of microcystin-LR (MC-LR) is presented. In the presence of MC-LR and ATP molecules, nucleic acid sensing structures fabricated with anti-MC-LR aptamer (T1) and ATP aptamer (T2) were triggered to release the remaining ATP. In addition, DNA origami-assisted assembly results in the formation of homogeneous plasmonic nanostructures for Raman enhancement via strong plasmonic coupling. After the binding in the gaps of functionalized DNA origami/AuNPs, the Raman shift of the ATP molecules becomes detectable, leading to increased SERS intensity in 734 cm-1. A linear response to MC-LR was obtained in the concentration range of 1.56-50 μg·L-1, and the limit of detection (LOD) was 0.29 μg·L-1. Combined with the solid-phase extraction sample pretreatment for extraction and 10-fold concentration, this proposed method was successfully used to detect MC-LR type in real lake-water samples with good recoveries of 98.4-116% and relative standard deviations of 1.9-6.7%. Furthermore, for the detection of MC-LR in contaminated lake-water samples, the results of the developed method and ultrahigh-performance liquid chromatography-tandem mass spectrometry were found to be in agreement with relative errors between -12 and 2.4%. The proposed strategy provides a sensitive recognition and signal amplification platform for trace MC-LR analysis as well as innovative nucleic acid sensing structures for toxin analysis more generally.

Characterization and determination of benvitimod, an unknown risk substance in cosmetics, using nuclear magnetic resonance spectroscopy and HPLC-MS/MS.

Ultrahigh-performance liquid chromatography-tandem high-resolution mass spectrometry, combined with preparative chromatography and nuclear magnetic resonance spectroscopy, a new method for identifying unknown risk substance structures in cosmetics has been established. Moreover, HPLC-MS/MS was developed for the determination of benvitimod in cosmetics. The sample was collected in ultrahigh-performance liquid chromatography-tandem high-resolution mass spectrometry, and the molecular formula of the unknown was obtained as C17 H18 O2 . After preparative chromatography enrichment and purification, the enriched compound was scanned by nuclear magnetic resonance spectroscopy, and the chemical structure of the unknown compound was confirmed to be benvitimod. Subsequently, the separation was determined in multiple reaction monitoring mode. The results showed that the linearity of benvitimod was good in the range of 1-100 μg/L with a correlation coefficient r2 >0.999; the limits of detection and quantification were 0.02 mg/kg and 0.067mg/kg; the precision and stability were good; and the average recoveries were 104.2%, 108.2%, and 108.7% for low, medium, and high spiked concentrations, respectively. Forty batches of cosmetics were screened, of which two batches were detected with the illegal addition of benvitimod at 2.48 and 3.13 g/kg. The method effectively solved the loopholes in regulation and provided a research basis for the qualitative identification of structurally unknown compounds in cosmetics.

Reduction in residual cyantraniliprole levels in spinach after various washing and blanching methods

Pesticides are used to protect crops from pests and diseases. However, as many pesticides are toxic to humans, it is necessary to assess methods that can remove pesticide residues from agricultural products before human consumption. Spinach is consumed immediately after a relatively simple washing and heating process in the Republic of Korea. Cyantraniliprole is used as a systemic insecticide during spinach cultivation, which means it might remain in the crop after processing. Consequently, it is important to assess whether residues can be reduced to levels that are harmless to the human body after processing. This study investigated lowering the residual cyantraniliprole levels in spinach after washing and blanching. The amount of cyantraniliprole residue in the spinach samples sprayed with cyantraniliprole during cultivation was analyzed using ultrahigh-performance liquid chromatography-tandem mass spectrometry (UHPLC–MS/MS). The time of each washing and blanching method was set at 1, 3, and 5 min. The residual levels of cyantraniliprole decreased by 15.1–54.6% and 60.1–93.5% based on the washing and blanching methods employed. The most effective washing method to lower residual cyantraniliprole levels was steeping with a neutral detergent, resulting in cyantraniliprole reduction by 42.9–54.6%. When spinach was blanched after steeping washing with a neutral detergent, the largest removal rates of 77.9 and 91.2% were observed after 1 and 3 min of blanching, respectively. Blanching for 5 min after steeping and running washing exhibited the highest reduction rate of 93.5%. Therefore, a considerable amount of cyantraniliprole residue in spinach could be removed by washing or blanching. Based on the results of this study, blanching after steeping washing can be implemented as an effective method of lowering pesticide concentrations in spinach and other crops, thereby reducing their potential toxicity to humans upon consumption.

Identification of Metabolite Markers Associated with Kidney Function.

Background Chronic kidney disease (CKD) is a global public health problem. Identifying new biomarkers that can be used to calculate the glomerular filtration rate (GFR) would greatly improve the diagnosis and understanding of CKD at the molecular level. A metabolomics study of blood samples derived from patients with widely divergent glomerular filtration rates could potentially discover small molecule metabolites associated with varying kidney function. Methods Using ultrahigh-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS), serum was analyzed from 53 participants with a spectrum of measured GFR (by iohexol plasma clearance) ranging from normal to severe renal insufficiency. An untargeted metabolomics assay (N ¼ 214) was conducted at the Calibra-Metabolon Joint Laboratory. Results From a large number of metabolomics-derived metabolites, the top 30 metabolites correlated to increasing renal insufficiency according to mGFR were selected by the random forest method. Significant differences in metabolite profiles with increasing stages of CKD were observed. Combining candidate lists from six other unique statistical analyses, six novel, potential metabolites that were reproducibly strongly associated with mGFR were selected, including erythronate, gulonate, C-glycosyltryptophan, N-acetylserine, N6-carbamoylthreonyladenosine, and pseudouridine. In addition, hydroxyasparagine were strongly associated with mGFR and CKD, which were unique to this study. Conclusions Global metabolite profiling of serum yielded potentially valuable biomarkers of different stages of CKD. Additionally, these potential biomarkers might provide insight into the underlying pathophysiologic processes that contribute to the progression of CKD as well as improve GFR estimation.

Ammonization of the R- and S-Epimers of Ergot Alkaloids to Assess Detoxification Potential.

Detoxification of ergot-contaminated feed by ammonia would be a practical application, given that ammonia is routinely used in the agriculture industry. To assess the effects of ammonia on ergot alkaloids, natural ergot-contaminated wheat was ammoniated. The total concentration of ergot alkaloids (R- and S-epimers) decreased after exposure to ammonia (8–29%). Separately, the total R-epimers decreased in concentration (40–66%), whereas the total S-epimers increased (21–81%). Specific ergot alkaloids demonstrated degradation and/or epimerization after exposure to ammonia, potentially associated with structural differences, and influenced the total concentrations observed. Ammonization of ergot standards resulted in potential degradation products and epimerization, supporting the above results. The use of ultrahigh-performance liquid chromatography–tandem mass spectrometry provides an updated assessment of the detoxification potential of ammonia for ergot alkaloids and the quantification of the S-epimers. Ammonia alters the R- and S-epimers of ergot alkaloids, which may lead to a potential practical detoxification process of ergot-contaminated feed.

Associations of plasma carnitine, lysine, trimethyllysine and glycine with incident ischemic stroke: Findings from a nested case-control study

Carnitine biosynthesis has been related to fatty acid oxidation, a process probably exerting neuroprotective effects. However, the role of carnitine biosynthesis in the development of ischemic stroke (IS) remains unclear. We aimed to examine the associations between plasma markers of carnitine biosynthesis and the IS risk. We performed a case-control study nested in a community-based cohort (2013-2018, n=16457). The study included 321 incident cases of IS and 321 controls matched by age and gender. Carnitine, lysine, trimethyllysine (TML), glycine, and their ratios were measured/calculated in the baseline plasma samples using ultra-high performance liquid chromatography-tandem mass-spectrometry (UHPLC-MS/MS). Conditional logistic regression analyses were used to calculate odds ratios (ORs) and their 95% confidence intervals (CIs). Plasma carnitine, lysine, TML, and glycine were not significantly associated with the IS risk, although a gradually reduced risk was observed across the increasing tertiles of glycine. Notably, the ratios of glycine/carnitine, glycine/lysine, and glycine/TML were all inversely associated with the IS risk. Compared to the lowest tertiles, the corresponding odds ratios for the highest tertiles were 0.60 (95% CI: 0.40-0.91), 0.63 (95% CI: 0.42-0.94), and 0.63 (95% CI: 0.42-0.95), respectively, after adjustment for body mass index, smoking, hypertension, family history of stroke, estimated glomerular filtration rate and total cholesterol. Repeating the analyses by excluding the first two years of follow-up did not materially alter the risk associations for the ratios of glycine/lysine and glycine/carnitine. Increased ratios of plasma glycine to carnitine, lysine, and TML were associated with a lower risk of incident IS. Our observational findings suggest that the homeostasis of circulating carnitine, lysine, TML, and glycine may involve in the pathogenesis of IS.

Simultaneously Determining Seven Second-Line Anti-TB Drugs by UHPLC- MS: Application for TDM in HIV-TB Patients

Background: To optimize therapy for patients with human immunodeficiency virus-tuberculosis (HIV-TB) coinfection, we developed an ultra-high-performance liquid chromatography/- tandem mass spectrometry (UHPLC-MS) method to monitor seven second-line anti-tuberculosis drugs. Methods: Blood samples (n = 70) were collected from 35 patients with HIV-TB coinfection; the plasma sample was protein-precipitated and diluted with a solution containing heptafluorobutyric acid. The plasma concentrations of rifabutin (RBT), clofazimine (CLO), moxifloxacin (MFX), prothionamide (PTH), levofloxacin (LFX), amikacin (AMK), and para-aminosalicylic acid (PAS) were detected by UHPLC-MS/MS method. Results: In these 70 samples, the mean concentrations of RBT, CLO, MFX, PTH, LFX, and AMK were 173.8 (10.0–550.0), 61.1 (54.4–67.7), 646.6 (25.0–2480.0), 120.5 (50.0–597.0), 1565.9 (100.0–3480.0), and 10753.0 (400.0–76 700.0) μg/L, respectively. Only one sample was detected to have PAS with a concentration less than the lower limit of quantification. Most of the drug concentrations detected in these patients were lower than the targeted concentrations in TB patients. Conclusion: We created a simple UHPLC-MS method for simultaneously quantifying anti-TB drugs. The plasma concentrations in HIV-TB co-infected patients were lower than the targeted concentrations. It is important to monitor anti-TB drugs in the future. This method will facilitate the monitoring of anti-TB drugs in the future.

Stereoselective bioactivity, toxicity and degradation of novel fungicide sedaxane with four enantiomers under rice-wheat rotation mode

Sedaxane was a novel chiral fungicide that contains four enantiomers. Unfortunately, the stereoselective bioactivity, toxicity and degradation of sedaxane have not been clarified. In this study, we identified the absolute configuration of the four sedaxane enantiomers at first time. The stereoselective bioactivity toward three wheat and rice pathogens, stereoselective acute toxicity to aquatic organisms (Selenastrum capricornutum and Daphnia magna), and stereoselective degradation of sedaxane were studied. The 1 S,2S-(+)-sedaxane possessed 5.4–7.3 times greater bioactivity than 1 R,2R-(-)-sedaxane to Rhizoctonia solani and Rhizoctonia cerealis. Contrarily, the 1 R,2S-(+)-sedaxane had 4.2 times greater activity than 1 S,2S-(+)-sedaxane against Fusarium graminearum. The 1 R,2R-(-)-sedaxane had 2.8 times greater toxicity than 1 S,2S-(+)-sedaxane to S. capricornutum. The chiral determination method used ultrahigh-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The recovery of sedaxane stereoisomers ranged from 83.1 % to 98.2 %, with RSDs (Relative standard deviations) of 1.2 %− 8.4 %. The trans-sedaxane existed stereoselective degradation phenomenon in the rice-wheat rotation mode, and 1 S,2S-(+)-sedaxane was preferentially degraded. Our results would provide scientific importance and practical guidance to the safety evaluation of chiral pesticides

Association of serum bisphenol AF concentration with depressive symptoms in adolescents: A nested case–control study in China

BackgroundAs an important alternative to bisphenol A (BPA), bisphenol AF (BPAF) is widely used and can be detected in multiple human biological samples. However, there are few studies on neurotoxicity of BPAF at present. In particular, no epidemiological studies have investigated BPAF in relation to depressive symptoms in adolescents. Here, our study aimed to evaluate the associations between serum BPAF concentrations and depressive symptoms in adolescents. MethodsA nested case-control study within an ongoing longitudinal prospective adolescent cohort that was established in Huaibei, China was conducted. A total of 175 participants who had new-onset depressive symptoms (cases) and 175 participants without depressive symptoms (controls) were included. Serum BPAF concentrations was measured using ultra–high–performance liquid chromatography–tandem mass spectrometry. The associations between BPAF exposure and the risk of depressive symptoms in adolescents were assessed using conditional logistic regression. The dose-response relationship between BPAF level and depressive symptoms was estimated using restricted cubic spline analyses. ResultsIn this study, the detection rate of serum BPAF was 100%, and the median (interquartile range, IQR) serum BPAF concentration was 5.24 (4.41–6.11) pg/mL in the case group and 4.86 (4.02–5.77) pg/mL in the control group (P = 0.009). Serum BPAF exposure was a risk factor for depressive symptoms (odds ratio (OR)= 1.132, 95% confidence interval (CI):1.013–1.264). After adjustment for all for confounders, compared with the low–exposure group, the high–exposure group had a 2.806-fold increased risk of depressive symptoms (OR=2.806, 95% CI: 1.188–6.626). Stratified analysis by sex revealed that males were more vulnerable to BPAF exposure than females. After adjustment for all confounders, compared with the low-exposure group, the relative risk of depressive symptoms in the high-exposure group was 3.858 (95% CI: 1.118–12.535) for males, however, no significant association between BPAF exposure and depressive symptoms was found in females. In addition, there was a marked linear association between BPAF exposure and the risk of depressive symptoms in the total population and in males. ConclusionsThe adolescents in this study were widely exposed to low levels of BPAF. A significant positive association was found between serum BPAF levels and the risk of depressive symptoms. The association was significantly modified by sex, and males were more vulnerable to BPAF exposure than females.

Hierarchical Porous Nanocellulose Aerogels Loaded with Metal-Organic Framework Particles for the Adsorption Application of Heterocyclic Aromatic Amines.

This work overcomes the long-standing challenge of cumbersome pretreatment methods in the detection of heterocyclic aromatic amines (HAAs). A UiO-66/nanocellulose composite aerogel (CMC-CNC-UiO-66) with layered pores and low density prepared by a self-cross-linking method is applied as a simple and rapid adsorbent for capturing 14 HAAs via strong electrostatic interactions, van der Waals force, and the steric effect. The adsorption capacity of CMC-CNC-UiO-66 to 14 HAAs reached 98.00-188.00 nmol/mg at equilibrium within 10 min. The adsorption and desorption abilities of CMC-CNC-UiO-66 were retained with values of 93.36 and 97.34% after two cycles. In the meantime, the kinetics study demonstrated the chemisorption between HAA molecules and CMC-CNC-UiO-66 due to the excellent agreement with the pseudo-second-order adsorption models. The fit with the Freundlich isotherm models suggested a multilayer adsorption mechanism between HAA molecules and materials with heterogeneous surfaces. Moreover, coupled with the ultrahigh-performance liquid chromatography-tandem mass spectrometry detection, the CMC-CNC-UiO-66 extraction process can be completed with a high average recovery ranging from 86.68 to 115.33%, indicating a potential application of CMC-CNC-UiO-66 in HAA adsorption for further quantitative analysis.

Development and Validation of Benzophenone Derivatives in Packaged Cereal-Based Foods by Solid-Liquid Extraction and Ultrahigh-Performance Liquid Chromatography-Tandem Mass Spectrometry.

We established and validated a sensitive multi-residue analytical method for identifying benzophenone (BP) and nine BP derivatives (2,4-dihydroxybenzophenone [BP-1], 2,2′,4,4′-tetrahydroxydroxybenzophenone, 2-hydroxy-4-methoxy benzophenone, 2,2′-dihydroxy 4-methoxy benzophenone, 2-hydroxybenzophenone [2-OHBP], 4-hydroxybenzophenone, 4-methylbenzophenone [4-MBP], methyl-2-benzoylbenzoate, and 4-benzoylbiphenyl). Solid–liquid extraction pretreatment and ultra-high-performance liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS) were employed in an analysis of 85 packaged cereal-based food samples (25 pastry, 50 rice, and 10 noodle samples). The method had satisfactory linearity (R2 ≥ 0.995), low limits of detection (pastry: 0.02–4.2 ng/g; rice and noodle: 0.02–2 ng/g), and favorable precision, with within-run and between-run coefficient of variation ranges of 1–29% and 1–28%, respectively. BP and 4-MBP were detected in 100% of the pastry samples, and BP-1 and 2-OHBP were found in 76% and 56% of the pastry samples, respectively. BP and 2-OHBP were found in 92% and 38% of the rice samples, respectively. BP was found in 50% of the noodle samples. BP contributed the most to the total level of BPs in pastries, with significantly higher mean ± standard deviation (range) levels for pastries (26.8 ± 32.6 [1.8–115.4] ng/g) than rice (1.2 ± 2.0 [0.4–13.4] ng/g) and noodles (0.7 ± 0.7 [0.4–1.9] ng/g); p < 0.0001). The trace levels of 4-MBP identified in the samples demonstrate the need for the development of analytical methods with high sensitivity and specificity; the proposed method satisfies this need.

Simultaneous determination of pyflubumide and its metabolite in vegetables and fruits by ultrahigh performance liquid chromatography-tandem mass spectrometry.

A rapid and cost-effective analytical method based on ultrahigh-performance liquid chromatography-tandem mass spectrometry was designed and verified for simultaneously monitoring the novel acaricide pyflubumide and its metabolite (pyflubumide-des(2-methyl-1oxopropyl)) in vegetables and fruits. After the extraction with acetonitrile, the samples were purified by dispersive solid-phase extraction with multi-walled carbon nanotubes. Detection of the two target analytes was achieved within 3.0min using a positive electrospray ionization mode. The average recovery, intra-day precision, and inter-day precision of the two analytes at three spiked levels (2, 20, and 100 μg/kg) were 75.0-101.0, 0.4-4.4, and 0.6-5.3%, respectively. The limit of quantification of two compounds was 2 μg/kg, which was far below the maximum residue limits of pyflubumide in foods established by Japan and South Korea. Finally, the concentrations of pyflubumide and its metabolite in the samples were 16.6 and 7.8 μg/kg respectively, which verified the practicability and reliability of the method. The method was used to efficiently detect pyflubumide and its metabolite in real samples and was confirmed to be robust and effective for routinely analyzing both pyflubumide and its metabolite in vegetable and fruit samples.

Dissipation behavior, residue transfer, and safety evaluation of chlorantraniliprole and indoxacarb during tea growing and brewing by ultrahigh-performance liquid chromatography-tandem mass spectrometry.

A reliable and simple analytical method was developed and validated to simultaneously determine chlorantraniliprole and indoxacarb in tea using ultrahigh-performance liquid chromatography-tandem mass spectrometry. The average recoveries of chlorantraniliprole were in the range of 86-110%, with the precision of intraday (n = 5) and interday (n = 15) ranging from 1.9 to 8.4% and from 2.4 to 8.8%, respectively. The average recoveries of indoxacarb were in the range 81-105%, with the precision of intraday (n = 5) and interday (n = 15) ranging from 2.0 to 9.8% and from 2.7 to 9.1%, respectively. The limits of quantification (LOQs) were all 0.01mg/kg. The results based on the supervised field trials showed that chlorantraniliprole and indoxacarb in two tea samples followed first-order kinetics models with half-lives of 2.2-4.7days and 2.5-3.5days, which could be regarded as a moderately degrading pesticide. The terminal residues of chlorantraniliprole and indoxacarb in made tea were below 6.7 and 4.5mg/kg, respectively, lower than their corresponding maximum residue limits (MRLs) established by several major countries and organizations (50 and 5mg/kg). The leaching rates of chlorantraniliprole and indoxacarb during the tea brewing ranged from 4.78 to 12.62% and 4.13 to 10.67%, respectively. The chronic intake risk quotient (RQc) values for chlorantraniliprole and indoxacarb were 0.24% and 35.10%, while the acute dietary risk assessment (RQa) value of indoxacarb was 5.8%, which were all much lower than 100%. The results in the present study indicated that the health risk posed by the chlorantraniliprole and indoxacarb mixture pesticides was negligible in tea for consumers at the recommended dosages.

Vascular endothelium as a target for perfluroalkyl substances (PFAs)

IntroductionPerfluoroalkyl substances (PFAs) are ubiquitous, anthropogenic organic compounds that have been linked with cardiovascular disease and cardiovascular risk factors. Older, long-chain PFAs have been phased out due to adverse cardiometabolic health effect and replaced by newer short-chain PFAs. However, emerging research suggests that short-chain PFAs may also have adverse cardiovascular effects. Non-invasive measures of vascular function can detect preclinical cardiovascular disease and serve as a useful surrogate for early CVD risk. We hypothesized that serum concentrations of PFAs would be associated with noninvasive measures of vascular function, carotid-femoral pulse wave velocity (PWV) and brachial artery reactivity testing (BART), in adults with non-occupational exposure to PFAs. MethodsWe measured serum concentrations of 14 PFAs with hybrid solid-phase extraction and ultrahigh-performance liquid chromatography–tandem mass spectrometry in 94 adult outpatients with no known cardiovascular disease. We collected clinical and demographic data; and measured vascular function, PWV and BART, using standard protocols. We assessed associations of individual PFAs with log-transformed BART and PWV using linear regression. We used weighted quantile sum regression to assess effects of correlated PFA mixtures on BART and PWV. ResultsTen PFAs were measured above the limit of detection in >50% of participants. Each standard deviation increase in concentration of perfluoroheptanoic acid (PFHpA) was associated with 15% decrease in BART (95% CI: −28.5, −0.17). The weighted index of a mixture of PFAs with correlated concentrations was inversely associated with BART: each tertile increase in the weighted PFA mixture was associated with 25% lower BART, with 73% of the effect driven by PFHpA. In contrast, no PFAs or mixtures were associated with PWV. ConclusionsSerum concentration of PFHpA, a new, short-chain PFA, was associated with impaired vascular function among outpatients without CVD. Our findings support a potential adverse cardiovascular effect of newer, short-chain PFAs.

A New LC-MS Method for Evaluating the Efficacy of Pesticide Residue Removal from Fruit Surfaces by Washing Agents

Modern agriculture uses pesticides to improve the quality and quantity of crops. However, pesticide residues can remain on agricultural products, posing very serious risks to human health and life. It is recommended to wash fruits and vegetables before consumption. To assess the removal efficacy of pesticide residue, a sensitive and reliable method based on ultrahigh-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) was developed and optimized for the simultaneous determination of four pesticide residues (acetamiprid, boscalid, pyraclostrobin, and pendimethalin). Isotope-labeled standards were used to validate the method in terms of recovery, linearity, matrix effects, precision, and sensitivity. The mean recovery values for both low-quality control (LQC) and high-quality control (HQC) transitions were in the range of 89–105%, and the intra-day precision was less than 13.7%. The limits of detection (LOD) and quantification (LOQ) were 0.003 mg/kg and 0.01 mg/kg, respectively. The proposed method is suitable for evaluating the quality of detergents for removing pesticide residues from fruit surfaces.

A strategy for the rapid discovery and validation of active diterpenoids as quality markers in different habitats of Langdu using ultrahigh-performance liquid chromatography-tandem mass spectrometry with multivariate statistical analysis.

Langdu, known as a traditional Chinese medicine, was identified as the roots of species of Euphorbia ebracteolata Hayata and Euphorbia fischeriana Steud, displaying anti-tuberculosis activity. To clarify the potent quality markers of Langdu, this research first developed a fast and sensitive ultrahigh-performance liquid chromatography-tandem mass spectrometry method for the quantification of 13 diterpenoids in Langdu. The developed method was further applied in the analyses of 12 authentic E. ebracteolata and E. fischeriana samples collected in northern and southeastern China. Then, the anti-tuberculosis evaluation of 12 batches of Langdu samples was performed in vitro. Finally, partial least squares discrimination analysis was used in the discrimination of E. ebracteolata and E. fischeriana from different origins and processing methods. Jolkinolide A (1), jolkinolide E (3), yuexiandajisu D (6), and ebractenone A (11) were identified as key, potent diterpenoids for the quality control of E. ebracteolata Hayata and E. fischeriana Steud. The present study established a qualitative chemical analysis method for Langdu (E. ebracteolata and E. fischeriana) and suggested the key bioactive components that will improve qualitative control methodology for this important medicine.

Development and validation of a sensitive and reliable targeted metabolomics method for the quantification of cardiovascular disease-related biomarkers in plasma using ultrahigh-performance liquid chromatography-tandem mass spectrometry.

Cardiovascular disease, as a multifactorial disease caused by genetics and environment, has emerged as a leading cause of mortality. The discovery of metabolic biomarkers for the clinical diagnosis, early warning and elucidation of the molecular pathogenesis of cardiovascular disease, using metabolomics, has attracted broad interest. Therefore, this work aimed to develop a sensitive and reliable targeted metabolomics method for the quantification of cardiovascular disease-related biomarkers in plasma. The method was developed and validated using ultrahigh-performance liquid chromatography augmented with tandem mass spectrometry (UHPLC/MS/MS). The LC conditions and MS parameters were optimized using selected reaction monitoring scanning mode to high-throughput and sensitive separation, and could detect 20 metabolic biomarkers in a single experiment. And the linearity, selectivity, accuracy, precision, stability and recovery of the developed method were assessed according to the Bioanalytical Method Validation guidelines of the United States Food and Drug Administration. These quantified metabolic biomarkers are involved in pathways such as aromatic amino acid catabolism (e.g. phenylalanine, tryptophan, tyrosine), trimethylamine N-oxide (TMAO) biosynthesis (e.g. TMAO, choline, carnitine, betaine) and histidine metabolism (e.g. histidine), among others. All analytes exhibited excellent linearities with coefficients of determination greater than 0.99. Accuracies deviated by less than 15% for medium- and high-concentration samples and less than 20% for low-concentration samples, with intra- and inter-day precisions of 1.12-14.12% and 0.30-13.74%, respectively. Recoveries and stabilities also met the analysis requirements of biological samples. The targeted metabolomics method was shown to have a powerful ability to accurately analyze metabolic biomarkers, thereby providing valuable information for large-scale biomarker validation and clarifying the potential material basis of cardiovascular disease for clinical diagnosis or early warning.

Development and validation of a liquid chromeatography-tandem mass spectrometry method for simultaneous quantification of medium- and long-chain saturated fatty acids in hamster plasma samples.

Saturated fatty acids (SFAs) are associated with many diseases in humans. Developing a reliable analytical method to analyze SFAs in plasma is essential to understand their biological activities. An ultrahigh-performance liquid chromatography-tandem mass spectrometry (UHPLC/MS/MS) method has been developed for the quantification of medium- and long-chain SFAs (M/LCSFAs) in hamster plasma. We compared three methods (DOLE, Folch and MTBE) for extracting M/LCSFAs from plasma. The M/LCSFA derivatives were separated using a C18 column. The method was validated and applied to analyze M/LCSFA concentrations in normal-fat diet (NFD) and high-fat diet (HFD) hamster plasma. Among the three extraction methods, the DOLE method had the highest extraction recovery and was simple to operate with a short incubation time. All of the calibration curves exhibited good linear relationships (r ≥ 0.9958). The results for selectivity, accuracy, precision, matrix effects and recovery were all within the acceptance criteria. In HFD hamster plasma, the concentration of M/LCSFAs with even-carbon chain length was significantly increased. A simple, robust and reproducible method for the simultaneous quantification of M/LCSFAs by UHPLC/MS/MS was developed and validated. The method gave successfully quantification of M/LCSFAs in plasma samples from NFD and HFD hamsters.
