Label-free rat brain traumatic penumbra imaging based on multiphoton fluorescence microscopy

Traumatic penumbra (TP) is a region with recoverable potential around the primary lesion of brain injury. Rapid and accurate imaging for identifying TP is essential for treating traumatic brain injury (TBI). In this study, we first established traumatic brain injuries (TBIs) in rats using a modified Feeney method, followed by label-free imaging of brain tissue sections with multiphoton fluorescence microscopy. The results showed that the technique effectively imaged normal and traumatic brain tissues, and revealed pathological features such as extracellular matrix changes, vascular cell proliferation, and intracellular edema in the traumatic penumbra. Compared with normal brain tissue, the extracellular matrix in the TP was sparse, cells were disorganized, and hyperplastic vascular cells emitted higher two-photon excited fluorescence (TPEF) signals. Our research demonstrates the potential of multiphoton fluorescence technology in the rapid diagnosis and therapeutic evaluation of TBI.

Reactive Stroma and Acinar Morphology in Prostate Cancer: Implications for Progression and Prognostic Assessment.

Prostate cancer (PC) remains a significant global health concern, with prognostic assessments largely reliant on the Gleason Classification System. While it has proven effective, subjectivity in interpretation persists, prompting the need for complementary approaches. Reactive stroma (RS) has emerged as a potential candidate for enhancing PC characterization, as it reflects intricate interactions among stromal, epithelial, and extracellular matrix components. To shed light on this, we conducted a comprehensive study. Two expert pathologists independently analyzed consecutive prostate biopsies (n = 120 patients), categorized into four groups based on Gleason scores. Four acinar patterns were described, denoted as A, B, C, and D. Our study uncovered a noteworthy presence of RS, predominantly within poorly differentiated tumors. Stromogenic tumors, characterized by high RS content, were particularly associated with Gleason scores of 4 + 3 and ≥ 8. Intriguingly, acinar patterns, including the distinctive B and D patterns, exhibited strong correlations with stromogenic tumors. Incorporating quantitative imaging techniques (Second Harmonic Generation and Two-Photon Excitation Fluorescence Microscopy), we examined collagen fiber density within the stroma. Our analysis revealed a direct relationship between RS intensity and collagen fiber counts, particularly prominent in patterns B and D. These findings suggest that the stromal reaction in PC is closely linked to acinar morphology and collagen deposition. Moreover, rudimentary microacini at the tumor periphery, associated with intense RS and patterns B and D, may signify an unfavorable prognosis. Our study highlights the potential of RS as an additional prognostic factor in PC. It underscores the intricate interplay between acinar patterns, RS intensity, and collagen fiber density, providing valuable insights for future prognostic assessments and therapeutic strategies. Further exploration of these relationships is essential for a comprehensive understanding of PC progression and management.

Label-Free Detection of Breast Phyllodes Tumors Based onMultiphoton Microscopy.

Phyllodes tumors (PTs) are rare breast stroma neoplasms, and their accurate identification at various stages is essential for personalized patient treatment. In this study, multiphoton microscopy (MPM) with two-photon excited fluorescence (TPEF) and second harmonic generation (SHG) imaging was used for label-free detection and differentiation of PTs and normal breast tissue. An automated image processing strategy was developed to quantify changes in collagen fiber morphology within the stroma and boundary of PTs, establishing optical diagnostic characteristics of PTs using MPM. The results demonstrated that MPM could be used for the detection of different stages of PTs, and the morphological alterations in collagen fibers could serve as critical indicators of PT malignancy, offering new insights for the diagnosis and grading of benign, borderline, and malignant PTs. It lays the groundwork for the future application of compact MPM for the rapid detection and diagnosis of PTs.

Complex-valued scatter compensation in nonlinear microscopy

Nonlinear, i.e., multiphoton microscopy is a powerful technique for imaging deep into biological tissues. Its penetration depth can be increased further using adaptive optics. In this work, we present a fast, feedback-based adaptive-optics algorithm, termed C-DASH, for multiphoton imaging through multiple-scattering media. C-DASH utilizes complex-valued light shaping (i.e., joint shaping of amplitude and phase), which offers several advantages over phase-only techniques: it converges faster, it delivers higher image quality enhancement, it shows a robust performance largely insensitive to the axial position of the correction plane, and it has a higher ability to cope with, on the one hand, scattering media that vary over time and, on the other hand, scattering media that are partially absorbing. Furthermore, our method is practically self-aligning. We also present a simple way to implement C-DASH using a single reflection off a phase-only spatial light modulator. We provide a thorough characterization of our method, presenting results of numerical simulations as well as two-photon excited fluorescence imaging experiments. Application Exploration of the Photon Chip Technology in Real time Processing of Biomedical Images

In the biomedical field, photonic chip technology is gradually becoming the key to improving the speed and accuracy of medical imaging. The development of this technology has given us new hope in early disease diagnosis and cell monitoring. Recent research has not only sorted out the hotspots and future trends of biomedical photonics, but also paid special attention to the application of photon technology in these fields. Specifically, photonic chips have demonstrated their unique capabilities in multi-color two-photon excitation fluorescence microscopy imaging, two-photon fluorescence lifetime imaging, and two-photon fiber endoscopic imaging. These applications not only improve the clarity of imaging, but also accelerate processing speed, which is crucial for rapid diagnosis and treatment. At the end of the article, the author summarized the contributions of photonic chip technology in the field of biomedical imaging and provided prospects for future development directions.

Small molecule probes with strong one and two-photon excited fluorescence for bioimaging

Two-photon excited fluorescence (TPEF) is widely sought after in bioimaging applications due to its exceptional deep tissue penetration and excellent high spatiotemporal resolution. In this paper, benzothiazole and benzimidazole are linked to N, N-diphenylaniline, respectively, through pyridine vinyl. Two new D-π-A (electron donor-π conjugated bridges-electron acceptor) probes (P1, P2) were designed and synthesized. The molecular structures were completely characterized through FT-IR, 1H NMR, 13C NMR, ESI-MS, and X-ray single crystal diffraction. The probes have good one-photon excited fluorescence and two-photon excited fluorescence. Experimental test results indicate that they can be fluorescence probes for biological imaging of HePG2 and juvenile zebrafish. In addition, the fluorescent probes also have the ability to target and locate cancer cell mitochondria.

Interference patterns of two-photon excited fluorescence by spatial beam shaping

We report on two-photon excited fluorescence interference patterns produced by spatial laser beam shaping. Thereto, a focused spatially modulated laser beam component is overlapped with an unfocused Gaussian beam component, and both are directed on a cuvette filled with dye. The phase retardance between the two common path laser components is precisely adjusted by a 2D liquid crystal modulator. With this method it is possible to generate various fluorescence patterns which exhibit sharp features and a high contrast due to constructive and destructive interference of a two-photon excitation process. The developed spatial shaping interference technique with two-photon excitations has a high potential for optical and biophotonic imaging applications.

Photo-modulation of the two-photon excited fluorescence of dithienylethene bis-(1-pyrenyl) compounds: An experimental and theoretical study

Photomodulation of two-photon excited fluorescence has been effectively controlled in 1-pyrene-based dithienylethenes. The specific molecular design enables photocommutation of two-photon excited fluorescence during sequential one- and two-photon excitations without interference nor destructive optical readout. Theoretical studies have concluded that the internal functionalization of pyrene is responsible for the photoactivity in these systems. Photophysical studies indicate that the presence of a second laterally introduced pyrene is crucial for maintaining high photocommutation contrast and durability.

Enhancing Two-Photon Excited Fluorescence of Metal-Organic Framework Single Crystals through Modulation of Inorganic Nodes.

Regulation of the two-photon excited fluorescence (TPEF) emission intensity and wavelength of metal-organic framework (MOF) crystals with similar constitutions presents a significant challenge. In this study, two MOFs, Zn-BTPPA and Cd3-BTPPA, were constructed using tetrakis(1,1'-biphenyl-4-carboxylic acid)-1,4-benzenediamine (H4BTPPA) as the organic ligand and mononuclear Zn and trinuclear Cd3 inorganic nodes, respectively. The incorporation of H4BTPPA within the MOF structures enables effective TPEF emission in both Zn-BTPPA and Cd3-BTPPA. The TPEF results show that Zn-BTPPA and Cd3-BTPPA exhibited strong emissions at 523 and 463 nm, respectively, when excited with a 780 nm laser. Moreover, Zn-BTPPA and Cd3-BTPPA exhibited much higher two-photon absorption cross sections, approximately 4.9 and 5.2 times higher than that of the reported dinuclear MOF, Cd2-BTPPA, with a similar composition, respectively. With different inorganic nodes, the stacking of chromophores, π···π interactions, and ligand geometry were found to correlate with the enhanced TPEF in Cd3-BTPPA and the blue-shifted TPEF in Zn-BTPPA. This work serves as an inspiration for designing efficient TPEF MOF materials based on the structure-property relationship.

Photobleaching analysis of fluorescent proteins in two-photon microscopy at high repetition rates

Severe photobleaching in two-photon excitation fluorescence microscopy (TPM) has limited its potential for long-term and quantitative imaging of live cells and tissues. One solution is to excite fluorophores with a high-repetition rate which reduces the peak intensity of irradiance. However, there is a lack of knowledge about the general utility of this strategy for fluorescent proteins. Here, using simple photobleaching assay, we studied the photobleaching of EGFP and tdTomato at 80 MHz and 640 MHz repetition rates. Our analysis shows that the impact of high repetition rate excitation on reducing photobleaching in TPM is highly dependent on the excitation wavelength and fluorophores. Our results are useful for selecting optimal parameters to minimize photobleaching and photodamage in TPM.

Multi-modal, Label-free, Optical Mapping of Cellular Metabolic Function and Oxidative Stress in 3D Engineered Brain Tissue Models.

Brain metabolism is essential for the function of organisms. While established imaging methods provide valuable insights into brain metabolic function, they lack the resolution to capture important metabolic interactions and heterogeneity at the cellular level. Label-free, two-photon excited fluorescence imaging addresses this issue by enabling dynamic metabolic assessments at the single-cell level without manipulations. In this study, we demonstrate the impact of spectral imaging on the development of rigorous intensity and lifetime label-free imaging protocols to assess dynamically over time metabolic function in 3D engineered brain tissue models comprising human induced neural stem cells, astrocytes, and microglia. Specifically, we rely on multi-wavelength spectral imaging to identify the excitation/emission profiles of key cellular fluorophores within human brain cells, including NAD(P)H, LipDH, FAD, and lipofuscin. These enable development of methods to mitigate lipofuscin's overlap with NAD(P)H and flavin autofluorescence to extract reliable optical metabolic function metrics from images acquired at two excitation wavelengths over two emission bands. We present fluorescence intensity and lifetime metrics reporting on redox state, mitochondrial fragmentation, and NAD(P)H binding status in neuronal monoculture and triculture systems, to highlight the functional impact of metabolic interactions between different cell types. Our findings reveal significant metabolic differences between neurons and glial cells, shedding light on metabolic pathway utilization, including the glutathione pathway, OXPHOS, glycolysis, and fatty acid oxidation. Collectively, our studies establish a label-free, non-destructive approach to assess the metabolic function and interactions among different brain cell types relying on endogenous fluorescence and illustrate the complementary nature of information that is gained by combining intensity and lifetime-based images. Such methods can improve understanding of physiological brain function and dysfunction that occurs at the onset of cancers, traumatic injuries and neurodegenerative diseases.

Variational calculus approach to Zernike polynomials with application to FCS

Zernike polynomials are a sequence of orthogonal polynomials that play a crucial role in optics and, in particular, modeling microscopy systems. Introduced by Frits Zernike in 1934, they are particularly useful in expressing wavefront aberrations and, thus, imperfections of imaging systems. However, their origin and properties are rarely discussed and proven. Here, we present a novel approach to Zernike polynomials using variational calculus and apply them to describe aberrations in fluorescence microscopy. In particular, we model the impact of various optical aberrations on the performance of one-photon and two-photon excitation fluorescence microscopy.

Smart design of highly luminescent octupolar mesogenic tetra styryl-alkynyl bipyrimidine-based chromophores presenting non-linear optical properties

The research investigations reported in this paper focuses on the influence of rational design on the photophysical properties of new all-organic multifunctional chromophores. Nonlinear optical (NLO) behavior, optical properties and mesogenicity of several new π-conjugated octupolar chromophores, presenting alkoxy styryl-alkynyl donor groups carrying aliphatic achiral and chiral chains in the 3,4/3,5/2,4 position, connected to electron-accepting 2,2-bipyrimidine cores are investigated by various techniques (light-transmission measurements, polarized-light optical microscopy, differential scanning calorimetry measurements, two-photon excited fluorescence and Hyper Rayleigh Scattering). The mesogenic properties were investigated depending on the position of the alkoxy chain and the 3,4 derivatives were found to exhibit liquid-crystal properties with the formation columnar phases over large temperature ranges which were confirmed by small-angle X-ray scattering analysis. All the molecules exhibit exceptional high fluorescence quantum yields values around 80 % and excellent nonlinear optical properties.

Application of the Knife-Edge Technique on Transition Metal Dichalcogenide Monolayers for Resolution Assessment of Nonlinear Microscopy Modalities.

We report application of the knife-edge technique at the sharp edges of WS2 and MoS2 monolayer flakes for lateral and axial resolution assessment in all three modalities of nonlinear laser scanning microscopy: two-photon excited fluorescence (TPEF), second- and third-harmonic generation (SHG, THG) imaging. This technique provides a high signal-to-noise ratio, no photobleaching effect and shows good agreement with standard resolution measurement techniques. Furthermore, we assessed both the lateral resolution in TPEF imaging modality and the axial resolution in SHG and THG imaging modality directly via the full-width at half maximum parameter of the corresponding Gaussian distribution. We comprehensively analyzed the factors influencing the resolution, such as the numerical aperture, the excitation wavelength and the refractive index of the embedding medium for the different imaging modalities. Glycerin was identified as the optimal embedding medium for achieving resolutions closest to the theoretical limit. The proposed use of WS2 and MoS2 monolayer flakes emerged as promising tools for characterization of nonlinear imaging systems.

Localization of protoporphyrin IX during glioma-resection surgery via paired stimulated Raman histology and fluorescence microscopy.

The most widely used fluorophore in glioma-resection surgery, 5-aminolevulinic acid (5-ALA), is thought to cause the selective accumulation of fluorescent protoporphyrin IX (PpIX) in tumour cells. Here we show that the clinical detection of PpIX can be improved via a microscope that performs paired stimulated Raman histology and two-photon excitation fluorescence microscopy (TPEF). We validated the technique in fresh tumour specimens from 115 patients with high-grade gliomas across four medical institutions. We found a weak negative correlation between tissue cellularity and the fluorescence intensity of PpIX across all imaged specimens. Semi-supervised clustering of the TPEF images revealed five distinct patterns of PpIX fluorescence, and spatial transcriptomic analyses of the imaged tissue showed that myeloid cells predominate in areas where PpIX accumulates in the intracellular space. Further analysis of external spatially resolved metabolomics, transcriptomics and RNA-sequencing datasets from glioblastoma specimens confirmed that myeloid cells preferentially accumulate and metabolize PpIX. Our findings question 5-ALA-induced fluorescence in glioma cells and show how 5-ALA and TPEF imaging can provide a window into the immune microenvironment of gliomas.

Label-Free Monitoring of Endometrial Cancer Progression Using Multiphoton Microscopy

Endometrial cancer is the most common gynecological cancer in the developed world. However, the accuracy of current diagnostic methods is still unsatisfactory and time-consuming. Here, we presented an alternate approach to monitoring the progression of endometrial cancer via multiphoton microscopy imaging and analysis of collagen, which is often overlooked in current endometrial cancer diagnosis protocols but can offer a crucial signature in cancer biology. Multiphoton microscopy (MPM) based on the second-harmonic generation and two-photon excited fluorescence was introduced to visualize the microenvironment of endometrium in normal, hyperplasia without atypia, atypical hyperplasia, and endometrial cancer specimens. Furthermore, automatic image analysis based on the MPM image processing algorithm was used to quantify the differences in the collagen morphological features among them. MPM enables the visualization of the morphological details and alterations of the glands in the development process of endometrial cancer, including irregular changes in the structure of the gland, increased ratio of the gland to the interstitium, and atypical changes in the glandular epithelial cells. Moreover, the destructed basement membrane caused by gland proliferation and fusion is clearly shown in SHG images, which is a key feature for identifying endometrial cancer progression. Quantitative analysis reveals that the formation of endometrial cancer is accompanied by an increase in collagen fiber length and width, a progressive linearization and loosening of interstitial collagen, and a more random arrangement of interstitial collagen. Observation and quantitative analysis of interstitial collagen provide invaluable information in monitoring the progression of endometrial cancer. Label-free multiphoton imaging reported here has the potential to become an in situ histological tool for effective and accurate early diagnosis and detection of malignant lesions in endometrial cancer.

Structural changes in the crystalline lens as a function of the postmortem interval assessed with two-photon imaging microscopy.

The properties and structure of the crystalline lens change as time after death passes. Some experiments have suggested that these might be used to estimate the postmortem interval (PMI). In this study, the organization and texture of the rabbit lens were objectively evaluated as a function of the PMI using two-photon excitation fluorescence (TPEF) imaging microscopy. Between 24 h and 72 h, the lens presented a highly organized structure, although the fiber delineation was progressively vanishing. At 96 h, this turned into a homogeneous pattern where fibers were hardly observed. This behaviour was similar for parameters providing information on tissue texture. On the other hand, the fiber density of the lens is linearly reduced with the PMI. On average, density at 24 h was approximately two-fold when compared to 96 h after death. The present results show that TPEF microscopy combined with different quantitative tools can be used to objectively monitor temporal changes in the lens fiber organization after death. This might help to estimate the PMI, which is one of the most complex problems in forensic science.

High-throughput volumetric mapping of synaptic transmission.

Volumetric imaging of synaptic transmission in vivo requires high spatial and high temporal resolution. Shaping the wavefront of two-photon fluorescence excitation light, we developed Bessel-droplet foci for high-contrast and high-resolution volumetric imaging of synapses. Applying our method to imaging glutamate release, we demonstrated high-throughput mapping of excitatory inputs at >1,000 synapses per volume and >500 dendritic spines per neuron in vivo and unveiled previously unseen features of functional synaptic organization in the mouse primary visual cortex.

Multimodal 2D and 3D microscopic mapping of growth cartilage by computational imaging techniques – a short review including new research

Being able to image the microstructure of growth cartilage is important for understanding the onset and progression of diseases such as osteochondrosis and osteoarthritis, as well as for developing new treatments and implants. Studies of cartilage using conventional optical brightfield microscopy rely heavily on histological staining, where the added chemicals provide tissue-specific colours. Other microscopy contrast mechanisms include polarization, phase- and scattering contrast, enabling non-stained or ‘label-free’ imaging that significantly simplifies the sample preparation, thereby also reducing the risk of artefacts. Traditional high-performance microscopes tend to be both bulky and expensive. Computational imaging denotes a range of techniques where computers with dedicated algorithms are used as an integral part of the image formation process. Computational imaging offers many advantages like 3D measurements, aberration correction and quantitative phase contrast, often combined with comparably cheap and compact hardware. X-ray microscopy is also progressing rapidly, in certain ways trailing the development of optical microscopy. In this study, we first briefly review the structures of growth cartilage and relevant microscopy characterization techniques, with an emphasis on Fourier ptychographic microscopy (FPM) and advanced x-ray microscopies. We next demonstrate with our own results computational imaging through FPM and compare the images with hematoxylin eosin and saffron (HES)-stained histology. Zernike phase contrast, and the nonlinear optical microscopy techniques of second harmonic generation (SHG) and two-photon excitation fluorescence (TPEF) are explored. Furthermore, X-ray attenuation-, phase- and diffraction-contrast computed tomography (CT) images of the very same sample are presented for comparisons. Future perspectives on the links to artificial intelligence, dynamic studies and in vivo possibilities conclude the article.
