Prevalence and risk factors of bovine tuberculosis in cattle in selected districts of Fafan pastoral settings, Eastern Ethiopia

Cross-sectional study was conducted from October 2021 to August 2022 to investigate the prevalence and associated risk factors of bovine tuberculosis in cattle in selected districts of the pastoral settings of Fafan zone, Somali region, eastern Ethiopia. A comparative intradermal tuberculin test was performed using purified protein derivatives. Animal-related characteristics, and the owner's knowledge on the importance of BTB were collected using a structured questionnaire. The prevalence was 11.24 % (95 % CI, 8.61–14.35) and 43.3 % (95 % CI, 33.27–53.75) at the individual and herd levels, respectively. There were statistically significant differences in the proportions of positive reactor animals according to body condition score (P = 0.000), age (P = 0.048), seasonal migration (P = 0.038), parity number (P = 0.005), and reproductive status (P = 0.037). Animals with poor body condition scores had a significantly higher likelihood of testing positive, with their odds being 11.4 times greater (COR = 11.408, CI = 3.43–37.94, P < 0.001). In multivariate logistic regression, poor body condition score remained significantly associated with the odds of a positive reaction to tuberculosis (AOR = 0.137, CI = 0.053–0.356, P < 0.001). Similarly, the analysis showed that seasonal migration (AOR = 2.882, CI = 1.155–7.191, P = 0.023) and parity number (AOR = 11.64, CI = 1.818–74.464, P = 0.010) were significant predictors of bovine tuberculosis infection in cattle. According to the questionnaire, 14.2 % (17 of 120) and 13.3 % (16 of 120) of the respondents were knowledgeable about bovine tuberculosis and its transmission from animals to humans, and vice versa, respectively. The general judgment of herders' understanding of bovine tuberculosis transmission methods to humans was very low. The study findings showed a high prevalence of bovine tuberculosis in the study area, emphasizing the need for an effective control and prevention strategy.

Histopathological and Molecular Investigation of Natural Cases of Bovine Tuberculosis Infection in Cattle

Bovine tuberculosis (bTB), a chronic infection in cattle caused by Mycobacterium tuberculosis/bovis, that impacts productivity and represents a major public health threat. Although the considerable economic costs and zoonotic risk consequences associated with the disease, accurate estimates of bTB prevalence are lacking in many countries, including India. Therefore, in the current study for collection of tubercular lesions the postmortem examination of 100 cattle was conducted. All major viscera and regional lymph nodes were examined and incised. Histopathology was performed in the cases where gross lesions were suggestive of tuberculosis. PCR was performed on the tissue and faecal samples by using IS6110 insertion sequence, Mycobacterium tuberculosis/bovis complex PCR kit. In 12 animals, nodular lesions with casseating mass suggestive of tuberculosis were observed in the lung tissue. All the 12 lung impression smear and only five faecal smear showed acid fast bacilli stained by Ziehl-Neelsen staining. Histologic features comprised a classic granuloma as a characteristic lesion of tuberculosis composed of a central caseous necrosis with mantle of macrophages, lymphocytes, plasma cells, epithelioid macrophages and Langhan's giant cells and were observed in all 12 cases. All the tissue samples and 11 faecal samples were positive for the Mycobacterium tuberculosis complex using IS6110 sequence. 8 tissue samples and 4 faecal samples were positive by using Mycobacterium tuberculosis/bovis complex PCR kit. It can be concluded that there was good agreement between histopathology, acid fast staining and PCR. It can also be concluded that faecal samples which are easier to collect should be preferred for diagnosis of TB by PCR in cattle.

Isolation and comparative genomics of Mycobacterium tuberculosis isolates from cattle and their attendants in South India

The major human pathogen Mycobacterium tuberculosis is rarely reported to cause disease in other animals. Cases in livestock are thought to occur through contact with infected handlers, but previous studies evaluating putative livestock-human transmission used typing techniques with limited resolution. Here, we undertook cross-sectional surveillance for tuberculosis in 271 livestock handlers and 167 cattle on three farms in Chennai, India and defined the relatedness of cultured isolates using whole genome sequencing. Humans and livestock were screened for active mycobacterial infection, and opportunistic post-mortem examination was performed on comparative intradermal test-positive cattle that died. Four cattle and 6 handlers on two farms were culture-positive for M. tuberculosis; M. bovis was not isolated. All 10 isolates (one from each case) belonged to Lineage 1. Pairwise genome comparisons of single nucleotide polymorphism (SNP) differences ranged from 1 to 600 SNPs, but 3 isolate pairs were less than 5 SNPs different. Two pairs were from handlers and the third pair were from two cattle on the same farm. The minimum pairwise SNP difference between a cattle and human isolate was >250 SNPs. Our study confirms the presence of M. tuberculosis infection in cattle in India, sequencing of which characterised relatedness between human and cattle-derived isolates.

Prevalence of bovine tuberculosis in the Aksu Region of Xinjiang, China, between 1985 and 2016

ABSTRACT Prevalence of bovine tuberculosis infection in cattle in Aksu Prefecture determined by intradermal tuberculin skin test (TST), between 1985 and 2016. Cattle were analyzed according to region, feeding pattern, herds and age. A total of 890,009 cattle were tested, with overall bovine tuberculosis prevalence of 0.13% (1172/890009). Statistically significant difference was found in feeding pattern and herds. Prevalence in cows (0.19%, 615/327022) was higher than that in beeves (P&lt; 0.01, OR= 1.903, 95% CI = 1.696 to 2.134). Significant difference (P&lt; 0.01; OR= 2.238, 95%; CI= 1.937 to 2.585) was evident for rates for bovine tuberculosis in the peasant household (0.12%, 942/802343) and farm groups (0.26%, 230/87666). The overall prevalence of bTB was decreased in the Aksu Prefecture, especially the positive rate was under 0.1% in 2010s. We concluded that the control measures forbovine tuberculosis in the Aksu region cattle herds are effective.

Association of single nucleotide polymorphisms in IFNGR1 and IFNGR2 genes with bovine tuberculosis

Interferon Gamma Receptor (IFNGR) genes play an important role in the immune response against mycobacteria by regulating the proinflammatory cytokine Interferon Gamma (IFNG) alongwith subsequent mycobactericidal milieu and are potential strong candidates for investigating genetic basis of disease resistance. Present investigation was aimed at exploring the association of one SNP in IFNGR1 gene and two SNPs in IFNGR2 gene with susceptibility/resistance against bovine tuberculosis infection in cattle. All the three SNPs under investigation (rs109049057, rs109579937 and rs110689128) revealed polymorphism. SNP loci rs109049057 was found to be significantly (P less than 0.01) associated with susceptibility to bovine tuberculosis in cattle in our case control population. The SNP was non-synonymous, suggesting its functional role in the immune response against bovine tuberculosis.

Different lesion distribution in calves orally or intratracheally challenged with Mycobacterium bovis: implications for diagnosis

Animal tuberculosis (TB) remains a major problem in some countries despite the existence of control programmes focused mainly on cattle. In this species, aerogenous transmission is accepted as the most frequent infection route, affecting mainly the respiratory system. Under the hypothesis that the oral route could be playing a more relevant role in transmission, diagnosis and disease persistence than previously thought, this study was performed to assess the course of TB infection in cattle and its effects on diagnosis depending on the route of entry of Mycobacterium bovis. Two groups of five calves each were either endotracheally (EC) or orally (OC) challenged. Necropsies were carried out 12 weeks after challenge except for three OC calves slaughtered 8 weeks later. All animals reacted to the tuberculin skin test and the entire EC group was positive to the interferon-gamma release assay (IGRA) 2 weeks after challenge and thereafter. The first positive IGRA results for OC calves (3/5) were recorded 4 weeks after challenge. Group comparison revealed significant differences in lesion and positive culture location and scoring. TB-compatible gross lesions and positive cultures were more frequently found in the thorax (p < 0.001) and lung (p < 0.05) of EC animals, whereas OC animals presented lesions (p = 0.23) and positive cultures (p < 0.05) mainly located in the abdomen. These results indicate that the infection route seems to be a determining factor for both the distribution and the time needed for the development of visible lesions. Our study suggests that confirmation of TB infection in some skin reactor animals can be problematic if current post-mortem examination and diagnostics are not improved.

Risk-based strategies for surveillance of tuberculosis infection in cattle for low-risk areas in England and Scotland

Disease surveillance can be made more effective by either improving disease detection, providing cost savings, or doing both. Currently, cattle herds in low-risk areas (LRAs) for bovine tuberculosis (bTB) in England are tested once every 4 years. In Scotland, the default herd testing frequency is also 4 years, but a risk-based system exempts some herds from testing altogether. To extend this approach to other areas, a bespoke understanding of at-risk herds and how risk-based surveillance can affect bTB detection is required. Here, we use a generalized linear mixed model to inform a Bayesian probabilistic model of freedom from infection and explore risk-based surveillance strategies in LRAs and Scotland. Our analyses show that in both areas the primary herd-level risk factors for bTB infection are the size of the herd and purchasing cattle from high-risk areas of Great Britain and/or Ireland. A risk-based approach can improve the current surveillance system by both increasing detection (9% and 7% fewer latent infections), and reducing testing burden (6% and 26% fewer animal tests) in LRAs and Scotland, respectively. Testing at-risk herds more frequently can also improve the level of detection by identifying more infected cases and reducing the hidden burden of the disease, and reduce surveillance effort by exempting low-risk herds from testing.

Mycobacterium tuberculosis infection in cattle from the Eastern Cape Province of South Africa

BackgroundMycobacterium tuberculosis is the main causative agent of tuberculosis (TB) in human and Mycobacterium bovis commonly causes tuberculosis in animals. Transmission of tuberculosis caused by both pathogens can occur from human to animals and vice versa.ResultsIn the current study, M. tuberculosis, as confirmed by polymerase chain reaction (PCR) using primers targeting 3 regions of difference (RD4, RD9 and RD12) on the genomes, was isolated from cattle originating from two epidemiologically unrelated farms in the Eastern Cape (E.C) Province of South Africa. Although the isolates were genotyped with variable number of tandem repeat (VNTR) typing, no detailed epidemiological investigation was carried out on the respective farms to unequivocally confirm or link humans as sources of TB transmission to cattle, a move that would have embraced the ‘One Health’ concept. In addition, strain comparison with human M. tuberculosis in the database from the E.C Province and other provinces in the country did not reveal any match.ConclusionsThis is the first report of cases of M. tuberculosis infection in cattle in South Africa. The VNTR profiles of the M. tuberculosis strains identified in the current study will form the basis for creating M. tuberculosis VNTR database for animals including cattle for future epidemiological studies. Our findings however, call for urgent reinforcement of collaborative efforts between the veterinary and the public health services of the country.

Abundance of badgers (Meles meles) in England and Wales

The European badger (Meles meles) is of considerable interest in the UK as it is both a protected species and the main wildlife reservoir for bovine tuberculosis infection in cattle. While there have been three national badger surveys in the 1980s, 1990s and 2011–13, using the number of badger main setts as a proxy for the abundance of badger social groups, none has combined contemporary data on social group size at landscape and national scales. We estimated social group size by genotyping hair samples collected at 120 main setts across England and Wales and employing a capture-mark-recapture method based on genotypes. The estimated mean social group size in England and Wales was 6.74 (±0.63) badgers. There was considerable variation in badger social group size among Land Class Groups (LCGs), with a low of 2.67 in LCG3 and a high of 7.92 in LCG4. Combining these results with the recent Badger Sett Survey of England and Wales, we estimate there are approximately 485,000 badgers (95% confidence intervals 391,000–581,000) in England and Wales. Although direct comparison with previous estimates is not ideal owing to methodological differences, our results are consistent with a marked increase in the badger population of England and Wales since the 1980s.

Association study of Single Nucleotide Polymorphisms (SNP) in Toll-likeReceptor 9 gene with bovine tuberculosis

Toll-like Receptor 9 (TLR9) play an important role in recognition of components of intracellular pathogens like Mycobacterium bovis and subsequent activation of both innate and adaptive immune response and is potential strong candidates for investigating genetic basis of disease resistance. Present investigation was aimed at exploring the association of four SNPs in TLR9 gene with susceptibility/resistance against bovine tuberculosis infection in cattle. Three of SNPs under investigation (rs210982793, rs207807011, rs209190268) revealed polymorphism whereas monomorphism was observed in SNP rs55617140. SNP loci rs210982793 and rs207807011 were significantly (P less than 0.01) associated with susceptibility to bovine tuberculosis in the case control population. Both these SNPs loci were non-synonymous, thus suggestive of their functional role in the immune response against bovine tuberculosis.

A review of risk factors for bovine tuberculosis infection in cattle in the UK and Ireland.

Bovine tuberculosis (bTB) is an important disease of cattle caused by infection with Mycobacterium bovis, a pathogen that may be extremely difficult to eradicate in the presence of a true wildlife reservoir. Our objective was to identify and review relevant literature and provide a succinct summary of current knowledge of risk factors for transmission of infection of cattle. Search strings were developed to identify publications from electronic databases to February 2015. Abstracts of 4255 papers identified were reviewed by three reviewers to determine whether the entire article was likely to contain relevant information. Risk factors could be broadly grouped as follows: animal (including nutrition and genetics), herd (including bTB and testing history), environment, wildlife and social factors. Many risk factors are inter-related and study designs often do not enable differentiation between cause and consequence of infection. Despite differences in study design and location, some risk factors are consistently identified, e.g. herd size, bTB history, presence of infected wildlife, whereas the evidence for others is less consistent and coherent, e.g. nutrition, local cattle movements. We have identified knowledge gaps where further research may result in an improved understanding of bTB transmission dynamics. The application of targeted, multifactorial disease control regimens that address a range of risk factors simultaneously is likely to be a key to effective, evidence-informed control strategies.

Tuberculosis Infection in Cattle and Cattle Owners in North Eastern Parts of Ethiopia

Introduction: Ethiopia is one of the highest TB burden countries in the world. Transmission of M. tuberculosis between humans is well known, however, little is known about the transmission of M. tuberculosis complex species between human and animals, in particular cattle. The purpose of this study was, therefore, to investigate the status of bovine tuberculosis infection in cattle owned by tuberculosis suspected households and isolate M. tuberculosis complex species from sputum of chronic coughers in North Wollo Zone of the Amhara National Regional State, Ethiopia. Method: A cross-sectional study was conducted in selected Kebeles of North Wollo Zone. An intradermal tuberculin skin test was carried out on 381 cattle and sputum samples collected from 124 chronic coughers. Sputum samples were cultured and positive ones were identified by polymerase chain reaction using RD4 and RD9 as a marker. Result: Of the 124 sputum samples cultured, 4/ 124 (3.2%) were positive. Polymerase Chain Reaction using has confirmed that 2/4 (50%) were found to be M. tuberculosis while the rest was atypical Mycobacterial species. Of the 381 tested cattle 5/381 (1.31%) and 10/38 (2.63%) were found positive according to manufacturer recommendation (with a cutoff value > 4 mms) and a cutoff value > 2 mms, respectively. About 67% (6/9) of the individuals who owned positive cattle were having the habit of drinking raw milk. Among the ten positives, five of them resided in lowland (Kolla) areas (1300-1500 mabs). However, none of the owners of tuberculin positive cattle were found to be TB positive. Conclusion: This study has shown 3.2% of culture positivity and of these two were found to be M. tuberculosis. The sputum sample culturing from suspected chronically coughing individuals would help detection of the TB causative agents. According to the current findings BTB is considerable in Ethiopia and cattle should be seen as a potential source of zoonotic TB to humans

Isolation of Mycobacterium tuberculosis complex (MTBC) from dairy cows in China

Eleven thousand five hundred and eighty non-blood samples from dairy cows were subjected to mycobacterium culture and genotyping. As a result, a total of 142 isolates of Mycobacterium tuberculosis complex (MBTC) were identified. Among them, 65 were Mycobacterium tuberculosis , while 77 Mycobacterium bovis . The genotype of M. tuberculosis strains was mainly Beijing family. In addition, the isolation rates of MTBC were 33.89% for lung lymph nodes, 2.81% for nasal swabs, and 3.95% for pharyngeal swabs from cattle positive to tuberculin skin test, respectively. This evidence implied that M. tuberculosis infection in cattle is a new risk to public health and should be paid more attention. Tuberculosis Infection in Animal and Human Populations in Three Districts of Western Gojam, Ethiopia

Tuberculosis concurrent infection in cattle and their respective owners in North-western Ethiopia had been investigated. Two hundred and ten cattle owners and 1220 heads of their cattle were included in the study to determine degree of tuberculosis infection in cattle owned by tuberculosis patients and tuberculosis patients. Comparative intradermal tuberculin test, bacteria culturing, acid fast staining and biochemical tests were used to conduct the study. The prevalence of tuberculosis was significantly (P < 0.001) higher in cattle owned by tuberculosis patients than in cattle owned by non-tuberculosis owners, and infection with tuberculosis was threefold greater in cattle owned by tuberculosis-positive owners. Further more, cattle owners who consumed raw milk were at higher risk (P < 0.001, OR = 3.23) for tuberculosis infection than those who consumed boiled milk. Mycobacterium tuberculosis (15.4%), Mycobacterium bovis (44.1%) and atypical mycobacteria (38.5%) were identified from milk collected from tuberculin-positive cows using biochemical tests. Similarly M. tuberculosis (74.5%), M. bovis (14.9%) and atypical mycobacteria (8.5%) were identified from sputum and fine needle aspiration specimens of tuberculosis patient cattle owners. Mutual transmission of mycobacterium from animals to humans and vice versa has been signified.

Prevalence of tuberculosis in cattle in the Dangme-West district of Ghana, public health implications

Bovine tuberculosis due to Mycobacterium bovis is a major cause of human gastrointestinal tuberculosis in developing countries where bovine milk is often not pasteurised before use. Little information is available on the prevalence of the disease in African cattle and its zoonotic impact. The Ghanaian Government, through its peri-urban dairy cattle development project, is promoting the use of milk and dairy products from local cows in selected districts of the country, including the Dangme-West district of the Greater Accra region. A survey was, therefore, undertaken to determine the prevalence of tuberculosis infection in cattle in this district and to assess the level of awareness with regard to the risks through milk consumption. The standard single intradermal comparative tuberculin test (SCITT) using purified protein derivative (PPD) of M. bovis and M. avium was used to detect cattle infected with M. bovis. The study established the prevalence of a 13.8% infection in cattle in the district although the prevalence was as high as 50% in some kraals. Prevalence was highest in the Ningo sub-district (19.0%), followed by Dodowa, the district capital (14.0%), while the other two sub-districts, Prampram and Osudoku had lower rates of 11.3 and 10.8%, respectively. Cattle of all ages and both sexes were affected, but the prevalence in cows was twice as high as that in heifers or bulls. The study also established that there is a considerable lack of knowledge about bovine tuberculosis among cattle owners and herdsmen in the community and that milk is often used untreated, thus increasing the risk of human infection.

Badgers (Meles meles), cattle and bovine tuberculosis (Mycobacterium bovis): a hypothesis to explain the influence of habitat on the risk of disease transmission in southwest England.

Badgers are believed to be responsible for a high proportion of the cases of bovine tuberculosis in cattle in southwest England where, despite the onset of badger control operations in 1975, comparatively high numbers of cattle continue to fail the tuberculin test. To determine why the disease remains a problem in these areas, data on badger densities and patterns of land use were examined. Areas subject to repeated badger control operations had greater landscape heterogeneity and a higher density of linear habitat features. These habitat features were not related to badger density, measured as the mean number of social groups per square kilometre. Environmental contamination by infected badger urine is thought to be the main mode for the transmission of bovine tuberculosis. Field studies in an area with tuberculosis in both badgers and cattle showed that badgers may urinate on pasture after crossing through a linear feature, and that the number of these crossing-point urinations increases with the number of linear features crossed. The hypothesis is presented that these crossing-point urinations are a major source of bovine tuberculosis infection in cattle, and that areas with increased numbers of linear features have greater levels of contamination of pasture with badger urine and hence increased opportunities for disease transmission.

Mycobacteria in pond waters as a source of non-specific reactions to bovine tuberculin in New Zealand

When 71 samples were collected from ponds throughout New Zealand, 35 (49.3%) were found to contain mycobacteria. The majority of these strains (62.9%) belonged to a homogeneous group (tentative designation H-Group, which differed from any known mycobacterial species. Mycobacteria of this H-group had also been found in sphagnum vegetation growing in the immediate vicinity of many of the ponds. H-Group mycobacteria induce sensitization in guinea pigs against bovine tuberculin. The PPD sensitin prepared from these mycobacteria gave rise to larger reactions in guinea pigs than did bovine tuberculin when used in the same concentrations (500 and 50 TU). The possible sensitization of cattle to bovine tuberculin via drinking water containing H-Group mycobacteria, is discussed. The larger size of the delayed hypersensitivity reactions in guinea pigs using the same concentrations of bovine and homologous tuberculin, suggests that comparative intradermal testing might enable this non-specific reaction to be distinguished from the specific reaction developed during bovine tuberculosis infection in cattle.

Tuberculosis infection in cattle and badgers in an area of Staffordshire.

The recent history of tuberculosis infection in cattle in Staffordshire is described, showing how a problem area in the north of the county was identified. The subsequent culture of Mycobacterium bovis from badger faeces and a badger carcase is reported together with the operations to remove badgers from the area.
