Measurement of Serum Vitamins B2 and B6 in Patients with Hashimoto’s Thyroiditis by LC–MS/MS Method

Vitamin B2 and vitamin B6 are cofactors and coenzymes in many biochemical reactions, and their sufficient serum concentrations may be crucial for the proper functioning of the thyroid gland. The aim of the study was to develop and validate a novel high-performance liquid chromatography–tandem mass spectrometry (LC–MS/MS) method for the determination of vitamin B2 and vitamin B6 in the serum of patients with Hashimoto’s thyroiditis (HT). Detection of the analytes was performed on a triple-quadrupole MS with multiple reaction monitoring using electrospray ionization. The mobile phase consisted of water and methanol, both containing 0.1% (v/v) formic acid. The analytes were isolated from serum by the one-step sample preparation of the trichloroacetic acid precipitation. Validation parameters, including linearity, accuracy, precision, limits of detection and quantification, and stability, were determined and fulfilled the criteria for analyzing compounds in biological fluids. The method was applied for the determination of the above vitamin concentrations in serum samples of 74 patients with HT and 33 healthy controls. The concentration of vitamin B2 was significantly lower in patients with HT compared to the control group (7.20 ± 7.86 ng/mL vs. 12.03 ± 9.62 ng/mL) and the concentration of vitamin B6 was also lower, but without a statistically significant difference (30.09 ± 19.16 ng/mL vs. 37.29 ± 23.02 ng/mL). We concluded that the vitamins concentration measurement may be helpful in the diagnosis of vitamin deficiencies and the supplementation strategy.

Comparison of Extraction Assays and Quantification of Protein from Ulva anandii (Cholorophycota)

Seaweeds contain many macronutrients including protein, therefore they can be utilized to fulfil the protein requirements of human beings. This research focused on extracting total protein in green seaweed Ulva anandii (Amjad et Shameel 1993), from the crude extracts, by using the trichloroacetic acid (TCA) and acetone precipitation methods, and the estimation of crude extract (water-soluble proteins), and those obtained from the two above-mentioned methods. The results indicate that the water-soluble proteins had the highest quantity (949.75µg/mL) followed by the TCA precipitation method (831µg/mL), while the acetone precipitation method had the least concentration of total protein (100 µg/mL). The study concludes that treatment with organic solvents lowers the quantity of protein extracted from U. anandii.

In-Depth Quantitative Proteomics Analysis of the Pseudomonas aeruginosa Secretome.

Secreted proteins play vital roles in bacterial communication, metabolism, and virulence. However, analysis of the bacterial secretome can be challenging, especially if bacteria require rich media for growth or secretion. In this protocol, we describe an efficient and sensitive method to analyze the secretome by shotgun proteomics, using a combination of trichloroacetic acid (TCA) precipitation and single-pot solid-phase-enhanced sample preparation (SP3) for the preparation of the samples. The method was used to identify and quantify proteins secreted by wildtype Pseudomonas aeruginosa PAO1, highlighting its applicability for proteins secreted in limited amounts and in rich media.

β-lactamase (Bla) Reporter-based System to Study Flagellar Type 3 Secretion in Salmonella.

Export of type 3 secretion (T3S) substrates is traditionally evaluated using trichloroacetic acid (TCA) precipitation of cultured cell supernatants followed by western blot analysis of the secreted substrates. In our lab, we have developed β-lactamase (Bla), lacking its Sec secretion signal, as a reporter for the export of flagellar proteins into the periplasm via the flagellar T3S system. Bla is normally exported into the periplasm through the SecYEG translocon. Bla must be secreted into the periplasm in order to fold into an active conformation, where it acts to cleave β-lactams (such as ampicillin) to confer ampicillin resistance (ApR) to the cell. The use of Bla as a reporter for flagellar T3S allows the relative comparison of translocation efficiency of a particular fusion protein in different genetic backgrounds. In addition, it can also be used as a positive selection for secretion. Protein sample preparation for tissue distribution study.

The distribution and expression level of a protein among animal tissues is indicative of its possible roles. It is important to establish a generally applicable method to prepare protein samples with high-quality and achieve near 100% recovery of proteins from animal tissues. During preparation, to sufficiently dissolve and maintain stability of almost all proteins from tissues, as well as to avoid most contaminations affecting protein detection, 2×SDS Sample Buffer, sonication and trichloroacetic acid precipitation are applied. Here we provide a relatively simple, reproducible, and broadly applicable method for studying protein distribution in most tissues, in which the issues resulting from protein degradation and modification during sample preparation and assay interference by other cellular components like neutral lipids and glycogen could be overcome. Furthermore, this method represents the protein content by equal wet tissue mass, which is a better means to present the expression level of a protein in various tissues. High-quality protein samples from almost all tissues could be prepared. The samples produced are amenable to tissue distribution analysis by Western blotting and for silver/Coomassie staining, proteomics, and other protein analyses, which would contribute to potential biomarkers or treatments for a disease.

PROTEIN EXTRACT OF BOVINE HEART DISPLAYS ANTIBACTERIAL ACTIVITY ON HUMAN PATHOGENIC BACTERIA IN In-vitro STUDY

A typical type of animal proteins known as antimicrobial proteins (AMPs) actively taking part in innate immune system are recognized as suitable alternatives to classical antibiotics to combat the development multidrug resistant bacterial strains. The protein fractions extracted from bovine heart tissue by different protein precipitating agents after acid extraction were screened for their bactericidal activity against four clinical isolates. Amongst eleven protein fractions including the acid extract, only three fractions (Phosphate buffered saline filtrate, 10% Trichloroacetic Acid (TCA) precipitate and 30% Ammonium sulphate precipitate) showed antibacterial activities. The Phosphate buffered saline filtrate showed activity on two organisms (Salmonella paratyphi ,A, and Pseudomonas aeruginosa) and the zone of inhibitions were found to be 8mm size for both organisms. The protein fraction extracted by 10% TCA exerted activity on Salmonella typhimurium and P. aeruginosa with 12 and10 mm of inhibitory zones respectively. The 30% ammonium sulphate precipitate inhibited the growth of all the four test organisms strongly with 14-23 mm of inhibitory zones. All protein fractions exhibiting bactericidal activities did not cause haemolysis on human red blood cell of ABO blood groups. Hence further studies in this regard would be much helpful for pharmaceutical applications.

Effect of Over‐the‐Counter Tooth Whitening Consumer Products of Release of Collagen and Protein from Teeth

While tooth whitening used to consist of either whitening strips or professional treatments: there are now a variety of products available to consumers in the United States. While most involve either hydrogen peroxide or its precursor, carbamide peroxide, as the active ingredients, there are alternatives. Carbamide peroxide can be administered in gel form in a tray with or without an LED light. Instead of a strip, hydrogen peroxide can now by applied directly to the teeth in form of pen and remains overnight. The goal of this project was to examine the effect of whitening agents on the release of both collagen and other proteins. Extracted molars from patients who signed informed consent forms were placed in artificial saliva and treated with activated charcoal, carbamide peroxide with (35%) or without (44%) an LED light, or a whitening pen with 14% hydrogen peroxide according to manufacturer’s instructions. Artificial saliva was changed daily, and all teeth received six days of no treatment and four rounds of 6 treatments with a break between the second and third. The fluid surrounding the teeth underwent a trichloroacetic acid precipitation to remove the non‐ collagen protein. The supernatant was adjusted to the proper pH and collagen was measured using a modified version of the Lowry assay. In addition, the proteins were separated using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS PAGE). Molars that received no treatment, as well as prior to treatment, show no protein. All treated teeth showed collagen and non‐collagen protein released whenever treatments were applied as well as in between rounds of treatment. The amount of collagen released from activated charcoal, 35% carbamide peroxide with LED, 44% carbamide peroxide and 14% hydrogen peroxide pen in mg/ml were 1.048, 1.994, 1.530, and 0.9034 respectively. For non‐collagen, the average amount of protein released for activated charcoal, 35% carbamide peroxide with LED, and 44% carbamide peroxide in mg/ml respectively were: 0.5714, 0.4155, and 0.6363. Both collagen and non‐collagen were released from all treated teeth in between rounds of treatments. The size of the proteins released was characterized using SDS PAGE. Consistent with the TCA and modified Lowry results, no bands were observed in artificial saliva prior to treatment but all treatments resulted in bands at molecular weight (MW) of 129 and 75 kDa among others. The MW of former band matches the expected subunit MW of collagen which is found in the dentin of teeth. These results demonstrate that treatment of teeth with hydrogen peroxide, carbamide peroxide with or without LED light or activated charcoal results in release of various proteins from dentin and enamel. In addition, these proteins continued to be released after treatments were applied. Hydrogen peroxide was measured using a colorimetric test. The amounts of peroxide in other samples was consistent with manufacturer’s claims.

UV-VIS spectrophotometric-assisted chemometric calibration models for simultaneous determination of thiamine and pyridoxine vitamins in powdered infant formula

Thiamine and pyridoxine, which are hydrophilic B vitamins, are compounds that play an important role in many biochemical events for the health of infants and adults. Recently, because of the complexity of food and pharmaceutical composition, chemometric models are preferred to obtain relevant information from multivariate measured data. The aim of this study was to determine the simultaneous quantitative analysis of thiamine and pyridoxine in powdered infant formula and pharmaceutical formulations by the UV-visible spectrophotometric method coupled with chemometric calibration models (partial least squares (PLS) and principal component regression (PCR)). These vitamins were extracted by addition of trichloroacetic acid (precipitation of proteins) after samples of powdered infant formula was dissolved in Milli-Q water. Factorial design was used to prepare the calibration (25 samples) and validation (8 samples) sets containing mixture of thiamine and pyridoxine within their linear range (2.5-25 μg/mL). Spectra of obtained mixtures and samples were recorded between wavelengths of 200-360 nm with the intervals Δλ=1 nm. In regression models created by using PLS and PCR algorithm, the results had acceptable recovery 98.14% for both compounds. These chemometric models predict the quantification of both, thiamine and pyridoxine in powdered infant formula samples via interpreting the UV-Vis spectrum. The average thiamine and pyridoxine contents of powdered infant formula samples were found to be 5.02±0.065 μg/g and 4.32±0.086 μg/g for PLS model and 4.98±0.078 μg/g and 4.03±0.091 μg/g for PCR model, respectively.

Genotoxicity of advanced glycation end products in vitro is influenced by their preparation temperature, purification and cell exposure time.

Advanced glycation end products (AGEs) are formed via non-enzymatic reactions between amino groups of proteins and the carbonyl groups of reducing sugars. Previous studies have shown that highly glycated albumin prepared using a glucose-bovine serum albumin (Glu-BSA) model system incubated at 60°C for 6 weeks induces genotoxicity in WIL2-NS cells at 9 days of exposure measured by the cytokinesis-block micronucleus cytome (CBMNcyt) assay. However, this AGE model system is not physiologically relevant as normal body temperature is 37°C and the degree of glycation may exceed the extent of albumin modification in vivo. We hypothesised that the incubation temperature and purification method used in these studies may cause changes to the chemical profile of the glycated albumin and may influence the extent of genotoxicity observed at 3, 6 and 9 days of exposure. We prepared AGEs generated using Glu-BSA model systems incubated at 60°C or 37°C purified using trichloroacetic acid (TCA) precipitation or ultrafiltration (UF) and compared their chemical profile (glycation, oxidation, and aggregation) and genotoxicity in WIL2-NS cells using the CBMNcyt assay after 3, 6 and 9 days of exposure. The number of micronuclei (MNi) was significantly higher for cells treated with Glu-BSA incubated at 60°C and purified via TCA (12 ± 1 MNi/1000 binucleated cells) compared to Glu-BSA incubated at 37°C and purified using UF (6 ± 1 MNi/1000 binucleated cells) after 9 days (P < 0.0001). The increase in genotoxicity observed could be explained by a higher level of protein glycation, oxidation, and aggregation of the Glu-BSA model system incubated at 60°C relative to 37°C. This study highlighted that the incubation temperature, purification method and cell exposure time are important variables to consider when generating AGEs in vitro and will enable future studies to better reflect in vivo situations of albumin glycation.

Comparison of the performance of secretome analysis based on metabolic labeling by three unnatural sugars

分泌蛋白质是调控细胞间信号转导的重要生物大分子。由于分泌蛋白的丰度相比于胞内蛋白以及培养基添加剂更低,因此分泌蛋白的高通量鉴定是目前蛋白质组学界研究的热点和难点。目前,基于生物质谱的分泌蛋白质组学分析一般均需要从无血清的条件培养基中获得分泌蛋白质,再对其进行富集和分析。该流程操作步骤繁琐,易造成分泌蛋白质的损失和降解。本工作采用基于生物正交化学生物学技术实现对分泌蛋白质的高选择性标记和高效富集。通过结合点击化学技术,综合评估了分泌蛋白质分析中用于代谢标记的不同糖类似物。采用3种最常用的商品化糖类似物,N-叠氮乙酰甘露糖胺(ManNAz)、N-叠氮乙酰半乳糖胺(GalNAz)和N-叠氮乙酰葡萄糖胺(GlcNAz)分别对HeLa细胞进行代谢标记,之后通过炔基生物素探针对条件培养基中的分泌蛋白进行富集,结合质谱分析来对比3种糖类似物对分泌蛋白的标记效率。最后通过无标定量蛋白质组学分析,系统评估了3种糖类似物用于分泌蛋白质组分析的性能。结果表明,基于ManNAz的分泌蛋白标记方法鉴定到了282个分泌蛋白、224个细胞质膜蛋白以及846个N-糖基化位点;对分泌蛋白的富集效率分别较GalNAz和GlcNAz提高了130%和67.2%;对细胞质膜蛋白的富集效率较GalNAz和GlcNAz分别提高了273.3%和148.7%,体现出了明显的优势。本研究的实验结果为分泌蛋白高选择性富集和系统分析提供了有益的对比分析和新技术策略。

Study on Sialic Acid Binding State in Stewed Bird's Nest and Optimization of Enzymatic Extraction of Free and Oligosaccharide-Bound Sialic Acid.

As a traditional Chinese health food, edible bird's nest (EBN) has high medicinal value, which is mostly attributed to the high content of sialic acid (SA). SA mainly exists in protein-bound, oligosaccharide-bound, and free forms and the binding forms of SA are closely related to the functions of EBN. To establish a simple but robust method to distinguish and determinate the free and oligosaccharide-bound SA content and the protein-bound SA content, and investigate the changes to SA binding states in EBN during different processing. Protein-bound SA in EBN was separated from other forms of SA by trichloroacetic acid precipitation, and SA content was determined by HPLC. The effects of stewing conditions on the distribution of SA in EBN were investigated and response surface methodology was used to explore the optimal conditions for enzymatic extraction of free and oligosaccharide-bound SA from EBN. The average recoveries of free and oligosaccharide-bound SA and protein-bound SA were 97.82-98.92% and 94.67-95.75%, respectively. The content of free and oligosaccharide-bound SA in stewed EBN was proportional to the stewing temperature, stewing time, and liquid to material ratio, while that of protein-bound SA was inversely proportional to those factors. Through response surface analysis, we found that the optimum technological parameters were as follows: liquid to material ratio, 60:1; enzymolysis time, 2 h; enzyme dosage; 12000 U/g (alkaline protease); pH, 11; enzymolysis temperature, 60°C. This method can not only distinguish between free and oligosaccharide-bound SA and protein-bound SA effectively, but can also determine the contents of them. The results of the investigation on stewing conditions and response surface analysis can be used as the theoretical basis for further pharmacological research of EBN, and can also provide theoretical guidance for the development of EBN products. A method for the determination of free and oligosaccharide-bound SA and protein-bound SA in EBN by HPLC was established, and the extraction process of free and oligosaccharide-bound SA was optimized.

Effect of sample preparation on analysis of human milk endogenous peptides using liquid chromatography-tandem mass spectrometry

人乳内源肽是乳蛋白在乳腺中被降解形成的具有生理功能的肽,是人乳的重要组成部分,研究人乳内源肽对于婴儿健康具有重要的意义。高效液相色谱-串联质谱(LC-MS/MS)联用技术的应用,促使人乳内源肽的研究取得了突破性的进展。人乳中内源肽含量低、干扰组分多,样品制备方法是影响分析结果的关键步骤。为了研究样品制备方法对分析结果的影响,分别采用不变性超滤法(UF 1)、热变性超滤法(UF 2)、化学变性超滤法(UF 3)、三氯乙酸沉淀法(PCPN 1)、乙醇沉淀法(PCPN 2)、强疏水性碳介孔材料(highly ordered mesoporous carbon, OMC)富集法等6种方法从人乳中提取内源肽,利用LC-MS/MS研究样品制备方法对人乳内源肽分析结果的影响。结果表明,UF 1和UF 2法制备的样品中可鉴定到的肽段数目分别为1161±8条和1017±91条,两种方法制备的样品中肽序列的重合率大于70%, UF 1在所有方法中鉴定到的肽的数目最多。UF 3法制备的样品所能鉴定到的肽段数目最少,仅为366±18条。PCPN 1和PCPN 2两种沉淀法制备样品中的内源肽分别为779±69和876±55条,但内源肽差异较大,仅有约50%肽段序列重合。OMC法制备样品中肽的数目为549±151条,与其他方法相比,虽然鉴定的肽数量上没有优势,但该方法制备的样品中肽在等电点(pI)和亲水性平均系数(GRAVY)等性质上没有偏倚,说明该法可用于制备特定人乳内源肽。6种方法制备的样品中鉴定到来源于β-酪蛋白、免疫球蛋白受体、骨桥蛋白、αS1-酪蛋白、κ-酪蛋白和胆盐激活脂肪酶的肽,并且源于以上蛋白质的肽段总数在该样品中均超过88%,说明6种方法制备的样品都可以满足鉴定一般人乳内源肽的需求。UF 2、UF 3和OMC法制备的样品中鉴定到源于乳铁蛋白的内源肽的数目分别为21、38和19条,内源肽在乳铁蛋白上的覆盖率分别为14%、16%和19%,而文献常用的PCPN 1法制备的样品则会丢失此类内源肽。综上,UF 2法制备的样品不仅肽段数量多、母体蛋白质种类丰富,还可鉴定到源于乳铁蛋白的肽,可作为人乳内源肽组学研究中的首选方法。

Role of zonulin and GLP-1/DPP-IV in alleviation of diabetes mellitus by peptide/polypeptide fraction of Aloe vera in streptozotocin- induced diabetic wistar rats

Ethno-pharmacological relevanceThe genus Aloe has a long history of usage in medicine. Aloe barbadensis Miller, commonly known as Aloe vera, is said to possess anti-diabetic, anti-inflammatory, anti-cancer, anti-microbial, immunomodulation, wound healing properties. Aim of the studyIn diabetes mellitus, loss in intestinal permeability is observed with high levels of zonulin and low levels of glucagon-like peptide-1 (GLP-1) leading to hyperglycemia. The aim of the study was to understand the role of peptide/polypeptide fraction (PPF) of Aloe vera in the alleviation of diabetes through maintaining the intestinal permeability by regulating the zonulin and GLP-1 levels. Materials and methodsThe PPF of Aloe vera was obtained through trichloroacetic acid precipitation. The anti-diabetic potential of the PPF was tested through DPP-IV inhibition, glucose diffusion assay, and by using Rin-m5F cells. The anti-diabetic potential of the PPF was tested at a dose of 0.450 mg/kg bw in vivo using streptozotocin-induced diabetic Wistar rats. The effect of PPF on fasting plasma glucose, insulin, glucagon, Zonulin, GLP-1, DPP-IV, levels were studied in diabetic rats. The histopathological studies of the pancreas, small intestine, and liver were carried out for organ-specific effects. ResultsPPF has the ability to reduce fasting plasma glucose levels with concomitant increase in insulin levels in streptozotocin-induced diabetic rats. It was also observed that increase in GLP-1 levels with a decrease in DPP-IV and zonulin levels thereby mitigating the loss of intestinal permeability. These findings correlate with the small intestine's histopathological observation where the excessive proliferation of epithelium in the small intestine of diabetic rats was reduced after PPF treatment. ConclusionThese results suggest that the PPF of Aloe vera alleviates diabetes through islet cell rejuvenation via GLP-1/DPP-IV pathway and thereby suggesting the usage of PPF as an alternate medicine for diabetes mellitus with the possibility to reduce the intestinal permeability and zonulin levels.

A method to improve quantitative radiotracing-based analysis of the in vivo biodistribution of drug carriers.

Biodistribution studies are essential in drug carrier design and translation, and radiotracing provides a sensitive quantitation for this purpose. Yet, for biodegradable formulations, small amounts of free‐label signal may arise prior to or immediately after injection in animal models, causing potentially confounding biodistribution results. In this study, we refined a method to overcome this obstacle. First, we verified free signal generation in animal samples and then, mimicking it in a controllable setting, we injected mice intravenously with a radiolabeled drug carrier formulation (125I‐antibody/3DNA) containing a known amount of free radiolabel (125I), or free 125I alone as a control. Corrected biodistribution data were obtained by separating the free radiolabel from blood and organs postmortem, using trichloroacetic acid precipitation, and subtracting the confounding signal from each tissue measurement. Control free 125I‐radiolabel was detected at ≥85% accuracy in blood and tissues, validating the method. It biodistributed very heterogeneously among organs (0.6–39 %ID/g), indicating that any free 125I generated in the body or present in an injected formulation cannot be simply corrected to the free‐label fraction in the original preparation, but the free label must be empirically measured in each organ. Application of this method to the biodistribution of 125I‐antibody/3DNA, including formulations directed to endothelial target ICAM‐1, showed accurate classification of free 125I species in blood and tissues. In addition, this technique rendered data on the in vivo degradation of the traced agents over time. Thus, this is a valuable technique to obtain accurate measurements of biodistribution using 125I and possibly other radiotracers.

Analysis of Protein Stability by Synthesis Shutoff.

In this protocol, we describe the analysis of protein stability over time, using synthesis shutoff. As an example, we express HA-tagged yeast mitofusin Fzo1 in Saccharomyces cerevisiae and inhibit translation via cycloheximide (CHX). Proteasomal inhibition with MG132 is performed, as an optional step, before the addition of CHX. Proteins are extracted via trichloroacetic acid (TCA) precipitation and subsequently separated via SDS-PAGE. Immunoblotting and antibody-decoration are performed to detect Fzo1 using HA-specific antibodies. We have adapted the method of blocking protein translation with cycloheximide to analyze the stability of high molecular weight proteins, including post-translational modifications and their impact on protein turnover.

Evaluation of Heating and Irradiation Methods for Production of Purified Protein Derivative (PPD) of Mycobacterium Tuberculosis

Tuberculin skin test, also known as the tuberculin or purified protein derivative (PPD) test, is an extensively applied diagnostic test for the detection of primary infection with Mycobacterium tuberculosis (Mtb). The production of PPD is accompanied by some difficulties that require a series of modifications in the production and purification processes. The present study aimed to determine the facilitation level of the manufacturing process by modifying evaluation methods for the production of PPD tuberculin. Mtb strains were cultured in Lowenstein-Jensen media, and the cultured strains were inoculated into the Dorset-Henley liquid medium by the biphasic medium of potato-Dorset-Henley. After incubation, flasks containing cultured strain were selected for bacterial inactivation, and the optimal gamma radiation dose(s) was determined. Tuberculoproteins were precipitated by ammonium sulfate (AS) and Trichloroacetic acid (TCA). Protein concentration was determined using the Bradford and Kjeldahl protein assay methods. Finally, the lymphocyte transformation test and potency test were performed. Based on the results, the Dorset-Henley liquid medium is suitable for the massive growth of the bacterium. The transferal of Mtb from solid to liquid medium was directly carried out without intermediate culture. It was found that during tuberculoprotein production, heating at 100&amp;deg;C for 3 h would be safe for killing mycobacterium. Furthermore, the simultaneous use of heating and gamma irradiation (8 kGgy) killed all of the mycobacteria, while doses of 1, 1.5, and 7 kGy decreased a significant number of bacterial cells. The results also indicated that the concentration of tuberculoprotein extracted by TCA precipitation method was higher than that obtained by AS precipitation. The tuberculoproteins which were produced by these two methods in the lymphocyte transformation test were not significantly different in terms of potency (P&amp;gt;0.05). Moreover, due to the high volume of produced protein, the protein measurement was more efficiently carried out by the Kjeldahl method, compared to the Bradford method. Finally, the results of the present study demonstrated that in addition to the novel approach of gamma irradiation, optimum methods are efficient and applicable in the production of PPD tuberculin.

Pea GH3 acyl acid amidosynthetase conjugates IAA to proteins in immature seeds of Pisum sativum L. – A new perspective on formation of high-molecular weight conjugates of auxin

Gretchen Hagen 3 (GH3) acyl acid amidosynthetases are encoded by early auxin-responsive genes and catalyze an ATP-dependent biosynthesis of IAA-amino acid conjugates. An amide conjugate of IAA, indole-3-acetyl-aspartate (IAA-aspartate, IAA-Asp), is a predominant form of bound auxin in immature seeds of pea. However, there is some evidence that IAA is also able to form high molecular weight amide conjugates with proteins in pea and other plant species. In this short study we report that recombinant PsGH3 IAA-amino acid synthetase, which exhibits a preference for the formation of IAA-Asp, can also conjugate IAA with the protein fraction from immature seeds of pea (S-10 fraction). We studied [14C]IAA incorporation to the S-10 protein fraction by two assays: TLC method and protein precipitation by trichloroacetic acid (TCA). In both cases, radioactivity of [14C]IAA in the protein fraction increases in comparison to the control (without PsGH3), about 9.3- and 3.17-fold, respectively. l-Asp, as a preferred substrate in the IAA conjugation catalyzed by PsGH3, down-regulates [14C]IAA conjugation to the proteins as shown by the TLC assay (∼2.8-fold decrease) and the TCA precipitation variant (∼2-fold decrease). Moreover, l-Trp that competes with Asp for the catalytic site of PsGH3 and inhibits activity of the enzyme, diminished radioactivity of [14C]IAA-proteins about 1.2- and 2.8-fold, respectively. Taking into account that amino group of an amino acid or a protein acts as an acceptor of the indole-3-acetyl moiety from IAA-AMP intermediate during GH3-dependent conjugation, we masked amine groups (α- and ε-NH2) of the S-10 protein fraction from pea seeds by reductive alkylation. The alkylated proteins revealed about 3- and 2.8-fold lower radioactivity of [14C]IAA than non-alkylated fraction for TLC and TCA precipitation variant, respectively. This is a first study demonstrating that formation of high molecular weight IAA conjugates with proteins is catalyzed by a GH3 acyl acid amidosynthetase.

Systematic examination of protein extraction, proteolytic glycopeptide enrichment and MS/MS fragmentation techniques for site-specific profiling of human milk N-glycoproteins

Human milk contains numerous N-glycoproteins with functions that provide protection to the infant. Increasing understanding of the functional role of human milk glycoproteins within the infant requires toolsets to comprehensively profile their site-specific glycosylation patterns. However, optimized methods for site-specific glycosylation analysis across the entire human milk proteome are not available. Therefore, we performed a systematic analysis of techniques for profiling the sites and compositions of N-glycans in human milk using liquid chromatography/mass spectrometry. To decrease interference from non-target molecules, we compared techniques for protein extraction, including ethanol (EtOH) precipitation, trichloroacetic acid precipitation, molecular weight cut-off filtration and techniques for tryptic glycopeptide enrichment, including C18-, porous graphitized carbon and hydrophilic interaction liquid chromatography (HILIC)-solid phase extraction (SPE) and acetone precipitation. We compared the capacity of higher-energy collision dissociation, electron-transfer dissociation and electron-transfer/higher-energy collision dissociation (EThcD) to produce fragment ions that would enable effective identification of the glycan composition, peptide sequence and glycosylation site. Of these methods, a combination of EtOH precipitation, HILIC-SPE and EThcD-fragmentation was the most effective for human milk N-glycopeptide profiling. This optimized approach significantly increased the number of N-glycopeptides and precursor N-glycoproteins (246 N-glycopeptides from 29 glycoproteins) compared with a more common extraction approach with no protein extraction and C18 clean-up (62 N-glycopeptides from 11 glycoproteins). The advancement in methods for human milk N-glycoproteins provided by this study represents a key step for better understanding the function of glycoproteins within the breast milk-fed infant.

Development of an alternative approach for detecting botulinum neurotoxin type A in honey: Analysis of non-toxic peptides with a reference labelled protein via liquid chromatography-tandem mass spectrometry

ABSTRACT In this study, we developed a reference labelled protein containing the partial amino acid sequence of botulinum neurotoxin type A (BoNTA). We also applied it as an internal standard to detect specific and non-toxic peptides originated from BoNTA in honey with the use of liquid chromatography–tandem mass spectrometry (LC–MS/MS). Original proteins in the honey sample were collected through a two-step process that included solubilisation and trichloroacetic acid (TCA) precipitation. Solubilisation by adding water enabled processing of proteins in honey. TCA precipitation collected proteins without specific binding. The combination of protein alkylation and an appropriate enzyme-to-protein ratio ensured feasibility of tryptic digestion. A desalting process eliminated a large amount of salts and other tryptic peptides in the honey sample. The use of the reference labelled protein enabled compensation for tryptic digestion efficiency and electrospray ionisation efficiency based on LC-MS/MS measurement. After the peptide selection and protein BlastP analysis, five unique peptides were chosen. The non-toxic peptides originating from BoNTA were reliably detected using LC–MS/MS based on a multiple-reaction monitoring mode. Detection of several peptides ensured screening of BoNTA in honey samples. Based on the responses, the proteotypic peptide LYGIAINPNR was selected as the quantitative peptide. Due to maintaining the relative ion ratios, the selective transition completely identified the non-toxic peptides. The intensity of the transitions established a detection limit of BoNTA estimated to be 9.4 ng mL−1. Although extraction efficiency was not evaluated using the BoNTA standard, the results suggested this method may be used for quantification of BoNTA in honey. The method was applied to 19 honey samples purchased in Tokyo; none of them was found to contain the target toxin. Overall, the method is expected to accelerate BoNTA monitoring for food safety.
