Comparison of the Shear Bond Strength of Orthodontic Composites Containing Silver and Amorphous Tricalcium Phosphate Nanoparticles: an ex vivo Study.

It is important to use orthodontic composites with favorable properties, which are easily removed after the end of the treatment but not easily debonded during treatment. Nanoparticles have drawn attention for their antibacterial properties when added to composite resins. However, the effect of addition of nanoparticle on shear bond strength is not broadly discussed. The present study was designed to compare the shear bond strength of orthodontic brackets bonded by orthodontic composite containing silver nanoparticles with orthodontic composite containing amorphous tricalcium phosphate nanoparticles. In this ex vivo study, 36 sound extracted human premolars were used and randomly divided into three groups. The brackets were bonded in the first group by composite without nanoparticles, in the second group by composite containing 3% amorphous tricalcium phosphate nanoparticles and in the third group by composite containing 0.3% silver nanoparticles at the buccal surface of the teeth. The shear bond strengths of the samples were measured 24 hours after preparation by a universal testing machine. Data were analyzed using SPSS 21 software through one-way ANOVA and Tamhane's T2 multiple comparison tests. pValues under 0.05 were considered significant. There was no significant difference between the mean shear bond strength of composite containing amorphous tricalcium phosphate nanoparticles with composite without nanoparticles (p= 0.142). However, the mean shear bond strength in the composite containing silver nanoparticles was significantly lower than the other two groups (p< 0.001). According to the results of this study, the addition of amorphous tricalcium phosphate nanoparticles to orthodontic composite does not significantly decrease the shear bond strength while silver nanoparticles reduce the shear bond strength of orthodontic composite.

Poly (acrylic acid)/tricalcium phosphate nanoparticles scaffold enriched with exosomes for cell-free therapy in bone tissue engineering: An in vivo evaluation

Introduction: This study aimed to assess the potential of poly (acrylic acid)/tricalcium phosphate nanoparticles (PAA/triCaPNPs) scaffold in terms of biocompatibility and osteoconductivity properties the in-vivo evaluation as well as to investigate the performance of PAA/triCaPNPs scaffold (with or without exosomes derived from UC-MSCs) for bone regeneration of rat critical-sized defect. Methods: PAA/triCaPNPs scaffold was made from acrylic acid (AA) monomer, N,N’-methylenebisacrylamide (MBAA), sodium bicarbonate (SBC), and ammonium persulfate (APS) through freeze-drying method. For in vivo evaluation, we randomly divided 24 rats into three groups. The rat calvarial bone defects were treated as follows: (1) Control group: defects without any treatment, (2) scaffold group: defects treated with scaffold only, (3) scaffold+exo group: defects treated with scaffold enriched with exosomes (1 μg/μl ,150 μg/per rat). Eight- and 12-weeks post-surgery, half of the animals were sacrificed and bone regeneration was examined through micro-computerized tomography (µ-CT), histological staining, and immunohistochemistry (IHC). Results: Quantitative analysis based on µ-CT scan images at 8 and 12 weeks post-implantation clearly indicated that healing rate for defects that were filled with scaffold enriched with exosome was significantly higher than defects filled with scaffold without exosome. The H&amp;E and Masson staining results revealed that more new bone-like form developed in the scaffold+exo group than that in control and scaffold groups. Further, IHC staining for osteocalcin and CD31 confirmed that more bone healing in the scaffold+exo group at 12 weeks could be associated with osteogenesis and angiogenesis concurrently. Conclusion: In the present study, we aimed to investigate the therapeutic potential of PAA/triCaPNPs scaffold as a carrier of human UC-MSC-derived exosome to achieve the exosome-controlled release on calvarial bone defect. The in vivo results indicated that the exosome-enriched scaffold could effectively minify the defect area and improve the bone healing in rat model, and as such it could be an option for exosome-based therapy.

Evaluation of the osteoconductive properties of scaffold containing platete-enriched-fibrin (PRF) with three calcium phosphate (TCP) in the alveolar socket repair after tooth extraction: An animal study.

Bone regeneration can be accomplished through osteogenesis, osteoinduction, and osteoconduction mechanisms. This study aimed to investigate the properties of the PRF scaffold with tricalcium phosphate nanoparticles in socket preservation in an animal model. Fabrication of PRF performed. In this experimental study, 18 rats were divided into three negative control, PRF, and PRF/TCP groups. The mechanical and chemical tests including swelling rate, degradation time, and MTT tests were applied to the scaffolds. In each animal, the first maxillary right molar was extracted, and extraction sites of test groups were filled with a resorbable biocompatible biomaterial in situ hardening bone substitute. After 2 and 4 weeks all animals were sacrificed and examined histopathologically and with qRT-PCR. Histological results showed TCP in combination with PRF accelerates bone regeneration with the highest amount of lamellar bone and collagen formation compared to the control and PRF alone. Mechanical and chemical tests on the scaffolds showed the addition of TCP to the PRF scaffold decreases the swelling rate and increases the degradation time. qRT-PCR showed expression of osteogenic genes increased significantly (p < 0.05) in PRF/TCP and PRF, respectively. In conclusion, the gelatin hydrogel containing PRF/TCP scaffold led to more bone formation after tooth extraction. Therefore, the injectable PRF\TCP hydrogel is a promising candidate for bone repair and regeneration.

Effects of tricalcium phosphate-titanium nanoparticles on mechanical performance after friction stir processing on titanium alloys for dental applications

Friction Stir Processing (FSP) is a solid-state method that is widely used in titanium and its alloys to enhance their strength. The FSP technique produces low-defect-density materials with improved mechanical properties, making it potentially valuable for orthopedic and dental applications. It is primarily used for low-melting-point materials such as titanium but can also be applied to high-melting-point metals such as steel, nickel, and titanium alloys. When conducting an FSP test, it is crucial to select appropriate tool geometry, rotational speed, tool speed, and workpiece thickness. Scanning electron microscopy (SEM) are commonly used to analyze the phase and morphology of samples containing Tricalcium Phosphate (TCP) and Titanium oxide nanoparticles (Ti-NPs) ceramic. The effect of hydrogenation on the mechanical properties of titanium alloys mixed with titanium during FSP is also investigated. The composition of TCP and Ti-NP is diffused onto the titanium substrate, and it is recommended that Ti-NP be joined to the substrate using FSP. Using FSP, the microstructure can be altered, and the properties of the new design tool can be studied using mechanical testing. The effects of hydrogenation during FSP of two non-homogeneous metals with Ti-NP can be investigated to evaluate the mechanical properties of the joint area and the worn tool mechanism. The pin tool mainly alters the direction of the secondary metal, allowing for an increase in the number and size of this secondary material attached to titanium. FSP parameters can be optimized based on the volume of the joined area using various established techniques.

Enhanced in vitro inhibition of MCF-7 and magnetic properties of cobalt incorporated calcium phosphate (HAp and β-TCP) nanoparticles

The Co2+ (0.1 M) incorporated hydroxyapatite (HAp) and beta tricalcium phosphate (β-TCP) nanoparticles were synthesized by the microwave assisted technique and sintering of HAp respectively. The samples were thermally treated at temperatures ranging from 200 to 1000°C. The partial substitutions of Co2+ at the Ca2+ site of HAp were confirmed from the slight shift (∼0.2°) in the (002) and (211) XRD peaks. The morphology of the nanoparticles was transformed from nanospheres to twinned particles on thermal treatment. In addition, the particle size of Co-600 was increased (from ∼50 nm to ∼100 nm) due to the recrystallization process. Further, the thermal treatment enhanced the crystallinity (41.15 to 90.16%), retentivity (Mr) and coercivity (Hc) of the nanoparticles. The cobalt incorporated HAp and β-TCP possessed paramagnetic property. The excellent bioactivity of β-TCP has been confirmed by the mineralization in simulated body fluid (SBF). Compared to HAp, β-TCP possessed better compatibility towards C2C12 cells on cobalt incorporation as evidenced by the in vitro cell viability. Moreover, both HAp and β-TCP have significantly inhibited the growth of MCF-7 on increasing the interaction time (72 h). Hence, the inhibition characteristics of Co2+ incorporated calcium phosphate (CaP) towards MCF-7 (without affecting the normal cells) demonstrate its competency as a potential material for cancer therapy over the already existing nanoparticles.

The Medical Study of Denture Base Resin Poly(Methyl Methacrylate) Reinforced by ZnO and TCP Nanoparticles

Denture base poly (methyl methacrylate (PMMA) resin is one of the most frequently used materials in denture base synthesis, but due to its poor mechanical properties, PMMA can be considered a medium for the attachment and growth of a variety of pathogenic bacteria and fungi, particularly due to PMMA's pores and rough surface. The porosity percentage and surface roughness of the PMMA resin sample was lowered in this study, which resulted in a reduction in microorganisms' surface adhesion by varying the ratios of additives such as zinc oxide (ZnO) and tri-calcium phosphate (TCP) nanoparticles with (1, 2, 3, and 10% wt percent) for each additive separately, and 3% as a combination of ZnO and TCP nanoparticles in an equal ratio. Additionally, mechanical features such as surface hardness are developed, which is a critical attribute for polishing and easy finishing, as well as offering great scratch resistance during denture base cleaning. These results indicated that when compared to the other groups, PMMA (ZnO wt. 1%) and TCP-wt. 1%) reinforced composite resins demonstrated the best optimum properties. Additionally, it was discovered that adding 1% of NPs improved the mechanical qualities, which benefited the biological properties by reducing bacterial adherence to the PMMA composite resin.

Preparation of poly(acrylic acid)/tricalcium phosphate nanoparticles scaffold: Characterization and releasing UC-MSCs derived exosomes for bone differentiation.

This study focused on preparing a multiscale three-dimensional (3D) scaffold using tricalcium phosphate nanoparticles (triCaPNPs) in a substrate of poly(acrylic acid) (PAA) polymer for controlled release of exosomes in bone tissue engineering. A scaffold was fabricated with a material mixture containing acrylic acid (AA) monomer, N,N'-methylenebisacrylamide (MBAA), ammonium persulfate (APS), sodium bicarbonate (SBC), and triCaPNPs called composite scaffold (PAA/triCaPNPs) via cross-linking and freeze-drying methods. The synthesis process was easy and without complex multi-steps. Through mimicking the hybrid (organic-inorganic) structure of the bone matrix, we here chose triCaPNPs for incorporation into the PAA polymer. After assessing the physicochemical properties of the scaffold, the interaction of the scaffold with human umbilical cord mesenchymal stem cells (UC-MSCs) such as attachment, proliferation, and differentiation to osteoblast cells was evaluated. In addition, we used DiI-labeled exosomes to verify the exosome entrapment and release from the scaffold. The polymerization reaction of 3D scaffold was successful. Based on results of physicochemical properties, the presence of nanoparticles in the composite scaffold enhanced the mechanical stiffness, boosted the porosity with a larger pore size range, and offered better hydrophilicity, all of which would contribute to greater cell penetration, proliferation, and then better bone differentiation. In addition, our results indicated that our scaffold could take up and release exosomes, where the exosomes released from it could significantly enhance the osteogenic commitment of UC-MSCs. The current research is the first study fabricating a multiscale scaffold using triCaPNPs in the substrate of PPA polymer using a cross-linker and freeze-drying process. This scaffold could mimic the nanoscale structure and chemical combination of native bone minerals. In addition, our results suggest that the PAA/triCaPNPs scaffold could be beneficial to achieve controlled exosome release for exosome-based therapy in bone tissue engineering.

Composite Hydrogels of Alkyl Functionalized Gellan Gum Derivative and Hydroxyapatite/Tricalcium Phosphate Nanoparticles as Injectable Scaffolds for bone Regeneration.

An alkyl functionalized gellan gum derivative is here used to produce hydrogels containing hydroxyapatite and tricalcium phosphate nanoparticles as injectable nanostructured scaffolds for bone regeneration. The amphiphilic nature of the polysaccharide derivative along with its thermotropic behavior and ionotropic crosslinking features make possible to produce injectable bone mimetic scaffolds that can be used to release viable cells and osteoinductive biomolecules. The influence of different nanoparticles concentration on the rheological and physicochemical properties of the injectable systems is studied. It is found that the presence of inorganic nanoparticles reinforces the 3D hydrated polymeric networks without influencing their injectability but improving the physicochemical properties of ionotropic crosslinked hydrogels produced with two different curing media. Preliminary cytocompatibility tests performed with murine preosteoblast cells revealed that gellan gum based hydrogels can safely encapsulate viable cells. Loading and release experiments for dexamethasone and stromal cell-derived factor-1 demonstrate the drug delivery features of the obtained injectable systems.

Incorporation of Hydroxyapatite and Calcium Triphosphate in the Epoxy Resin-Based Sealer

Abstract Objective This study aims to assess and compare the influence of hydroxyapatite (HA) and tricalcium phosphate (TCP) nanoparticles on a commercially available epoxy resinbased sealer, focusing on porosity and push-out bond strength. Materials and Methods This work was classified into a control group and two experimental groups. In each experimental group, the sealer was mixed with 2.5 wt.% of HA and TCP nanoparticles. Thirty extracted single-rooted teeth were utilized. After sectioning the crowns, the remaining roots of 15 teeth were used, up to 40 to 0.06, using a K3 rotary system. Smear layers were removed with 3 mL of 17% EDTA applied for 60 seconds. Then, the canals were irrigated with 3 mL of 2.25% NaOCl and 5 mL of distilled water. The strength of push-out bonds was tested via an Instron universal testing machine on a 2 mm section acquired from obturated canals. Data were assessed using one-way ANOVA and post hoc tests at a significance level of p &lt; 0.05. Results A nonsignificant difference (p &gt; 0.05) was evident when the three groups were crosschecked in terms of void volume and bond strength. Micro-CT evaluations revealed the lowest volume of voids to be 0.1152 mm3 (2.69%) for the HA group compared with the control group 0.1818 mm3 (3.9%) and the TCP group 0.2194 mm3 (4.33%). Mean bond strength values were 4.18 ± 1.77 MPa for group 1 (control), 4.19 ± 1.54 MPa for group 2 (HA 2.5%) and 3.76 ± 1.95 MPa for group 3 (TCP 2.5%). Groups 1 and 3 showed both cohesive and a mixed type of failure, while group 2 showed adhesive and a mixed type of bond failure. Conclusion Within the limitations of the study, incorporation of 2.5 wt% HA and TCP nanoparticles into AH Plus did not significantly affect the percentage volume of voids and the bond strength negatively.

Novel injectable and self-setting composite materials for bone defect repair

Although calcium sulfate bone cements possess favorable characteristics of excellent clinical handling and osteoconductivity, the rapid degradation and poor bioactivity limit their wide applications. This study reports a type of novel injectable and self-setting composite materials for bone defect repair. Magnesium substituted tricalcium phosphate nanoparticles (43.8±9.0 nm) and calcium sulfate hemihydrate micro-size particles (5–21 µm) were mixed with a polyvinyl alcohol solution with optimal compositions to obtain the composite materials, which showed reasonable initial setting time (11.7–19.2 min) and suitable compressed strength (2.28–6.33 MPa). By utilizing magnesium powder as a porogen, macro-pores (>100 µm) together with micro-pores were created in the final product after setting. In addition, MC3T3E1 cells extended well and exhibited numerous lamellipodia and long filopodia when cultured with the composite materials, indicating that they had no cytotoxicity. The in vivo results indicated that the injectable composite materials could achieve bone defect repair, when implanted in beagle femoral condyle defects for 10 months. Our results show that the new injectable composite materials are biocompatible and biodegradable, which possess great potential for bone defect repair.

The Viability of Human Testis-Derived Cells on Human Serum Albumin-Based Scaffold as An Artificial Male Germ Cell Niche.

Azoospermia is one of the challenging disorders affecting couples who are afflicted with infertility. Human testisderived cells (hTCs) are suitable candidates for the initiation of in-vitro spermatogenesis for these types of patients. The current study aimed to assess the proliferation of hTCs through the cell culture on the three-dimensional (3D) porous scaffolds. Cells harvested from the testicular sperm extraction (TESE) samples of the azoospermic patients were cultured on the 3D porous scaffolds containing human serum albumin (HSA)/tri calcium phosphate nanoparticles (TCP NPs) for two weeks. The proliferation/viability of the cells was assessed using the MTT assay, along with H&E histological staining method. The MTT assay showed that hTCs could stay alive on this scaffold with 50 and 66.66% viability after 7 and 14 days, respectively. Such viability was not significantly different when compared with cells grown on monolayer flask culture (P>0.05). Therefore, 3D HSA/TCP NPs scaffolds could be used for the reconstitu- tion of the artificial human somatic testicular niche for future applications in regenerative medicine for male infertility.

Hydrothermal synthesis of Mg-substituted tricalcium phosphate nanocrystals

Abstract

The efficacy of Copper Oxide, Tri-calcium Phosphate and Silicon Dioxide Nanoparticles in Controlling Black Scurf Disease of Potato

Black scurf disease caused by Rhizoctonia solani, was a serious and common disease on potato worldwide. Effect of nanoparticles forms of Tri–calcium phosphate, copper oxide and silicon dioxide nanoparticle was evaluated at five concentrations to control black scurf disease in vitro and in vivo. Examination of treated R. solani with different tested nanoparticle was done using Transmission Electron Microscope (TEM). Activities of defense related enzyme were determined in artificially inoculated potato plants and tubers (cv. Spunta) with R solani. Physical characteristics of tested nanoparticles exhibit that all of them are spherical in shape and varied in their sizes. Also, all tested nanoparticles exhibited inhibitory effect of R. solani. Non-copper oxide was the most effective one (56.42 %) in suppressing the mycelial growth of R. solani at concentration 250 µl/L. The TEM examination of treated R. solani with different nanoparticles exhibit that instability they caused in cell wall thickness, abnormalities of nucleus and disappearance of nuclear membrane. All tested nanoparticles reduced of incidence and severity% of black scurf disease at concentrations 150 and 200 µl/L. during 2014 and 2015 seasons. Results cleared also that treating potato plants with different tested nanoparticles affected positively the activities of poly phenoloxidase, peroxidase, catalase and chitinase enzymes in leaves and tubers.

Future of Spermatogonial Stem Cell Culture: Application of Nanofiber Scaffolds.

Spermatogonial stem cells (SSCs) are unique in mammals because they can transmit genetic information from generation to generation and it is of significant importance. In testes, Sertoli cells, peritubular myoid cells, Leydig cells and other interstitial cells contribute to the spermatogonial stem cell "niche". So, creation of niche in an in vitro condition that mimics the in vivo environment is essential to maintain functional characteristic of SSCs. In this review, we describe the impact of nanofiber scaffolds on the culture of SSCs derived from human-to-mouse. Nanofiber Matrices mimic the architecture and size scale of the natural extracellular matrix (ECM). The scaffold provides more three-dimensional (3D), biomimicking and topographical signals to the cells and results in a more physiologically relevant cellular phenotype. Several investigators use different nanofiber scaffold-like carbon nanotubes (CNTs) scaffold, poly-L-lactic acid (PLLA) nanofiber scaffold, 3D soft agar culture system, human serum albumin (HSA)/tri calcium phosphate nanoparticles (TCPNPs) and electrospun polyamide nanofiber for proliferation and maintenance of self-renewal activity of the SSCs. Application of nanofiber scaffolds for in vitro culture of the SSCs may produce spermatogonial stem cells that can be used in regenerative medicine, tissue engineering, assisted reproductive technology and in the treatment of infertility in pre-pubertal cancer patients.

The effect of magnesium alloy wires and tricalcium phosphate particles on apatite mineralization on polylactide-based composites

Biodegradable magnesium alloy wires and tricalcium phosphate nanoparticles were used as a modifying phase of novel polylactide based composite system. The aim of this study was to determine the effect of those two components on apatite mineralization and thus potential bioactivity of the composite material. Studied samples were incubated in Simulated Body Fluid and Phosphate Buffer Saline to simulate biological environment. It was revealed that coexisting modification of polymer matrix with Mg and TCP resulted in formation of apatites with magnesium substitutions that have superior biological properties. Results of the study confirmed that chemical composition of the incubation medium affects composition of the precipitates. Designed multiphase composite can be potentially used for bioactive bone implants.

Extracellular Calcium Modulates Chondrogenic and Osteogenic Differentiation of Human Adipose-Derived Stem Cells: A Novel Approach for Osteochondral Tissue Engineering Using a Single Stem Cell Source.

We have previously shown that elevating extracellular calcium from a concentration of 1.8 to 8 mM accelerates and increases human adipose-derived stem cell (hASC) osteogenic differentiation and cell-mediated calcium accretion, even in the absence of any other soluble osteogenic factors in the culture medium. However, the effects of elevated calcium on hASC chondrogenic differentiation have not been reported. The goal of this study was to determine the effects of varied calcium concentrations on chondrogenic differentiation of hASC. We hypothesized that exposure to elevated extracellular calcium (8 mM concentration) in a chondrogenic differentiation medium (CDM) would inhibit chondrogenesis of hASC when compared to basal calcium (1.8 mM concentration) controls. We further hypothesized that a full osteochondral construct could be engineered by controlling local release of calcium to induce site-specific chondrogenesis and osteogenesis using only hASC as the cell source. Human ASC was cultured as micromass pellets in CDM containing transforming growth factor-β1 and bone morphogenetic protein 6 for 28 days at extracellular calcium concentrations of either 1.8 mM (basal) or 8 mM (elevated). Our findings indicated that elevated calcium induced osteogenesis and inhibited chondrogenesis in hASC. Based on these findings, stacked polylactic acid nanofibrous scaffolds containing either 0% or 20% tricalcium phosphate (TCP) nanoparticles were electrospun and tested for site-specific chondrogenesis and osteogenesis. Histological assays confirmed that human ASC differentiated locally to generate calcified tissue in layers containing 20% TCP, and cartilage in the layers with no TCP when cultured in CDM. This is the first study to report the effects of elevated calcium on chondrogenic differentiation of hASC, and to develop osteochondral nanofibrous scaffolds using a single cell source and controlled calcium release to induce site-specific differentiation. This approach holds great promise for osteochondral tissue engineering using a single cell source (hASC) and single scaffold.

Calcium and phosphate release from resin-based materials containing different calcium orthophosphate nanoparticles.

The study compared ion release from resin-based materials containing calcium orthophosphates. Amorphous calcium phosphate (ACP), dicalcium phosphate anhydrous (DCPA), dicalcium phosphate dihydrate (DCPD), and tricalcium phosphate (β-TCP) nanoparticles were characterized by X-ray diffraction (XRD), transmission electron microscopy (TEM), dynamic light scattering (DLS), and surface area (nitrogen adsorption isotherms, BET method). Nanoparticles were added to a dimethacrylate-based resin and materials were tested for degree of conversion (DC) and calcium/phosphate release up to 28 days under pH 5.5 and 7.0. Data were analyzed by ANOVA/Tukey test (alpha: 0.05).The crystallinity of DCPA, DCPD, and β-TCP were confirmed, as well as the ACP amorphous nature. DCPD and β-TCP presented larger agglomerates than DCPA and ACP. The surface area of ACP was 5-11 times higher than those of the other nanoparticles. Materials showed similar DC. The material containing ACP released significantly more ions than the others, which released similar amounts of calcium and, in most cases, phosphate. Ion release was not affected by pH. Calcium release decreased between 7 and 21 days, while phosphate levels remained constant after 14 days. In conclusion, ACP higher ion release can be ascribed to its high surface area. DCPA, DCPD, and β-TCP had similar performances as ion-releasing fillers.

Surface modification of tri-calcium phosphate nanoparticles by DOPE and/or anti-E6 antibody to enhance uptake of antisense of E6 mRNA

The main aim of this study was to evaluate the uptake of E6 mRNA antisense into cervical cancer cells, induced by human papilloma virus (HPV). In this study, the carrier of the antisense was tri-calcium phosphate nanoparticles (TCP NPs) conjugated with dioleoyl phosphatidyl ethanolamine (DOPE) and/or anti-E6 antibody. At first, TCP NPs were synthesized, coated with carboxy-polyethylene glycol, and then conjugated with anti-E6 antibody and/or DOPE by carbodiimide cross-linker. Then, a single stranded DNA, which was complementary (antisense) of E6 mRNA, was attached to each one. Finally, the uptake of conjugated and unconjugated TCP NPs into HelaS3 cells was separately evaluated by Fourier transform infrared spectroscopy, optical microscopy, and fluorescent microscopy. Also, the cytotoxicity of these carriers was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay. Overall, 4 types of TCP NPs were used in this study, including 1) TCP NPs conjugated with DOPE (TCP NPs/DOPE), 2) TCP NPs conjugated with DOPE and antibody (TCP NPs/DOPE/Anti-E6 Ab), 3) TCP NPs conjugated with antibody (TCP NPs/Anti-E6 Ab), and 4) TCP NPs which not conjugated with DOPE and antibody (unconjugated TCP NPs). Uptake tests showed that although all types of TCP NPs could transfer antisense of E6 mRNA into HelaS3 cells, TCP NPs/DOPE and TCP NPs/DOPE/Anti-E6 Ab had more uptake than TCP NPs/Anti-E6 Ab and unconjugated TCP NPs. Moreover, MTT assay showed that TCP NPs/DOPE was more toxic than TCP NPs/DOPE/Anti-E6 Ab, TCP NPs/Anti-E6 Ab, and unconjugated TCP NPs. It can be concluded that TCP NPs/DOPE/Anti-E6 Ab is a good choice for oligonucleotide delivery, because of higher uptake and less toxicity, compared with other formulations.
