Advancing biodegradation of petroleum contaminants by indigenous microbial consortia through assembly strategy innovations

Our demand for petroleum has challenged Earth's ecosystems, causing extensive water and soil pollution. Petroleum contamination treatment technology has relied on a range of biochemical processes to treat hydrocarbon-contaminated wastes, of which microbial degradation is one of the most economical and ecological. Engineered microbialconsortia can solve complex and difficult-to-degrade contaminants. However, the functional stability of consortia is challenged during long-term co-cultivation. Here, we introduced a novel strategy to design a stable consortium for degrading hydrocarbon pollutants, developed from ecological coexistence theory and trait-based method. Specifically, the degradation rate of the consortium constructed using the five strains was 31.54% higher than that of a single strain in a 6% (m/v) petroleum concentration after 16 days of co-culture. Besides, molecular characterization of the composition and transformation of petroleum-based substances further reveals the functional stability of consortium and the degradation mechanism and pattern of hydrocarbons by the consortium. Two independent soil experiments showed that the treatment of oil-contaminated soil by consortia from the indigenous microbiota maximizes the removal of hydrocarbon contaminants. Collectively, this strategy is generally applicable, facilitates the design of engineered microbial consortia, and provides a foundation for solving environmental pollution.

Comprehensive treatment of oil-contaminated soils using CO2-Responsive O/W microemulsions

In exploration of petroleum resources, a considerable amount of soils are inevitably contaminated, leading to many long-term environmental impacts. However, the treatment of oil-contaminated soils to reduce the residual oil for safe disposal at low energy intensity is a challenging task. In this study, a CO2-responsive microemulsion was formulated to treat oil-contaminated soils effectively at ambient temperatures of low energy consumption. Such effective treatment reduced the oil level of the contaminated soils from ∼15 wt% to below the discharge limit of 1 wt%. Purging of CO2 through the spent microemulsions for 30 s caused a rapid dissociation of the superamphiphile in the microemulsions, leading to a complete phase separation with the cleaned soils settled to the bottom for safe disposal, followed by an aqueous phase in the middle for recycling and a value-added oil product on top. The rapid and deep cleaning of oil-contaminated soils was attributed to an ultra-low interfacial tension and high level solubilization of oily contaminants in the O/W microemulsions. Such approach leads to a zero-waste discharge operation of remediating oil-contaminated soils while recovering value-added oil. Compared with other treatment methods operated at high temperature and/or pressure conditions, treatment of oil-contaminated soils using CO2-responsive microemulsions is easier to reach the discharge limit of residual oil while operating at ambient temperatures without any expensive equipment or extreme working conditions, making this microemulsion method a promising candidate for commercial applications of treating oil-contaminated soils in the oil field.

Influence of oil pollution on the activity of hydrolytic enzymes of sod-podzolic soil against the background of the use of biological products

The article presents the results of studies of the effect of biological products based on oil-oxidizing bacteria on the enzymatic activity of soddy-podzolic soil contaminated with oil. Barley (Hordeum sativum L) and wheat (Triticum vulgare L.) were used as phytomeliorants. The effect of different concentrations of oil on the activity of enzymes such as invertase, phosphatase, urease is shown. It was found that the activity of the studied enzymes depended on the concentration of the pollutant and the duration of oil exposure to the soil environment. It is noted that the treatment of oil-contaminated soil with biological products contributes to the restoration of its biological activity.

In march 10 1991 more than 700 oil wells have been damage by Iraqi troops in Kuwait,71,428,571 tons of oil expelled on land and sea of Kuwait .low cost of cleanup contaminated Raduhtain oil field soil was implemented the authors introduced and used washing the contaminated soil by date seeds technique. This work deals with the treatment of oil-contaminated soil, by using different concentration of date seeds. Soil washing is one of the sustainable technologies at present to remove oil from soil. From lab test, it seems to be an excellent way to treat contaminated soil. Samples of soil and wastewater were taken and analyzed in order to evaluate TOC concentration and removal value. The original TOC values of control water samples in (day 1st, day 3rd and day 5th) are (319925, 319896, 319680, µg/g) respectively. The experiment with 50g date seeds in (day 1, day 3 and day 5) shows the result are (275285.7, 98714.29, 91642.8, µg/g) respectively. Whereas with 100 g date seed with 100g in (day1, day 3, day 5) the TOC concentration is (289428.57, 249642.9, 71714.29, µg/g) respectively.

Use of Silicon Dioxide Encapsulation Method for Restoration of Oil-Polluted Soils

Purpose. Approbation of the method of encapsulation of silicon dioxide to restore the biological value of oil-contaminated soil. Methods. The encapsulating solution was prepared using sodium silicate (7% w. / vol.) as the main component and a synthetic surfactant (sodium dodecyl sulfate). To restore the contaminated soil, a treatment solution ratio of 1:1, 1:2, 1:3 and 1:4 was used for sodium silicate and sodium dodecyl sulfate, respectively. Phytotoxicity of oil-contaminated soil was determined by biotesting aqueous extracts from the soil. Results. The most optimized for use from the studied ratios of substances is a solution consisting of 2 parts: sodium silicate and sodium dodecyl sulfate. The lowest phytotoxic effect (17%) was recorded at pH of 5 of the treated soil and the ratio of solution components 1:2 (sodium silicate / sodium dodecyl sulfate). In the experiments, 2 species of monocotyledons (oats, corn) and 2 species of dicotyledonous plants (lettuce, black radish) were used. Conclusions. The technology of encapsulation of silicon dioxide in the treatment of oil-contaminated soils with a solution of sodium silicate and sodium dodecyl sulfate is quite economically attractive. The material formed as a result of the encapsulation process dries, forming an amorphous silica material, within which, in our opinion, hydrocarbons and heavy metals accumulate, but further research is needed for such a statement.

D-optimal Design Application to Study Enhanced Biostimulation of Used Motor Oil Contaminated Soil

The indiscriminate disposal of fresh and used motor oil into the environment (land and water) is a common practice in Nigeria and this practice has contributed to its soil contamination and makes up a greater percentage of polluted ground in the world. Over the years, the contamination of auto-mechanic workshops by used oil and fresh oil has been left uncared for, Soil contaminated with fresh and used motor oil creates a serious effect on plant tissues, soil components, and its microorganisms, human and other animal health. Optimization of a biostimulation process used in the treatment of used motor oil contaminated soil has been carried out. D-optimal design of response surface methodology with one numerical factor and a categorical factor at two levels was employed to generate 10 experimental runs for the biostimulation system. The input variables of the system were palm bunch ash (a stimulant) and pollution level. The two levels of the latter factor considered were 5% and 10 % (w/w) pollution level, while the response of the system was oil and grease (O&G) removal. The biostimulation experiments were carried out according to the RSM design, and the experimental data obtained were analyzed using the quadratic model. The results of the analyses showed that the developed quadratic model was able to represent the system with both input factors affecting the system significantly since p-values for the model was found to be less than 0.05. The results also showed that O&G removal was affected by both the PBA and the pollution level. It was also observed from the numerical optimisation carried out that 100 % and 83.36 % O&G removal was attained at 5 % and 10 % pollution level respectively after ten weeks of the study. The findings of this work revealed that palm bunch ash (PBA) can be used as a stimulant for the bioremediation of used motor oil contaminated soil. Thus, its application in the treatment of oil-contaminated soil is highly recommended.

Physical and Biological Treatment of Oil-Contaminated Soil in a Baffled Roller Bioreactor

ABSTRACT Physical and biological removal of diesel oil from contaminated soil was studied in a baffled roller bioreactor. Initially, the effects of four factors (soil loading, temperature, pH, and surfactant) on physical removal of diesel oil were investigated. Only the presence of a surfactant (sodium dodecyl sulfate [SDS]) demonstrated a significant effect on diesel oil removal. Diesel oil removal efficiency was increased from 32.0% to 63.9% in the presence of 100 mg/L SDS. Using a microbial culture enriched from contaminated soil, biological treatment of diesel oil polluted soil under different soil loadings (15% to 50%), different diesel oil concentrations (1 to 50 g/L), and different types of soil (sand, silt, and clay) was then investigated in the baffled roller bioreactor. Biodegradation consisted of both fast and slow stages for degradation of light and heavy compounds, respectively. All biodegradation experiments demonstrated significant decreases in diesel oil concentrations (88.3% in 14 days for initial diesel oil concentrations of 1000 mg/L and a wide range of soil loadings). The presence of silty or sandy soils enhanced the biodegradation rate compared to the control bioreactor (without soil). The sandy soil loading had no effect on the biodegradation results. Using the enriched culture, the baffled roller bioreactor was able to biodegrade high diesel concentrations (up to 50 g/L) with biodegradation rates of 112.2 and 39.3 mg/L· h during fast and slow stages, respectively.
