Two-dimensional persistent luminescence “optical battery” for autophagy inhibition-augmented photodynamic tumor nanotherapy

Aiming at solving the critical issue of traditional photodynamic therapy (PDT) that requires long-lasting external light irradiation, in this work, we develop a distinct irradiation-free PDT nanosystem based on the rational combination of two-dimensional (2D) CaAl2O4:Eu,Nd persistent luminescence nanosheets (CAO PLNSs) and photosensitizer verteporfin with mitochondrial-targeting function. 2D CAO PLNSs are served as internal “optical battery” for the continuous output of cytotoxic singlet oxygen (1O2). Notably, in addition to triggering cancer-cell apoptosis, the characteristic of 1O2-induced cell autophagy changes from pro-survival to pro-death due to the blockage downstream of autophagy flow originating from lysosomal de-functionalization. The 1O2 generated by 2D PDT nanosystem with mitochondrial-targeting feature activates mitophagy-autophagy dual self-protective pathways, and the subsequent excessive production of autophagosome and mitophagosome causes the formation of overloaded lysosome. Additionally, the strong coordination ability of Nd3+ ions with phosphates disrupts the lysosomal phosphoprotein membrane structure. This 2D PDT nanosystem-mediated autophagy inhibition would amplify apoptosis-dominant therapeutic efficacy. Both in vitro and in vivo assay results positively affirm the antineoplastic effect of autophagy inhibition-sensitized PDT. This work emphasizes the considerable role of 2D persistent luminescence PDT nanoplatform-induced pro-death autophagy in cancer treatment.

Semiconductor Quantum Dots (CdX, X = S, Te, Se) Modify Titanium Dioxide Nanoparticles for Photodynamic Inactivation of Leukemia HL60 Cancer Cells

Titanium dioxide nanoparticles (TiO2-NPs) are highly efficient photosensitizers in traditional photodynamic therapy (PDT). The particle size of TiO2-NPs is small, only about 20 nm. However, the demands of ultraviolet light (UV) excitation feature shallow tissue penetration depth and may lead to severe tissue photon damage. Thus, in this research, TiO2-NPs are modified with semiconductor quantum dots (QDs) CdX (X = S, Te, Se) in various methods, such as ultrasonic, hydrothermal, sol-gel, aqueous phase, and hydrolysis precipitation. The transmission electron microscopy (TEM) images show that the size of CdSe-TiO2 is ranging from 6 to 14 nm. The ultraviolet-visible (UV-Vis) spectrum demonstrates that the CdX (X = S, Te, Se) modification can successfully extend the absorption range of TiO2-NPs into a different visible light region. CdSe QDs have the narrowest band gap compared with CdX (X = S, Te, Se) QDs. Visible light-activated CdSe-TiO2 nanocomposite shows the highest PDT inactivation efficiency toward HL60 cells compared with CdX-TiO2. The photogenerated carrier separation efficiency of CdSe-TiO2 nanocomposite is the highest shown in a fluorescence spectrum (FS). Furthermore, when conjugated with folic acid (FA), the prepared FA-CdX-TiO2 (X = S, Se) exhibits excellent cancer-targeting ability during PDT treatment. Optimum PDT efficiency of FA-CdSe-TiO2 indicates that photocatalytic and targeting ability is much higher than pure TiO2 and CdSe-TiO2. Our results provided a detailed investigation on the PDT performance of CdX (X = S, Te, Se) modified TiO2 and may act as a guide for further design of highly targeted performance visible-light response TiO2-NPs.

Nanoparticle-Mediated Delivery Systems in Photodynamic Therapy of Colorectal Cancer.

Colorectal cancer (CRC) involving a malignant tumour remains one of the greatest contributing causes of fatal mortality and has become the third globally ranked malignancy in terms of cancer-associated deaths. Conventional CRC treatment approaches such as surgery, radiation, and chemotherapy are the most utilized approaches to treat this disease. However, they are limited by low selectivity and systemic toxicity, so they cannot completely eradicate this disease. Photodynamic therapy (PDT) is an emerging therapeutic modality that exerts selective cytotoxicity to cancerous cells through the activation of photosensitizers (PSs) under light irradiation to produce cytotoxic reactive oxygen species (ROS), which then cause cancer cell death. Cumulative research findings have highlighted the significant role of traditional PDT in CRC treatment; however, the therapeutic efficacy of the classical PDT strategy is restricted due to skin photosensitivity, poor cancerous tissue specificity, and limited penetration of light. The application of nanoparticles in PDT can mitigate some of these shortcomings and enhance the targeting ability of PS in order to effectively use PDT against CRC as well as to reduce systemic side effects. Although 2D culture models are widely used in cancer research, they have some limitations. Therefore, 3D models in CRC PDT, particularly multicellular tumour spheroids (MCTS), have attracted researchers. This review summarizes several photosensitizers that are currently used in CRC PDT and gives an overview of recent advances in nanoparticle application for enhanced CRC PDT. In addition, the progress of 3D-model applications in CRC PDT is discussed.

Near infrared light activated upconversion nanoparticles (UCNP) based photodynamic therapy of prostate cancers: An in vitro study

Photodynamic therapy (PDT), has a potential to cure cancerous prostate tissue with minimal side effects. Traditional PDT, however, mostly utilized visible (VIS) light range with direct application of hydrophobic photosensitizers which may not be adequate in clinical practice for especially deep-seated cancer cells because of poor penetration of VIS wavelengths. Here, we report near infrared light (NIR) induced and dual photosensitizers (PS) encapsulated PDT strategy to reduce prostate cancer cells - PC3. The designed nanoplatform (MC540/ZnPc-UCNP@Au), in this study, include upconversion nanoparticles (UCNP) synthesis to convert NIR light into multiple VIS wavelengths, porous silica coating to upload dual photosensitizers (MC540/ZnPc), and gold (Au) functionalization to enhance PDT treatment. High chemical stabilization provided MC540/ZnPc-UCNP@Au show excellent biocompatibility, and efficient PDT treatment for prostate cancer cells. In fact, the fluorescence of the synthesized nanoplatforms, upon NIR light excitation, can produce considerable amount of ROS in 5 min, as it is well matched with the absorption of MC540, ZnPc and Au nanoparticles (np). In addition, the easy visualization of cellular internalized/adsorbed nanoplatforms reveal the in situ cell imaging possibility for diagnosis. Based on the evidence of the results, NIR light activated MC540/ZnPc-UCNP@Au may offer a PDT technique for the treatment of prostate cancer.

A Regioselectively Oxidized 2D Bi/BiOx Lateral Nano-Heterostructure for Hypoxic Photodynamic Therapy.

Optoelectronic science and 2D nanomaterial technologies are currently at the forefront of multidisciplinary research and have numerous applications in electronics and photonics. The unique energy and optically induced interfacial electron transfer in these nanomaterials, enabled by their relative band alignment characteristics, can provide important therapeutic modalities for healthcare. Given that nano-heterostructures can facilitate photoinduced electron-hole separation and enhance generation of reactive oxygen species (ROS), 2D nano-heterostructure-based photosensitizers can provide a major advancement in photodynamic therapy (PDT), to overcome the current limitations in hypoxic tumor microenvironments. Herein, a bismuthene/bismuth oxide (Bi/BiOx)-based lateral nano-heterostructure synthesized using a regioselective oxidation process is introduced, which, upon irradiation at 660nm, effectively generates 1 O2 under normoxia but produces cytotoxic •OH and H2 under hypoxia, which synergistically enhances PDT. Furthermore, this Bi/BiOx nano-heterostructure is biocompatible and biodegradable, and, with the surface molecular engineering used here, it improves tumor tissue penetration and increases cellular uptake during in vitro and in vivo experiments, yielding excellent oxygen-independent tumor ablation with 660nm irradiation, when compared with traditional PDT agents.

Soft X-Ray Stimulated Lanthanide@MOF Nanoprobe for Amplifying Deep Tissue Synergistic Photodynamic and Antitumor Immunotherapy.

Combining photodynamic therapy (PDT) and immunotherapy has shown profound impact for synergistic treatment of malignant tumors. However, the shallow penetration depth of the traditional visible light activated PDT, immunosuppressive tumor microenvironment (TME), and poor immunogenicity of deep-seated solid tumors have significantly impeded the therapeutic efficiency. Herein, a soft X-ray activated nanoprobe is rationally engineered via integrating porphyrin Zr-based metal-organic framework with lanthanide NaYF4 :Gd,Tb@NaYF4 scintillator nanoparticles (SNPs) by a new in situ growth strategy for synergistic PDT and immunotherapy of tumor. The nanoprobe possesses remarkably enhanced reactive oxygen species (ROS) generation triggered by soft X-ray via further covalently grafting rose bengal on the nanoprobe, even at tissue depths of 3cm. Moreover, the soft X-ray induced ROS can act as potential immunogenic cell death (ICD) trigger, subsequently leading to the activation of the adaptive antitumor immune-response. Significantly, the boosted ROS generation can further modulate the immunosuppressive TME. This work provides new strategy of designing antitumor nanoprobes for soft X-ray triggered deep-tissue PDT and immune response, breaking the depth barriers suffered by the traditional photoactivated PDT or ICD using visible and near infrared light.

Smart 131 I-Labeled Self-Illuminating Photosensitizers for Deep Tumor Therapy.

Upon intravenous injection, 131 I-sPS NPs showed great tumor inhibition capability in subcutaneous 4T1-tumor-bearing Balb/c mice and orthotopic VX2 liver tumor bearing rabbits. We believed 131 I-sPS NPs could expand the application of CR and provide an effective strategy for deep tumor theranostics.

The triplet formation originates from an effective excited‐state relaxation from the initially populated singlet (S1) to triplet (T1) states via an intermediate triplet (T2) state. The low reduction potential and ultralong lifetime of the T1 state facilitate the efficient generation of O2−. by inter‐molecular charge transfer to molecular oxygen. The energy gap of T1‐S0 is smaller than that between 3O2 and 1O2 thereby precluding the generation of singlet oxygen by the Type‐II process. The trimer exhibits superior PDT performance under the hypoxic environment.

A feasible molecular engineering for bright Π-conjugation free radical photosensitizers with aggregation-induced emission

Fluorescence imaging-guided tumor photodynamic therapy (PDT) has received increasing attention due to its higher resolution of fluorescence imaging and noninvasive treatment of PDT. Two problems of fluorescence aggregation-caused quenching (ACQ) and tumor microenvironment hypoxia are urgently resolved in traditional PDT with fluorescence imaging function, but there is still a lack of effective molecular engineering to design delicate photosensitizers (PSs) to overcome fluorescence ACQ and tumor hypoxia, simultaneously. Herein, we develop a feasible molecular engineering to design aggregation-induced emission (AIE)-active PSs with unique free radical (type I) reactive oxygen species (ROS) by researching molecular structure-property relationship, which possesses bright fluorescence at aggregation and low O2-dependent under the process of generating free radical ROS. The intersystem crossing (ISC) channel is activated when frequently enhance intramolecular charge transfer (ICT) effect in electron-rich AIEgens with typical heavy atoms, which ensure enough triplet energy generation to induce superoxide anion free radical (O2−·) generation. In vitro fluorescence imaging and photo triggering biotoxicity evaluation both revealed that new AIE-active photogenerator is a promising candidate for fluorescence imaging-guided PDT.

Combretastatin A4-combined photodynamic therapy for enhanced tumor therapeutic efficacy

The limitation of traditional photodynamic therapy (PDT) such as the inherent hypoxia, the light penetration and the photosensitizer’s activity make it difficult to achieve a satisfactory therapeutic effect. To overcome these restrictions and enhance the antitumor therapeutic effect, the combination of PDT with other means offers a promising strategy to improve the efficacy of PDT. Herein, biodegradable poly (L-glutamic acid) (PLG) was used to chemically conjugate hydrophobic photosensitizers (TPP-NH2) and vascular disrupting agents (CA4) to prepare polymer-drug conjugates PT and PC, respectively. Both the solubility of drug and the endocytosis ability of nanoparticles have been enhanced. When the tumor was treated by PC and PT simultaneously, the conjugated TPP-NH2 and CA4 would be released from the internalized PC and PT due to the degradation of PLG. Under laser irradiation, the released TPP-NH2 could mainly kill the marginal tumor cells by generating cytotoxic singlet oxygen, while the released CA4 caused necrosis in the central region of the tumor by blocking the blood vessels. In vivo and in vitro analysis showed that this combined administration strategy exhibited more satisfied anti-proliferative effect compared to monotherapy, providing an effective strategy to develop synergistic cancer therapy.

Supramolecular Assemblies for Photodynamic Therapy

In recent years, supramolecular systems for nano-medicine, and in particular for photodynamic therapy, have gained great attention for their uses as smart and engineered therapeutic agents. We proposed a collection of very recent articles on supramolecular complexes for photodynamic therapy based on different photosensitizers assembled with cyclodextrins, cucurbiturils, calixarenes, pillararenes, or involved in nanobox and tweezer structures, nanoparticles, aggregates and micelles, that are dynamic assemblies inspired to biological systems. Despite the advantages of traditional Photodynamic therapy (PDT), which is a non-invasive, reliable and highly selective clinical treatment for several pathological conditions, different drawbacks are still smothering the applicability of this clinical treatment. In this contest, a new supramolecular approach is emerging, in fact, the reversible formation of these supramolecular assemblies, combined with the possibility to modify their dimensions and shapes in the presence of a guest make them similar to biological macromolecules, such as proteins and enzymes. Furthermore, due to the relatively weak and dynamic nature of supramolecular assemblies, they can undergo assembly and disassembly very fast as well as responses to external stimuli, such as biological (e.g. enzyme activation), chemical (e.g. redox potential or pH), and physical (e.g. temperature, light or magnetic fields). Therefore, the responsiveness of these supramolecular assemblies represents a highly promising approach to obtain potentially personalized PDT.

Bioorthogonal Coordination Polymer Nanoparticles with Aggregation-Induced Emission for Deep Tumor-Penetrating Radio- and Radiodynamic Therapy.

Radiodynamic therapy (RDT), an emerging therapeutic approach for cancer treatment by employing ionizing irradiation to induce localized photodynamic therapy (PDT) can overcome the drawbacks of the limited penetration depth for traditional PDT and the unconcentrated energy in the tumor for traditional radiotherapy (RT). Taking advantage of aggregation-induced emission (AIE) photosensitizers with bright fluorescence and efficient singlet oxygen production in the aggregate state, Hf-AIE coordination polymer nanoparticles (CPNs), which show both strong RT and RDT effect under X-ray irradiation, are developed. Furthermore, to enhance the tumor accumulation and prolong the tumor retention of the CPNs, bioorthogonal click chemistry is applied in the system through coupling between dibenzocyclooctyne (DBCO)-modified CPNs (Hf-AIE-PEG-DBCO) (PEG: poly(ethylene glycol)) and azide groups on the cell membrane formed by metabolic glycoengineering. Thanks to the high penetration of X-ray irradiation, the bioorthogonal-assisted RT and RDT combination therapy realizes significant killing of cancer cells without showing noticeable biotoxicity after intravenous administration of CPNs.

Enhancement of tumor lethality of ROS in photodynamic therapy.

In the process of photodynamic therapy (PDT) treatment of tumors, reactive oxygen species (ROS) plays a key role in destroying tumor tissues. However, traditional PDT often has limited ROS killing capacity due to hypoxia in the tumor microenvironment (TME) or obstruction by the ROS defense system, resulting in poor efficacy. Therefore, enhancing the killing effect of ROS on tumors is the core of enhancing the anti‐tumor effect of PDT. In recent years, many studies have developed a series of strategies to enhance the ability of ROS to kill tumors in view of the limitations of the TME on PDT. This article summarizes the commonly used or innovative strategies in recent years, including not only frequently used methods for hypoxia in the TME but also innovative strategies to inhibit the ROS defense system.

Self-Delivery Photodynamic Nanoinhibitors for Tumor Targeted Therapy and Metastasis Inhibition.

Simultaneous inhibitions of primary tumor growth and distant metastasis are very critical for cancer patients to improve their survival and cure rates. Although photodynamic therapy (PDT) shows great potential for primary tumor treatment, it often exacerbates hypoxia with a reduced therapeutic efficacy and subsequently contributes to carcinoma progression and metastatic dissemination. To solve these issues, self-delivery photodynamic nanoinhibitors (PNI) are developed for tumor targeted therapy and metastasis inhibition. PNI are composed of a carbonic anhydrase inhibitor (CAi), a hydrophilic poly(ethylene glycol) (PEG) linker, and a hydrophobic photosensitizer protoporphyrin IX (PpIX). Such self-delivery design of PNI avoids the premature release and heterogeneous distribution of CAi and PpIX to enhance the availability and synergism. Briefly, the CAi-based nanoinhibitors improve the selectivity of CAi for specific recognition and inhibition of tumor-associated isoform carbonic anhydrase (CA) IX, which would not only facilitate the targeted drug delivery of PNI but also regulate the hypoxia-induced signaling cascade and PDT resistance. Benefiting from the CA IX inhibition and targeted PDT, PNI exhibit a robust inhibitory effect on primary tumor growth and distant metastasis. This targeted self-delivery strategy sheds light on the photosensitizer-based molecular design to overcome the defect of traditional PDT.

Oxygen Self-Sufficient Core-Shell Metal-Organic Framework-Based Smart Nanoplatform for Enhanced Synergistic Chemotherapy and Photodynamic Therapy.

The abnormal angiogenesis and insufficient oxygen supply in solid tumors lead to intratumoral hypoxia, which severely limits the efficacy of traditional photodynamic therapy (PDT). Here, a multifunctional nanoplatform (ZDZP@PP) based on a zeolitic imidazolate framework-67 (ZIF-67) core as a hydrogen peroxide catalyst, a zeolitic imidazolate framework-8 (ZIF-8) shell with a pH-responsive property, and a polydopamine-poly(ethylene glycol) (PDA-PEG) layer for improving the biocompatibility is fabricated for not only relieving tumor hypoxia but also enhancing the efficacy of combination chemo-photodynamic therapy. The chemotherapy drug doxorubicin (DOX) and photosensitizer protoporphyrin IX (PpIX) are encapsulated in different layers independently; thus, a unique two-stage stepwise release becomes possible. Moreover, the nanoplatform can effectively decompose hydrogen peroxide to produce oxygen and thus relieve tumor hypoxia, which further facilitates the production of cytotoxic reactive oxygen species (ROS) by PpIX under laser irradiation. Both in vitro and in vivo experimental results confirm that the combination chemo-photodynamic therapy with the ZDZP@PP nanoplatform can provide more effective cancer treatment than chemotherapy or PDT alone. Consequently, the oxygen self-sufficient multifunctional nanoplatform holds promising potential to overcome hypoxia and treat solid tumors in the future.

Light-assisted gadofullerene nanoparticles disrupt tumor vasculatures for potent melanoma treatment.

The traditional photodynamic therapy (PDT) using a photosensitizer and oxygen under light generates reactive oxygen species (ROS) to kill tumor cells. However, its treatment efficiency is limited by insufficient oxygen in tumor cells. Herein, β-alanine modified gadofullerene nanoparticles (GFNPs) were explored to disrupt tumor vasculatures assisted by light for potent melanoma treatment. As tumor vasculatures are oxygen-rich, the yields of photo-induced singlet oxygen (1O2) by GFNPs are not subjected to the hypoxemia of tumor tissues. Different from the small molecule photosensitizer Chlorin e6 (Ce6), GFNPs realize high-efficiency tumor vascular disruption under light observed by using the mice tumor vascular dorsal skin fold chamber (DSFC) model. The tumor vascular disruption efficiency of GFNPs is size-dependent, and the smallest one (hydration diameter of ca. 126 nm) is more efficient. Mechanistically, the high yields of photo-induced 1O2 by GFNPs can lead to the destruction of the tumor vascular endothelial adherent junction protein-VE cadherin and the decrease of tumor vascular endothelial cells-CD31 proteins, inducing rapid tumor necrosis. In conclusion, our work provides an insight into the design of well-sized nanoparticles to powerfully treat melanoma assisted by light, as well as greatly extending the applications of PDT for robust tumor therapy.

Nanoscintillator-Mediated X-Ray Induced Photodynamic Therapy for Deep-Seated Tumors: From Concept to Biomedical Applications.

Photodynamic therapy (PDT) has shown great effectiveness in oncotherapy but has not been implemented in broad clinical applications because the limited penetration depth of the light used has been unable to reach deep-seated tumors. However, X-rays have been widely used in the clinical field for imaging and radiation therapy due to their excellent tissue penetration depth. Recently, X-rays have been established as an ideal excitation source for PDT, which holds great promise for breaking the depth limitation of traditional PDT for treatment of deep-seated tumors. This review aims to provide an overview of nanoscintillator-mediated X-ray induced PDT (X-PDT) including the concept, the design considerations of nanosensitizers for X-PDT, the modelling of nanosensitizer energy deposition, the putative mechanism by which X-PDT kills cells, and the prospects of future directions. We attempt to summarize the main developments that have occurred over the past decades. Possibilities and challenges for the clinical translation of X-PDT are also discussed.

Biomimetic Platinum Nanozyme Immobilized on 2D Metal-Organic Frameworks for Mitochondrion-Targeting and Oxygen Self-Supply Photodynamic Therapy.

Photodynamic therapy (PDT) as a noninvasive therapy mode has attracted considerable attention in the field of oncotherapy. However, the PDT efficacy is restricted either by the tumor hypoxia environment or the inherent properties of photosensitizers (PSs) including bad water solution, photobleaching, and easy aggregation. Herein, we designed and synthesized a new two-dimensional (2D) metal-organic framework, Sm-tetrakis(4-carboxyphenyl)porphyrin (TCPP) nanosheets, by assembling transition metal ions (Sm3+) and PSs (TCPP), on which the catalase (CAT)-mimicking platinum nanozymes were then in situ grown for sufficient oxygen supply during PDT. The prepared Sm-TCPP with nanoplate morphology (∼100 nm in diameter) and ultrathin thickness (<10 nm) showed significantly enhanced 1O2 generation capacity due to the improved physicochemical properties and the enhanced intersystem crossing from heavy Sm nodes. More importantly, the CAT-mimicking Pt nanozyme on the Sm-TCPP nanosheets could effectively convert over-expressed H2O2 in the tumor microenvironment into O2 to relieve tumor hypoxia. Further, the triphenylphosphine (TPP) molecule was introduced to Sm-TCPP-Pt to develop a mitochondrion-targeting and O2 self-supply PDT system. The in vitro and in vivo experimental results based on the MCF-7 breast cancer model revealed that Sm-TCPP-Pt/TPP could relieve tumor hypoxia and the generated reactive oxygen species nearby intracellular mitochondria significantly induced cell apoptosis. This study offers an engineering strategy to integrate 2D PS-based metal-organic frameworks and nanozymes into a nanoplatform to surmount the pitfalls of traditional PDT.
