In Vitro and In Vivo Ruminal and Physiological Responses to Endophyte-Infected Tall Fescue

Relationships between alkaloid compounds in endophyte-infected tall fescue and ruminal metabolism were studied in two experiments. In the first experiment, different combinations of the pyrrolizidine alkaloids, N-formyl and N-acetyl loline, were incubated with ruminal fluid for 0, 24, or 48h. Rate of disappearance of N-formyl and N-acetyl loline increased over time. After 48h, disappearance of N-formyl loline and combinations of N-formyl and N-acetyl loline was greater than N-acetyl loline. Significant amounts of N-formyl and N-acetyl loline were metabolized and converted to loline. In the second experiment, abomasally cannulated sheep were fed increasing amounts of endophyte-infected feed to compare diet digestibility, alkaloid metabolism, and physiological responses. Total tract DM digestibility was greatest for the endophyte-free diet, as were ruminal and total tract ADF, ruminal NDF, and total tract CP digestibilities. N-Formyl and N-acetyl loline recoveries averaged 5% from abomasal contents and 0% in feces. Sixty-eight percent of the pyrrolizidine alkaloids recovered in abomasal contents had been metabolizable to loline. Ergot alkaloids administered in the diet were recovered (50 to 60%) in the abomasal contents, but recovery was only 5% in fecal collections. No significant differences occurred in the physiological parameters measured. Results indicate that response to endophyte-infected tall fescue may be influenced by ruminal metabolism.

Effects of urea and sodium bicarbonate supplementation of a high-fiber diet on nutrient digestion and ruminal characteristics of defaunated sheep.

Five sheep (average BW 48 kg) with ruminal, duodenal, and ileal cannulas were fed 63% roughage: 37% concentrate diets (CP = 14.5%) in a 5 x 5 Latin square design to study effects of urea and sodium bicarbonate supplementation on nutrient digestion and ruminal characteristics of defaunated sheep. Diets were fed twice daily (DMI = 1,076 g/d). Defaunation was accomplished with 25-ml doses of alkanate 3SL3/sheep daily for 3 d. Control sheep were faunated (Treatment 1) and fed soybean meal as the major N supplement. Remaining sheep were maintained defaunated and fed either the same diet as Treatment 1 (Treatment 2), Treatment 1 with urea replacing 30% of the soybean meal N (Treatment 3), or Treatment 1 with 2% sodium bicarbonate in the diet (Treatment 4). Treatment 5 was a combination of Treatments 3 and 4. Compared with the faunated control, defaunation decreased (P less than .05) total tract DM, OM, NDF, ADF, and CP digestibilities (71.5 vs 69.4, 73.8 vs 71.7, 64.6 vs 61.4, 58.7 vs 55.8, and 74.2 vs 70.6%, respectively) and average (2 to 12 h postfeeding) ruminal fluid ammonia (23.5 vs 13.7 mg/dl) and isobutyrate (.9 vs .7 mM) concentrations. However, defaunation increased (P less than .05) linoleic and linolenic acid flows (.58 vs .45 g C18:2/d; .17 vs .14 g C18:3/d) to and disappearance (.50 vs .39 g C18:2/d; .14 vs .11 g C18:3/d) from the small intestine. Urea supplementation increased (P less than .05) total tract DM (70.2 vs 68.6%) and OM (72.3 vs 71.0%) digestibilities of defaunated sheep but lowered (P less than .05) ruminal fluid isobutyrate concentration (.6 vs .8 mM). Sodium bicarbonate supplementation increased (P less than .05) ruminal fluid pH (6.4 vs 6.2), isobutyrate concentration (.75 vs .60 mM), total tract ADF digestibility (57.6 vs 54.2%), and ruminal NDF (41.6 vs 28.5%), ADF (36.6 vs 22.8%), and CP (-5.5 vs -26.8%) digestibilities in defaunated sheep. Dietary supplementation of urea or sodium bicarbonate increased nutrient digestion by defaunated sheep.

Digestion of soybean meal and canola meal protein and amino acids in the digestive tract of young ruminants.

Eight male Holstein calves (body weight 68 +/- 5 kg; age 75 +/- 6 d), each with a permanent re-entrant pancreatic cannula and T-type ileal and duodenal cannulas, were used in a crossover design with four animals per group to determine amino acid kinetics and digestibilities in the digestive tract of calves fed soybean meal (SBM) and canola meal (CM) protein. The SBM and CM diets were fed twice daily at a level of 900 g at each feeding time (air-dry basis). With the exception of methionine, crude protein and amino acid flows at the proximal duodenum, expressed as a percentage of intake, were not influenced by dietary protein source. Apparent ileal and total tract digestibilities of CP and amino acids were reduced (P less than .05) by feeding CM compared to SBM, but apparent ileal digestibility of methionine was not affected by dietary protein source. Except for methionine, net disappearance of all amino acids in the small intestine, relative to the amount fed, was higher for the SBM diet than for the CM diet. Net disappearance or synthesis of amino acids in the large intestine were not affected by dietary protein source. Similarly, dietary protein source did not affect (P greater than .05) the secretion of pancreatic juice or concentrations of protein, chymotrypsin and trypsin in pancreatic juice. Soybean meal protein has higher ileal and total gastrointestinal tract digestibility than CM protein for young, growing calves.

Intestinal acid flow, dry matter, starch and protein digestibility and amino acid absorption in beef cattle fed a high-concentrate diet with defluorinated rock phosphate, limestone or magnesium oxide.

Five Angus heifers (285 kg) with duodenal and ileal cannulas were used to study the effects of feeding mineral acid neutralizers on digesta and fecal pH, intestinal acid flow, DM, starch and CP digestion and amino acid absorption in cattle fed a high-concentrate diet (7.5 kg/d). The experimental design was a 5 x 5 latin square. Treatments included control diet alone or control diet with the addition of 1.60% defluorinated rock phosphate-medium (DRP-M, 77% greater than 150 mu but less than 1,180 mu), 1.60% defluorinated rock phosphate-coarse (DRP-C, 85% greater than 850 mu but less than 1,700 mu), 1.28% limestone (92% greater than 150 mu but less than 850 mu) or .50% MgO, (81% greater than 250 mu but less than 1,180 mu), as an as-fed basis. Ileal pH was increased (P less than .05) from 7.49 to 7.85 by MgO. Fecal pH was increased (P less than .05) in this order: MgO (7.18) greater than DRP-M (6.86) = limestone (6.86) = DRP-C (6.75) greater than control (6.54). Liquid flow to the duodenum was increased (P less than .05) an average of 19.7% by the mineral treatments. Duodenal acid concentrations (mEq/liter) were greatest (P less than .05) for DRP-M (65.4) and DRP-C (64.0) vs limestone (55.5) and MgO (54.4). Acid flow to the intestine was increased (P less than .05) to 4.02, 4.53 and 4.82 Eq/d for animals fed limestone, DRP-M and DRP-C, respectively, from 3.51 Eq/d for control animals. Total tract DM, starch and CP digestibilities were similar (P greater than .10) among treatments. Limestone and DRP-M increased (P less than .10) the partial digestibility of most amino acids by about 6.8%. Mineral treatments did not increase total tract digestion of starch or CP. However, cattle fed DRP-M and limestone may have used protein more efficiently.
