Identification of host antioxidant effectors as thioridazine targets: Impact on cardiomyocytes infection and Trypanosoma cruzi-induced acute myocarditis

Phenothiazines inhibit antioxidant enzymes in trypanosomatids. However, potential interferences with host cell antioxidant defenses are central concerns in using these drugs to treat Trypanosoma cruzi-induced infectious myocarditis. Thus, the interaction of thioridazine (TDZ) with T. cruzi and cardiomyocytes antioxidant enzymes, and its impact on cardiomyocytes and cardiac infection was investigated in vitro and in vivo. Cardiomyocytes and trypomastigotes in culture, and mice treated with TDZ and benznidazole (Bz, reference antiparasitic drug) were submitted to microstructural, biochemical and molecular analyses. TDZ was more cytotoxic and less selective against T. cruzi than Bz in vitro. TDZ-pretreated cardiomyocytes developed increased infection rate, reactive oxygen species (ROS) production, lipid and protein oxidation; similar catalase (CAT) and superoxide dismutase (SOD) activity, and reduced glutathione's (peroxidase - GPx, S-transferase – GST, and reductase – GR) activity than infected untreated cells. TDZ attenuated trypanothione reductase activity in T. cruzi, and protein antioxidant capacity in cardiomyocytes, making these cells more susceptible to H2O2-based oxidative challenge. In vivo, TDZ potentiated heart parasitism, total ROS production, myocarditis, lipid and protein oxidation; as well as reduced GPx, GR, and GST activities compared to untreated mice. Benznidazole decreased heart parasitism, total ROS production, heart inflammation, lipid and protein oxidation in T. cruzi-infected mice. Our findings indicate that TDZ simultaneously interact with enzymatic antioxidant targets in cardiomyocytes and T. cruzi, potentiating the infection by inducing antioxidant fragility and increasing cardiomyocytes and heart susceptibility to parasitism, inflammation and oxidative damage.

SB431542 partially inhibits high glucose-induced EMT by restoring mitochondrial homeostasis in RPE cells

BackgroundThe epithelial–mesenchymal transition (EMT) of retinal pigment epithelial (RPE) cells participated in the development of retinal fibrosis. SB431542 is a small molecule inhibitor with inhibitory effects on the ALK4, ALK5 and ALK7. Our study aimed to explore the effect of SB431542 on the EMT of RPE cells and to provide new ideas for the treatment of retinal fibrosis.MethodsWe performed fundus fluorescein angiography, optical coherence tomography and hematoxylin–eosin staining in vivo to observe the effect of SB431542 on choroidal neovascularization (CNV)-induced retinopathy. The proliferation, migration, cytoskeleton, adhesion, reactive oxygen species (ROS), mitochondrial morphology and membrane potential of RPE cells were observed in vitro through fluorescein diacetate staining, Cell Counting Kit-8 experiment, wound healing assay, phalloidin staining, immunofluorescence, MitoSOX, DCFH-DA, MitoTracker and JC-10 staining. Western blot, reverse transcription quantitative and immunofluorescence were used to detect the expression of EMT–related markers, pERK1/2, pGSK3β and β-catenin.ResultsSB431542 significantly alleviated retinopathy in the CNV model. The proliferation, migration and adhesion in RPE cells decreased to a certain extent in SB431542 treatment. SB431542 partially normalized the structure of RPE cells. The expression levels of E-cadherin increased, while the expression levels of laminin and N-cadherin decreased with SB431542 treatment. SB431542 reduced the production of total ROS, mitochondrial SOX and recovered the mitochondrial membrane potential to a certain degree. Enhancement of Human Neutrophil Bactericidal Activity through ROS Regulation

Antimicrobial agents are vital for treating infectious diseases, but their widespread use has led to a concerning rise in drug-resistant bacteria. The development and overuse of new antimicrobial agents have paradoxically given rise to even more drug-resistant strains. If current trends persist, drug-resistant infections may become the leading global cause of mortality by 2050. However, current measures such as restricting antimicrobial use and promoting appropriate usage have not provided a definitive solution. Therefore, exploring alternative treatment approaches is essential. In this study, our research was based on a novel strategy that shifts the focus from targeting specific bacterial species to leveraging the defense mechanisms of human neutrophils. Neutrophils play a critical role in innate immunity by engulfing bacteria and generating reactive oxygen species (ROS) in phagosomes. The primary ROS producer in neutrophils is the NADPH oxidase (NOX) complex centered on gp91 phox (NOX2), a membrane protein of the NOX family. However, excessive ROS are harmful, necessitating negative regulation. Neutrophils contain a negative regulator of reactive oxygen species (NRROS), which regulates ROS production by competing with p22 phox for binding to NOX2 when inflammation go into ending. NRROS knockout mice have shown increased ROS production and bactericidal activity during bacterial infection, resulting in significantly prolonged survival compared to wild-type mice. If this mechanism could be applied to human cells to enhance bactericidal activity, it would eliminate the need to target specific bacterial species and could revolutionize the treatment of infectious diseases. We used human induced pluripotent stem cells (iPSC) as a model to validate the NRROS knockout effect in human cells, overcoming challenges in isolating peripheral blood neutrophils due to their limited lifespan, poor in vitro culture tolerance, and genetic editing complexities. We successfully generated NRROS-heterozygous knockout (hetero-KO) and NRROS-homozygous knockout (homo-KO) iPSC clones using genome editing techniques. Additionally, using the PiggyBac system, we established doxycycline-inducible NRROS expression clones (homo-KO+Dox) based on homo-KO clones. These iPSC were differentiated into neutrophils and evaluated for function. The isolated neutrophil fraction exhibited characteristic segmented nuclei in all clones. We measured ROS production and examined changes induced by NRROS knockout after stimulation. Immediate superoxide production showed no difference between wild-type and NRROS-KO clones. However, after 2 hours of stimulation, the NRROS-KO clones exhibited significantly elevated hydrogen peroxide production, indicating increased total ROS production, possibly due to delayed NOX complex degradation. Furthermore, we assessed the bactericidal capacity of iPSC-derived neutrophils against multidrug-resistant bacteria, revealing significantly enhanced bactericidal activity in NRROS-KO clones compared to wild-type clones. In conclusion, NRROS knockout in human neutrophils enhances bactericidal capacity through heightened ROS production, offering promising prospects for innovative infectious disease treatments. If successfully harnessed, this innovative treatment modality could revolutionize infectious disease management, combating drug-resistant infections, and improving global health outcomes.

Tigecycline Suppresses the Virulence Factors of Multidrug-Resistant Acinetobacter baumannii Allowing Human Neutrophils to Act.

PurposeTo determine the ability of human neutrophils to kill multidrug-resistant Acinetobacter baumannii (MDRAB) in the presence of tigecycline (TGC).MethodsClinical isolates of MDRAB were cultured with human neutrophils and H2O2 in the presence of TGC. The numbers of viable bacteria, catalase activity, gene expression at the K locus of the MDRAB, reactive oxygen species (ROS) production, and granule exocytosis in human neutrophils were determined.ResultsThere was a time-dependent increase in the numbers of MDRAB after co-culturing with human neutrophils, whereas there was a significant decrease in the MDRAB numbers when co-cultured with both, human neutrophils and TGC for 6 h. The presence or absence of TGC did not affect total ROS production or the expression of CD11b, CD15, and CD63 on human neutrophils occurred when co-cultured with MDRAB. TGC significantly suppressed catalase activity and gene expression at the K locus of MDRAB, and significantly reduced the thickness of the capsule. Additionally, the bacterial viability of TGC-treated MDRAB cultured with H2O2 was lower than that without H2O2 after 6 h of culture.ConclusionTGC significantly suppressed the expression of catalase and the capsule in MDRAB without adverse effects on neutrophil function, allowing human neutrophils to kill MDRAB. TGC is an effective antibiotic for treating MDRAB infections.

Fructose Induces Pulmonary Fibrotic Phenotype Through Promoting Epithelial-Mesenchymal Transition Mediated by ROS-Activated Latent TGF-β1.

Fructose is a commonly used food additive and has many adverse effects on human health, but it is unclear whether fructose impacts pulmonary fibrosis. TGF-β1, a potent fibrotic inducer, is produced as latent complexes by various cells, including alveolar epithelial cells, macrophages, and fibroblasts, and must be activated by many factors such as reactive oxygen species (ROS). This study explored the impact of fructose on pulmonary fibrotic phenotype and epithelial-mesenchymal transition (EMT) using lung epithelial cells (A549 or BEAS-2B) and the underlying mechanisms. Fructose promoted the cell viability of lung epithelial cells, while N-Acetyl-l-cysteine (NAC) inhibited such. Co-treatment of fructose and latent TGF-β1 could induce the fibrosis phenotype and the epithelial-mesenchymal transition (EMT)-related protein expression, increasing lung epithelial cell migration and invasion. Mechanism analysis shows that fructose dose-dependently promoted the production of total and mitochondrial ROS in A549 cells, while NAC eliminated this promotion. Notably, post-administration with NAC or SB431542 (a potent TGF-β type I receptor inhibitor) inhibited fibrosis phenotype and EMT process of lung epithelial cells co-treated with fructose and latent TGF-β1. Finally, the fibrosis phenotype and EMT-related protein expression of lung epithelial cells were mediated by the ROS-activated latent TGF-β1/Smad3 signal. This study revealed that high fructose promoted the fibrotic phenotype of human lung epithelial cells by up-regulating oxidative stress, which enabled the latent form of TGF-β1 into activated TGF-β1, which provides help and reference for the diet adjustment of healthy people and patients with fibrosis.

The Effect of Pristine and Pegylated Graphene Oxide Nanosheets on the Functions of Human Neutrophils

Graphene oxide (GO) is very useful for biomedicine, due to its physicochemical properties; therefore, its interaction with cells of the immune system has beenextensively studied. Many studies have aimed toreduce the undesirable effects of GO through chemical modification, including through polyethylene glycol (PEG) coating. Neutrophils are the first to respond to foreign object invasion in the body. Their main functions are the uptake and destruction of foreign particles, including with the help of reactive oxygen species (ROS).Our study aimed to investigate theengulfment of unmodified graphene oxide (GO) and graphene oxide coated with polyethylene glycol (GO-PEG) by human neutrophils and the effect of nanosheets on the production of ROS.We used sheets of GO (Ossila, Great Britain, average plate size 1-5 μm) and GO-PEG (569 ± 14 nm, PEG coating≈ 20%) at concentrations of 12.5μg/mL, 25μg/mL, and 50 μg/mL. The uptake of nanosheets was assessed by flow cytometry, taking into account the level of background adhesion of nanoparticles. ROS production was evaluated by luminol-dependent chemiluminescence (LCL).It was found that GO (12.5μg/mL, 25μg/mL, and 50 μg/mL) was actively internalized by neutrophils, while the uptake of GO-PEG was not detected. GO and GO-PEG particles (25 μg/mLand 50 μg/mL) reduced the total production of ROS by human leukocytes.Thus, the modifying of GOnanosheets with PEG resulted in the abolishment of their active uptake by neutrophils but did not affect the GO inhibitory effect on their oxidative activity.

Intracellular Amplifiers of Reactive Oxygen Species Affecting Mitochondria as Radiosensitizers.

Simple SummaryProdrugs that increase the level of reactive oxygen species (ROS) specifically in cancer cells while not affecting normal cells can potentially act as radiosensitizers for side effect free radiotherapy (RT). However, previously known ROS-amplifying prodrugs were found to not beuseful for this purpose. Since functional mitochondria are necessary for RT-resistance, we assumed that the problem could be solved by using dual prodrugs as radiosensitizers, both targeting mitochondria and simultaneously inducing ROS. In this paper, we explored this possibility. In particular, we developed an N-alkylaminoferrocene-based prodrug (2c) effective at low μMolar concentrations. Upon conversion to its active form 2c_BA in aqueous solution, it is efficiently taken up by cancer cells. This leads to the decrease of their mitochondrial membrane potential and the amplification of both, intracellular mitochondrial and total ROS generation. We found that 2c_BA acts as an efficient radiosensitizer in human head and neck squamous carcinoma cells in vitro.Radiotherapy (RT) efficacy can be improved by using radiosensitizers, i.e., drugs enhancing the effect of ionizing radiation (IR). One of the side effects of RT includes damage of normal tissue in close proximity to the treated tumor. This problem can be solved by applying cancer specific radiosensitizers. N-Alkylaminoferrocene-based (NAAF) prodrugs produce reactive oxygen species (ROS) in cancer cells, but not in normal cells. Therefore, they can potentially act as cancer specific radiosensitizers. However, early NAAF prodrugs did not exhibit this property. Since functional mitochondria are important for RT resistance, we assumed that NAAF prodrugs affecting mitochondria in parallel with increasing intracellular ROS can potentially exhibit synergy with RT. We applied sequential Cu+-catalyzed alkyne-azide cycloadditions (CuAAC) to obtain a series of NAAF derivatives with the goal of improving anticancer efficacies over already existing compounds. One of the obtained prodrugs (2c) exhibited high anticancer activity with IC50 values in the range of 5–7.1 µM in human ovarian carcinoma, Burkitt’s lymphoma, pancreatic carcinoma and T-cell leukemia cells retained moderate water solubility and showed cancer specificity. 2c strongly affects mitochondria of cancer cells, leading to the amplification of mitochondrial and total ROS production and thus causing cell death via necrosis and apoptosis. We observed that 2c acts as a radiosensitizer in human head and neck squamous carcinoma cells. This is the first demonstration of a synergy between the radiotherapy and NAAF-based ROS amplifiers.

ARMC5 modifies cell redox state to regulate steroidogenesis and lipid metabolism in the adrenal cortex

ARMC5 is a putative tumor suppressor gene that is frequently mutated in primary bilateral macronodular adrenal hyperplasia (PBMAH), a rare cause of Cushing's syndrome. The function of ARMC5 is poorly known, aside the fact that it regulates cell apoptosis and adrenal steroidogenesis in by mechanisms still unknown. Tumor suppressor genes play an important role in oxidative stress. In this study we used as model the adrenocortical carcinoma cell line H295R. In order to investigate ARMC5 response to stress, cells were treated with Menadione and reactive oxygen species (ROS) production was measured by Cellrox assays with flow cytometry. Oxidative response genes and steroidogenic enzymes expression were investigated by qPCR, whereas p38 phosphorylation was investigated by western blotting. ARMC5 protein is differentially regulated in response to menadione-induced stress in H295R adrenocortical cells. Moreover, ARMC5 depletion increases total intracellular ROS production (P < 0.05) and causes an imbalanced transcription of pro and anti-oxidant genes leading to increased phosphorylation of p38 (P < 0.05). ROS production and p38 pathway activation alter steroidogenesis. These effects are partially reversed by the anti-oxidant compound N-acetylcysteine (NAC) or the p38 inhibitor (SB203580). Altogether, this study describes a new function of ARMC5 as regulator of the redox homeostasis in adrenocortical cells.

Ivermectin has New Application in Inhibiting Colorectal Cancer Cell Growth.

Colorectal cancer (CRC) is the third most common cancer worldwide and still lacks effective therapy. Ivermectin, an antiparasitic drug, has been shown to possess anti-inflammation, anti-virus, and antitumor properties. However, whether ivermectin affects CRC is still unclear. The objective of this study was to evaluate the influence of ivermectin on CRC using CRC cell lines SW480 and SW1116. We used CCK-8 assay to determine the cell viability, used an optical microscope to measure cell morphology, used Annexin V-FITC/7-AAD kit to determine cell apoptosis, used Caspase 3/7 Activity Apoptosis Assay Kit to evaluate Caspase 3/7 activity, used Western blot to determine apoptosis-associated protein expression, and used flow cytometry and fluorescence microscope to determine the reactive oxygen species (ROS) levels and cell cycle. The results demonstrated that ivermectin dose-dependently inhibited colorectal cancer SW480 and SW1116 cell growth, followed by promoting cell apoptosis and increasing Caspase-3/7 activity. Besides, ivermectin upregulated the expression of proapoptotic proteins Bax and cleaved PARP and downregulated antiapoptotic protein Bcl-2. Mechanism analysis showed that ivermectin promoted both total and mitochondrial ROS production in a dose-dependent manner, which could be eliminated by administering N-acetyl-l-cysteine (NAC) in CRC cells. Following NAC treatment, the inhibition of cell growth induced by ivermectin was reversed. Finally, ivermectin at low doses (2.5 and 5 µM) induced CRC cell arrest. Overall, ivermectin suppressed cell proliferation by promoting ROS-mediated mitochondrial apoptosis pathway and inducing S phase arrest in CRC cells, suggesting that ivermectin might be a new potential anticancer drug therapy for human colorectal cancer and other cancers.

Toxicity of Water- and Organic-Soluble Wood Tar Fractions from Biomass Burning in Lung Epithelial Cells.

Widespread smoke from wildfires and biomass burning contributes to air pollution and the deterioration of air quality and human health. A common and major emission of biomass burning, often found in collected smoke particles, is spherical wood tar particles, also known as “tar balls”. However, the toxicity of wood tar particles and the mechanisms that govern their health impacts and the impact of their complicated chemical matrix are not fully elucidated. To address these questions, we generated wood tar material from wood pyrolysis and isolated two main subfractions: water-soluble and organic-soluble fractions. The chemical characteristics as well as the cytotoxicity, oxidative damage, and DNA damage mechanisms were investigated after exposure of A549 and BEAS-2B lung epithelial cells to wood tar. Our results suggest that both wood tar subfractions reduce cell viability in exposed lung cells; however, these fractions have different modes of action that are related to their physicochemical properties. Exposure to the water-soluble wood tar fraction increased total reactive oxygen species production in the cells, decreased mitochondrial membrane potential (MMP), and induced oxidative damage and cell death, probably through apoptosis. Exposure to the organic-soluble fraction increased superoxide anion production, with a sharp decrease in MMP. DNA damage is a significant process that may explain the course of toxicity of the organic-soluble fraction. For both subfractions, exposure caused cell cycle alterations in the G2/M phase that were induced by upregulation of p21 and p16. Collectively, both subfractions of wood tar are toxic. The water-soluble fraction contains chemicals (such as phenolic compounds) that induce a strong oxidative stress response and penetrate living cells more easily. The organic-soluble fraction contained more polycyclic aromatic hydrocarbons (PAHs) and oxygenated PAHs and induced genotoxic processes, such as DNA damage.

Photobiomodulation of oral fibroblasts stimulated with periodontal pathogens

Photobiomodulation (PBM) utilises light energy to treat oral disease, periodontitis. However, there remains inconsistency in the reporting of treatment parameters and a lack of knowledge as to how PBM elicits its molecular effects in vitro. Therefore, this study aimed to establish the potential immunomodulatory effects of blue and near infra-red light irradiation on gingival fibroblasts (GFs), a key cell involved in the pathogenesis of periodontitis. GFs were seeded in 96-well plates in media + / − Escherichia coli lipopolysaccharide (LPS 1 μg/ml), or heat-killed Fusobacterium nucleatum (F. nucleatum, 100:1MOI) or Porphyromonas gingivalis (P. gingivalis, 500:1MOI). Cultures were incubated overnight and subsequently irradiated using a bespoke radiometrically calibrated LED array (400–830 nm, irradiance: 24 mW/cm2 dose: 5.76 J/cm2). Effects of PBM on mitochondrial activity (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and adenosine triphosphate (ATP) assays, total reactive oxygen species production (ROS assay) and pro-inflammatory/cytokine response (interleukin-8 (IL-8) and tumour growth factor-β1 (TGFβ1)) were assessed 24 h post-irradiation. Data were analysed using one-way ANOVA followed by the Tukey test. Irradiation of untreated (no inflammatory stimulus) cultures at 400 nm induced 15%, 27% and 13% increases in MTT, ROS and IL-8 levels, respectively (p < 0.05). Exposure with 450 nm light following application of P. gingivalis, F. nucleatum or LPS induced significant decreases in TGFβ1 secretion relative to their bacterially stimulated controls (p < 0.001). Following stimulation with P. gingivalis, 400 nm irradiation induced 14% increases in MTT, respectively, relative to bacteria-stimulated controls (p < 0.05). These findings could identify important irradiation parameters to enable management of the hyper-inflammatory response characteristic of periodontitis.

Mitochondria-Rich Fraction Isolated From Mesenchymal Stromal Cells Reduces Lung and Distal Organ Injury in Experimental Sepsis.

To ascertain whether systemic administration of mitochondria-rich fraction isolated from mesenchymal stromal cells would reduce lung, kidney, and liver injury in experimental sepsis. Animal study. Laboratory investigation. Sixty C57BL/6 male mice. Sepsis was induced by cecal ligation and puncture; sham-operated animals were used as control. At 24 hours after surgery, cecal ligation and puncture and Sham animals were further randomized to receive saline or mitochondria-rich fraction isolated from mesenchymal stromal cells (3 × 106) IV. At 48 hours, survival, peritoneal bacterial load, lung, kidney, and liver injury were analyzed. Furthermore, the effects of mitochondria on oxygen consumption rate and reactive oxygen species production of lung epithelial and endothelial cells were evaluated in vitro. In vitro exposure of lung epithelial and endothelial cells from cecal ligation and puncture animals to mitochondria-rich fraction isolated from mesenchymal stromal cells restored oxygen consumption rate and reduced total reactive oxygen species production. Infusion of exogenous mitochondria-rich fraction from mesenchymal stromal cells (mitotherapy) reduced peritoneal bacterial load, improved lung mechanics and histology, and decreased the expression of interleukin-1β, keratinocyte chemoattractant, indoleamine 2,3-dioxygenase-2, and programmed cell death protein 1 in lung tissue, while increasing keratinocyte growth factor expression and survival rate in cecal ligation and puncture-induced sepsis. Mitotherapy also reduced kidney and liver injury, plasma creatinine levels, and messenger RNA expressions of interleukin-18 in kidney, interleukin-6, indoleamine 2,3-dioxygenase-2, and programmed cell death protein 1 in liver, while increasing nuclear factor erythroid 2-related factor-2 and superoxide dismutase-2 in kidney and interleukin-10 in liver. Mitotherapy decreased lung, liver, and kidney injury and increased survival rate in cecal ligation and puncture-induced sepsis.

MITOCHONDRIA ISOLATED FROM MESENCHYMAL STROMAL CELLS REDUCE LUNGAND DISTAL ORGAN INJURY IN EXPERIMENTAL SEPSIS

<h3>Background</h3> Although mesenchymal stromal cells (MSC) administration has demonstrated beneficial effects in experimental sepsis, cell-free therapeutics based on biologically active products from MSCs have been proposed to circumvent inherent risks related to whole-cell transplantation. As mitochondria transfer has emerged as one of the key mechanisms by which MSCs induce therapeutic actions, we asked whether MSC-isolated mitochondria could reduce lung, kidney, and liver injury, thus improving survival in experimental sepsis. <h3>Methods</h3> Experimental sepsis was induced by cecal ligation and puncture (CLP), while sham-operated animals were used as control. Twenty-four hours after surgery, CLP and Sham animals were further randomized to receive saline or MSC-isolated mitochondria (MT, from 3 × 106 MSCs) intravenously. After additional 24 h, survival rate, peritoneal bacterial load, lung mechanics and histology, kidney and liver injury, kidney function, as well as expression levels of selected mediators in tissue homogenates were analyzed. Moreover, the uptake and effects mitochondria on oxygen consumption rate (OCR) and reactive oxygen species (ROS) production of lung epithelial and endothelial cells were evaluated in vitro. <h3>Results</h3> In vitro exposure of lung epithelial and endothelial cells from CLP animals to MSC-isolated mitochondria resulted in mitochondrial uptake by endothelial cells and restored OCR and reduced total ROS production. Systemic administration of MSC-isolated mitochondria led to reduction of bacterial load, static lung elastance, alveolar collapse, neutrophil count, collagen and elastic fiber content as well as expression of interleukin (IL)-1β, keratinocyte chemoattractant, indoleamine 2,3-dioxygenase (IDO)-2 and programmed cell death protein 1 (PD-1) in lung tissue, while increasing KGF levels and survival rate in CLP-induced sepsis. Mitochondria therapy also reduced kidney and liver injury, decreasing plasma creatinine levels, mRNA expression of IL-18 in kidney, IL-6, IDO-2 and PD-1 in liver and increasing mRNA levels of nuclear factor erythroid 2-related factor-2 and superoxide dismutase-2 in kidney and IL-10 in liver. <h3>Conclusions</h3> Mitochondria therapy enabled the recovery of host mitochondrial function and reduced inflammation and oxidative stress, thus decreasing lung, liver, and kidney injury and increasing survival rate in CLP-induced sepsis.

MiR-204 silencing reduces mitochondrial autophagy and ROS production in a murine AD model via the TRPML1-activated STAT3 pathway

Mitochondrial dysfunction is an early feature of Alzheimer’s disease (AD), whereby accumulation of damaged mitochondria in conjunction with impaired mitophagy contributes to neurodegeneration. Various non-transcribed microRNAs (miRNAs) are involved in this process. In the present study, we aimed to decipher the participation of miR-204 in a murine AD model. Primary hippocampal neurons were isolated from mice and treated with β-amyloid 1-42 (Aβ1-42) to establish a cell model of AD. Dichloro-dihydro-fluorescein diacetate and dihydrorhodamine 123 staining assays were performed to measure total reactive oxygen species (ROS) and mitochondrial ROS production in neurons, and MitoSOX staining was done to analyze mitochondrial ROS production in hippocampus. Furthermore, mitochondrial autophagy was observed in hippocampus from amyloid precursor protein/pesenilin-1 AD modeled mice, and their cognitive function was assessed by Morris water maze. Mitochondrial damage, ROS production, and mitochondrial autophagy were observed in AD cell model induced by Aβ1-42. In AD, signal transducer and activator of transcription 3 (STAT3) and transient receptor potential mucolipin-1 (TRPML1) expression was downregulated, although miR-204 expression was upregulated. TRPML1 overexpression, downregulation of miR-204, or STAT3 pathway activation reduced the Aβ1-42-induced mitochondrial damage, along with ROS production and mitochondrial autophagy in vivo and in vitro. Silencing of miR-204 could upregulate TRPML1 expression, thus suppressing ROS production and mitochondrial autophagy in AD through STAT3 pathway.

ВЛИЯНИЕ НАНОЧАСТИЦ НА ОСНОВЕ ОКСИДА ЖЕЛЕЗА, МОДИФИЦИРОВАННЫХ РАЗЛИЧНЫМИ ОБОЛОЧКАМИ, НА ГЕНЕРАЦИЮ АКТИВНЫХ ФОРМ КИСЛОРОДА СТИМУЛИРОВАННЫМИ КЛЕТКАМИ КРОВИ ЧЕЛОВЕКА В УСЛОВИЯХ IN VITRO

Among the variety of nanoparticles, those based on iron oxide are of particular interest due to their biological safety, magnetic properties, as well as the possibility to give them the required physico-chemical properties and to modifying their surface with biocompatible, bioactive materials, ligands and antibodies. An important aspect that determines the possibility and range of clinical applications of nanoparticles is their biocompatibility profile. Considering the mechanism mediated by reactive oxygen species (ROS) and involved in the implementation of the toxic effect of nanoparticles, special attention at the stage of their development for biomedical applications should be paid to assessing their safety in terms of the development of oxidative stress. In this regard, we studied the dose-dependent effect of magnetite nanoparticles (MNPs) modified with various shells (based on polylactide, polysaccharide, and albumin) on the generation of ROS in stimulated human blood cells, as well as on the dynamics of induced oxidative hemolysis of erythrocytes. It was found that MNPs with a shell of polylactide, albumin, and polysaccharide, in the range of all the used concentrations (0.1–2.0 mg/mL) and throughout the entire incubation period (0–180 min), do not affect the kinetics of the chemiluminescent response, while providing a unidirectional but differently pronounced decrease in the maximum intensity of induced chemiluminescence and total ROS production. All types of investigated nanoparticles in the range of concentrations from 1.0 to 2.0 mg/mL provide a dose-dependent enhancement of this effect. Under conditions of induced ROS generation, various MNP shells do not modify the effects of these nanoparticles and only regulate their intensity. MNPs with a polylactide shell have maximum effect.

Nanoengineering of a newly designed chlorin e6 derivative for amplified photodynamic therapy via regulating lactate metabolism.

Chlorin e6 (Ce6) is a widely utilized photosensitizer in photodynamic therapy (PDT) against tumor growth, but its hydrophobic feature and the hypoxia in the tumor microenvironment greatly compromise its therapeutic efficacy. To address the issues, here we designed a new Ce6 derivative (TCe6) by coupling Ce6 with amphiphilic d-α-tocopherol polyethylene glycol 1000 succinate (TPGS), endowing Ce6 with an excellent amphiphilic feature. In particular, the overall reactive oxygen species (ROS) generation by TCe6 was significantly enhanced because TPGS could interact with mitochondrial complex II to induce extra ROS production, amplifying the total ROS production under PDT. Inspired by the unique property of α-cyano-4-hydroxycinnamate (CHC) in regulating lactate metabolism to spare more intracellular oxygen for PDT, TCe6 was further co-assembled with CHC to construct TCe6/CHC nanoparticles (NPs) for addressing the insufficient oxygen issue in PDT. The as-prepared TCe6/CHC NPs not only increased the efficiency of cell internalization but also improved the solubility and stability of Ce6 and CHC. Thanks to the extra ROS production by the TPGS unit, the amphiphilic feature of TCe6 and the CHC-mediated hypoxia microenvironment, the TCe6/CHC NPs demonstrated excellent PDT against tumor growth. This work provided a versatile strategy to solve the current bottleneck in photosensitizer-based PDT, holding great promise for the design of advanced photodynamic nanoplatforms.

Baicalin ameliorates atherosclerosis by inhibiting NLRP3 inflammasome in apolipoprotein E-deficient mice.

Background:NLR family pyrin domain containing 3 (NLRP3) inflammasome has been implicated in the development of atherosclerosis and several studies have suggested that inhibiting NLRP3 inflammasome could be a potential therapeutic approach to treat atherosclerosis. Baicalin is a flavone glycoside with anti-inflammation, anti-oxidative activities. The inhibition of NLRP3 inflammasome activation by baicalin has also been described. Therefore, the effects of baicalin on NLRP3 inflammasome activation and atherosclerosis were evaluated in present study.Methods:We established the apolipoprotein E-deficient atherosclerosis mice model. After baicalin treatment, the IL-1, IL-18, and reactive oxygen species (ROS) production, and the plaque area was monitored. We also measured the NLRP3, ASC, caspase-1, ICAM-1, and VCAM-1 expression in atherosclerosis mice after baicalin treatment. We silenced NLRP3 by administration of lentivirus expressing NLRP3 shRNA to atherosclerosis mice and monitored the IL-1, IL-18, and ROS production, and NLRP3 inflammasome activation.Results:Baicalin remarkably inhibited the production of IL-1, IL-18, mitochondria ROS, total ROS, ICAM-1, and VCAM-1. Baicalin reduced the expression of NLRP3 inflammasome and suppressed its activation. Baicalin significantly reduced the plaque area. Silencing NLRP3 resulted in decreased production of IL-1, IL-18, mitochondria ROS, total ROS, ICAM-1, and VCAM-1, and inhibition of NLRP3 inflammasome activation.Conclusion:Baicalin ameliorated atherosclerosis by inhibiting NLRP3 inflammasome.

Oleic acid magnetic iron oxide nanoparticles improve iron uptake by the modification of NADH-HCF (III) oxidoreductase without affecting cellular viability

Iron (Fe) is an essential micronutrient that plays a vital role in cell proliferation and metabolism and is required in highest amounts among micronutrients. The uptake of Fe involves the reduction of Fe3+ to Fe2+ before uptake through the plasma membrane. Fe is also a Fenton active metal and is responsible for reactive oxygen species (ROS) in the presence of hydrogen peroxide. Fe uptake in animals is associated with digestive difficulties and in all organisms, with ROS generation. In the present study, we have used yeast Saccharomyces cerevisiae to test the delivery of Fe as Oleic Acid coated Magnetic Iron Oxide Nanoparticles (OA-MION) and investigated the accompanying changes in membrane phenomena and ROS levels. Our study strongly indicates that up to a reasonably high concentration of 20 μg/ml OA-MION do not alter cell viability. OA-MION treated yeast cells exhibit unaltered morphology indicating low or no interaction of Fe nanoparticles with cell-wall components, possibly due to oleic acid coating. OA-MION treatment stimulated transplasma membrane electron transport, total ROS production, and H+ extrusion measured as a rise in intracellular pH, in a strictly concentration-dependent manner. The higher ROS, however, does not impact cellular viability and could be due to higher metabolic rates. Overall, the study proposes OA-MION as a vector of choice for the delivery of iron into cells.

Effects of melatonin to arecoline-induced reactive oxygen species production and DNA damage in oral squamous cell carcinoma

Arecoline, the major alkaloid of areca nut, is known to induce reactive oxygen species (ROS) and DNA damage during oral cancer progression. This study aim to evaluate whether melatonin, an antioxidant, supported or repressed the arecoline-induced carcinogenesis phenotypes in oral squamous cell carcinoma (OSCC). The cytotoxicity of arecoline or melatonin treatment alone and their co-treatment in the OSCC cell line OEC-M1 were analyzed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The cell cycle, cell death, and total ROS production were analyzed using flow cytometer. The protein expression was determined using western blot analysis. The genotoxicity and mutation rate were determined using micronucleus assay and hypoxanthine phosphoribosyl transferase (HPRT) forward mutation assay, respectively, in CHO-K1 cells. The ataxia telangiectasia mutated (ATM) promoter activity and DNA repair ability were determined through reporter assay. The result showed that both the arecoline and melatonin induced ROS production and antioxidant enzymes expression. Melatonin treatment enhanced arecoline-induced ROS production, cytotoxicity, G2/M phase arrest, and cell apoptosis in OSCC cells. On the other hand, melatonin treatment activated DNA repair activity to reverse arecoline-induced DNA damage and mutation. These results indicated that melatonin is a potential chemopreventive agent for betel quid chewers to prevent OSCC initiation and progression.

Mitochondrial Alterations and Enhanced Human Leukocyte/Endothelial Cell Interactions in Type 1 Diabetes

Type 1 diabetes has been associated with oxidative stress. This study evaluates the rates of oxidative stress, mitochondrial function, leukocyte–endothelium interactions and adhesion molecules in type 1 diabetic patients. The study population consisted of 52 diabetic patients and 46 body-composition and age-matched controls. We assessed anthropometric and metabolic parameters, oxidative stress and mitochondrial function by evaluating reactive oxygen species (ROS) production, mitochondrial ROS production, mitochondrial membrane potential and superoxide dismutase (SOD) and catalase (CAT) expression in polymorphonuclear leukocytes from type 1 diabetic patients. In addition, we evaluated interactions between leukocytes and human umbilical vein endothelial cells (HUVEC), and serum expression of adhesion molecules (P-selectin, VCAM-1 and ICAM-1), proinflammatory cytokines (IL-6 and TNFα) and myeloperoxidase (MPO). HbA1C and glucose levels were higher in diabetic patients than in control subjects, as expected. Mitochondrial function was altered and leukocyte–endothelium interactions were enhanced in diabetic patients, which was evident in the increase in total and mitochondrial ROS production, higher mitochondrial membrane potential, enhanced leukocyte rolling and adhesion, and decreased rolling velocity. Furthermore, we observed an increase in levels of adhesion molecules P-selectin, VCAM-1, and ICAM-1 in these subjects. In addition, type 1 diabetic patients exhibited an increase in proinflammatory mediators TNFα and MPO, and a decreased expression of SOD. The enhancement of leukocyte–endothelium interactions and proinflammatory markers correlated with glucose and HbA1Clevels. Mitochondrial alteration, oxidative stress, and enhanced leukocyte–endothelium interactions are features of type 1 diabetes and may be related to cardiovascular implications.
