External aluminium supply regulates photosynthesis and carbon partitioning in the Al-accumulating tropical shrub Melastoma malabathricum.

Aluminium (Al) is toxic to most plants, but recent research has suggested that Al addition may stimulate growth and nutrient uptake in some species capable of accumulating high tissue Al concentrations. The physiological basis of this growth response is unknown, but it may be associated with processes linked to the regulation of carbon assimilation and partitioning by Al supply. To test alternative hypotheses for the physiological mechanism explaining this response, we examined the effects of increasing Al concentrations in the growth medium on tissue nutrient concentrations and carbon assimilation in two populations of the Al-accumulator Melastoma malabathricum. Compared to seedlings grown in a control nutrient solution containing no Al, mean rates of photosynthesis and respiration increased by 46% and 27%, respectively, total non-structural carbohydrate concentrations increased by 45%, and lignin concentration in roots decreased by 26% when seedlings were grown in a nutrient solution containing 2.0 mM Al. The concentrations of P, Ca and Mg in leaves and stems increased by 31%, 22%, and 26%, respectively, in response to an increase in nutrient solution Al concentration from 0 to 2.0 mM. Elemental concentrations in roots increased for P (114%), Mg (61%) and K (5%) in response to this increase in Al concentration in the nutrient solution. Plants derived from an inherently faster-growing population had a greater relative increase in final dry mass, net photosynthetic and respiration rates and total non-structural carbohydrate concentrations in response to higher external Al supply. We conclude that growth stimulation by Al supply is associated with increases in photosynthetic and respiration rates and enhanced production of non-structural carbohydrates that are differentially allocated to roots, as well as stimulation of nutrient uptake. These responses suggest that internal carbon assimilation is up-regulated to provide the necessary resources of non-structural carbohydrates for uptake, transport and storage of Al in Melastoma malabathricum. This physiological mechanism has only been recorded previously in one other plant species, Camellia sinensis, which last shared a common ancestor with M. malabathricum more than 120 million years ago.

Integrated small RNA, transcriptome and physiological approaches provide insight into Taxodium hybrid 'Zhongshanshan' roots in acclimation to prolonged flooding.

Although Taxodium hybrid 'Zhongshanshan' 406 (Taxodium mucronatum Tenore×Taxodium distichum; Taxodium 406) is an extremely flooding-tolerant woody plant, the physiological and molecular mechanisms underlying acclimation of its roots to long-term flooding remain largely unknown. Thus, we exposed saplings of Taxodium 406 to either non-flooding (control) or flooding for 2months. Flooding resulted in reduced root biomass, which is in line with lower concentrations of citrate, α-ketoglutaric acid, fumaric acid, malic acid and adenosine triphosphate (ATP) in Taxodium 406 roots. Flooding led to elevated activities of pyruvate decarboxylase, alcohol dehydrogenase and lactate dehydrogenase, which is consistent with higher lactate concentration in the roots of Taxodium 406. Flooding brought about stimulated activities of superoxide dismutase and catalase and elevated reduced glutathione (GSH) concentration and GSH/oxidized glutathione, which is in agreement with reduced concentrations of O2- and H2O2 in Taxodium 406 roots. The levels of starch, soluble protein, indole-3-acetic acid, gibberellin A4 and jasmonate were decreased, whereas the concentrations of glucose, total non-structural carbohydrates, most amino acids and 1-aminocyclopropane-1-carboxylate (ACC) were improved in the roots of flooding-treated Taxodium 406. Underlying these changes in growth and physiological characteristics, 12,420 mRNAs and 42 miRNAs were significantly differentially expressed, and 886 miRNA-mRNA pairs were identified in the roots of flooding-exposed Taxodium 406. For instance, 1-aminocyclopropane-1-carboxylate synthase 8 (ACS8) was a target of Th-miR162-3p and 1-aminocyclopropane-1-carboxylate oxidase 4 (ACO4) was a target of Th-miR166i, and the downregulation of Th-miR162-3p and Th-miR166i results in the upregulation of ACS8 and ACO4, probably bringing about higher ACC content in flooding-treated roots. Overall, these results indicate that differentially expressed mRNA and miRNAs are involved in regulating tricarboxylic acid cycle, ATP production, fermentation, and metabolism of carbohydrates, amino acids and phytohormones, as well as reactive oxygen species detoxification of Taxodium 406 roots. These processes play pivotal roles in acclimation to flooding stress. These results will improve our understanding of the molecular and physiological bases underlying woody plant flooding acclimation and provide valuable insights into breeding-flooding tolerant trees.

Contrasting coordination of non-structural carbohydrates with leaf and root economic strategies of alpine coniferous forests.

Non-structural carbohydrates (NSCs), as the labile fraction and dominant carbon currency, are essential mediators of plant adaptation to environments. However, whether and how NSC coordinates with plant economic strategy frameworks, particularly the well-recognized leaf economics spectrums (LES) and root economics space (RES), remains unclear. We examined the relationships between NSC and key plant economics traits in leaves and fine roots across 90 alpine coniferous populations on the Tibetan Plateau, China. We observed contrasting coordination of NSC with economics traits in leaves and roots. Leaf total NSC and soluble sugar aligned with the leaf economic spectrum, conveying a trade-off between growth and storage in leaves. However, NSC in roots was independent of the root economic spectrum, but highly coordinated with root foraging, with more starch and less sugar in forage-efficient, thinner roots. Further, NSC-trait coordination in leaves and roots was, respectively, driven by local temperature and precipitation. These findings highlight distinct roles of NSC in shaping the above- and belowground multidimensional economics trait space, and NSC-based carbon economics provides a mechanistic understanding of how plants adapt to heterogeneous habitats and respond to environmental changes.

In Situ Sucrose Injection for Alteration of Carbohydrate Reserve Dynamics in Grapevine

Inconsistent yield of subtropical table grape across seasons is often associated with low carbohydrate reserves during flowering. In an attempt to increase TNC and thus yield, sucrose was injected into treated trunks during periods of high carbohydrate demand (i.e., between budburst and flowering). Total non-structural carbohydrate (TNC) concentration dynamics were assessed in the grapevine root and trunk tissues of both control and treated vines. In the control (untreated) vines, the TNC concentration in root and trunk tissues was 13.5% and 7.5% w/dw at leaf fall and 7.2% and 3.7% w/dw at flowering, respectively. This decrease in carbohydrate reserve was estimated at ~500 g/vine and is associated with the re-establishment of the plant canopy in early spring. Carbohydrate reserves remained stable or rose slightly between flowering and harvest and recovered between crop harvest and leaf fall. In treated vines, a constantly pressurised low-pressure in situ trunk injection system (69 kPa) with 5% w/v sucrose solution over 45 days from budburst (to flowering), in each of the two seasons, delivered a widely variable amount of sucrose into each vine with variation ascribed to the amount of internal dead wood in the trunk. In the best circumstances, an average of 150 g sucrose/vine/season was injected, and sucrose-injected vines had higher trunk TNC reserve (4.1% compared to 3.6% w/dw in the control) at flowering. A δ13C (‰) analysis confirmed the presence of injected sucrose in the shoot at flowering. However, the correlation between the amount of loaded sucrose and δ13C in young shoot tissue was poor, indicative of variable partitioning patterns. Inflorescence number per vine and berry yield were markedly higher in sucrose-injected vines, but differences were not significant given the high variation between vines. The addition of KCl to the sucrose solution and use of the healthy vines are recommended to increase sucrose loading using the injection method to address inconsistent yielding of subtropical table grape.

Seed mass and seed resources – testing the defense trade-off hypothesis in woody plants

Physical and chemical defenses of seeds are essential for plant survival and reproduction. By protecting their seeds from herbivores and other threats, plants can ensure that their offspring have a bet­ter chance of surviving and growing into adult plants. Plant seed-size variation can affect their ability to allocate defense resources and nutrients. Smaller seeds tend to contain fewer resources and are likely to be more constrained in resource allocation to defense or to nutrient supply for the germinating seedling than larger seeds. In this study, we hypothesized that (1) the trade-off between physical and chemical defenses and reserve materials (in this study TNC content) in seeds is related to the allocation of those resources to seeds, 2) the direction of allocation of these resources is mediated by seed size. Based on seeds of 77 woody plant species collected in the Kórnik Arboretum located in western Poland, we measured several physical and chemical seed traits: seed mass, total phenols, tannin content, crude fiber, lignin content, and total non-structural carbohydrates. We revealed that chemical defense (phenol content) and total non-structural carbohydrates increased with seed mass, whereas physical defense (fiber content) had the opposite effect. We found that the trade-off between physical and chemical defense in plants was often mediated by seed size.

Understory seedlings of Quercus mongolica survive by phenological escape

Understanding understory seedling regeneration mechanisms is important for the sustainable development of temperate primary forests in the context of increasingly intense climate warming events. The poor regeneration of dominant tree species, however, is one of the biggest challenges it faces at the moment. Especially, the regeneration of the shade-intolerant Quercus mongolica seedling is difficult in primary forests, which contrasts with the extreme abundance of understory seedlings in secondary forests. The mechanism behind the interesting phenomenon is still unknown. This study used in-situ monitoring and nursery-controlled experiment to investigate the survival rate, growth performance, as well as nonstructural carbohydrate (NSC) concentrations and pools of various organ tissues of seedlings for two consecutive years, further analyze the understory light availability and simulate the foliage carbon (C) gain in the secondary and primary forest. Results suggested that seedlings in the secondary forest had greater biomass allocation aboveground, height and specific leaf area (SLA) in summer, which allowed the seedling to survive longer in the canopy closure period. High light availability and positive C gain in early spring and late autumn are key factors affecting the growth and survival of understory seedlings in the secondary forest, whereas seedlings in the primary forest had annual negative carbon gain. Through the growing season, the total NSC concentrations of seedlings gradually decreased, whereas those of seedlings in the secondary forest increased significantly in autumn, and were mainly stored in roots for winter consumption and the following year's summer shade period, which was verified by the nursery-controlled experiment that simulated autumn enhanced light availability improved seedling survival rate and NSC pools. In conclusion, our results revealed the survival trade-off strategies of Quercus mongolica seedlings and highlighted the necessity of high light availability during the spring and autumn phenological periods for shade-intolerant tree seedling recruitment.

Effects of mepiquat chloride and plant population density on leaf photosynthesis and carbohydrate metabolism in upland cotton

BackgroundMepiquat chloride (MC) application and plant population density (PPD) increasing are required for modern cotton production. However, their interactive effects on leaf physiology and carbohydrate metabolism remain obscure. This study aimed to examine whether and how MC and PPD affect the leaf morpho-physiological characteristics, and thus final cotton yield. PPD of three levels (D1: 2.25 plants·m−2, D2: 4.5 plants·m−2, and D3: 6.75 plants·m−2) and MC dosage of two levels (MC0: 0 g·ha−2, MC1: 82.5 g·ha−2) were combined to create six treatments. The dynamics of nonstructual carbohydrate concentration, carbon metabolism-related enzyme activity, and photosynthetic attributes in cotton leaves were examined during reproductive growth in 2019 and 2020.ResultsAmong six treatments, the high PPD of 6.75 plants·m−2 combined with MC application (MC1D3) exhibited the greatest seed cotton yield and biological yield. The sucrose, hexose, starch, and total nonstructural carbohydrate (TNC) concentrations peaked at the first flowering (FF) stage and then declined to a minimum at the first boll opening (FBO) stage. Compared with other treatments, MC1D3 improved starch and TNC concentration by 5.4% ~ 88.4%, 7.8% ~ 52.0% in 2019, and by 14.6% ~ 55.9%, 13.5% ~ 39.7% in 2020 at the FF stage, respectively. Additionally, MC1D3 produced higher transformation rates of starch and TNC from the FF to FBO stages, indicating greater carbon production and utilization efficiency. MC1D3 displayed the maximal specific leaf weight (SLW) at the FBO stage, and the highest chlorophyll a (Chl a), Chl b, and Chl a + b concentration at the mid-late growth phase in both years. The Rubisco activity with MC1D3 was 2.6% ~ 53.2% higher at the flowering and boll setting stages in both years, and 2.4% ~ 52.7% higher at the FBO stage in 2020 than those in other treatments. These results provided a explanation of higher leaf senescence-resistant ability in MC1D3.ConclusionIncreasing PPD coupled with MC application improves cotton yield by enhancing leaf carbohydrate production and utilization efficiency and delaying leaf senescence.

Responses of non-structural carbohydrates and biomass in plant to heavy metal treatment

The contamination of heavy metals profoundly impacts plant metabolic processes and various physiological indicators, such as non-structural carbohydrates (NSC). However, a comprehensive understanding of how NSC in plants respond to heavy metal treatment and how different experimental setting and plant types affect the response of plant NSC is still lacking. Here, we compiled data of 2084 observations of NSC from 85 published studies and conducted a meta-analysis to investigate the responses of soluble sugars, starch, the ratio of soluble sugar to starch, and total non-structural carbohydrates (TNSC) to heavy metal treatment. Our results showed that, under heavy metal treatment, foliar soluble sugars, foliar TNSC, and the ratio of soluble sugars to starch in both foliage and root increased significantly by 21.6 %, 11.6 %, 55.9 %, and 65.1 %, respectively; and foliar starch, root starch, and root TNSC decreased significantly by 10 %, 23.3 %, and 11 %, respectively; while root soluble sugars remained unchanged. The treatment of heavy metals significantly diminished the biomass of foliage, above-ground, and root by 12.3 %, 29.5 %, and 34.3 %, respectively. The responses of foliar NSC to heavy metal treatment were strongly dependent on leaf habit, the duration and concentration of heavy metal treatment, and soil pH value. The magnitude of the response of NSC to heavy metals increased with the duration and concentration of heavy metal treatment. Furthermore, the types of heavy metals modulated the magnitude of the response of foliar NSC to heavy metal treatment. Overall, our findings provide valuable insights into the responses of plant NSC to heavy metal stress and contribute to a comprehensive understanding of this crucial aspect of plant physiology.

Genome size is strongly linked to carbohydrate storage and weakly linked to root sprouting ability in herbs.

Several lines of evidence indicate that carbohydrate storage in plant below-ground organs might be positively related to genome size because both these plant properties represent resource sinks and can affect cell size, cell cycle time, water-use efficiency and plant growth. However, plants adapted to disturbance, such as root sprouters, could be an exception because their strategy would require higher carbohydrate reserves to fuel biomass production but small genomes to complete their cell cycles faster. We used data from a field survey to test the relationship between genome size and the probability of root sprouting ability in 172 Central European herbaceous species. Additionally, we conducted a pot experiment with 19 herbaceous species with different sprouting ability (nine congeneric pairs plus one species), and measured root non-structural carbohydrate concentrations and pools at the end of a growing season. In the Central European flora, the probability of root sprouting ability was lower in large-genome species but this pattern was weak. In the pot experiment, both total non-structural and water-soluble carbohydrates (mainly fructans) were positively and non-linearly related to genome size, regardless of sprouting strategy. The concentrations of mono- and disaccharides and all carbohydrate pools showed no link to genome size, and starch was absent in large-genome species. The link between genome size and carbohydrate storage was less apparent at a small phylogenetic scale because we only observed a higher carbohydrate concentration in species with larger genomes for four of the species pairs. Root sprouters may have smaller genomes because of their frequent occurrence in dry and open habitats. Large-genome species with presumably large cells and vacuoles could accumulate more water-soluble carbohydrates at the end of the growing season to fuel their growth and perhaps protect vulnerable organs from freezing early in the next season.

Shrubs have a greater influence on the nonstructural carbohydrates of desert mosses along precipitation decreased

Biological soil crusts and the mosaic distribution of vascular plants have become an important landscape feature on the desert surface owing to limited water. The desert shrub, as one of the best-known vascular plants, exerts a notable influence on multiple facets, encompassing the growth and development of biological soil crusts as well as the coverage, biomass, physiology, and biochemical properties of mosses. Therefore, to comprehend how plants react and adapt to drought conditions, it is necessary to investigate the concentration and allocation patterns of nonstructural carbohydrates (NSCs). Nevertheless, the phenomenon still remains unclear whether the influence of shrubs on biological soil crusts follows any pattern concerning the alteration in precipitation gradients on a broader spatial scale. In this investigation, Syntrichia caninervis was gathered from two distinct microenvironments within the Gurbantunggut Desert spanning the gradient of natural precipitation. The aim was to examine the reaction of NSCs within S. caninervis to the presence of shrubs under varying precipitation circumstances. The amount of total NSCs and their components in S. caninervis were determined by the shrub effect, mean annual precipitation (MAP), and mean annual temperature (MAT). The presence of shrubs led to a remarkable rise in the content of fructose, starch, and total NSCs in S. caninervis, but a decline in the ratio of soluble sugar to starch. Nonetheless, variations were observed in the overall NSCs content, the individual constituents of NSCs, and the proportion of soluble sugars to starch within S. caninervis across diverse microenvironments and sampling locations. The content of total NSC and its components, as well as the ratio of soluble sugar to starch, substantially increased with a decrease in MAP (negative correlation) in S. caninervis. Conversely, they significantly decreased as MAT decreased (positive correlation). With the amplifying MAP, the disparities in these parameters (total NSC content, its components, and the ratio of soluble sugar to starch) between two distinct microhabitats diminished gradually, whereas these disparities progressively intensified with rising mean annual temperature (MAT). The outcomes derived from the structural equation model (SEM) underscored the direct and indirect influence of climate and the presence of shrubs influenced the content of NSCs directly or indirectly by enhancing soil nutrients. As a conclusion, the impact of shrub presence, followed by mean annual precipitation (MAP), had the most notable effect on NSCs in S. caninervis. Consequently, despite the observed spatial pattern in NSCs content, shrubs emerged as the most critical factor, in speaking of their effect continuous raising with the declining MAP. These findings provide new insights into the mechanisms of NSCs distribution and stress resistance in desert moss in different microhabitats.

Mechanical thinning restores ecological functions in a seasonally dry ponderosa pine forest in the inland Pacific Northwest, USA

An increasingly important goal of federal land managers in seasonally dry forests of the western US is restoring forest resilience. In this study, we quantified the degree to which a thinning treatment in a dry forest of eastern Oregon restored aspects of forest resilience by focusing on key functional attributes of our study system. First, we measured several physiological responses of overstory trees that are associated with resilience, including radial growth, resin production, abundance of non-structural carbohydrates (NSC), and leaf area. Second, we investigated understory vegetation responses including species diversity, composition, and cover by growth form that influence fire behavior and resilience to disturbance. We found that tree radial growth was greater in trees in thinned stands beginning three years post-treatment. The abundance of key chemical compounds found in trees, including resin, starch, total NSC and sucrose did not differ between treatments; however, abundance of glucose plus fructose was lower in treated stands, suggesting mobilization and use of carbon reserves for foliar and wood production following thinning. We observed an increase in species richness and diversity within thinned stands three to four years after thinning, primarily due to the release of forbs and reestablishment of graminoids following ground disturbance. Here, we demonstrate that elements of forest resilience can be restored in dry forest systems via selective thinning to promote historical forest structure. In forests where thinning reduces stand density, vigorous overstory trees and increased herbaceous cover can help facilitate the re-establishment of low intensity surface fire regimes that maintain stable and persistent vegetative states. Understanding the ecological effects of fuel reduction treatments allows land managers to assess potential forest resiliency and adapt future treatments based on the observed results of previous activities.

Estimation of the Total Nonstructural Carbohydrate Concentration in Apple Trees Using Hyperspectral Imaging

The total nonstructural carbohydrate (TNC) concentration is an important indicator of the growth period and health of fruit trees. Remote sensing can be applied to monitor the TNC concentration in crops in a non-destructive manner. In this study, hyperspectral imaging from an unmanned aerial vehicle was applied to estimate the TNC concentration in apple trees. Partial least-squares regression, ridge regression, and Gaussian process regression (GP) were used to develop estimation models, and their effectiveness using selected key bands as opposed to full bands was evaluated in an effort to reduce computational costs and improve reproducibility. Nine key bands were identified, and the GP-based model using these key bands performed almost as well as the models using full bands. These results can be combined with previous studies on estimating the nitrogen concentration to provide useful information for more precise nutrient management to improve the yield and quality of apple trees.

Effects of drought hardening on the carbohydrate dynamics of Quercus acutissima seedlings under successional drought.

As precipitation patterns are predicted to become increasingly erratic, the functional maintenance of warm-temperate forests constitutes a key challenge for forest managers. In this study, 2-year-old Quercus acutissima seedlings were selected to elucidate the mechanisms whereby they respond to soil water fluctuations and the drought hardening effects on plant carbohydrate dynamics. Seedlings were trained under different soil water conditions for 2 months: drought (D), well-watered (W), 1-month drought and then 1-month well-watered (D-W), and 1-month well-watered and then 1-month drought (W-D). The functional traits involved in water- and carbon-use strategies were explored at the end of the hardening period. Compared with seedlings in group W, seedlings in groups D, D-W, and W-D had increased potential for carbon uptake (i.e., light saturated point, maximum ribulose-1,5-bisphosphate (RuBP) saturated rate, and electron transport rate) and water uptake (i.e., fine root-to-coarse root ratio) and downregulated growth and mitochondrial respiration to decrease carbon consumption. After water fluctuation hardening, we performed a successional dry-down experiment for 1 month to detect carbohydrate dynamics and explore the acclimation caused by prior hardening. Our results revealed that there were more soluble sugars allocated in the leaves and more starch allocated in the stems and roots of seedlings hardened in the D, W-D, and D-W treatments than that of seedlings hardened in the W treatment. No significant changes in total non-structural carbohydrates were found. In addition, we found near-zero (seedlings trained by D and D-W treatments) or negative (seedlings trained by W-D treatment) growth of structural biomass at the end of the dry-down experiment, which was significantly lower than that of W-hardened seedlings. This suggests that there was a shift in allocation patterns between carbon storage and growth under recurrent soil drought, which can be strengthened by drought memory. We conclude that Q. acutissima seedlings adjusted water- and carbon-use strategies in response to water fluctuations, whereas stress memory can enhance their overall performance in reoccurring drought. Therefore, taking advantage of stress memory is a promising management strategy in forest nurseries, and drought-trained seedlings might be more suitable for afforestation practices in sites characterized by fluctuating soil water content, considering the ongoing global climatic changes.

Tropical forest lianas have greater non-structural carbohydrate concentrations in the stem xylem than trees.

Lianas (woody vines) are important components of tropical forests and are known to compete with host trees for resources, decrease tree growth and increase tree mortality. Given the observed increases in liana abundance in some forests and their impacts on forest function, an integrated understanding of carbon dynamics of lianas and liana-infested host trees is critical for improved prediction of tropical forest responses to climate change. Non-structural carbohydrates (NSC) are the main substrate for plant metabolism (e.g., growth, respiration), and have been implicated in enabling tree survival under environmental stress, but little is known of how they vary among life-forms or of how liana infestation impacts host tree NSC. We quantified stem total NSC (NSC) concentrations and its fractions (starch and soluble sugars) in trees without liana infestation, trees with more than 50% of the canopy covered by lianas, and the lianas infesting those trees. We hypothesized that i) liana infestation depletes NSC storage in host trees by reducing carbon assimilation due to competition for resources; ii) trees and lianas, which greatly differ in functional traits related to water transport and carbon uptake, would also have large differences in NSC storage, and that As water availability has a significant role in NSC dynamics of Amazonian tree species, we tested these hypotheses within a moist site in western Amazonia and a drier forest site in southern Amazonia. We did not find any difference in NSC, starch or soluble sugar concentrations between infested and non-infested trees, in either site. This result suggests that negative liana impact on trees may be mediated through mechanisms other than depletion of host tree NSC concentrations. We found lianas have higher stem NSC and starch than trees in both sites. The consistent differences in starch concentrations, a long term NSC reserve, between life forms across sites reflect differences in carbon gain and use of lianas and trees. Soluble sugar concentrations were higher in lianas than in trees in the moist site but indistinguishable between life forms in the dry site. The lack of difference in soluble sugars between trees and lianas in the dry site emphasize the importance of this NSC fraction for plant metabolism of plants occurring in water limited environments. Effects of elevated CO2 concentration and experimental warming on morphological, physiological, and biochemical responses of winter wheat under soil water deficiency.

Global climate change and freshwater scarcity have become two major environmental issues that constrain the sustainable development of the world economy. Climate warming caused by increasing atmospheric CO2 concentration can change global/regional rainfall patterns, leading to uneven global seasonal precipitation distribution and frequent regional extreme drought events, resulting in a drastic reduction of available water resources during the critical crop reproduction period, thus causing many important food-producing regions to face severe water deficiency problems. Understanding the potential processes and mechanisms of crops in response to elevated CO2 concentration and temperature under soil water deficiency may further shed lights on the potential risks of climate change on the primary productivity and grain yield of agriculture. We examined the effects of elevated CO2 concentration (e[CO2]) and temperature (experimental warming) on plant biomass and leaf area, stomatal morphology and distribution, leaf gas exchange and mesophyll anatomy, rubisco activity and gene expression level of winter wheat grown at soil water deficiency with environmental growth chambers. We found that e[CO2] × water × warming sharply reduced plant biomass by 57% and leaf photosynthesis (P n) 50%, although elevated [CO2] could alleviated the stress from water × warming at the amount of gene expression in RbcL3 (128%) and RbcS2 (215%). At ambient [CO2], the combined stress of warming and water deficiency resulted in a significant decrease in biomass (52%), leaf area (50%), P n (71%), and G s (90%) of winter wheat. Furthermore, the total nonstructural carbohydrates were accumulated 10% and 27% and increased R d by 127% and 99% when subjected to water × warming and e[CO2] × water × warming. These results suggest that water × warming may cause irreversible damage in winter wheat and thus the effect of "CO2 fertilization effect" may be overestimated by the current process-based ecological model.

Moisture stress limits radial mixing of non-structural carbohydrates in sapwood of trembling aspen.

Dynamics in non-structural carbohydrate (NSC) pools may underlie observed drought legacies in tree growth. We assessed how aridity influences the dynamics of different-aged NSC pools in tree sapwood at two sites with differing climate conditions ('wet' vs. 'dry') that also experienced widespread regional drought five years earlier. We used an incubation method to measure radiocarbon (Δ14C) in CO2 respired from Populus tremuloides (aspen) tree rings to evaluate NSC storage and mixing patterns, coupled with measurements of NSC (soluble sugars, starch) concentrations and respired δ13C-CO2. At a wet site, CO2 respired from rings formed during 1962-1967 was only ~ 11years old, suggesting deep sapwood mixing of NSCs as starch. At a dry site, total NSC was about one-third of wet site totals, maximum ages in deep rings were lower, and ages more rapidly increased in shallow rings then plateaued. These results suggest historically shallower mixing and/or relatively higher consumption of NSCs under dry conditions. Both sites, however, had similar aged NSC (<1yr) in the most recent six rings, indicative of deep radial mixing following relatively wet conditions during the sampling year. We suggest significant differences in NSC mixing among sites are driven by moisture stress, where aridity reduces NSC reserves and restricts the depth of radial mixing. However, dynamic climate conditions in the southwestern US resulted in more complex radial patterns of sapwood NSC age than previously described. We suggest a novel conceptual framework to understand how moisture variability might influence the dynamics of NSC mixing in the sapwood.

Effects of Drought Stress on Non-Structural Carbohydrates in Different Organs of Cunninghamia lanceolata.

The Chinese fir Cunninghamia lanceolata (Lamb.) Hook. is an important timber conifer species in China. Much has been studied about Chinese fir, but the distribution of non-structural carbohydrates (NSCs) among different organs (needles, branch, stem, and roots) under drought stress remains poorly understood. In this study, we used one-year-old C. lanceolata plantlets to evaluate the effects of simulated drought under four water regimes, i.e., adequate water or control, light drought, moderate drought, and severe drought stress corresponding to 80%, 60%, 50%, and 40%, respectively of soil field maximum capacity on various NSCs in the needles, branch, stem and roots. The degree and duration of drought stress had significant effects on fructose, glucose, sucrose, soluble sugar, starch, and NSC content in various organs (p < 0.05). Fructose content increased in stem xylem, stem phloem, and leaves. Glucose and sucrose content declined in stem and branch xylem under light drought stress and moderate drought stress, and increased under severe drought stress conditions. Soluble sugars content declined, and starch content increased in leaf and branch phloem, but the latter could not compensate for soluble sugar consumption in the whole plant, and therefore, total NSCs decreased. Correlation analysis showed that a significant positive correlation existed in the soluble sugar content between leaves and roots, and between xylem and phloem in the stems and branches. Chinese fir appears to have different NSCs distribution strategies in response to drought stress, viz., allocating more soluble sugars to fine roots and increasing starch content in the needles, as well as ensuring osmosis to prevent xylem embolism. Our study may broaden the understanding of the various mechanisms that Chinese fir and other plants have to enhance their tolerance to drought stress.

Local adaptation to aridity in a widely distributed angiosperm tree species is mediated by seasonal increase of sugars and reduced growth.

Trees in dry climates often have higher concentrations of total non-structural carbohydrates (NSC=starch + soluble sugars) and grow less than conspecifics in more humid climates. This pattern might result from growth being more constrained by aridity than the carbon (C) gain, or reflect local adaptation to aridity, since NSC fuel metabolism and ensure adequate osmoregulation through the supply of soluble sugars (SS), while low growth reduces water and C demands. It has been further proposed that C allocation to storage could come at the expense of growth (i.e., a growth-storage trade-off). We examined whether NSC and growth reflect local adaptation to aridity in Embothrium coccineum (Proteaceae), a species with an exceptionally wide niche. To control for any influence of the phenotypic plasticity on NSC and growth, we collected seeds from dry (500mmyear-1) and moist (> 2500mmyear-1) climates and grew seedlings in a common garden experiment for 3years. We then compared NSC and SS concentrations and pools (i.e., total contents), and the biomass of seedlings at spring, summer, and fall. Seedlings from the dry climate had significantly lower biomass and similar NSC concentrations and pools than seedlings from moist climate, suggesting that reduced growth in arid environments does not result from a prioritization of C allocation to storage but it confers advantages under aridity (e.g., lower transpiration area). Across organs, starch and NSC decreased similarly in seedlings from both climates from spring onward. However, root and stem SS concentrations increased during the growing season, and these increases were significantly higher in seedlings from the dry climate. The greater SS accumulation in seedlings from the dry climate compared to those from the moist climate demonstrates ecotypic differentiation in the seasonal dynamics of SS, suggesting that SS underlie local adaptation to aridity. Effect of Rootstock on Vineyard Establishment Using Green-Growing Benchgrafts

Demand for vine plant material has increased drastically due to the ongoing expansion of viticulture, and recent widespread replanting efforts. Nurseries and growers are turning to green-grafted vines to meet demand. Unfortunately, most vineyard establishment studies have centered around dormant benchgrafted vines. Thus, little is known regarding the specific establishment trends of green-growing benchgrafts. This study aimed to explore the role rootstock selection has in green-growing benchgraft establishment and development over the first four years post-planting. Vitis vinifera L. cv. Sauvignon blanc was grafted onto multiple rootstocks of varying parentage, including ‘101-14MGT’ (V. riparia × V. rupestris), ‘1103P’ (V. berlandieri × V. rupestris), ‘110R’ (V. berlandieri × V. rupestris), ‘420A MGT’ (V. berlandieri × V. riparia), and ‘Teleki 5C’ (V. berlandieri × V. riparia). The experimental site was organized using a completely randomized design (n = 12) with all vines managed to industry-standard cultural practices. Vines grafted onto 1103P had the largest average trunk diameter (p = 0.0012) and circumference (p &lt; 0.0001) at 22.2 mm and 7.57 cm, respectively. Vines grafted onto 110R had the second-largest trunk circumference at 6.65 cm. Vines grafted onto 110R had the largest concentration of total non-structural carbohydrates at planting at 1.47 g/L, followed by 1103P at 1.25 g/L (p &lt; 0.0001). Total non-structural carbohydrate concentrations in the trunk during dormancy was the highest in 110R at 16.0% total dry weight (p = 0.0008). The larger trunk size and more extensive carbohydrate reserves suggest that green-growing benchgrafts using 110R or 1103P have a higher capacity and likelihood of establishment success.

The potential conservation value of anthropogenically modified habitat for the Endangered moor macaque Macaca maura in Sulawesi, Indonesia

AbstractHuman-induced land-use change has resulted in substantial loss and degradation of habitat for forest-dwelling wildlife. The moor macaque Macaca maura, an Endangered primate endemic to Sulawesi, Indonesia, has been observed in a wide array of habitats, including heavily modified areas, but little is known about the quality of these habitats. Here we compared the habitat quality of two areas occupied by moor macaque populations: a previously human-modified karst forest now located within a national park and a heavily modified forest located in an unprotected area. We assessed habitat quality by measuring specific indicators of potential food availability derived from vegetation data and quantified the nutritional environment based on macronutrient analysis of fruit samples collected at the two sites. Although the species richness, diversity and overall stem density of macaque food trees were greater in the protected area forest, total basal area and mean diameter at breast height were greater in the heavily modified forest. Mean metabolizable energy concentrations of fruits eaten by macaques were similar between forests, as were the proportions of protein, lipids and total non-structural carbohydrates. These results provide further support for the notion that heavily modified habitats should not be overlooked for their potential conservation value. To further augment their value, conservation efforts should focus on forest restoration, specifically the planting of fast-growing species that are utilized by both wildlife and people.
