The effect of prescribed fires on abiotic and biotic factors in the southern region of Puerto Rico

Field fires can modify soil nutrient cycling and alter soil microbial communities (SMC), although the latter is not well understood. In the southern region of Puerto Rico, field fires have become a significant problem during the dry season. To mimic the effects of a field fire, we performed prescribed fires on a hillside at the Juana Díaz Agricultural Experiment substation in October 2015 and March 2017. A complete randomized block design was established in Yauco soil (Typic calciustolls) that included the following treatments: negative control (unburned), positive control (burned plots, no remediation), mulching treatment (burned plots remediated with Leucaena spp. mulch), and surfactant treatment (burned plots remediated with a surfactant). In the first burning (2015), soil samples were collected before burning and at 30, 180, and 420 days after burning (DAB). In the second burning (2017), soil samples were collected at 30, 90, and 270 DAb. soil physicochemical properties and microbial community structure were assessed using phospholipid fatty acid (PLFA) analysis. Overall, burning increased soil exchangeable Ca2+ (except after 30 DAB in the second burning) and decreased exchangeable K+ when compared to unburned soils. compared to unburned plots, total fungal PLFA was significantly lower in burned plots with or without mulch and surfactant treatments, and total bacterial PLFA did not differ between burned and unburned plots after 30 days. Total microbial biomass was significantly (P<0.05) higher in mulch and surfactant treated burned soil compared to unburned and burned plots without treatment after 90 DAB (2017) and 420 (2015) DAB. The use of mulch and surfactant treatments in prescribed burning fields increased microbial communities 90 DAB. This study emphasizes short-term changes in microbial communities and suggests they are highly resilient to disturbances after prescribed fires.

Dynamic changes in microbial communities during the bioremediation of herbicide (chlorimuron-ethyl and atrazine) contaminated soils by combined degrading bacteria

Chlorimuron-ethyl and atrazine are two herbicides with long half-lives in soil; their long-term and excessive application has led to a series of environmental problems. In this study, the strains Chenggangzhangella methanolivorans CHL1 and Arthrobacter sp. ART1 were combined and used for the remediation of chlorimuron-ethyl, atrazine and combined contaminated soils in a microcosm experiment. Changes in chlorimuron-ethyl and atrazine concentrations in soils were monitored, and variations in the soil microbial community were studied by phospholipid fatty acid (PLFA) analysis. The two inoculated degrading strains accelerated the degradation of chlorimuron-ethyl and atrazine in soil, especially in the combined contaminated soil. Addition of the two herbicides and their combination generally decreased the concentrations of total PLFAs, total bacterial PLFAs, Gram-negative and Gram-positive bacterial PLFAs and Shannon-Wiener indices, and changed microbial community composition, whilst stimulating fungal PLFA concentrations. In addition, the combined herbicide treatment had more impact on microbial biomass than the single herbicide treatments. Inoculation treatments significantly relieved the effects of herbicides on soil microbial biomass, diversity and community structure. This study demonstrated that strains CHL1 and ATR1 have the potential to remediate chlorimuron-ethyl, atrazine and combined contaminated soils, and provided valuable information for remediation of chlorimuron-ethyl, atrazine and combined contaminated soils in situ.

Nitrogen additions affect litter quality and soil biochemical properties in a peatland of Northeast China

Nitrogen (N) is a limiting nutrient in many peatland ecosystems. Enhanced N deposition, a major component of global climate change, affects ecosystem carbon (C) balance and alters soil C storage by changing plant and soil properties. However, the effects of enhanced N deposition on peatland ecosystems are poorly understood. We conducted a two-year N additions field experiment in a peatland dominated by Eriophorum vaginatum in the Da Xing’an Mountains, Northeast China. Four levels of N treatments were applied: (1) CK (no N added), (2) N1 (6gNm−2yr−1), (3) N2 (12gNm−2yr−1), and (4) N3 (24gNm−2yr−1). Plant and soil material was harvested at the end of the second growing season. N additions increased litter N and phosphorus (P) content, as well as β-glucosidase, invertase, and acid-phosphatase activity, but decreased litter C:N and C:P ratios. Litter carbon content remained unchanged. N additions increased available NH4+–N and NO3−–N as well as total Gram-positive (Gram+), Gram-negative (Gram−), and total bacterial phospholipid fatty acids (PLFA) in shallow soil (0–15cm depth). An increase in these PLFAs was accompanied by a decrease in soil labile organic C (microbial biomass carbon and dissolved organic carbon), and appeared to accelerate decomposition and reduce the stability of the soil C pool. Invertase and urease activity in shallow soils and acid-phosphatase activity in deep soils (15–30cm depth) was inhibited by N additions. Together, these findings suggest that an increase in N deposition in peatlands could accelerate litter decomposition and the loss of labile C, as well as alter microbial biomass and function.

Impact of tropical lowland rainforest conversion into rubber and oil palm plantations on soil microbial communities

Focusing on Sumatra, a hotspot of tropical lowland rainforest transformation, we investigated effects of the con- version of rainforests into rubber agroforests (Bjungle rubber^), intensive rubber, and oil palm plantations on the communities of litter and soil microorganisms and identified factors responsible for these changes. Litter basal respiration, microbial biomass, total bacterial phospholipid fatty acids (PLFAs), and fungal PLFAs did not vary significantly with rainforest conversion. In litter of converted ecosystems, the concentration of certain PLFAs including the Gram-negative bacteria marker PLFA cy17:0 and the Gram-positive bacteria marker PLFA i17:0 was reduced as compared to rainforest, whereas the concentration of the arbuscular mycorrhizal fungi (AMF) marker neutral lipid fatty acid (NLFA) 16:1ω5c in- creased. As indicated by redundancy analysis, litter pH and carbon concentration explained most of the variation in litter microbial community composition. In soil, microbial biomass did not vary significantly with rainforest conversion, whereas basal respiration declined. Total PLFAs and especially that of Gram-negative bacteria decreased, whereas PLFA i17:0 in- creased with rainforest conversion. The concentration of fun- gal PLFAs increased with rainforest conversion, whereas NLFA 16:1ω5c did not change significantly. Redundancy analysis indicated that soil pH explained most of the variation in soil microbial community composition. Overall, the data suggest that conversion of rainforests into production systems results in more pronounced changes in microbial community composition in soil as compared to litter. In particular, the response of fungi and bacteria was more pronounced in soil, while the response of AMF was more pronounced in litter. Notably, only certain bacterial markers but not those of saprotrophic fungi and AMF were detrimentally affected by rainforest conversion.

Dynamics of Microbial Community in Japanese Andisol of Apple Orchard Production Systems

In this study, two fields of temperate Andisols from high‐input and low‐input (zero‐input) management practices of an apple orchard were selected to assess microbial community dynamics based on environmental variables. Soils from an Ap horizon were sampled in five consecutive months from May to September and assessed for phospholipid fatty acids as a biomarker of soil microbial community, soil hardness, bulk density, porosity, pH, electrical conductivity (EC), organic carbon (C), available nitrogen (N) and phosphorus (P), and exchangeable cations as soil environmental variables. For all sample dates, total phospholipid fatty acids (PLFAs), total bacterial PLFAs, fungal PLFA, mycorrhizal PLFA, PLFA for actinomycetes, and earthworm were higher in low‐input management than high‐input management. Total PLFAs showed a high degree of seasonality, having August maxima and May minima. Significant effect on the relationships among soil environmental variables and microbiological attributes were observed. Soil management practices also showed a remarkable effect on the relationships among microbiological traits, indicating that some mechanism regulated soil microbial dynamics under two soil management practices. Comparatively higher correlations among the microbiological attributes were observed in low‐input management than high‐input management. Irrespective of soil management practices, bacterial and fungal lipid biomarkers were negatively correlated, suggesting that these subsets of fatty acids are contrasting components of the microbial biomass. Bulk density has negative influence on all soil microbial communities except fungi. On the other hand, linoleic acid and organic C were positively correlated, referring to the distribution of soil organic C implying an upper layer of soils. Microbial community composition and structure were greatly affected by sampling date and to a lesser extent by long‐term management practice. In this study, both ecosystems were characterized by a very diverse microbial community.
