Biomedical and therapeutic potential of marine-derived Pseudomonas sp. strain AHG22 exopolysaccharide: A novel bioactive microbial metabolite

Abstract Microbial exopolysaccharides (EPSs) are gaining interest as alternatives to chemical antioxidants and pharmaceuticals. This study mines the promising biomedical and antimicrobial potential of a marine bacterium, a prolific EPS producer, isolated from the Red Sea. Pseudomonas sp. strain AHG22 generated an EPS weighing 6.98 g·L−1, coded EPSF8, subjected to FT-IR and HPLC chemical analysis. EPSF8 was then investigated for antioxidant assessment by 2,2-diphenyl-1-picrylhydrazyl (DPPH), H2O2, ABTS˙ + , nitric oxide, total antioxidant capacity (TAC), and ferric reducing antioxidant power (FRAP). EPSF8 had an IC50 of 46.99 μg·mL−1 in the DPPH antioxidant assay and antioxidant capacities of 219.45 μg·mg−1 ascorbic acid equivalent (AAE) in the TAC assay and 54.15 μg·mg−1 AAE in the FRAP assay. The in vitro anti-inflammatory effect of EPSF8 was tested against 5-lipoxygenase (5-LOX) and cyclooxygenase-2 (COX-2) enzymes and compared with the drugs ibuprofen and celecoxib used as controls. The IC50 values of 5-LOX, COX-2, ibuprofen, and celecoxib were found to be 14.82, 15.49, 1.5, and 0.28 μg·mL−1, respectively. Additionally, EPSF8 revealed antidiabetic activity toward α-amylase and α-glucosidase, and the IC50 values were 93.1 and 127.28 μg·mL−1, compared to those of acarbose (50.93 and 4.13 μg·mL−1, respectively). Anti-obesity activity of EPSF8 by lipase inhibition revealed IC50 = 56.12 μg·mL−1 compared to orlistat (IC50 = 20.08 μg·mL−1) as a control. EPSF8 displayed antibiofilm and bactericidal activity against Gram-positive (G +ve) and Gram-negative (G −ve) ATCC pathogenic bacterial strains. It had a minimum bactericidal concentration/minimum inhibitory concentration ratio ≤2, indicating a broad bactericidal spectrum. Furthermore, EPSF8 is evidenced to have a promising anti-butyrylcholinesterase activity for the control of Alzheimer’s disease. The findings of the present analysis suggest that the isolated Pseudomonas sp. strain AHG22 EPS can potentially be explored as a promising green therapeutic compound.

In vitro examination of the antioxidants, osajin and pomiferin, in cultured rat mesangial cells

Purpose: The goal of this study was to identify concentrations of osajin and pomiferin, purified compounds originating from Maclura pomifera, that work as effective antioxidants while maintaining cell viability in cultured rat mesangial cells. Rationale and Significance: Diabetic nephropathy (DN) is a serious complication faced by type 1 and type 2 diabetic patients alike. Oxidative stress has emerged as an important pathogenic mechanism in the development of glomerular and tubular injury in DN. Identifying novel antioxidants found in natural compounds could serve as an effective treatment in DN to prevent renal cell damage. Methodology: Rat mesangial cells (NMS) were grown to confluency in RPMI 1640 medium with 17% fetal bovine serum and 1x antibiotic/antimycotic. NMS cells were exposed to vehicle (control) or varying concentrations of pomiferin or osajin. MTT cell survival assays were used to assess cell viability for the selected concentrations. Antioxidant status was assessed using the Total Antioxidant Capacity Assay from Sigma-Aldrich and quercetin was selected as a positive control. Findings: Data from MTT cell viability assays demonstrated several concentrations (25, 50, and 100 mM) of both osajin and pomiferin maintained cell viability in NMS cells. Antioxidant capacity assay plans will be discussed as this is an ongoing project and we hope to identify the total antioxidant capacity and antioxidant response in NMS cells. Future plans also include moving forward with selected concentrations to identify the effectiveness of osajin and/or pomiferin in preventing high glucose-induced damage to glomerular cells and proximal tubular epithelial cells as this will be essential for investigating the benefit of using either compound in diabetic nephropathy. Synthesis, molecular docking studies and biological evaluation of N-(4-oxo-2-(trifluoromethyl)-4H-chromen-7-yl) benzamides as potential antioxidant, and anticancer agents

A series of novel chromone derivatives of (N-(4-oxo-2-(trifluoromethyl)-4H-chromen-6-yl) benzamides) were synthesized by treating 7-amino-2-(trifluoromethyl)-4H-chromen-4-one with K2CO3 and/or NaH, suitable alkyl halides and acetonitrile and/or 1,4-dioxane. The obtained products are in high yields (87 to 96%) with various substituents in short reaction times with no more by-products and confirmed by FT-IR, 1H, and 13C-NMR Spectral data. The in vitro cytotoxic activity was examined against two human cancer cell lines, namely the human lung adenocarcinoma (A-549) and the human breast (MCF-7) cancer cell line. Compound 4h showed promising cytotoxicity against both cell lines with IC50 values of 22.09 and 6.40 ± 0.26 µg/mL respectively, compared to that of the standard drug. We also performed the in vitro antioxidant activity by DPPH radical, hydrogen peroxide, NO scavenging, and total antioxidant capacity (TAC) assay methods, and they showed significant activities. The possible binding interactions of all the synthesized chromone derivatives are also investigated against selective pharmacological targets of human beings, such as HERA protein for cytotoxic activity and Peroxiredoxins (3MNG) for antioxidant activity which showed closer binding free energies than the standard drugs and evidencing the above two types of activities.

Exploring the multifaceted bioactivities of Lavandula pinnata L. essential oil: promising pharmacological activities.

Introduction: This study investigates the biological activities of Lavandula pinnata essential oil (LPEO), an endemic lavender species from the Canary Islands, traditionally used in treating various ailments. Methods: LPEO was extracted by hydrodistillation and analyzed using GC-MS. Antioxidant activity was assessed by DPPH radical scavenging and total antioxidant capacity assays. Antimicrobial activity was evaluated by disc diffusion, MIC, MBC, and MFC determination against bacterial (Staphylococcus aureus, Micrococcus luteus, Escherichia coli, Pseudomonas aeruginosa) and fungal (Candida glabrata, Rhodotorula glutinis, Aspergillus niger, Penicillium digitatum) strains. Antidiabetic and anti-gout potential were investigated through α-amylase, α-glucosidase, and xanthine oxidase inhibition assays. Antityrosinase activity was determined using a modified dopachrome method. Cytotoxicity was assessed by MTT assay against breast (MCF-7, MDA-MB-468), liver (HepG2), colon (HCT-15) cancer cells, and normal cells (PBMCs). Results and discussion: LPEO exhibits potent antiradical activity (IC50 = 148.33 ± 2.48μg/mL) and significant antioxidant capacity (TAC = 171.56 ± 2.34μg AA/mg of EO). It demonstrates notable antibacterial activity against four strains (Staphylococcus aureus, Micrococcus luteus, Escherichia coli, and Pseudomonas aeruginosa) with inhibition zones ranging from 18.70 ± 0.30mm to 29.20 ± 0.30mm, along with relatively low MIC and MBC values. LPEO displays significant antifungal activity against four strains (Candida glabrata, Rhodotorula glutinis, Aspergillus niger, and Penicillium digitatum) with a fungicidal effect at 1mg/mL, surpassing the positive control (cycloheximide), and MIC and MFC values indicating a fungicidal effect. It exhibits substantial inhibition of xanthine oxidase enzyme (IC50 = 26.48 ± 0.90μg/mL), comparable to allopurinol, and marked inhibitory effects on α-amylase (IC50 = 31.56 ± 0.46μg/mL) and α-glucosidase (IC50 = 58.47 ± 2.35μg/mL) enzymes.The enzyme tyrosinase is inhibited by LPEO (IC50 = 29.11 ± 0.08mg/mL). LPEO displays moderate cytotoxic activity against breast, liver, and colon cancer cells, with low toxicity towards normal cells (PBMC). LPEO exhibits greater selectivity than cisplatin for breast (MCF-7) and colon (HCT-15) cancer cells but lower selectivity for liver (HepG2) and metastatic breast (MDA-MB-468) cancer cells. These findings suggest the potential of LPEO as an antioxidant, antimicrobial, anti-gout, antidiabetic, and anticancer agent.

Distribution of antioxidants and phenolic compounds in flour milling fractions from hard red winter wheat

Mature wheat kernels contain three main parts: endosperm, bran, and germ. Flour milling results in multiple streams that are chemically different; however, the distribution of antioxidants and phenolic compounds has not been well documented in terms of conventional milling by-product streams. In this study, multiple analytical methods were used to investigate antioxidant activity and phenolic compound compositions of hard red winter wheat (whole ground wheat), the parts of a wheat kernel (bran, flour, germ), and wheat by-product streams (mill feed, red dog, shorts) for the first time. For each mill stream, phenolic compounds (total, flavonoid, and anthocyanin contents) were determined and antioxidant activities were evaluated with 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity, ferric reducing/antioxidant power (FRAP), and total antioxidant capacity assays. Significant differences (P < 0.05) were observed in phenolic concentrations among fractions of bran, flour, and germ milled from the same kernels and noted that germ accounts for the majority of antioxidant properties, whereas bran contains a substantial portion of phenolic compounds and anthocyanins. Mill feed was high in phenolic content (5.29 mg FAE/g), total antioxidant capacity (866 mg/g), and antioxidant activity (up to 75% DPPH inhibition and 20.26 μmol FeSO4/g). The comprehensive information on distribution of antioxidants and phenolic compounds provides insights for future human consumption of commonly produced co-products from flour milling, and for selecting and using different milling fractions to make foods with improved nutritional properties.

Quality Evaluation and Identification of Phyllanthi fructus (Yuganzi) Based on the Spectrum-Effect Relationship

To establish a quality evaluation and identification method for Phyllanthi fructus (Yuganzi), the spectrum-effect relationship was explored. A high-performance liquid chromatography (HPLC) fingerprint was established using ultraviolet spectrophotometry, and the in vitro antioxidant activity was determined using a total antioxidant capacity assay kit. Similarity analysis, hierarchical cluster analysis (HCA), and partial least squares regression (PLSR) were performed to establish the spectrum-effect relationships. Thirteen batches of Yuganzi were collected for testing. The results revealed that the optimal chromatographic conditions for the HPLC fingerprint were as follows: the mobile phase consisted of 0.1% phosphoric acid solution (A) and acetonitrile (B), the detection wavelength was 214 nm, the column temperature was 30 °C, and the flow rate was 0.8 mL/min. Among the batches of samples, the similarity values of 10 samples (S1–S10) from Yunnan were larger than 0.995; the similarity values of 3 samples (S11–S13) from India, Gaoligong Mountain, and Fujian were less than or equal to 0.986. Furthermore, nineteen characteristic peaks of Yuganzi were calibrated using fingerprint evaluation software. The study on the spectrum-effect relationship further revealed that compounds corresponding to peaks 5 and 8 were potentially key ingredients for the quality evaluation and identification of Yuganzi, closely related to the stable antioxidant activities of Yuganzi. The spectrum-effect relationship is an agile and efficient approach that can ensure the intra-assay stability of Yuganzi from same region and identify Yuganzi from different regions. Compounds with antioxidant activity can be identified as quality markers for Yuganzi.

Colorimetric Sensing of Antioxidant Capacity via Auric Acid Reduction Coupled to ABTS Oxidation.

In this study, a simple and sensitive colorimetric assay has been developed for total antioxidant capacity measurement. The assay is based on the absorption measurement of the bluish-green oxidized product (ABTS·+) formed as a result of the oxidation reaction of the chromogenic reagent ABTS (2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) with gold(III). However, in the presence of antioxidants, the ABTS oxidation process is effectively suppressed due to the reduction of gold(III) ions to the zerovalent state forming gold nanoparticles (AuNPs). Relatively lighter colors and a significant decrease in absorbance are observed depending on the total antioxidant capacity. Taking advantage of this situation, qualitative and quantitative total antioxidant capacity (TAC) measurements, with the naked eye and UV-vis spectroscopy, respectively, could be successfully performed. The assay is named "auric reducing antioxidant capacity" (AuRAC) because the gold(III) ion-reducing ability of antioxidants is measured. The AuRAC assay was applied to dietary polyphenols, vitamin C, thiol-type antioxidants, and their synthetic mixtures. Trolox equivalent antioxidant capacity (TEAC) values obtained with the AuRAC assay were found to be compatible with those of the reference CUPRAC (cupric reducing antioxidant capacity) assay. The AuRAC assay was validated through linearity, additivity, precision, and recovery, demonstrating that the assay is reliable and robust. Compared to the simple TAC assays in the literature based on AuNP formation with subsequent surface plasmon resonance (SPR) absorbance measurement, this indirect assay has a smoother linear range starting from lower antioxidant concentrations. This method displays much higher molar absorption coefficients for antioxidant compounds than other conventional single electron transfer (SET) assays because 3-e- reduction of trivalent gold (i.e., Au(III) → Au(0)) produces three chromophore cation radicals (ABTS·+) of the assay reagent. The sensor has been successfully applied to complex matrices, such as tea infusions and pharmaceutical samples. The AuRAC assay stands out with its high molar absorptivity connected to enhanced sensitivity as well as its potential to convert into a paper-based colorimetric sensor.

Chemical profiling of volatile compounds of the essential oil of grey-leaved rockrose (Cistus albidus L.) and its antioxidant, anti-inflammatory, antibacterial, antifungal, and anticancer activity in vitro and in silico.

Cistus albidus: L., also known as Grey-leaved rockrose and locally addressed as šṭab or tûzzâla lbîḍa, is a plant species with a well-established reputation for its health-promoting properties and traditional use for the treatment of various diseases. This research delves into exploring the essential oil extracted from the aerial components of Cistus albidus (referred to as CAEO), aiming to comprehend its properties concerning antioxidation, anti-inflammation, antimicrobial efficacy, and cytotoxicity. Firstly, a comprehensive analysis of CAEO's chemical composition was performed through Gas Chromatography-Mass Spectrometry (GC-MS). Subsequently, four complementary assays were conducted to assess its antioxidant potential, including DPPH scavenging, β-carotene bleaching, ABTS scavenging, and total antioxidant capacity assays. The investigation delved into the anti-inflammatory properties via the 5-lipoxygenase assay and the antimicrobial effects of CAEO against various bacterial and fungal strains. Additionally, the research investigated the cytotoxic effects of CAEO on two human breast cancer subtypes, namely, MCF-7 and MDA-MB-231. Chemical analysis revealed camphene as the major compound, comprising 39.21% of the composition, followed by α-pinene (19.01%), bornyl acetate (18.32%), tricyclene (6.86%), and melonal (5.44%). Notably, CAEO exhibited robust antioxidant activity, as demonstrated by the low IC50 values in DPPH (153.92 ± 4.30μg/mL) and β-carotene (95.25 ± 3.75μg/mL) assays, indicating its ability to counteract oxidative damage. The ABTS assay and the total antioxidant capacity assay also confirmed the potent antioxidant potential with IC50 values of 120.51 ± 3.33TEμmol/mL and 458.25 ± 3.67µg AAE/mg, respectively. In terms of anti-inflammatory activity, CAEO displayed a substantial lipoxygenase inhibition at 0.5mg/mL. Its antimicrobial properties were broad-spectrum, although some resistance was observed in the case of Escherichia coli and Staphylococcus aureus. CAEO exhibited significant dose-dependent inhibitory effects on tumor cell lines in vitro. Additionally, computational analyses were carried out to appraise the physicochemical characteristics, drug-likeness, and pharmacokinetic properties of CAEO's constituent molecules, while the toxicity was assessed using the Protox II web server.

Iridium nanoparticles supported on polyaniline nanotubes for peroxidase mimicking towards total antioxidant capacity assay of fruits and vegetables

In this study, a straightforward approach is presented for the first time to anchor Ir nanoparticles on the surface of uniform polyaniline (PANi) nanotubes (NTs), which can be used as an efficient peroxidase (POD)-like catalyst. The morphology and chemical structure of the PANi-Ir nanocomposite are characterized by scanning electron microscopy (SEM), high-resolution transmission electron microscopy (HRTEM), X-ray diffractometer (XRD), Raman and X-ray photoelectron spectroscopy (XPS) measurements. Owing to the strong interaction between Ir nanoparticles and PANi, a remarkable catalytic enhancement is achieved compared to the bare Ir black catalyst and individual PANi NTs, dominating withan electron transfer mechanism. Furthermore, an efficient colorimetric sensor for ascorbic acid (AA) is developed with a low detection limit of 1.0 μM (S/N = 3), and a total antioxidant capacity (TAC) sensing platform is also constructed for the rigorous detection and analysis of fruits and vegetables.

Organic wastes of Citrus sinensis Peels- a source of eco-friendly and sustainable bioactive compounds for promoting health

To determine the phytochemicals, radical scavenging and antioxidant potential of orange peel extract. Citrus sinensis were subjected to extraction with ethanol. Gas chromatography (GC) was utilized to determine the phytochemical composition of orange peel extract. Hydrogen peroxide, superoxide, nitric oxide, and hydroxyl radical scavenging assays were conducted to assess radical scavenging potential of the extract. Antioxidant activities of the peel extracts were determined via the 2,2-diphenylpicrylhydrazyl (DPPH) free radical scavenging activity, ferric reducing antioxidant power (FRAP) assay, ABTS scavenging, and total antioxidant capacity (TAC) assay. The GC-FID analysis revealed the presence of alkaloids, flavonoids, polyphenols, tannins, sapogenin, and steroids in the orange peel extract. The results of radical scavenging assays demonstrated the extract’s ability to scavenge hydrogen peroxide, superoxide, nitric oxide and hydroxyl radicals. The scavenging capacity of the extract was observed to be concentration-dependent, with comparisons made to standard antioxidants ascorbic acid and BHT. Peels from citrus sinensis represent a valuable source of phytochemicals, demonstrating significant antioxidant and radical scavenging activities.

Role of hazelnut skin supplementation on plasma antioxidant status and cytokine profile in growing lambs.

In this study, the effect of hazelnut skin dietary supplementation on antioxidant status and cytokine profile was evaluated in growing lambs. A total of 22 male lambs at the age of 2 months, balanced for their initial live weight (15.33 ± SD 1.79 kg), were selected and allocated into two experimental groups: the control group (CON) receiving a maize-barley-based concentrated diet, and the hazelnut group (HS) receiving supplementation with hazelnut skin (150 g/kg on the dry matter) as a maize substitute for the concentrate diet. The experiment lasted for 56 days. Peripheral blood was collected at 7, 35, and 56 days of the experiment. The free radical scavenging activity using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, the total antioxidant capacity assay (TAC), the reactive oxygen species (ROS), and reactive nitrogen species (RNS) were determined in plasma. The secretion of IL-1β, IL-6, and IL-10 cytokines was also determined by ELISA. The DPPH was affected by the interaction between feeding strategy and time of sampling (p = 0.039) with a higher level of DPPH at 7 days in the HS group than the CON group. The time of sampling affected the levels of plasma TAC (p = 0.016), while the ROS/RNS levels showed a higher value in the HS group (p < 0.001), on average. The antioxidant/oxidant index, which combines the TAC and the ROS/RNS levels, was not affected by the inclusion of hazelnut skin in the diet (p = 0.394). The cytokine profile showed a lower IL-6 secretion at both 35 and 56 days than at 7 days, on average. Furthermore, the feeding treatment affected the IL-1β level, showing a lower level in the HS group than in the CON group on average. Lambs from the HS group had higher IL-10 plasma levels than the CON group at 7 days of the experiment. The present data highlight an antioxidant effect and a modulatory role in the cytokine profile of HS supplementation in growing lambs.

Methylene Blue Pretreatment Protects Against Repeated Neonatal Isoflurane Exposure-Induced Brain Injury and Memory Loss.

Perioperative neurocognitive impairment (PND) is a common medical complication in the postoperative period. General anesthesia through volatile anesthetics poses a high risk of POCD. Moreover, the developing brain is especially vulnerable to anesthesia-induced neurotoxicity. Therefore, finding a practical approach to prevent or alleviate neonatal isoflurane (ISO) exposure-induced brain injury and cognitive decline is essential for reducing medical complications following major surgery during the early postnatal period. Using a repeated neonatal ISO exposure-induced PND rat model, we investigated the effects of methylene blue (MB) pretreatment on repeated neonatal isoflurane exposure-induced brain injury and memory loss. Intraperitoneal injection of low-dose MB (1mg/kg) was conducted three times 24h before each ISO exposure. The Barnes maze and novel objection test were conducted to assess learning and memory. Immunofluorescence staining, F-Jade C staining, TUNEL staining, and Western blot analysis were performed to determine mitochondrial fragmentation, neuronal injury, degeneration, and apoptosis. Evans blue extravasation assay, total antioxidant capacity assay, MDA assay kit, and related inflammatory assay kits were used to test blood-brain barrier (BBB) disruption, antioxidant capacity, and neuroinflammation. Behavioral tests revealed that MB pretreatment significantly ameliorated ISO exposure-induced cognitive deficits. In addition, MB pretreatment alleviates neuronal injury, apoptosis, and degeneration. Furthermore, the BBB integrity was preserved by MB pretreatment. Additional studies revealed that ISO-induced excessive mitochondrial fragmentation, oxidative stress, and neuroinflammation were significantly attenuated by MB pretreatment in the PND rat model. Our findings suggest that MB pretreatment alleviates ISO exposure-induced brain injury and memory loss for the first time, supporting MB pretreatment as a promising approach to protect the brain against neonatal ISO exposure-induced postoperative cognitive dysfunction.

Highly efficient persistent luminescent nanozymes-based luminescence-colorimetric dual-mode sensor for total antioxidant capacity assay

Total antioxidant capacity (TAC) is a vital parameter to assess the gross antioxidant activity in biological and food matrices. Accurate determination of TAC has become increasingly important for health monitoring and dietary guidance. Here, we report a persistent luminescent nanozyme (ZGO-Pt) based luminescence-colorimetric dual-mode sensor for selective determination of TAC. ZGO-Pt was fabricated based on ZnGa2O4:Cr nanoparticles (ZGO) with platinum nanoflower as synthetic additive. The prepared ZGO-Pt exhibited fascinating peroxidase-mimetic activity and persistent luminescence, and enabled effectively catalysis of the oxidation of colorless 3,3′,5,5′-tetramethylbenzidine (TMB) to blue oxTMB. Meanwhile, the generated oxTMB gave strong absorption at 652 nm to quench the luminescence of ZGO-Pt at 700 nm via the inner filter effect. Antioxidants could compete with the oxidable TMB or reduce oxTMB to TMB, resulting in the simultaneous variations of blue color and luminescence to provide the basis for dual-mode assay for TAC. The highly efficient catalysis of ZGO-Pt (Km, 3.12 mM for H2O2, 0.592 mM for TMB) allows the dual-mode sensor to realize precise determination of TAC with a wide linear range (0.5–200 μM and 1–300 μM for colorimetric and luminescence mode, respectively) and low limit of detection (LOD, 0.05 μM and 0.78 μM for colorimetric and luminescence mode, respectively). The developed dual-mode sensor was successfully applied to the determination of TAC in vitamin tablet, beverages and fruits. This work provides an efficient persistent luminescent nanoparticle-based nanozyme for a facile TAC assay with further vision in biosensing and food technology.

E-cadherin and TAC in GCF accurately discriminate periodontal health and disease.

To investigate the accuracy of gingival crevicular fluid (GCF) E-cadherin and total antioxidant capacity (TAC) to discriminate periodontal health from disease. GCF samples were collected from participants with periodontal health (control), gingivitis, and periodontitis (n = 25 each group). The latter group was further subdivided according to stage (S) and grade. Periodontal parameters were recorded then levels of biomarkers were assayed using ELISA and antioxidant status by use of the Total Antioxidant Capacity Assay for E-cadherin and TAC, respectively. All periodontal parameters were significantly higher in periodontally diseased groups than controls. The GCF E-cadherin significantly increased in gingivitis and periodontitis (S2 to S4) cases as compared to controls. Level of this protein in GCF samples from periodontitis S3 was significantly higher than in gingivitis and S2 groups. The GCF-TAC level was significantly higher in controls than in periodontally diseased groups. No significant differences were observed in the levels of these proteins between grade B and C periodontitis. Both molecules could discriminate periodontal health from gingivitis and periodontitis stages and differentiating periodontitis S3 from gingivitis and other periodontitis stages. Levels of TAC and unbounded E-cadherin in GCF samples exhibited promising diagnostic abilities to differentiate periodontal health and disease.

Antioxidant, photoprotective, cytotoxic, and antimicrobial activities of Albuca amoena Batt.

Introduction: Albuca amoena Batt. is an endemic plant from Morocco used for treating melanoma or leishmaniasis. The aim of this study is to test the antioxidant, photoprotective, cytotoxic, and antimicrobial properties of this plant. Methods: The extracts of the aerial and bulbous parts were obtained by hydroalcoholic maceration of the plant. Their antioxidant potentials were evaluated in vitro using four methods of 2,2-diphenyl-1picrylhydrazil (DPPH), 2,2’-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), ferric reducing antioxidant power (FRAP) assay, and total antioxidant capacity assay (TAC). The photoprotective properties were evaluated through the study of UVA and UVB absorbance and antityrosinase activity. The cytotoxic activity was determined against two breast cancer cell lines (MCF-7 and MDA-MB-231) and one human colorectal cancer cell line (HT-29) using the 3-[4,5-Dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) method. In addition, the antimicrobial activity was tested on six pathogens: Pseudomonas aeruginosa, Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis, Staphylococcus aureus, and Candida albicans by using the disc diffusion method. Results: The results showed that the UVA and UVB absorptive proprieties of the extracts were similar to those used as positive controls: Methyl salicylate and zinc oxide. Moreover, the antioxidant and antityrosinase properties of the studied extracts were found to be weakly active. The extracts had strong cytotoxicity activities, especially against HT-29 cell line. However, the extracts did not show any antibacterial activity. Conclusion: According to the results of this study, the extracts can be used as a sunscreen to avoid dermal anomalies and as a new source of cytotoxic bioactive compound.

Ultrathin porous PdCu metallenezymes as oxidase mimics for colorimetric analysis.

Ultrathin porous and highly curved two-dimensional PdCu alloy metalleneare shown to be highly efficient oxidase mimics. Serving as intrinsic oxidase mimic, the ultrathin porous structure of the PdCu metallenezymes could effectively utilize all the Pd atoms of the metallenezymes during catalytic reactions. By using the oxidation capability of 3,3'5,5'-tetramethylbenzidine as distinctive chromogenic substrate, the PdCu metallenezymes was used as oxidase-like mimics for determination of total antioxidant capacity (TAC) of vitamin C containing real products including fresh orange juice, commercial beverages, Vitamin C tablets and dermo-cosmetic products. AAP was hydrolyzed using ALP to generate AA and the corresponding ALP activity was successfully detected in the0-100 U/L rangewith a lowest detection limit of 0.9 U/L. This study demonstrates the significant catalytic performance and oxidase-like activity of PdCu metallene nanozyme providing astrategy to develop a TAC assay for the assessment of antioxidant food quality as well as oxidative stress in skin and health care products.

Antimicrobial, Antidiabetic, Antioxidant, and Anticoagulant Activities of Cupressus sempervirens In Vitro and In Silico.

In the last decade, the urgent need to explore medicinal plants or drug development has increased enormously around the world to overcome numerous health problems. In the present investigation, HPLC indicated the existence of 18 phenolic and flavonoid compounds in the Cupressus sempervirens extract. Hesperetin represents the greatest concentration (25,579.57 µg/mL), while other compounds, such as pyro catechol, rutin, gallic acid, chlorogenic acid, naringenin, and quercetin, were recognized in concentrations of 2922.53 µg/mL, 1313.26 µg/mL, 1107.26 µg/mL, 389.09 µg/mL, 156.53 µg/mL, and 97.56 µg/mL, respectively. The well diffusion method documented the antibacterial/antifungal activity of C. sempervirens extract against E. faecalis, E. coli, C. albicans, S. typhi, S.aureus, and M. circinelloid with 35, 33, 32, 25, 23, and 21 mm inhibition zones, respectively, more than the standard antibiotic/antifungal agent. Low values ranging from 7.80 to 15.62 µg/mL of MIC and MBC were recorded for E. faecalis, E. coli, and C. albicans. From the 1- diphenyl-2-picryl hydrazyl (DPPH) assay, promising antioxidant activity was recorded for C. sempervirens extract with IC50 of an 8.97 µg/mL. Moreover, ferric reducing antioxidant power (FRAP) and total antioxidant capacity assays (TAC) confirmed the antioxidant activity of the extract, which was expressed as the ascorbic acid equivalent (AAE) of 366.9 ± 0.2 µg/mg and 102 ± 0.2 µg/mg of extracts, respectively. α-amylase and α-glucosidase inhibition % were determined to express the antidiabetic activity of the extract in vitro, with promising IC50 value (27.01 µg/mL) for α-amylase compared to that of acarbose (50.93 µg/mL), while IC50 value of the extract for α-glucosidase was 19.21µg/mL compared to that of acarbose 4.13 µg/mL. Prothrombin time (PT) and activated partial thromboplastin time (APTT) revealed the role of C. sempervirens extract as an anticoagulant agent if compared with the activity of heparin. Binding interactions of hesperetin and gallic acid were examined via the Molecular Operating Environment (MOE) Dock software against E. faecalis (PDB ID: 3CLQ), C. albicans (PDB ID: 7RJC), α-amylase (PDB ID: 4W93), and α-glucosidase (PDB ID: 3TOP). The obtained results shed light on how molecular modeling methods might inhibit the tested compounds, which have the potential to be useful in the treatment of target proteins.

Screening of Phytochemical, Antimicrobial, and Antioxidant Properties of Juncus acutus from Northeastern Morocco.

Juncus acutus, acknowledged through its indigenous nomenclature "samar", is part of the Juncaceae taxonomic lineage, bearing considerable import as a botanical reservoir harboring conceivable therapeutic attributes. Its historical precedence in traditional curative methodologies for the alleviation of infections and inflammatory conditions is notable. In the purview of Eastern traditional medicine, Juncus species seeds find application for their remedial efficacy in addressing diarrhea, while the botanical fruits are subjected to infusion processes targeting the attenuation of symptoms associated with cold manifestations. The primary objective of this study was to unravel the phytochemical composition of distinct constituents within J. acutus, specifically leaves (JALE) and roots (JARE), originating from the indigenous expanse of the Nador region in northeastern Morocco. The extraction of plant constituents was executed utilizing an ethanol-based extraction protocol. The subsequent elucidation of chemical constituents embedded within the extracts was accomplished employing analytical techniques based on high-performance liquid chromatography (HPLC). For the purpose of in vitro antioxidant evaluation, a dual approach was adopted, encompassing the radical scavenging technique employing 2,2-diphenyl-1-picrylhydrazyl (DPPH) and the total antioxidant capacity (TAC) assay. The acquired empirical data showcase substantial radical scavenging efficacy and pronounced relative antioxidant activity. Specifically, the DPPH and TAC methods yielded values of 483.45 ± 4.07 µg/mL and 54.59 ± 2.44 µg of ascorbic acid (AA)/mL, respectively, for the leaf extracts. Correspondingly, the root extracts demonstrated values of 297.03 ± 43.3 µg/mL and 65.615 ± 0.54 µg of AA/mL for the DPPH and TAC methods. In the realm of antimicrobial evaluation, the assessment of effects was undertaken through the agar well diffusion technique. The minimum inhibitory concentration, minimum bactericidal concentration, and minimum fungicidal concentration were determined for each extract. The inhibitory influence of the ethanol extracts was observed across bacterial strains including Staphylococcus aureus, Micrococcus luteus, and Pseudomonas aeruginosa, with the notable exception of Escherichia coli. However, fungal strains such as Candida glabrata and Rhodotorula glutinis exhibited comparatively lower resistance, whereas Aspergillus niger and Penicillium digitatum exhibited heightened resistance, evincing negligible antifungal activity. An anticipatory computational assessment of pharmacokinetic parameters was conducted, complemented by the application of the Pro-tox II web tool to delineate the potential toxicity profile of compounds intrinsic to the studied extracts. The culmination of these endeavors underpins the conceivable prospects of the investigated extracts as promising candidates for oral medicinal applications.

The effect of Moringa oleifera crude extract on liver cell line, HepG2

BackgroundThe liver plays a crucial role in the body's metabolic and detoxification processes. Given its importance, compromised liver function can negatively impact the body’s metabolic and physiological function. Liver diseases can result from several factors, including exposure to toxins, alcohol consumption, and viral infections. Therefore, finding natural remedies for liver protection and treatment is important. Moringa oleifera is a tree known for its various medicinal properties, including hepatoprotective effects. This study aimed to investigate the potential of M. oleifera seed extract in protecting liver cells.MethodsIn this study, dried-seed powder of M. oleifera was extracted using extraction solvents, methanol, and ethanol. HepG2 cells were cultured and treated with different concentrations of the extracts. The antioxidative activity, cell viability, and antiproliferation were assessed using the total antioxidant capacity assay (TAC) and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. Additionally, liver enzyme activity was determined through alkaline phosphatase and aspartate aminotransferase activity assays.ResultsThe extracts had varying effects on liver cells depending on the concentration and time of exposure. Lower concentrations (50 mg/l and 100 mg/l) have mild stimulatory effects/minimal impact on metabolic activity, while higher concentrations (200 mg/l and 400 mg/l) tend to decrease metabolic activity, especially at later time points. Moreover, the extracts effectively reduced the levels of the liver enzyme AST, indicating their ability to mitigate liver injury.ConclusionThe study concludes that the crude extracts of M. oleifera seeds exhibit potential as a natural remedy for liver diseases. The effects of M. oleifera extract suggest that it has potential as a preventive and therapeutic agent for liver damage. This study highlights the importance of exploring natural remedies for liver protection and treatment.
