Molecular Cloning and Functional Characterization of a Lepidopteran Insect β4-N-Acetylgalactosaminyltransferase with Broad Substrate Specificity, a Functional Role in Glycoprotein Biosynthesis, and a Potential Functional Role in Glycolipid Biosynthesis

A degenerate PCR approach was used to isolate a lepidopteran insect cDNA encoding a beta4-galactosyl-transferase family member. The isolation and initial identification of this cDNA was based on bioinformatics, but its identification as a beta4-galactosyltransferase family member was experimentally confirmed. The newly identified beta4-galactosyltransferase family member had unusually broad donor and acceptor substrate specificities in vitro, as transferred galactose, N-acetylglucosamine, and N-acetylgalactosamine to carbohydrate, glycoprotein, and glycolipid acceptors. However, the enzyme preferentially utilized N-acetylgalactosamine as the donor for all three acceptors, and its derived amino acid sequence was closely related to a known N-acetylgalactosaminyltransferase. These data suggested that the newly isolated cDNA encodes a beta4-N-acetylgalactosaminyltransferase that functions in insect cell glycoprotein biosynthesis, glycolipid biosynthesis, or both. The remainder of this study focused on the role of this enzyme in N-glycoprotein biosynthesis. The results showed that the purified enzyme transferred N-acetylgalactosamine, but no detectable galactose or N-acetylglucosamine, to a synthetic N-glycan in vitro. The structure of the reaction product was confirmed by chromatographic, mass spectroscopic, and nuclear magnetic resonance analyses. Co-expression of the new cDNA product in insect cells with an N-glycoprotein reporter showed that it transferred N-acetylgalactosamine, but no detectable galactose or N-acetylglucosamine, to this N-glycoprotein in vivo. Confocal microscopy showed that a GFP-tagged version of the enzyme was localized in the insect cell Golgi apparatus. In summary, this study demonstrated that lepidopteran insect cells encode and express a beta4-N-acetylgalactosaminyltransferase that functions in N-glycoprotein biosynthesis and perhaps in glycolipid biosynthesis, as well. The isolation and characterization of this gene and its product contribute to our basic understanding of insect protein N-glycosylation pathways and to the growing body of evidence that insects can produce glycoproteins with complex N-glycans.

α1,3Fucosyltransferases in cystic fibrosis airway epithelial cells

Cystic fibrosis (CF) has a glycophenotype of aberrant sialylation and/or fucosylation. The CF glycophenotype is expressed on membrane glycoconjugates of CF airway epithelial cells as increased fucosyl residues in α1,3/4 linkage to N-acetyl glucosamine, decreased fucosyl residues in α1,2 linkage to galactose and decreased sialic acid. To define the cause of this phenotype, the enzyme activity of α1,3fucosyltransferase (FucT) was examined in extracts of CF airway epithelial cells with a variety of low molecular weight substrates. Using Galβ1,4GlcNAc as substrate, the activity was divided into 66% α1,3FucT and 34% α1,2FucT. mRNA expression examined with probes to FucTIII, IV, and VII showed that the highest expression of two CF cell lines was for FucTIV. Only one CF cell line expressed mRNA for FucTIII. The non CF airway epithelial cells had significant enzyme activity for α1,3FucT and strong mRNA expression for FucTIV. Thus as reported previously for α1,2FucT, the biochemical capacity for α1,3FucT was present in both the CF and non CF cells and can not be the cause of the CF glycophenotype. These results support the hypothesis that wild type CFTR acts in the Golgi and when mutated as in CF, faulty compartmentalization of terminal glycosyltransferases results, yielding the CF glycophenotype.

Sulphation of the salivary mucin MG1 (MUC-5B) is not correlated to the degree of its sialylation and is unaffected by cystic fibrosis.

Defective acidification of intracellular organelles, particularly the trans-Golgi network, has been proposed to explain the decreased sialylation and increased sulphation of secreted proteins in cystic fibrosis (CF). To test this hypothesis we compared expression of sulphate and sialic acid on three salivary mucins namely MG1 (MUC-5B), MG2 (MUC-7) and GL. Proteins in whole mouth saliva (WMS) from four individuals were separated by fast protein liquid chromatography (FPLC) on a Superdex 200 column and the partially purified mucins slot-blotted and assayed for sulphate content by staining with Alcian Blue. Sulphation varied with the individual and with the mucin: MG1 was the most sulphated and contributed almost the entire sulphate content of WMS. These results allowed us to test small volumes of WMS from 20 CF patients and age- and sex-matched controls for estimates of sulphate content on MG1. Wherever possible sulphate on MG1 was also visualised by staining washed SDS-PAGE gels with Alcian Blue at pH 1.0. To assess the sialic acid content of salivary mucins, electroblots of SDS-PAGE gels were probed with labelled Triticum vulgaris agglutinin. In summary, our results show MG1 to be the main sulphated protein in whole mouth saliva and there are large differences in the expression of sulphate and of sialic acid on this mucin, both in control and CF groups. CF led neither a decrease in sialylation nor an increase in sulphation and direct comparisons of sialic acid content with sulphate in MG1 failed to reveal any obvious link between the two in health or in disease. Our data thus do not support the hypothesis of defective acidification as the underlying cause of altered glycosylation in CF, but point instead to inter-individual differences in expression/functioning of terminal glycosyltransferases for published observations. The Gene Encoding Murine α1,3-Galactosyltransferase Is Expressed in Female Germ Cells but Not in Male Germ Cells

An essential step in murine fertilization is the binding of acrosome-intact sperm to specific O-linked glycans on zona pellucida glycoprotein 3 (ZP3). While there is agreement on the primary role of O-linked glycans in sperm-ZP3 binding, there is a striking lack of consensus on both the terminal monosaccharide(s) required for a functional binding site and the cognate protein on the sperm cell surface that recognizes this glycan. Much current debate centers on the essential role of nonreducing terminal N -acetyl-glucosaminyl or alternatively, α-galactosyl residues, to form a functional sperm binding ligand. Relevant to this debate, we demonstrated that α1,3-galactosyltransferase (α3-GT), which adds nonreducing terminal α-galactosyl residues to glycans, is not expressed in murine spermatocytes or spermatids. The objectives of this study were to determine whether α3-GT is expressed in female germ cells and to compare the pattern of expression of two other terminal glycosyltransferases, β1,4-galactosyltransferase (β4-GT) and α2,6-sialyltransferase (α6-ST), between male and female germ cells. Total RNA was isolated from growing oocytes obtained from 15-day-old animals, fully grown oocytes, and eggs as wall as spermatogonia, spermatocytes, and spermatids. The presence of α3-GT, β4-GT, and α6-ST mRNAs was analyzed by an RT-PCR-based assay. Our data demonstrate that the α3-GT gene is expressed in female germ cells, but not in male germ cells. In contrast, both β4-GT and α6-ST are expressed during oogenesis and spermatogenesis. This differential expression of α3-GT in female germ cells is consistent with the model of sperm-egg binding in which a nonreducing terminal α-galactosyl residue is required for a functional determinant on ZP3 and with our hypothesis that the biological significance for the suppression of α3-GT expression in male germ cells is to prevent sperm-sperm aggregation.

Biochemical Characterization of Liver Microsomal, Golgi, Lysosomal, and Serum β-Glucuronidases in Dibuty1 Phosphate-Treated Rats1

Organophosphate compounds are known to cause the selective release of rat liver microsomal beta-glucuronidase into plasma. To investigate the alterations of molecular forms and oligosaccharide moieties of liver beta-glucuronidase in organophosphate compound-administered rats, beta-glucuronidase was isolated from microsomal, Golgi, lysosomal, and serum fractions. In SDS-polyacrylamide gel electrophoresis, a single polypeptide band was observed on gels in Golgi and serum beta-glucuronidases. This result indicated that Golgi and serum beta-glucuronidases of treated rats did not undergo post-translational proteolytic processing, in contrast to those in control rat livers. Biochemical characterization of the isolated beta-glucuronidases by employing lectin affinity chromatography revealed that interaction of serum and Golgi enzymes with Ricinus communis agglutinin- and wheat germ agglutinin-Sepharose was fairly strong, and that microsomal and lysosomal enzymes were poorly retained on those columns. These results suggested that the serum and Golgi beta-glucuronidases are sialoglycoproteins. A clearance study also showed that infused serum beta-glucuronidase was slowly cleared from plasma with a half-life of about 60 min, but the asialo-serum enzyme was rapidly cleared with a half-life of about 5 min. These results imply that microsomal beta-glucuronidase undergoes extensive modification of the oligosaccharide moieties by terminal glycosyltransferases at the trans Golgi when it is destined for secretion into serum in response to treatment with an organophosphate compound.

Oligosaccharide reprocessing and recycling of a cell surface glycoprotein in cultured rat hepatocytes.

The metabolism of the cell surface glycoprotein dipeptidyl peptidase IV(DPPIV) was studied in cultured rat hepatocytes. In pulse-chase labelling experiments using L-[35S]methionine a 100-kDa high-mannose precursor polypeptide is converted into the mature complex-type 110-kDa glycoprotein. Digestion with exo- and endoglycosidases and metabolic labelling with radioactive sugars demonstrate that the 110-kDa form contains about 6 complex-type oligosaccharides which are fucosylated and sialylated. About 25 min after the beginning of the pulse-labelled glycoprotein appears in the sinusoidal membrane. Physiologically only the 110-kDa form is found in the cell surface. If cell surface DPP IV was desialylated by sialidase at 4 degrees C, it is resialylated during incubation at 37 degrees C. This oligosaccharide reprocessing indicates that the surface glycoprotein has been recycled to the cell compartment containing terminal glycosyltransferases (presumably the trans Golgi system). Two different methods demonstrate internalization of cell surface DPP IV: 1) The complex cell surface DPPIV -anti-DPP IV-antibody -L-[35S]methionine-labelled secondary goat-anti-mouse-antibody formed at 4 degrees C becomes less accessible to trypsin during incubation at 37 degrees C. 2) Part of the complex plasma membrane DPP IV-anti-DPP IV-antibody formed in the cold cannot be recognized by the radioactive secondary antibody after rewarming. Internalization is not blocked by inhibition of protein synthesis with cycloheximide. During internalization of plasma membrane DPP IV its concentration in the membrane remains constant.(ABSTRACT TRUNCATED AT 250 WORDS)

Regulated expression of alpha 2,6-sialyltransferase by the liver-enriched transcription factors HNF-1, DBP, and LAP.

Tissue-specific carbohydrate structures are thought to result from the selective expression of specific terminal glycosyltransferases responsible for their synthesis. However, little is known about the regulation of the expression of these enzymes. Previous analysis of the distribution of one such enzyme, beta-galactoside alpha 2,6-sialyltransferase, revealed that its expression is tissue restricted, with highest levels being found in the liver. Examination of the gene suggested that its expression is regulated at the level of transcription by multiple promoters, one of which is strongly active in the liver. In this present work, an analysis of the liver-restricted promoter was undertaken to identify the promoter elements necessary for liver-restricted expression. Footprinting studies, 5' deletion analysis, and site-directed mutagenesis identified two cis-elements which were potentially important in the tissue-specific expression of this promoter. One of these elements contains a consensus binding site for the liver-enriched transcription factor hepatocyte nuclear factor-1 alpha, while the other is a consensus binding site for the liver-specific factors D-binding protein and liver-enriched transcriptional activator protein. Expression vectors containing cDNAs of these factors are capable of trans-activating transcription of the alpha 2,6ST promoter, demonstrating their ability to regulate transcription of this promoter. Together, these results suggest that tissue-specific glycosylation can be regulated at the level of transcription by the same factors involved in the expression of a number of other tissue-specific genes.

Alteration of Terminal Glycosylation Sequences on N-linked Oligosaccharides of Chinese Hamster Ovary Cells by Expression of β-Galactosideα2,6-Sialyltransferase

Abstract In this report we describe the alteration of the N-linked oligosaccharide terminal sequences of Chinese hamster ovary cell glycoproteins by expression of a beta-galactoside alpha 2,6-sialyltransferase cDNA. While wild type cells normally produce sugar chains terminating in the NeuAc alpha 2,3Gal linkage, the expressed enzyme competes with the endogenous sialyltransferase to attach an alternative terminal sequence, NeuAc alpha 2,6Gal. Subcellular localization of the NeuAc alpha 2,6Gal product by lectin-gold electron microscopy revealed localization throughout the Golgi apparatus cis to trans, post-Golgi membranes and vesicular structures. The results demonstrate the potential for purposefully altering terminal carbohydrate structures in vivo by mis-expressing terminal glycosyltransferases that compete with the endogenous enzyme normally produced by the cells.

ATP-coupled transport of vesicular stomatitis virus G protein. Functional boundaries of secretory compartments.

The oligosaccharide processing intermediates of the vesicular stomatitis virus strain ts045 G protein were used to identify ATP- and temperature-sensitive steps in the constitutive pathway of protein transfer to the cell surface. In addition to the initial ATP-sensitive step required for export from the endoplasmic reticulum (Balch, W. E., Elliott, M. M., and Keller, D. S. (1986) J. Biol. Chem. 261, 14681-14689), two distinct ATP-sensitive steps functionally dissect the Golgi into at least 3 compartments: a cis compartment containing the trimming enzyme mannosidase I, a medial compartment conferring resistance to endoglycosidase H, and a trans compartment containing terminal glycosyl transferases. A fourth ATP-sensitive step is required for export of G protein from the trans Golgi to the cell surface. A high threshold of cellular ATP (70% of the control) was required for maximal rates of transport between Golgi compartments. Transport between compartments is inhibited at 40% of the normal cellular ATP pool. Only a single temperature-sensitive step localized to the endoplasmic reticulum inhibited transport of ts045 G protein to the cell surface. The data suggest that ATP-sensitive steps punctuate transport of protein between compartmental boundaries of the secretory pathway.

Differential subcompartmentation of terminal glycosylation in the Golgi apparatus of intestinal absorptive and goblet cells.

Two terminal glycosyltransferases, a sialyltransferase and the blood group A alpha 1,3 N-acetylgalactosaminyltransferase, were found to exhibit differential subcompartmentation in the Golgi apparatus of intestinal goblet and absorptive cells. As expected from their role in terminal glycosylation, the two glycosyltransferases and their products, sialic acid residues and blood group A substance, were localized in the trans cisternae of the Golgi apparatus of goblet cells. In contrast, however, they were found throughout the Golgi apparatus stack of adjacent absorptive cells, with the exception of the fenestrated first cis cisterna. The results are in contrast to the general view that enzymes in the glycosylation pathway are arranged in a cis to trans gradient across the Golgi apparatus and that such polarized distributions may instead be cell type-specific.

Responsiveness of glycosyltransferases to thyrotropin in a serum-free culture of porcine thyroid cells

Administration of thyrotropin to porcine thyroid follicles, obtained in a serum-free chemically defined medium, provoked marked increases in the activities of several glycosyltransferases involved in protein N-glycosylation. The coincidence of these effects with a previously demonstrated enhancement of thyroglobulin production renders a relationship between these events likely. The most important stimulation was for peptide oligosaccharyltransferase (3-fold). Among the enzymes involved in the synthesis of the lipid oligosaccharide donor, Dol-P glycosyl- and mannosyltransferases were increased 1.5-fold, and Dol-P N-acetylglucosaminylphosphotransferase only 1.15-fold. As regards terminal glycosyltransferases, asialofetuin sialyltransferase was increased 2-fold and ovomucoid galactosyltransferase only 1.2-fold. There was a continuous release of the latter two enzymes into the culture medium.

Effect of antimicrotubule agents on terminal glycosyltransferases and other enzymes associated with rat liver subcellular fractions.

Previous studies have shown that anti microtubule agents disrupt Golgi complexes in hepatocytes and other cells, causing breakdown or vesiculation of Golgi cisternal membranes. Whether this change in the structure of the Golgi membranes is associated with changes in Golgi membrane function is not known. The present study was initiated to investigate this issue; i.e., to determine whether anti-microtubule agents that cause structural changes in Golgi membranes in vivo would, at the same time, affect characteristic enzyme functions of Golgi membranes. To this end, colchicine was given to young rats in vivo and various hepatic subcellular membranes were subsequently isolated and utilized for enzyme assays. Initially it was shown that colchicine (2.5 mg/kg body wt.) given for 5h significantly decreased the activities of the Golgi membrane associated enzymes galactosyl-, sialyl- and N-acetylglucosaminyl-transferases. More detailed experiments indicated that low doses of colchicine (0.8 mg/kg body wt.), although less effective than higher doses, decreased the activities of the terminal glycosylating enzymes maximally at 5h, with partial and complete recovery at 12 and 24h respectively. Treatment in vivo of rats with vinblastine (20 mg/kg body wt.) for 5h mimicked the action of colchicine. Two microsomal glycosylating enzymes (mannosyl and N acetylglucosaminyl transferases) were unaffected by the treatment with colchicine, as were various hepatic 'marker' enzymes such as 5' nucleotidase, glucose 6 phosphatase and succinate: 2-(p iodophenyl)-3-(p nitrophenyl)-5-phenyltetrazolium reductase (succinate dehydrogenase; EC 1.3.99.1), which were found to be enriched in plasma membrane, endoplasmic-reticulum and mitochondrial-membrane fractions respectively. These results show that anti-microtubule agents specifically suppress the activity of Golgi-associated glycosyltransferases in liver. Although it seems likely that these changes are related to the previously observed structural changes in hepatocyte Golgi complexes after colchicine treatment, to what extent the results are linked to the interaction of colchicine with microtubule protein remains to be clarified.

Distribution of terminal glycosyltransferases in hepatic Golgi fractions.

The distribution of the three glycosyltransferases synthesizing the terminal trisaccharide sialic acid yields D-galactose yields N-acetylglucosamine present in many glycoproteins was determined in Golgi fractions prepared from rat liver homogenates by a modification of the procedure of Ehrenreich et al. (1973, J. Cell Biol. 70:671--684). The enzymes were assayed with asialofetuin, ovomucoid, and Smith-degraded ovomucoid as sugar acceptors. Careful adjustment of the pH of all sucrose solutions to 7.0 +/- 0.1 prevented enzyme inactivation, and allowed quantitative recoveries at every isolation step. The three morphologically and functionally different Golgi fractions GJ1, GF2, and GF3 showed (in that order) decreasing specific activities of all three enzymes, but the relative amounts and relative specific activities of the three transferases in any given fraction were nearly identical. Two marginal fractions, one extra heavy (collected on the gradient below GF3) and the other extra light (isolated by flotation from the postmicrosomal supernate) were found to contain recognizable Golgi elements. An enrichment of any transferase over the two others was not detected in either preparation. A partial release of content from a combined GF1+2 was achieved by treatment with the nonionic detergent Triton X-100. Low Triton/phospholipid ratios (less than 2 mg/mg) led to lysis of the vesicles and cisternae and loss of very low density lipoprotein particles (ascertained by electron microscopy), but failed to separate the transferases from each other; the three enzymes sedimented together with a population of empty vesicles to a density of approximately 1.08 g/ml.
