Advancing fishing technology education and research: a 65-year legacy at Nha Trang University, Vietnam

As a coastal country, Vietnam relies on marine exploitation for food security and livelihoods. Training human resources to research, exploit, and manage marine resources is an important strategy to maintain rapid and sustainable economic growth and development. Official training in fishing technology at Nha Trang University (NTU) started in 1959. With 65 years of teaching, the unique program only offered at NTU, has made significant contribution to social-economic development in general and sustainable marine fisheries in particular. In this paper, we review the major achievements in teaching and scientific research in fishing technology at NTU as well as highlight the challenges and progress. Over the years, thousands of students have been trained under the fishing technology program and they then have worked all over the country, working in fisheries management, science, and services. Hundreds of research projects and peer-reviewed papers have been conducted and published. Those have supported fishing efficiency, environmentally friendly fishing methods, and effective management. However, the number of fishing technology students has decreased during the past few years because of unfavorable study and working environments where students are often exposed to commercial fishing vessels that frequently operate under rough weather conditions. Despite challenges and difficulties, NTU is determined to maintain the program to support the ocean economic development of the nation.

Novel Application of Non-Invasive Methodological Approaches in Biomedical Sciences Towards Better Understanding of Marine Teleost Ocular Health and Disease.

Seafood is an important resource for global nutrition and food security, with both land and marine aquaculture playing pivotal roles. High visual acuity is key for health and survival of farmed, cultured, and wild fish. Cleaner fish technology to control parasite infestation has become important in marine aquaculture and highlights the importance of visual acuity in the efficacy of cleaner fish species. New clinical diagnostic approaches towards understanding and optimising fish visual health could benefit both aquacultured and wild fish populations. Opportunities for developing and using advanced non-invasive clinical assessment and diagnosis of ocular health in wild, cultured, and experimental fish are key to more rapidly realising how threats to eye health in these animals might be better understood and mitigated. Ophthalmoscopy can rapidly and non-invasively image anatomical aspects of retinal and anterior ocular tissues and has been used in mammalian biomedicine since the turn of the 20th century. More now than ever, labour-intensive post-mortem approaches for ocular analysis such as histology are increasingly being replaced or supplemented by application of various forms of optical coherence tomography (OCT) imaging of ocular tissues in mammalian biomedicine. Advances and availability of other methodological approaches such as three-dimensional printing and computer science make instrument customisation affordable and adaptable. This review article will outline how ophthalmoscopy, OCT, and other methodologies are being applied towards understanding ocular health in teleost fish species and will describe some of the future opportunities that technological advances might afford in advancing ocular imaging in fish health and disease in general.

Comprehensive mapping of molecular cytogenetic markers in pitaya (Hylocereus undatus) and related species.

Pitaya (Hylocereus undatus; 2n=22) is an important fruit crop from the Cactaceae family, originally domesticated in Mexico and the USA, and is now widely cultivated for its nutritional benefits. It is characterized by its distinctive triangular-shaped stems and large, showy flowers, thriving in arid and semi-arid environments, particularly in hot, dry climates. However, systematic chromosomal studies, including chromosomal mapping of cytogenetic markers in pitaya, are limited, presenting challenges for its cytogenetic improvement. To address this issue, we designed oligo-barcodes specific to thirty-three chromosome regions based on the pitaya reference genome and applied them to both pitaya and cactus (Selenicerus grandifloras; 2n=22) for oligo-barcodes mapping, karyotyping, and chromosome identification. We utilized FISH technology, employing oligo, rDNA, and tandem repeat probes for chromosomal mapping, identification, and karyotyping of pitaya and related species. We successfully localized oligo-barcodes on eleven pairs of chromosomes in both pitaya and cactus, demonstrating the effectiveness of the synthesized oligo-barcodes. We used two ribosomal DNA (rDNA) probes (45S and 5S) and two tandem repeat probes (GTR11 and STR3) in pitaya (both diploid and tetraploid) and two other Cactaceae species (S. grandifloras and Opuntia humifusa; 2n=40) for chromosomal mapping. The analysis of rDNA distribution and CMA (Chromomycin A3) banding across different chromosomes in pitaya and cacti highlights the concept of conserved rDNA. This study provides fundamental insights into cytogenetic markers and their localization across different chromosomes in pitaya and other Cactaceae species.

The Influence of Light Colour on the Behaviour of Atlantic Cod in an Experimental Setting

Fishing technologies often exploit the visual sensitivity of target species to alter their behaviours. Atlantic cod (Gadus morhua Linnaeus, 1758) are an economically important species, commonly targeted by fisheries in the North Atlantic, yet the behaviour of adult Atlantic cod in reaction to the simultaneous presentation of various light stimuli has not been assessed in an isolated setting to determine if there is a preference for certain light qualities. To assess the influence that artificial light may have on the behaviour of Atlantic cod, we investigated the movement of wild-caught cod in a laboratory setting with green, blue, and white light with a blank control presented in paired choice tests. We predicted that green light would have the greatest influence on cod behaviour and that they would preferentially spend more time in proximity to that light condition. The findings show that cod behaved consistently across trials and across experimental sessions, and that the right side of the arena was preferred regardless of the colours of light presented. These findings indicate that cod do not prefer to spend more time in proximity to certain colours of light in an artificial environment, which has direct implications for animal husbandry, fisheries research, and behavioural ecology.

Diversifikasi Produk Perikanan Lokal Berkelanjutan Melalui Inovasi Teknologi Berbasis Energi Terbarukan di Kelurahan Sibatua

This community service program aims to support the diversification of sustainable local fishery products through the application of renewable energy-based technological innovations in Kelurahan Sibatua. Using the Participatory Action Research (PAR) approach, this initiative actively involves the community in training, demonstration, and implementation of solar panel technology for fish cooling boxes and processing fishery products into various value-added products such as milkfish nuggets, fish balls, and crackers. The results indicate that the solar panel technology used in the cooling boxes effectively improves the quality and shelf life of fish catches while reducing operational costs that previously relied on fossil energy. Additionally, technical training and mentoring successfully enhanced the community’s skills in fish processing and the use of renewable energy technologies. Evaluation through pre-test and post-test assessments showed a significant improvement in participants' competencies, from basic understanding to the application of technology and sustainable fishery product production.

Carbon Footprint and Automation in Aquaculture

Objective: This paper proposes research on the state of the art of automation and carbon footprint in aquaculture. Theoretical Framework: The development of electronics and automation, which are so prevalent in everyday activities and industries, has contributed to the advancement of automation levels in aquaculture. The use of electrical sensors could be an alternative for estimating the carbon footprint of aquaculture production systems. Method: The methodology was based on scientific research with a qualitative approach on automation and the carbon footprint in aquaculture and observations made during visits to fish farms and fish technology fairs. Results and Discussion: The levels of automation used in aquaculture are still low compared to other economic activities, especially industry. The use of existing technology in fish farms would raise the level of automation in aquaculture, but this transformation faces challenges on the part of fish farmers, such as: cost of access, difficulty of operation and lack of interest in investing in automation. The use of automation and control techniques could corroborate methodologies for estimating the carbon footprint during the animal rearing phase in excavated ponds. Research Implications: Promote discussion on the use of new technologies and influence research on the theme of carbon footprint and automation in aquaculture. Advance research into the need to expand the use of automation in fish farms. Originality/Value: This study contributes to the literature by reviewing the state of the art of automation in aquaculture, demonstrating the need for research to expand the use of technologies both to automate aquaculture farming and to estimate the carbon footprint during farming.

Impact of Integrated Agricultural Productivity Project on the Smallholder Beneficiaries in Mithapukur Upazila, Rangpur District of Bangladesh

In Bangladesh, plenty of development projects are assumed, though success of very few is evaluated. This study was aimed to evaluate the impact of Integrated Agricultural Productivity Project (IAPP) on the smallholder beneficiaries. The proportionate random sampling technique was used to select 90 smallholder beneficiaries from seven project villages implemented. Data were collected during 2018 to 2019 using a predesigned interview schedule. The perceived advantages of the project and the beneficiaries' annual income change were utilized to assess the impact of the project on the smallholder beneficiaries. Majority (58.8%) of the smallholder project farmers were under young to middle age category, with literacy belonged to moderate family size, small farm category, average annual income of BDT 134989 and 227633 during before and after the project intervention, respectively. They showed moderate contact with the extension source (95.6%) and moderate organizational participation (64.5%). Benefits derived from different components of the project were caused change in the usages of different agricultural improved technologies by the beneficiaries. The technologies implemented were improved fish, crop, livestock and water technology. All technologies had statistically significant contributions to the beneficiaries. The changes were significant for all the cases, viz. improved fish technology (t=13.65), improved crop technology (t=24.36), improved livestock technology (t = 20.43) and improved water technology (t= 22.29). There was also significant change in annual income comparing before and after the project intervention. Major problems faced during the intervention of the project were poor coordination of activities followed by training, poor quality deliverables and inadequate supply of project incentives. It is recommended that the authority needs to consider necessary measures to sustain the tempo and resolve the problems experienced by the beneficiaries in the future interventions.

SnapFISH-IMPUTE: an imputation method for multiplexed DNA FISH data

Chromatin spatial organization plays a crucial role in gene regulation. Recently developed and prospering multiplexed DNA FISH technologies enable direct visualization of chromatin conformation in the nucleus. However, incomplete data caused by limited detection efficiency can substantially complicate and impair downstream analysis. Here, we present SnapFISH-IMPUTE that imputes missing values in multiplexed DNA FISH data. Analysis on multiple published datasets shows that the proposed method preserves the distribution of pairwise distances between imaging loci, and the imputed chromatin conformations are indistinguishable from the observed conformations. Additionally, imputation greatly improves downstream analyses such as identifying enhancer-promoter loops and clustering cells into distinct cell types. Analisis Faktor Penghambat Ekonomi Keluarga Nelayan Wilayah Pesisir Desa Bagan Percut, Kecamatan Percut Sei Tuan, Kabupaten Deli Serdang, Sumatera Utara

The aim of this research is to analyze the economic inhibiting factors for fishing families in the coastal area of ​​Bagan Percut Village, Percut Sei Tuan District, Deli Serdang Regency, North Sumatra. This research uses data analysis methods. Research results show that access to financial capital is one of the main challenges for fishermen in this region. The majority of fishermen depend on a loan system from local middlemen or "toke", which often provides fish selling terms and prices that are less favorable for fishermen. This causes fishermen to be trapped in a cycle of poverty and prolonged economic difficulties. The low level of education among fishermen has a significant impact on their ability to manage fisheries efficiently and increase the added value of their catch. Lack of knowledge and skills in post-harvest handling, marketing, and the adoption of modern fishing technology and practices are major obstacles in efforts to increase fishermen's productivity and income. Unpredictable weather conditions, limited access to modern technology and equipment, competition with large fishing industries, unsustainable natural resource management, and inadequate infrastructure are also significant economic inhibiting factors for fishing families in the Coastal Area of ​​Bagan Percut Village.

Cytoplasmic light-chain immunofluorescence combined with FISH in bone marrow smears to detect cytogenetic abnormalities in multiple myeloma

Objective: To analyze the sensitivity of cytoplasmic light-chain immunofluorescence with fluorescence in situ hybridization in bone marrow smears (new FISH) for detecting cytogenetic abnormalities in multiple myeloma (MM) . Methods: 42 MM patients admitted to the First Affiliated Hospital of Nanjing Medical University from April 2022 to October 2023 were enrolled. The patients with MM were detected by new FISH and CD138 immunomagnetic bead sorting technology combined with FISH (MACS-FISH) or cytoplasmic immunoglobulin FISH (cIg-FISH) to analyze cytogenetic detection results using combination probes which included 1q21/1p32, p53, IgH, IgH/FGFR3 [t (4;14) ], and IgH/MAF [t (14;16) ]. Results: In 23 patients with MM, the abnormality detection rates of cIg-FISH and new FISH were 95.7% and 100.0%, respectively (P>0.05). The detection rates of 1q21+, 1p32-, p53 deletion, and IgH abnormalities by cIg-FISH and new FISH were consistent, which were 52.2%, 8.7%, 17.4%, and 65.2%, respectively. The results of the two methods further performed with t (4;14) and t (14;16) in patients with IgH abnormalities were identical. The positive rate of t (4;14) was 26.7%, whereas t (14;16) was not detected. In 19 patients with MM, the abnormality detection rates of MACS-FISH and new FISH were 73.7% and 63.2%, respectively (P>0.05). The positivity rate of 1q21+, 1p32- and IgH abnormalities detected by MACS-FISH were slightly higher than those detected by new FISH; however, the differences were not statistically significant (all P values >0.05) . Conclusion: The new FISH method has a higher detection rate of cytogenetic abnormalities in patients with MM and has good consistency with MACS-FISH and cIg-FISH.

Rapid and accurate detection of fish allergen within 30 min using a food nucleic acid releaser combined with rapid real time PCR assay

To achieve rapid on-site nucleic acid detection of fish allergens in food, this study established a new direct and rapid qPCR detection technology for fish. The technology was able to complete nucleic acid extraction in 4 min using a food nucleic acid releaser, and the DNA samples were amplified by rapid qPCR within 25 min. The results showed that direct-rapid qPCR can specifically detect fish, and it can also detect 0.00001 % of fish components in artificially simulated samples, with high detection precision. This method can be used to effectively detect fish in various deep-processed foods. As a simple, rapid, and accurate molecular detection method for fish allergens, this method has broad application prospects for on-site food safety detection.

Pemberdayaan Pembudidaya Ikan di Desa Bukit Bamba melalui Pelatihan Pembuatan “Pahesang” Pakan Herbal Daun Sangkareho (Callicarpa longifolia Lam.)

In freshwater fish cultivation, information and technology on alternative feed, namely feed mixed with herbs, has developed rapidly, one of which is artificial herbal feed from the medicinal plant Sangkareho (Callicarpa longifolia Lam.). This medicinal plant is a typical plant of Central Kalimantan. The aim of implementing this activity is to increase the knowledge and skills of partners or the fish-cultivating community regarding techniques for making herbal feed for fish cultivation with nutritional content formulations that suit the needs of cultivated fish. This activity will be carried out in Bukit Bamba Village, Central Kahayan District, Pulang Pisau Regency, Central Kalimantan with the target partners being fish farmers in Bukit Bamba Village. The method used to achieve this goal is active participatory empowerment of the community through collaboration with partners, namely the transfer of knowledge and technology through training activities on techniques for making herbal feed for cultivating fish in the form of pellets using local forest products, namely Sangkareho leaves, through direct assistance in the field starting from the process of taking and selecting raw materials to the production of herbal feed.

Application and Development of Targeted Fishing Technology in Natural Product Screening - A Simple Minireview

Background: The screening of active ingredients in traditional Chinese medicine is an important task in the modernization of traditional Chinese medicine, and the commonly used analytical means in the past were mainly to screen the extracts of traditional Chinese medicine through pharmacological experiments, but the method has major defects. The target fishing strategy provides a new idea for the screening of active ingredients, and it has rapidly become a hot research direction, but there are many methods that need to be summarized and aggregated. Objective: It aims to provide readers with an understanding of the achievements, developments, and dilemmas of target fishing techniques over the past few years and to provide new ideas for subsequent research. Methods: Research articles in recent years using target fishing as an entry point are used as a basis to summarize the types of literature based on their principles and characteristics and to discuss the advantages and disadvantages of each method. Conclusion: This paper summarizes the classification and development of fishing techniques such as ultrafiltration, equilibrium dialysis, cell membrane chromatography, and immobilization of target molecules and target fishing and describes the principles and characteristics of these methods. The applications of these methods in the active ingredients of traditional Chinese medicine are summarized, and the problems and solutions of these methods are discussed.

When outsiders are resource users: Fishers’ migration in Southwest Panay, Philippines, and its impact on small-scale fisheries

This paper examined the fishers’ migration in Southwest Panay, Philippines, how it has shaped the small-scale fisheries management in the receiving area, influenced local resource access and utilization, and the potential impacts of the phenomenon on fishing technology and practices. The study was conducted in two municipalities in the province of Iloilo (Miagao and San Joaquin) and two in the province of Antique (Anini-y and San Jose de Buenavista). It utilized household surveys, key informant interviews (KII), and focus group discussion (FGD) as research methodologies. Results show that there are various push and pull factors that influence the migrant fishers (locally known as pangayaw) to migrate to other areas. The most common reason cited is the presence of a richer fishing ground or better livelihood opportunities in their host community.Migrant fishers play a vital role in the local fishing economy as sources of new fishing technology. However, results revealed that most of the technologies that they introduced were either more efficient gears (e.g., use of purse seine and gill nets) or modifications (e.g., use of kati or decoy) that would increase the efficiency of the fishing gears. This may have a consequent negative impact on the sustainability of the fishery resources in the host community.It is evident from the available data of the local government units and the observations of the participants through their local ecological knowledge that migration has a potential impact on local resource access and utilization patterns, and may influence the socio-cultural landscape of the receiving area. It poses a challenge to the sustainable utilization of the resources, and exerts additional pressure on the social services of the receiving local governments. Instead of treating them as “outsiders”, they should be treated as partners of the local community and must share the responsibilities of sustainable utilization and management of the fisheries resources.

Implementation Strategy For Fishery Processing Business Based On Blue Economy

This study investigates the readiness and capacity of the East Fakfak district community to transform fish into micro-small scale business products, aiming to contribute to the blue economy's development and enhance their welfare. Employing a semi-qualitative research approach through surveys, the research team, including surveyors, engaged in direct observation and conducted comprehensive interviews across all villages in East Fakfak. The objective was to assess the community's preparedness to process fish into preserved products that are marketable, high-quality, and possess unique organoleptic qualities characteristic of Fakfak. The analysis revealed a prevalent unfamiliarity among the East Fakfak district inhabitants with fish processing technologies. A traditional preference for consuming fresh fish, attributed to the ease of access to fish resources, has significantly hampered the advancement of fishery product processing in the region. This study underscores a critical need for introducing and fostering fish processing knowledge and technologies among the community. By educating and equipping the local populace with the necessary skills and resources to undertake sustainable fish processing, there lies a potential to unlock a new avenue for economic development. Such initiatives would not only diversify income sources but also ensure the longevity and sustainability of fishery resources, pivotal to the prosperity of East Fakfak's blue economy

Pengabdian Diversifikasi Olahan Ikan Slengek

Kawasan Kota Tuo is one of the coastal areas in Bengkulu City. Most of the livelihoods of the residents of Kawasan Kota Tuo are fishermen. One of the fish that many fishermen get from Tuo City is slengek fish. Slengek fish is a typical fish from the Bengkulu region which is usually obtained in large quantities. However, this fish has a fairly cheap price, which is around Rp. 5000, -/Kg. So it is necessary to have information and processing technology that can add added value and shelf life. Therefore, training and counseling were held on the processing of dimsum and nuggets made from slengek fish. After the service was carried out in Kawasan Kota Tuo, the community had obtained information and knowledge about the technology for processing slengek fish into products that have added value, namely dimsum and nuggets and the community had received digital marketing counseling that had never been received before and through this training partners were guided how to use social media as a forum for the promotion of its products so that it is expected to be a stimulant of economic growth.

Processing Trash Fish into Economically Valuable Animal Feed in Nagari Pilubang, Padang Pariaman Regency

There has been concealed on the processing of trash fish into animal feed in Nagari Pilubang, Padang Pariaman Regency. The majority of the Pilubang community are fishermen. However, so far the trash fish caught have not been processed properly. Meanwhile, trash fish has the potential to be used as animal feed because it contains high protein. Through this PKM activity, the community receives education and training regarding the application of technology for processing trash fish into animal feed using appropriate technology. The animal feed was made from trash fish is processed using a fermentation process. From the results of this activity, it is known that community knowledge has increased and is very useful for implementation by the community in Nagari Pilubang

Discovery of drug targets based on traditional Chinese medicine microspheres (TCM-MPs) fishing strategy combined with bio-layer interferometry (BLI) technology

Target discovery of natural products is a key step in the development of new drugs, and it is also a difficult speed-limiting step. In this study, a traditional Chinese medicine microspheres (TCM-MPs) target fishing strategy was developed to discover the key drug targets from complex system. The microspheres are composed of Fe3O4 magnetic nanolayer, oleic acid modified layer, the photoaffinity group (4- [3-(Trifluoromethyl)-3H-diazirin-3-yl] benzoic acid, TAD) layer and active small molecule layer from inside to outside. TAD produces highly reactive carbene under ultraviolet light, which can realize the self-assembly and fixation of drug active small molecules with non-selective properties. Here, taking Shenqi Jiangtang Granules (SJG) as an example, the constructed TCM-MPs was used to fish the related proteins of human glomerular mesangial cells (HMCs) lysate. 28 differential proteins were screened. According to the target analysis based on bioinformatics, GNAS was selected as the key target, which participated in insulin secretion and cAMP signaling pathway. To further verify the interaction effect of GNAS and small molecules, a reverse fishing technique was established based on bio-layer interferometry (BLI) coupled with UHPLC-Q/TOF-MS/MS. The results displayed that 26 small molecules may potentially interact with GNAS, and 7 of them were found to have strong binding activity. In vitro experiments for HMCs have shown that 7 active compounds can significantly activate the cAMP pathway by binding to GNAS. The developed TCM-MPs target fishing strategy combined with BLI reverse fishing technology to screen out key proteins that directly interact with active ingredients from complex target protein systems is significant for the discovery of drug targets for complex systems of TCM.
