Statistical Optimization of Fast Dissolving Tablet Contains Isosorbide Dinitrate for the Treatment of Angina Pectoris

ABSTRACT: The ultimate objective about this experiment was to create a fast-dissolving tablet Isosorbide dinitrate that released its medication quickly. The physical properties of tablets, such as hardness, angle of repose, weight variation, and friability were assessed. In-vitro release was investigated, and a 22-factorial design was created. The results show that the in-vitro release shows that the F2 formulation has the highest release at 20 min. Tablet disintegration time ranges from 31 to 48 s. The hardness ranges from 6-7 kg/cm3. The friability test shows the range from 0.49-0.68%. The weight variation test shows that all the formulation passes the tests. ANOVA statistical analysis revealed that the generated formulations are statistically significant. It is concluded that FDT formulations have optimum and reproducible disintegration time and increased dissolve characteristics, resulting in higher patient compliance.

Effects of Granulated Lactose Characteristics and Lubricant Blending Conditions on Tablet Physical Properties in Direct Powder Compression.

Lactose is an excipient used extensively for bulking, diluting, and molding active pharmaceutical ingredients in tablet manufacturing. Particularly, granulated lactose (GL) intended for direct powder compression has distinct properties due to differences in manufacturing methods. It contributes to handling blended powders for tableting and tablet quality. In this study, we aimed to compare the functions of different forms of GL added as excipients during direct powder compression on the tablet properties and the effect of magnesium stearate (Mg-S) used as a lubricant on each type of GL. Different GL types obtained using different manufacturing methods (agitated granulation, GL-AG; spray-dried granulation, GL-SD; fluidized bed granulation, GL-FB) were blended with maize starch, low-substituted hydroxypropyl cellulose, and paracetamol in a V-type blender for 10 min. Mg-S was added at varying amounts (0.1, 1.0, and 2.0%) and blending times (5, 10, and 30 min) for the nine types of blended powders for tableting formulation. The powders were tableted, and the tablets were evaluated for weight and drug loading variations, tensile strength, friability, and disintegration time. When tablets with the same blending conditions were compared, the tensile strength and disintegration time were in the order of GL-FB > GL-SD > GL-AG. For each GL, we analyzed the effects of changes in the added amount of Mg-S and blending time using contour plots, evaluated the effects of blending conditions on tablet properties, and determined the target tablet properties. We investigated the optimization of the lubricant blending conditions to obtain suitable tablets.

Formulasi Sediaan Tablet Ekstrak Daun Talas (Colocasia esculenta (L.) Schott.) Dengan Variasi Kadar Polivinil Pirolidon (PVP) Sebagai Bahan Pengikat

Taro leaves (Colocasia Esculenta (L.) Schott.) contain alkaloids, saponins, tannins, flavonoids and polyphenols. Utilization of taro leaves requires new innovations by developing ways of making tablets, which increase the convenience of use through more comfortable use and more stable storage compared to liquid pharmaceutical preparations or suspensions. The aim of this study was to determine the effect of the comparison of variations of Polyvinylpyrrolidone (PVP) binder on the physical properties of granules and tablets and to obtain the optimum formula. Tablets were made by wet granulation process with 1%, 3% and 5% PVP. Granules were tested for water content, flow time test, angle of repose and tapping. The resulting tablets were then tested for their physical properties. Research result; Weight uniformity with 5% PVP fulfills the physical requirement of 600 g. Size uniformity fulfills the physical requirements with a yield of 2.91 mm. Hardness with 3% PVP meets the physical requirements of 5.95 kg. Fragility meets the physical requirements of 1% PVP, namely 0.4%. The disintegration time with 1% PVP met the physical requirements, namely 7.88 minutes. The results stated that variations in the concentration of the PVP binder could be formulated into tablets and the tablet binder resistance was measured which met the requirements of the best tablet physical properties. Optimal formulation is achieved in formula III with binder containing 5% PVP.

Solid Dispersion of Acetosal Using Polyvinyl Pyrrolidone (PVP) K-30 in Tablets with Direct Compressing Method

Acetosal is classified in the Biopharmaceutical Classification System (BCS) class II (low solubility, high permeability). Low solubility causes a decreased dissolution rate. Polyvinyl pyrrolidone (PVP) K-30 is an inert carrier easily soluble in water and can influence the solubility of a drug substance. Efforts to increase the solubility of acetosal make a solid dispersion system. This study aims to determine the effect of the solid dispersion system of acetosal: PVP K-30 on dissolution rate, the ratio of the solid dispersion with the best dissolution rate, and the physical properties of acetosal tablets formed in the dispersion system. Solid dispersions using the dissolving method with variations in the concentration of acetosal: PVP K-30 1:1, 1:3, and 1:5. The results of the dissolution test of acetosal in solid dispersion powder, i.e., PVP Formula 1:5, which has the highest dissolution percentage compared to formula 1:1 and 1:3 with the concentration this formula was 140.96 mg, dissolution percentage was 28.19±0,63% in 30 minutes. Statistical results by ANOVA test show a significant difference of 0.044 (p<0.05). The physical properties of tablets with a dispersion system show higher addition of PVP K-30. This result is related to slower disintegration time and lower friability.

Formulation of a 3D Printed Biopharmaceutical: The Development of an Alkaline Phosphatase Containing Tablet with Ileo-Colonic Release Profile to Treat Ulcerative Colitis

Powder bed printing is a 3D-printing process that creates freeform geometries from powders, with increasing traction for personalized medicine potential. Little is known about its applications for biopharmaceuticals. In this study, the production of tablets containing alkaline phosphatase using powder bed printing for the potential treatment of ulcerative colitis (UC) was investigated, as was the coating of these tablets to obtain ileo-colonic targeting. The printing process was studied, revealing line spacing as a critical factor affecting tablet physical properties when using hydroxypropyl cellulose as the binder. Increasing line spacing yielded tablets with higher porosity. The enzymatic activity of alkaline phosphatase (formulated in inulin glass) remained over 95% after 2 weeks of storage at 45 °C. The subsequent application of a colonic targeting coating required a PEG 1500 sub-coating. In vitro release experiments, using a gastrointestinal simulated system, indicated that the desired ileo-colonic release was achieved. Less than 8% of the methylene blue, a release marker, was released in the terminal ileum phase, followed by a fast release in the colon phase. No significant impact from the coating process on the enzymatic activity was found. These tablets are the first to achieve both biopharmaceutical incorporation in powder bed printed tablets and ileo-colonic targeting, thus might be suitable for on-demand patient-centric treatment of UC.

Development and In-vitro, In-vivo Evaluation of Gastro Retentive Floating Tablets of Felodipine

Direct compression was utilized to develop the floating tablets, because of moisture sensitivity of Felodipine(FD), formulation included polymers such as CARBOPOL 934 & HPMC K 15 M. FD Floating tablets were designed to enhance drug availability by prolonging gastric retention time (GRT). Physical properties of tablets, such as hardness, thickness, friability, and weight variation as well as drug content and floating behaviors, were evaluated. Further, tablets were studied for In vitro drug release tests for 12 hours, while floating in the dissolution medium, In- vivo imaging studies were conducted. According to FTIR studies, there is no interaction between the drug and polymer, In-vitro buoyancy of tablets was 12 hours, the in-vitro dissolution release studies exhibited sustained and prolonged drug release profiles. The release mechanism from these tablets has been confirmed to be non-Fickian diffusion, which also fits the zero order and higuchi models, GRT of floating tablets was observed to be 4 hours. Based on in –vitro characteristics, F14 is the most efficient formulation. It was exploited in- vivo imaging studies by incorporating BaSo4, and the floating concept was used to boost gastric retention time, which was initially assumed.

Orally Disintegrating Tablet Manufacture via Direct Powder Compression Using Cellulose Nanofiber as a Functional Additive.

In recent years, orally disintegrating (OD) tablets have been continuously improved to increase efficacy. Herein, we focused on the benefits of cellulose nanofiber (CNF), a highly functional material, in OD tablet manufacturing. We studied its effects on the physical properties of tablets during manufacture. The analyzed tablet formulations included different content CNF (0-50%; 6 preparations), lactose hydrate, acetaminophen, and magnesium stearate (Mg-St). We measured the angles of repose and evaluated the flowability of the powder. Tablets were prepared on a tabletop and rotary tableting presses, whereafter their weight, drug content, hardness, friability, and disintegration time were evaluated. Although CNF addition slightly reduced powder flowability, continuous tableting was feasible via direct powder compression. Tablet hardness (~40 N) was comparable between CNF-containing (20%) tablets and those prepared with crystalline cellulose under 10 kN compression force. Disintegration time (~30 s) was similar between CNF-supplemented tablets and those supplemented with low-substituted hydroxypropyl cellulose, crospovidone, or croscarmellose sodium. At higher CNF fractions, tablet hardness increased, while friability decreased. Adding ≥30% CNF prolonged the tablet disintegration time. To set the optimized manufacturing condition for ensuring the desired tablet physical properties, we created contour plots for evaluating the effects of CNF concentration and compression force on hardness and disintegration time. A CNF concentration of 10-20% and a compression force of 12-13 kN would allow for the preparation of tablets with a hardness ≥30 N and a disintegration time ≤60 s. Altogether, addition of CNF to the OD tablet formulation for direct powder compression enhanced hardness and disintegration.

EFFECT OF DRYING TEMPERATURE AND TIME ON TABLET FORMULATION OF MORINGA LEAF (Moringa oleifera) EXTRACT WITH WET GRANULATION METHOD USING FLUID BED DRYER

Moringa leave is medicinal plant type that often used by society. Flavonoid and saponin are the content of moringa leave which have benefit as immunomodulator. Moringa leave using can be more practical and easy to used by making as pharmaceutical preparation. Drying temperature and time have a great effect on the physical properties of granule and tablet manufacturing. The objective of this research is to find out the effect of drying temperature and time on the physical-mechanical properties of granule and physical-disintegrant properties of moringa leave extract tablet by wet granulation method. The research method using experimental laboratory methods is correlational (causation). Moringa leave extract tablet was made with 6 formulations by various drying temperature and time. Drying temperature and time which used are FI 40°35', FII 50°35', FIII 60°35', FIV 40°45', FV 50°45', FVI 60°45'. Each formulation was carried out physical properties test of granule and tablet. The data obtained were compared to the pharmacopoeai and other literature and carried out statistical test. In order to know the effect of temperature using Shapiro-Wilk test, then continued ANOVA test, while for the effect of time using independent sample T-test with 95% confidence level, then continued Turkey-HSD test. The result of the test showed that the differences of drying temperature and time have effect on the physical properties of tablet, namely, hardness, brittleness, and disintegration time. The increasing of drying temperature and time are able to reduce the brittleness, increase the hardness, and prolong disintegration time of tablet.

Physical characteristics of effervescent tablet probiotics in various concentration of tapioca coatings

RICA (Research Institute for Coastal Aquaculture) Maros in South Sulawesi has developed 5 probiotic preparations, namely RICA-1, RICA-2, Rica-3, RICA-4 and RICA-5 in liquid form to improve shrimp farming in ponds. In practice, the use of liquid probiotics has been very optimal but the distribution is constrained because of its liquid form. For this reason, microencapsulation of liquid probiotics is carried out to protect from the external environment and maintain the viability of probiotic cells in the encapsulated matrix. The purpose of this study was to determine the physical properties of probiotic effervescent tablets using tapioca coating with different concentration variants (10 and 20%) with four effervescent tablet formulas. The results showed that the pH of the probiotic effervescent tablet Formula 3 with a coating concentration of 10% tapioca had a neutral pH compared to all existing formulas. However, the pH for probiotic effervescent tablets produced by all formulas can still be applied to shrimp ponds. Formula 4 with 10% tapioca coating has a mean weight and hardness that is close to the standard as well as a disintegration time that meets the specified standard of tablet physical properties. Meanwhile, the tablet friability value was met by Formula 3 with a concentration of 10% tapioca coating. It can be concluded that the best formula that meets the standard physical properties of tablets is Formula 4 with 10% tapioca coating. The probiotic effervescent tablet with tapioca coating has a weakness in the hardness value which causes the average weight to be not uniform. These results indicate that tapioca coating is not suitable for use as a coating for probiotic effervescent tablets.

PENGARUH KONSENTRASI HYDROXYPROPYL METHYLCELLULOSE SEBAGAI BAHAN PENGIKAT PADA SEDIAAN TABLET EKSTRAK RIMPANG JAHE MERAH (zingiber officinale Var. Rubrum.)

Lozenges are solid preparations made from aromatic and sweet taste that can dissolve slowly in the mouth, which are intended to treat infections in the mouth and throat. Hydroxypropyl Methylcellulose (HPMC) is a binder that can be used in lozenges.This study attempted to examine the effect of the concentration of HPMC as a binder in the tablet preparation of red ginger rhizome extract (Zingiber officinale Var Rubrum) by wet granulation method. This study was a laboratory experimental study by comparing the three concentrations of HPMC binder used in each formula i.e., FI (4%), FII (5%), FIII (6%).The resulting tablets were tested for physical properties including organoleptic tests, weight uniformity, tablet hardness, tablet friability and hedonic tests. Based on the results of the study, it was shown that with an increase in the concentration of the HPMC binder, the physical quality of the resulting tablet also increased. The higher the concentration of HPMC used, the better the uniformity of weight and the high level of tablet hardness. However, tablet friability will decrease. The results showed that the concentrations of 4%, 5%, and 6% had an effect on the tablet physical properties whichwas fragility. Further, formula II with a concentration of 5% had better tablet physical quality and was preferred by respondentsbased on hedonic tests.

Analysis of Continuous Manufacturing Process of Oral Solid Dosage Using Neural Network

In this study, a neural network (NN) was applied to a continuous manufacturing process of tablets to predict the tablet physical properties based on a twin-screw granulation operating conditions and tableting pressure. The hyperparameters in the NN model were optimized to accurately predict the tablet physical properties. The constructed NN model successfully demonstrated the predictive capability with the R2 of ca. 0.9 in both training and validation. The effects of the granulation operating conditions and the tableting pressure on the tablet physical properties were investigated. It was found that the tableting pressure was the most dominant factor for the tablet hardness and disintegration time. Among the granulation operating conditions, liquid solid ratio had the strongest impact on the tablet physical properties. Focusing on the tableting pressure and liquid solid ratio, the contour maps for the relationships between the operating conditions and the tablet physical properties were obtained by the NN model. It was suggested that the obtained contour maps can be helpful to predict the continuous manufacturing of tablets with the desired tablet physical properties.

Formulation of extended release orphenadrine citrate tablets to treat muscle spasm and pain

Objective: Orphenadrine is an anticholinergic, antimuscarinic, centrally acting skeletal muscle relaxant. It presentsin the form of citrate and HCl salts which are used in treatment of the symptoms of mild Parkinson's disease andalso it is used as adjuvant with other drugs in the therapy. Method: Many trials were made to formulate orphenadrine citrate as tablet using wet granulation or direct compression technique in order to get a satisfactoryformula through studying the effect of various factors such as binders, diluents and disintegrants types. The bestformula was obtained by using poly vinyl pyrrolidone (PVP) as a binder also the results indicated that starch andmannitol gave acceptable physical properties to the tablets when they were used as diluents. At the same time, theresults showed that avicel which was used as a disintegrant gave an acceptable disintegration and dissolution time in comparison with the reference tablet DISIPAL. In addition, the selected formula was used to study the effect ofmethod of incorporation of disintegrant on the physical properties of tablets. Result: It was found that theintragranular incorporation resulted in a shorter disintegration and dissolution times. The stability of orphenadrinecitrate prepared tablets was also studied upon storage at 50C, 60C and 70C for four months. Conclusion: Theoverall results of this study indicate that the drug can be prepared as tablets, which fit the requirements of BritishPharmacopoeia since the prepared tablets gave satisfactory results.

Formulasi dan Evaluasi Sediaan Tablet Nanopartikel Ekstrak Daun Salam (Syzygium Polyanthum) dengan Variasi Konsentrasi Na Alginat dan Avicel PH 102

The use of traditional medicines is an alternative treatment which is considered safer in terms of side effects and toxicity. One of the herbal plants that have properties that can reduce blood pressure is bay leaf (Syzigium polyanthum) because it contains essential oils (citral, eugenol), tannin, and flavonoids. Ethanol extract of bay leaves (Syzigium polyanthum) with antihypertensive potential needs to be made into dosage forms. This study aimed to determine the optimal formula of the preparation of bay leaf (Syzigium polyanthum) nanoparticles tablet with variations in the concentration of Na alginate and Avicel PH 102. Salam leaf extract was made using maceration method using ethanol 96%, followed by evaporation until thick extract was formed. Colloidal nanoparticles were prepared by mixing bay leaf extract into technical ethanol 96% and aquadest, chitosan solution in acetic acid and NaTTP solution. Further, it was measured using PSA (Particle Size Analyzer) to determine the particle size. Tablets were prepared using 4 formulas with variations in the concentration of sodium alginate and avicel PH 102 using the direct pressing method. The results of this study indicate that variations in the concentration of Na alginate and Avicel PH 102 affect the physical properties of tablets. The addition of Avicel PH 102 can increase the hardness and disintegration time of the tablet. While the addition of Na alginate can increase the fragility of tablets. Based on the evaluation results, the optimum formulation of tablet formulas is formula 4.

Influence of low-molecular-weight aggregates on aggregate growth kinetics and physical properties of solid-state proteins during storage

Low-molecular-weight (LMW) aggregate is a critical determinant of subsequent protein aggregation, but the aggregate growth kinetics of solid proteins have not been fully characterized. In this study, the high-molecular weight (HMW) aggregate formation process for solid-state proteins and the relationship between aggregation and physical properties of tablets were evaluated using proteins with various initial aggregate ratios. Quantitative changes in monomers, LMW aggregates, and HMW aggregates during storage were measured. The monomer amount decreased uniformly for all proteins. Proteins with low initial LMW aggregates showed remarkable increases in LMW aggregates but little increases in HMW aggregates during storage. Proteins with high initial LMW aggregates showed decreases in LMW aggregates but remarkable increases in HMW aggregates. A correlation analysis and logistic regression indicated that HMW aggregate formation depended on the initial quantity of LMW aggregates. Furthermore, the initial LMW aggregate ratio was related to the disintegratability of protein-containing tablets after storage. These results provide novel insight into solid-state protein aggregation and may guide the prediction of the long-term quality of solid protein-containing pharmaceuticals and foods without storage.

Formulation and Evaluation of Metoprolol Tartrate Sustained Release Matrix Tablets

The objective of the present work was to develop sustained release matrix tablets of Metoprolol tartrate using different polymers viz. Guar gum, Xanthan gum, Kondagogu gum and HPMC K100M. The release rates were modulated by combination of two different rates controlling material and triple mixture of two different rate controlling materials. After evaluation of physical properties of tablet, the in-vitro release study was performed in phosphate buffer pH 6.8 up to 12 hrs. Dissolution data was analyzed for release kinetics. It was observed that matrix tablets contained polymer Xanthan gum was successfully sustained the release of drug up to 12 hrs. Among all the formulations, F6 which contains 45 % of Xanthan gum, release of drug which follows zero order kinetics via, swelling, diffusion and the release profile of formulation F6 was compared with marketed product. The FTIR study revealed that there was no chemical interaction between drug and excipient.

FORMULASI DAN UJI STABILITAS FISIK SEDIAAN TABLET EKSTRAK BUAH PARE (Momordica charantia L.) SECARA GRANULASI BASAH

<p>Bitter melon fruit is one type of medicinal plant used by the community. Charantin is one of the efficacious compounds contained in bitter melon fruit and can be used to reduce glucose levels in the blood. Tablets have advantage that are easy to consume and exactly the size, so in this study bitter melon is made from tablet preparations. The purpose of this study was to determine the concentration of gelatin binder which can produce tablets of bitter fruit extract (<em>Momordica charantia</em> L.) with good physical quality, and to determine the effect of gelatin as a binder on the physical stability test of bitter melon extract tablets (<em>Momordica charantia</em> L.) making bitter melon extract tablets is by wet granulation method, because the active ingredient of bitter melon fruit is not resistant to warming above 600C. Formulation tablets of bitter melon extract (<em>Momordica charantia</em> L.) with gelatin binder at a concentration of 5%, 7.5%, 10% able to produce tablets that meet the requirements of tablet physical examination for 28 days. Gelatin binding concentration has an effect on the physical properties of tablets, the best concentration of gelatin binder is formula III with 10% gelatin concentration. Comparison of Linear and 4-Arm Star Poly(vinyl pyrrolidone) for Aqueous Binder Jetting Additive Manufacturing of Personalized Dosage Tablets

Fabrication of personalized dosage oral pharmaceuticals using additive manufacturing (AM) provides patients with customizable, locally manufactured, and cost-efficient tablets, while reducing the probability of side effects. Binder jetting AM has potential for fabrication of customized dosage tablets, but the resulting products lack in strength due to solely relying on the binder to produce structural integrity. The selection of polymeric binders is also limited due to viscosity restraints, which limits molecular weight and concentration. To investigate and ameliorate these limitations, this article reports a comprehensive study of linear and 4-arm star poly(vinyl pyrrolidone) (PVP) over a range of molecular weights as polymeric binders for binder jetting AM and their effect on physical tablet properties. Formulation of varying molecular weights and concentrations of linear and 4-arm star PVP in deionized water and subsequent jetting revealed relationships between the critical overlap concentrations (C*) and jettability on binder jetting systems with thermal inkjet printheads. After printing with a commercially available ZCorp Spectrum Z510 printer with an HP11 printhead with a lactose and powdered sugar powder bed, subsequent measurement of compressive strength, compressive modulus, and porosity revealed structure-property relationships between molecular weight, polymer concentration, and linear and 4-arm star architectures with physical properties of binder jetted tablets. This study elucidated that the dominating factor to increase compressive strength of a tablet is dependent on the weight percent of the polymer in the binder, which filled interstitial voids between powder particles. Because 4-arm star polymers have lower solution viscosities compared to linear analogues at the same molecular weights, they were jettable at higher concentrations, thus producing the strongest tablets at a compressive strength of 1.2 MPa. Finally, the inclusion of an active pharmaceutical ingredient (API), acetaminophen, revealed maintenance of the tablet physical properties across 5-50 total wt % API in each tablet.

FORMULATION OF CHLORPHENIRAMINE MALEATE TABLETS USING CO-PROCESSED EXCIPIENT AS A FILLER AND BINDER

Co-Processed Excipient (CPE) is technological innovation for tablet preparation through the direct compression method with a quick and straightforward manufacturing process. This research aimed to formulate and evaluate tablets prepared via a spray drying method with CPE used to improve the compressibility and flowability of formulations containing chlorpheniramine maleate. The spray dried CPE containing MCC PH 101, and Kollidon ® K30 was made into tablets through a direct compression method. Meanwhile, Ludipress ® and Avicel ® PH 102 were used as filler-binder comparators. All the prepared tablet formulations were then evaluated for weight variation, hardness, friability, disintegration time, content uniformity of active ingredient, and dissolution test. The physical properties of tablets with CPE as a filler and binder produced an average weight of 151.65 ± 1.53 mg, 5.92 ± 0.38 kg of hardness, 0.06 ± 0.051% friability, 520.00 ± 2.00 seconds of disintegration time, and 99.24 ± 0.15% content uniformity of active ingredient. The comparators indicated better disintegration time than CPE, while the dissolution test showed that more than 80% (Q) of the amount of active ingredient was dissolved in 30 minutes.CPE could be successfully used to prepare tablet dosage form, and the tablets had fulfilled the standards of pharmacopoeia.

Utility of microcrystalline cellulose to prevent drug segregation in direct powder compression

Direct powder compression is the simplest process method for tableting; however, when drug content is low, segregation tends to occur, making it more difficult to manufacture tablets with assured drug content uniformity. Using fine microcrystalline cellulose (MCCIM) which can influence drug dispersibility in pharmaceutical powders, we assessed the influence of the addition of MCCIM on drug content uniformity, and the utility of MCCIM as a segregation inhibitor. The formulated tablet had one of two types of acetaminophen of different particle sizes, lactose hydrate, cornstarch, MCCIM (0%, 10%, 20%), croscarmellose sodium, and magnesium stearate (Mg-St). Flowability index of the pharmaceutical powder was measured, and flowability evaluated. With MCCIM addition, flowability slightly decreased. Tableting using a rotary-type tableting press, and physical properties of tablets were evaluated. In the prepared tablets, hardness increased and disintegration time decreased with increasing amounts of added MCCIM. Adding MCCIM also decreased drug content variation, and this effect was remarkable for the drug with a large particle size. However, even when commonly used MCC was added, a decrease in drug content variation did not occur. Therefore, MCCIM in a tablet formulation may improve drug content uniformity and would be useful as an agent to prevent segregation.
