Synthesis of 1α and 1β‐acylamino and alkylamino‐1,2,3,4,5,6‐hexahydro‐2,6‐methano‐3‐benzazocines and related compounds

AbstractThe synthesis of series of 1α and 1β‐alkylamino‐1,2,3,4,5,6‐hexahydro‐2,6‐methano‐3‐benzazocines from the corresponding acylamino intermediates is described. Cyclization of the 1β‐chloromethylamido derivative 20 to the novel bridged benzomorphan 21 occurred whereas the corresponding 1β‐chloromethylamido derivative 19 failed to cyclize for steric reasons. In addition several related 1α‐acylamidomethyl‐ and 1β‐alkyl‐aminomethyl benzomorphans are reported.

Synthesis of 1α, 24 (R)- and 1α, 24 (S)-Dihydroxycholesterol from Cholic Acid Derivatives

From 5α-cholestane-3α, 24 (R)- or -3α, 24 (S)-diol (2a and 2b) obtained from lithocholic acid in the previous work, both 24-epimers of 1α, 24-dihydroxycholesterol (6a and 6b) were synthesized. Thus, syntheses of these compounds were accomplished by dehydrogenation of the diols with dichlorodicyanobenzoquinone followed by the 1α-hydroxylation of the resulted 1, 4-dien-3-ones (3a and 3b) by the three-step procedure originally uncovered by one of the present authors (C. K.). The last step in the above transformation, the modified procedure reported recently by Ikekawa's group : oxymercuration-demercuration, was used. An alternative preparation of 1α, 24ξ-dihydroxycholesterol (6) either from lithocholic acid or from hyodeoxycholic acid was also described.

Synthesis of 2α- and 2β-hydrqxycortisol

The synthesis (in 1 gm. amounts) of 2α-hydroxy- (I) and 2β-hydroxycortisol (II), their 2,21-diacetates (I-diac. II-diac.) and 2,11β,21-triacetates (I-triac. II-triac.) is described. Cortisol 21-acetate was acetoxylated with lead tetraacetate in glacial acetic acid. Following saponification in methanolic potassium hydroxide a mixture of I and II was obtained by partition chromatography in approximately 13% yield. Reacetylation of the purified mixture of I and II with acetic anhydride and pyridine at room temperature and subsequent partition chromatography yielded pure I- and II-diac. and 5–10% of I- and II-triac. I and II were obtained by saponification of I- and II-diac. in methanolic potassium hydroxide without inversion. The structure of the isolated compounds was assigned from their elemental analyses, molecular rotation, NMR, ultraviolet and infrared spectra.
