Integration of sulfate assimilation with carbon and nitrogen metabolism in transition from C3 to C4 photosynthesis.

The first product of sulfate assimilation in plants, cysteine, is a proteinogenic amino acid and a source of reduced sulfur for plant metabolism. Cysteine synthesis is the convergence point of the three major pathways of primary metabolism: carbon, nitrate, and sulfate assimilation. Despite the importance of metabolic and genetic coordination of these three pathways for nutrient balance in plants, the molecular mechanisms underlying this coordination, and the sensors and signals, are far from being understood. This is even more apparent in C4 plants, where coordination of these pathways for cysteine synthesis includes the additional challenge of differential spatial localization. Here we review the coordination of sulfate, nitrate, and carbon assimilation, and show how they are altered in C4 plants. We then summarize current knowledge of the mechanisms of coordination of these pathways. Finally, we identify urgent questions to be addressed in order to understand the integration of sulfate assimilation with carbon and nitrogen metabolism particularly in C4 plants. We consider answering these questions to be a prerequisite for successful engineering of C4 photosynthesis into C3 crops to increase their efficiency.

Ectopic Overexpression of bol-miR171b Increases Chlorophyll Content and Results in Sterility in Broccoli ( Brassica oleracea L var. italica).

MiR171 plays pleiotropic roles in the growth and development of several plant species. However, the mechanism underlying the miR171-mediated regulation of organ development in broccoli remains unknown. In this study, bol-miR171b was characterized and found to be differentially expressed in various broccoli organs. The ectopic overexpression of bol-miR171b in Arabidopsis affected the leaf and silique development of transgenic lines. In particular, the chlorophyll content of leaves from overexpressed bol-miR171b transgenic Arabidopsis was higher than that of the vector controls. The fertility and seed yield of Arabidopsis with overexpressed bol-miR171b were markedly lower than those of the vector controls. Similarly, overexpressed bol-miR171b transgenic broccoli exhibited dark green leaves with high chlorophyll content, and nearly all of the flowers were sterile. These results demonstrated that overexpression of bol-miR171b could increase the chlorophyll content of transgenic plants. Degradome sequencing was conducted to identify the targets of bol-miR171b. Two members of the GRAS gene family, BolSCL6 and BolSCL27, were cleaved by bol-miR171b-3p in broccoli. In addition to the genes targeted by bol-miR171b-3p, adenylylsulfate reductase 3 ( APSR3), which played important roles in plant sulfate assimilation and reduction, was speculated to be cleaved by bol-miR171b-5p, suggesting that the star sequence of bol-miR171b may also have functions in broccoli. Comparative transcriptome analysis further revealed that the genes involved in chloroplast development and sulfate homeostasis should participate in the bol-miR171b -mediated regulatory network. Taken together, these findings provided new insights into the function and regulation of bol-miR171b in broccoli and indicated the potential of bol-miR171b as a small RNA molecule that increased leaf chlorophyll in plants by genetic engineering.

Determination of Enzymes Associated with Sulfite Toxicity in Plants: Kinetic Assays for SO, APR, SiR, and In-Gel SiR Activity.

The amino acid cysteine plays a major role in plant response to abiotic stress by being the donor of elemental sulfur for the sulfuration of the molybdenum cofactor, otherwise the last step of ABA biosynthesis, the oxidation of abscisic aldehyde, is inactivated. Additionally, cysteine serves as a precursor for the biosynthesis of glutathione, the reactive oxygen species scavenger essential for redox status homeostasis during stress. Cysteine is generated by the sulfate reductive pathway where sulfite oxidase (SO; EC 1.8.3.1) is an important enzyme in the homeostasis of sulfite levels (present either as a toxic intermediate in the pathway or as a toxic air pollutant that has penetrated the plant tissue via the stomata). SO is localized to the peroxisomes and detoxifies excess sulfite by catalyzing its oxidation to sulfate. Here we show a kinetic assay that relies on fuchsin colorimetric detection of sulfite, a substrate of SO activity. This SO assay is highly specific, technically simple, and readily performed in any laboratory.5'-adenylylsulfate (APS) reductase (APR, E.C. 1.8.4.9) enzyme regulates a crucial step of sulfate assimilation in plants, algae and some human pathogens. The enzyme is upregulated in response to oxidative stress induced by abiotic stresses, such as salinity and hydrogen peroxide, to generate sulfite an intermediate for cysteine generation essential for the biosynthesis of glutathione, the hydrogen peroxide scavenger. Here we present two robust, sensitive, and simple colorimetric methods of APR activity based on sulfite determination by fuchsin.Sulfite reductase (SiR) is one of the key enzymes in the primary sulfur assimilation pathway. It has been shown that SiR is an important plant enzyme for protection plant against sulfite toxicity and premature senescence. Here we describe two methods for SiR activity determination: a kinetic assay using desalted extract and an in-gel assay using crude extract.Due to the energetically favorable equilibrium, sulfurtransferase (ST) activity measured as sulfite generation or consumption. Sulfite-generating ST activity is determined by colorimetric detection of SCN- formation at 460 nm as the red Fe(SCN)3 complex from cyanide and thiosulfate using acidic iron reagent. Sulfite-consuming (MST) activity is detected as sulfite disappearance in the presence of thiocyanate (SCN-) or as SCN- disappearance. To abrogate interfering SO activity, total ST activities is detected by inhibiting SO activity with tungstate.

An emphasis of hydrogen sulfide-cysteine cycle on enhancing the tolerance to chromium stress in Arabidopsis

Increasing attention has been focused on the health of vegetables and grains grown in the contaminated agricultural soil, it is thus meaningful to find ways to reduce the heavy metals (HMs) accumulation in plants. As sulfur is considered to be an essential macronutrient for plant stress defenses, the important role of sulfur assimilation in plants responding to HMs stress has been followed. However, the potential mechanism of the only sulfur-containing gasotransmitter hydrogen sulfide (H2S) and its main endogenously generated substrate, cysteine (Cys), in plant defense is poorly understood. The physiological and biochemical methods together with qRT-PCR were used to explore the response pattern of H2S-Cys cycle in plants resisting to chromium (Cr6+) stress. Our results suggested that Cr6+ stress inhibited Arabidopsis root elongation, increased the H2S and Cys contents time-dependently, and H2S production was activated earlier than Cys. Furthermore, H2S increased Cys accumulation more quickly than Cr6+ stress. The qRT-PCR results revealed that H2S up-regulated the Cys generation-related genes OASTLa, SAT1 and SAT5 expression levels, and that SAT1 and SAT5 expression was elevated for a longer duration. Data suggested that H2S might regulate Cys metabolism-related genes expression to participate in Cr6+-mediated Cys accumulation. H2S and Cys relieved the root elongation inhibition caused by Cr6+ in Arabidopsis. Both H2S and Cys enhanced glutathione generation and activated phytochelatins (PCs) synthesis by up-regulating PCS1 and PCS2 expression levels to fight against Cr6+ stress. Besides regulating the expression of PCs synthase encoding genes, H2S might promote metallothioneins accumulation by significantly increasing the MT2A gene expression. Overall, H2S and H2S-induced Cys accumulation (H2S-Cys system) was critical in imparting Cr6+ tolerance in Arabidopsis. This paper is the first to indicate that gasotransmitter H2S induced Cys accumulation in Arabidopsis Cr6+-stress defense and provides evidence for more extensive studies of the H2S signaling pathway.

Nitrogen and sulfur assimilation in plants and algae

Nitrogen and sulfur are abundant constituents of plant and algal cells that are assimilated at the lowest oxidation number, as NH4+ and S2−, although they can (in the case of sulfur, usually must) be acquired with their highest oxidation number, as NO3− and SO42−. Some occasional differences and variants exists for transport and assimilation systems; the greatest differences in the way vascular plants and algae use N and S, however, most probably resides in regulation. For instance, nitrate assimilation in plants is strongly regulated by phosphorylation. In algae, redox regulation appears to be more important. Similarly, sulfate reduction has its main control step at the level of APS reductase in higher plants, whereas in algae a redox regulation has been recently been hypothesized for ATP sulfurylase, the first step in sulfate assimilation. Unfortunately, the information on the regulation of N and S acquisition and assimilation is limited to very few species (e.g. Chlamydomonas reinhardtii, Arabidopsis thaliana) this is especially true in the case of sulfur. This review attempts to highlight the points of divergence in N and S utilization by plants and algae, leaving aside the biochemical details and the features that do not show any obvious difference.

Kinetic mechanism of the dimeric ATP sulfurylase from plants

In plants, sulfur must be obtained from the environment and assimilated into usable forms for metabolism. ATP sulfurylase catalyses the thermodynamically unfavourable formation of a mixed phosphosulfate anhydride in APS (adenosine 5′-phosphosulfate) from ATP and sulfate as the first committed step of sulfur assimilation in plants. In contrast to the multi-functional, allosterically regulated ATP sulfurylases from bacteria, fungi and mammals, the plant enzyme functions as a mono-functional, non-allosteric homodimer. Owing to these differences, here we examine the kinetic mechanism of soybean ATP sulfurylase [GmATPS1 (Glycine max (soybean) ATP sulfurylase isoform 1)]. For the forward reaction (APS synthesis), initial velocity methods indicate a single-displacement mechanism. Dead-end inhibition studies with chlorate showed competitive inhibition versus sulfate and non-competitive inhibition versus APS. Initial velocity studies of the reverse reaction (ATP synthesis) demonstrate a sequential mechanism with global fitting analysis suggesting an ordered binding of substrates. ITC (isothermal titration calorimetry) showed tight binding of APS to GmATPS1. In contrast, binding of PPi (pyrophosphate) to GmATPS1 was not detected, although titration of the E•APS complex with PPi in the absence of magnesium displayed ternary complex formation. These results suggest a kinetic mechanism in which ATP and APS are the first substrates bound in the forward and reverse reactions, respectively.

Effects of fou8/fry1 Mutation on Sulfur Metabolism: Is Decreased Internal Sulfate the Trigger of Sulfate Starvation Response?

The fou8 loss of function allele of adenosine bisphosphate phosphatase FIERY1 results in numerous phenotypes including the increased enzymatic oxygenation of fatty acids and increased jasmonate synthesis. Here we show that the mutation causes also profound alterations of sulfur metabolism. The fou8 mutants possess lower levels of sulfated secondary compounds, glucosinolates, and accumulate the desulfo-precursors similar to previously described mutants in adenosine 5′phosphosulfate kinase. Transcript levels of genes involved in sulfate assimilation differ in fou8 compared to wild type Col-0 plants and are similar to plants subjected to sulfate deficiency. Indeed, independent microarray analyses of various alleles of mutants in FIERY1 showed similar patterns of gene expression as in sulfate deficient plants. This was not caused by alterations in signalling, as the fou8 mutants contained significantly lower levels of sulfate and glutathione and, consequently, of total elemental sulfur. Analysis of mutants with altered levels of sulfate and glutathione confirmed the correlation of sulfate deficiency-like gene expression pattern with low internal sulfate but not low glutathione. The changes in sulfur metabolism in fou8 correlated with massive increases in 3′-phosphoadenosine 5′-phosphate levels. The analysis of fou8 thus revealed that sulfate starvation response is triggered by a decrease in internal sulfate as opposed to external sulfate availability and that the presence of desulfo-glucosinolates does not induce the glucosinolate synthesis network. However, as well as resolving these important questions on the regulation of sulfate assimilation in plants, fou8 has also opened an array of new questions on the links between jasmonate synthesis and sulfur metabolism.

The key enzyme of sulfate assimilation, adenosine 5′‐phosphosulfate reductase, is regulated by HY5 in Arabidopsis

Plant sulfate assimilation is regulated by demand for reduced sulfur, as is its key enzyme, adenosine 5'-phosphosulfate reductase (APR). In a genetic screen for mutants lacking this regulation, we identified the bZIP transcription factor LONG HYPOCOTYL 5 (HY5) as a necessary component of the regulatory circuit. Regulation of APR activity by the inhibitor of glutathione synthesis, buthionine sulfoximine, or by the precursor of cysteine, O-acetylserine, was disrupted in the hy5 mutant. When dark-adapted plants were re-illuminated, the rapid induction of APR1 and APR2 mRNA levels was attenuated in hy5 seedlings, but APR3 regulation was not affected. Chromatin immunoprecipitation revealed that HY5 binds directly to the APR1 and APR2 promoters but not to the APR3 promoter. Accordingly, the regulation of APR1 and APR2 by O-acetylserine was disturbed in hy5 roots. HY5 is also important for the coordination of nitrogen and sulfur assimilation, as, unlike the wild-type, hy5 mutants do not undergo a reduction in sulfate uptake and APR activity during nitrogen starvation. Altogether, these data show that HY5 plays an important role in regulation of APR gene expression and plant sulfate assimilation.

Sulfur Assimilation in Photosynthetic Organisms: Molecular Functions and Regulations of Transporters and Assimilatory Enzymes

Sulfur is required for growth of all organisms and is present in a wide variety of metabolites having distinctive biological functions. Sulfur is cycled in ecosystems in nature where conversion of sulfate to organic sulfur compounds is primarily dependent on sulfate uptake and reduction pathways in photosynthetic organisms and microorganisms. In vascular plant species, transport proteins and enzymes in this pathway are functionally diversified to have distinct biochemical properties in specific cellular and subcellular compartments. Recent findings indicate regulatory processes of sulfate transport and metabolism are tightly connected through several modes of transcriptional and posttranscriptional mechanisms. This review provides up-to-date knowledge in functions and regulations of sulfur assimilation in plants and algae, focusing on sulfate transport systems and metabolic pathways for sulfate reduction and synthesis of downstream metabolites with diverse biological functions.

Interplay of SLIM1 and miR395 in the regulation of sulfate assimilation in Arabidopsis

MicroRNAs play a key role in the control of plant development and response to adverse environmental conditions. For example, microRNA395 (miR395), which targets three out of four isoforms of ATP sulfurylase, the first enzyme of sulfate assimilation, as well as a low-affinity sulfate transporter, SULTR2;1, is strongly induced by sulfate deficiency. However, other components of sulfate assimilation are induced by sulfate starvation, so that the role of miR395 is counterintuitive. Here, we describe the regulation of miR395 and its targets by sulfate starvation. We show that miR395 is important for the increased translocation of sulfate to the shoots during sulfate starvation. MiR395 together with the SULFUR LIMITATION 1 transcription factor maintain optimal levels of ATP sulfurylase transcripts to enable increased flux through the sulfate assimilation pathway in sulfate-deficient plants. Reduced expression of ATP sulfurylase (ATPS) alone affects both sulfate translocation and flux, but SULTR2;1 is important for the full rate of sulfate translocation to the shoots. Thus, miR395 is an integral part of the regulatory circuit controlling plant sulfate assimilation with a complex mechanism of action.

The influence of sulfur limitation on expression levels of an o-acetylserine (thiol) lyase gene cloned from Vicia sativa

The o-acetylserine (thiol) lyase (OAS-TL) gene is part of the plant sulfate assimilation pathway. The synthesis of cysteine, the first product of the pathway containing organic sulfur, has been previously observed to be dependent on the availability of sulfur and activity of OAS-TL. In this study, the ability of Vicia sativa L. to alter its metabolism to compensate for deficiencies brought about by sulfur stress was utilized to elucidate the functionality of Voas-tl5 (GenBank Accession No. DQ456491), a gene cloned from Vicia sativa. The transcription levels of Voas-tl5 increased in response to sulfur deficiency, indicating that the gene was active in the cysteine biosynthetic pathway.

Plant sulfate assimilation genes: redundancy versus specialization

Sulfur is an essential nutrient present in the amino acids cysteine and methionine, co-enzymes and vitamins. Plants and many microorganisms are able to utilize inorganic sulfate and assimilate it into these compounds. Sulfate assimilation in plants has been extensively studied because of the many functions of sulfur in plant metabolism and stress defense. The pathway is highly regulated in a demand-driven manner. A characteristic feature of this pathway is that most of its components are encoded by small multigene families. This may not be surprising, as several steps of sulfate assimilation occur in multiple cellular compartments, but the composition of the gene families is more complex than simply organellar versus cytosolic forms. Recently, several of these gene families have been investigated in a systematic manner utilizing Arabidopsis reverse genetics tools. In this review, we will assess how far the individual isoforms of sulfate assimilation enzymes possess specific functions and what level of genetic redundancy is retained. We will also compare the genomic organization of sulfate assimilation in the model plant Arabidopsis thaliana with other plant species to find common and species-specific features of the pathway.

Transcriptional regulation of genes involved in sulfur assimilation in plants: Understanding from the analysis of high-affinity sulfate transporters

Sulfur is one of the essential macronutrients required for plant growth. Since the expression of several sulfur-assimilatory genes is stimulated under the condition of sulfur deficiency (−S), transcriptional regulation of these genes is considered to be critical for the control of sulfur assimilation. In the last several years, the author and coworkers have been investigated molecular mechanisms of −S-inducible expression of high-affinity sulfate transporters, SULTR1;1 and SULTR1;2 in Arabidopsis thaliana. SULTR1;1 and SULTR1;2 facilitate sulfate uptake in roots. This review summarizes the recent progress about the transcriptional regulation of these sulfate transporters by focusing on three major topics. 1) Identification of a cis-acting element involved in the −S-inducible expression of SULTR1;1. 2) Cytokinin-dependent repression of SULTR expression and sulfate uptake. 3) Central transcription regulator SLIM1 controlling −S responsive gene expression including sulfur assimilation and metabolism.

The role of the novel adenosine 5′-phosphosulfate reductase in regulation of sulfate assimilation of Physcomitrella patens

Sulfate assimilation provides reduced sulfur for the synthesis of the amino acids cysteine and methionine and for a range of other metabolites. The key step in control of plant sulfate assimilation is the reduction of adenosine 5'-phosphosulfate to sulfite. The enzyme catalyzing this reaction, adenosine 5'phosphosulfate reductase (APR), is found as an iron sulfur protein in plants, algae, and many bacteria. In the moss Physcomitrella patens, however, a novel isoform of the enzyme, APR-B, has recently been discovered lacking the co-factor. To assess the function of the novel APR-B we used homologous recombination to disrupt the corresponding gene in P. patens. The knock-out plants were able to grow on sulfate as a sole sulfur source and the content of low molecular weight thiols was not different from wild type plants or plants where APR was disrupted. However, when treated with low concentrations of cadmium the APR-B knockout plants were more sensitive than both wild type and APR knockouts. In wild type P. patens, the two APR isoforms were not affected by treatments that strongly regulate this enzyme in flowering plants. The data thus suggest that in P. patens APS reduction is not the major control step of sulfate assimilation.

Sulfate Assimilation in Basal Land Plants ‐ What Does Genomic Sequencing Tell Us?

Sulfate assimilation is a pathway providing reduced sulfur for the synthesis of cysteine, methionine, co-enzymes such as iron-sulfur centres, thiamine, lipoic acid, or Coenzyme A, and many secondary metabolites, e.g., glucosinolates or alliins. The pathway is relatively well understood in flowering plants, but very little information exists on sulfate assimilation in basal land plants. Since the finding of a putative 3'-phosphoadenosine 5'-phosphosulfate reductase in PHYSCOMITRELLA PATENS, an enigmatic enzyme thought to exist in fungi and some bacteria only, it has been evident that sulfur metabolism in lower plants may substantially differ from seed plant models. The genomic sequencing of two basal plant species, the Bryophyte PHYSCOMITRELLA PATENS, and the Lycophyte SELAGINELLA MOELLENDORFFII, opens up the possibility to search for differences between lower and higher plants at the genomic level. Here we describe the similarities and differences in the organisation of the sulfate assimilation pathway between basal and advanced land plants derived from genome comparisons of these two species with ARABIDOPSIS THALIANA and ORYZA SATIVA, two seed plants with sequenced genomes. We found differences in the number of genes encoding sulfate transporters, adenosine 5'-phosphosulfate reductase, and sulfite reductase between the lower and higher plants. The consequences for regulation of the pathway and evolution of sulfate assimilation in plants are discussed.

The Putative Moss 3′-Phosphoadenosine-5′-phosphosulfate Reductase Is a Novel Form of Adenosine-5′-phosphosulfate Reductase without an Iron-Sulfur Cluster

Sulfate assimilation provides reduced sulfur for synthesis of the amino acids cysteine and methionine and for a range of other metabolites. Sulfate has to be activated prior to reduction by adenylation to adenosine 5'-phosphosulfate (APS). In plants, algae, and many bacteria, this compound is reduced to sulfite by APS reductase (APR); in fungi and some cyanobacteria and gamma-proteobacteria, a second activation step, phosphorylation to 3'-phosphoadenosine 5'-phosphosulfate (PAPS), is necessary before reduction to sulfite by PAPS reductase (PAPR). We found previously that the moss Physcomitrella patens is unique among these organisms in possessing orthologs of both APR and PAPR genes (Koprivova, A., Meyer, A. J., Schween, G., Herschbach, C., Reski, R., and Kopriva, S. (2002) J. Biol. Chem. 277, 32195-32201). To assess the function of the two enzymes, we compared their biochemical properties by analysis of purified recombinant proteins. APR from Physcomitrella is very similar to the well characterized APRs from seed plants. On the other hand, we found that the putative PAPR preferentially reduces APS. Sequence analysis, analysis of UV-visible spectra, and determination of iron revealed that this new APR, named PpAPR-B, does not contain the FeS cluster, which was previously believed to determine the substrate specificity of the otherwise relatively similar enzymes. The lack of the FeS cluster in PpAPR-B catalysis is connected with a lower turnover rate but higher stability of the protein. These findings show that APS reduction without the FeS cluster is possible and that plant sulfate assimilation is predominantly dependent on reduction of APS.

DNA binding and partial nucleoid localization of the chloroplast stromal enzyme ferredoxin:sulfite reductase.

Sulfite reductase (SiR) is an important enzyme catalyzing the reduction of sulfite to sulfide during sulfur assimilation in plants. This enzyme is localized in plastids, including chloroplasts, and uses ferredoxin as an electron donor. Ferredoxin-dependent SiR has been found in isolated chloroplast nucleoids, but its localization in vivo or in intact plastids has not been examined. Here, we report the DNA-binding properties of SiRs from pea (PsSiR) and maize (ZmSiR) using an enzymatically active holoenzyme with prosthetic groups. PsSiR binds to both double-stranded and single-stranded DNA without significant sequence specificity. DNA binding did not affect the enzymatic activity of PsSiR, suggesting that ferredoxin and sulfite are accessible to SiR molecules within the nucleoids. Comparison of PsSiR and ZmSiR suggests that ZmSiR does indeed have DNA-binding activity, as was reported previously, but the DNA affinity and DNA-compacting ability are higher in PsSiR than in ZmSiR. The tight compaction of nucleoids by PsSiR led to severe repression of transcription activity in pea nucleoids. Indirect immunofluorescence microscopy showed that the majority of SiR molecules colocalized with nucleoids in pea chloroplasts, whereas no particular localization to nucleoids was detected in maize chloroplasts. These results suggest that SiR plays an essential role in compacting nucleoids in plastids, but that the extent of association of SiR with nucleoids varies among plant species.

Sulfur assimilation in plants and weed control: Potential targets for novel herbicides and action sites of certain safeners

Sulfur is an indispensable element for plants. It is found in sulfur‐containing amino acids, cysteine and methionine, and in various other important biochemical components and processes. Inhibitors of sulfur assimilation, or cysteine and methionine synthesis, could be potential herbicides. In the present paper, the sulfur assimilation pathway in plants is described, followed by the introduction of several compounds (inhibitors and safeners) acting on this pathway. Uptake of inorganic sulfate through the roots is the first step of sulfur assimilation in plants. Sulfate is reduced mainly in chloroplasts to sulfide by a multistep process, and sulfide is then incorporated into cysteine. Cysteine is converted to cystathionine, homocysteine and methionine. Cysteine is incorporated into glutathione (GSH) by γ‐glutamylcysteine synthetase and GSH synthetase. Three enzymes involved in cysteine and methionine biosynthesis, cysteine synthase, cystathionine γ‐synthase and cystathionine β‐lyase, have been investigated as target sites for herbicides. Several inhibitors of these enzymes (e.g. rhizobitoxine and propargylglycine) were also phytotoxic, suggesting that the synthetic pathway of sulfur‐containing amino acids could be a new target site for herbicides. Some safeners for herbicides were found to act on the sulfur assimilation pathway and on GSH synthesis to increase GSH, which can be involved in herbicide metabolism and detoxification. Several safeners elevate GSH levels by increasing the activities of enzymes involved in sulfur assimilation and GSH synthesis. Further studies on plant sulfur metabolism may lead to the discovery of new herbicides and to the comprehensive understanding of the mode of action of safeners.

Regulation of the plant-type 5'-adenylyl sulfate reductase by oxidative stress.

5'-Adenylyl sulfate (APS) reductase (EC 1.8.4.9) catalyzes a key reaction in the plant sulfate assimilation pathway leading to the synthesis of cysteine and the antioxidant glutathione. In Arabidopsis thaliana APS reductase is encoded by a family of three genes. In vitro biochemical studies revealed that the enzyme product derived from one of them (APR1) is activated by oxidation, probably through the formation of a disulfide bond. The APR1 enzyme is 45-fold more active when expressed in a trxB strain of Escherichia coli than in a trxB(+) wild type. The enzyme is inactivated in vitro by treatment with disulfide reductants and is reactivated with thiol oxidants. Redox titrations show that the regulation site has a midpoint potential of -330 mV at pH 8.5 and involves a two-electron redox reaction. Exposure of a variety of plants to ozone induces a rapid increase in APS reductase activity that correlates with the oxidation of the glutathione pool and is followed by an increase in free cysteine and total glutathione. During the response to ozone, the level of immunodetectable APS reductase enzyme does not increase. Treatment of A. thaliana seedlings with oxidized glutathione or paraquat induces APS reductase activity even when transcription or translation is blocked with inhibitors. The results suggest that a posttranslational mechanism controls APS reductase. A model is proposed whereby redox regulation of APS reductase provides a rapidly responding, self-regulating mechanism to control the glutathione synthesis necessary to combat oxidative stress.
