Elevated CO2 concentration enhance carbon and nitrogen metabolism and biomass accumulation of Ormosiahosiei

Elevated CO2 concentrations may inhibit photosynthesis due to nitrogen deficiency, but legumes may be able to overcome this limitation and continue to grow. Our study confirms this conjecture well. First, we placed the two-year-old potted saplings of Ormosia hosiei (O. hosiei) (a leguminous tree species) in the open-top chamber (OTC) with three CO2 concentrations of 400 (CK), 600 (E1), and 800 μmol·mol−1 (E2) to simulate the elevated CO2 concentration environment. After 146 days, the light saturation point (LSP), light compensation point (LCP), apparent quantum efficiency (AQE), and dark respiration rate (Rd) of O. hosiei were increased under increasing CO2 concentration and obtain the maximum ribulose diphosphate (RuBP) carboxylation rate (Vc max) and RuBP regenerated photosynthetic electron transfer rate (Jmax) were also significantly increased under E2 treatment (P < 0.05). This results in a significant increase of the maximum assimilation rate (Amax) under elevated CO2 concentrations. Sucrose phosphate synthase (SPS) activity in sucrose metabolism increased in the leaves, more soluble sugars, starches, and sucrose was produced, but sucrose content only in leaves increased at E2, and more carbon flows to the roots. The activity of the NH4+ assimilating enzymes glutamine synthetase (GS), glutamate synthetase (GOGAT), and glutamate dehydrogenase (GDH) in the leaves of O. hosiei increases under elevated CO2 concentrations to promote nitrogen synthesis that reduces the content of ammonium nitrogen and increases the content of nitrate nitrogen. In addition, under E1 conditions, sucrose synthase (SS), direction of synthesis activity was highest and sucrose invertase (INV) activity was lowest, this means that the balance of C and N metabolism is maintained. While under E2 conditions SS activity decreased and INV activity increased, this increased C/N and nitrogen use efficiency. So, the elevated CO2 concentration promotes the accumulation of O. hosiei biomass, especially in the aboveground part, but did not have a significant effect on the accumulation of root biomass. This means that O. hosiei is able to cope under the elevated CO2 concentration without showing photosynthetic adaptation during the experimental period.

Wheat response to winter night warming based on physiological and transcriptome analyses

Global warming is primarily characterized by asymmetric temperature increases, with higher temperature rises in winter/spring and at night compared to summer/autumn and daytime. We investigated the impact of winter night warming on wheat leaves using the spring wheat cultivar Yangmai 18 and the semi-winter wheat cultivar Yannong 19 during the 2020–2021 growing season. This study aimed to examine the effect of winter night warming on the top expanded leaf of wheat plants. The results showed that the night mean temperature in the treatment group increased by 1.27°C compared to the ambient temperature and winter night warming increased the yield of both wheat cultivars, the activities of sucrose synthase and sucrose phosphate synthase after anthesis, and the biosynthesis of sucrose and soluble sugars. The differentially expressed genes were identified using P-value<0.05 and fold change>2, and subjected to Gene Ontology annotation and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses. The genes differentially expressed in wheat leaves treated with night warming were primarily associated with starch and sucrose metabolism, amino acid biosynthesis, carbon metabolism, plant hormone signal transduction, and amino sugar and nucleotide sugar metabolism. Comparison between groups identified 14 differentially expressed genes related to temperature. These results highlight the effects of winter night warming on wheat development from various perspectives. Our results provide new insights into the molecular mechanisms of the wheat response to winter night warming and the candidate genes involved in this process.

Changes in carbohydrate distribution of cotton and increase in boll weight reduced yield loss under high temperature.

Cotton yield is sometimes unresponsive to high temperature (HT) that induce significant reductions in fruit retention. To investigate the underlying mechanisms, a greenhouse experiment was conducted with two temperature regimes (control, control treatment, 28 °C; HT, 34 °C) for 7 days. Results showed HT did not significantly influence cotton yield, but remarkably reduce boll number and increase boll weight. 13C distribution ratio of the leaf subtending to cotton boll (LSCB) decreased while that of cotton boll increased under HT. Transcriptomic and proteomic analyses of LSCB revealed up-regulated genes involved in cytokinin and jasmonic acid synthesis, as well as SWEET15 (GH_D01G0218), which positively regulated photosynthesis and transport photosynthate, ultimately leading to increased boll weight. After 7 days recovering from HT, 13C distribution ratio of LSCB increased while that of cotton boll decreased. However, the boll weight still increased, which was related to the increased amylase and sucrose phosphate synthase activities and up-regulated sucrose transport genes in main-stem leaf and capsule wall. Thus, both the accelerated sucrose synthesis and transport in LSCB under HT and the increased sucrose supply ability in main-stem leaf and capsule wall after recovering from HT contributed to an increased boll weight, which finally maintained cotton yield.

Rice husk ash based growing media impact on cucumber and melon growth and quality

Rice husk, an agricultural waste from the rice industry, can cause serious environmental pollution if not properly managed. However, rice husk ash (RHA) has been found to have many positive properties, making it a potential replacement for non-renewable peat in soilless planting. Thus, this study investigated the impact of a RHA composite substrate on the growth, photosynthetic parameters, and fruit quality of cucumber (Yuyi longxiang variety) and melon (Yutian yangjiaomi variety). The RHA, peat, vermiculite, and perlite were blended in varying proportions, with the conventional seedling substrate (peat:vermiculite:perlite = 1:1:1 volume ratio) serving as the control (CK). All plants were cultivated in barrels filled with 10L of the mixed substrates. The results from this study found that RHA 40 (RHA:peat:vermiculite:perlite = 4:4:1:1 volume ratio) significantly enhanced substrate ventilation and positively influenced the stem diameter, root activity, seedling index, chlorophyll content, net photosynthetic rate (Pn), stomatal conductance (Gs), and transpiration rate (Tr) of cucumber and melon plants. Additionally, plant planted using RHA 40, the individual fruit weight of cucumber and melon found to increase by 34.62% and 21.67%, respectively, as compared to the control. Aside from that, both cucumber and melon fruits had significantly higher sucrose, total soluble sugar, vitamin C, and soluble protein levels. This subsequently improved the activity of sucrose synthase and sucrose phosphate synthase in both cucumber and melon. In conclusion, the RHA 40 found to best promote cucumber and melon plant growth, increase plant leaf photosynthesis, and improve cucumber and melon fruit quality, making it a suitable substrate formula for cucumber and melon cultivation in place of peat.

Comparative Analysis of Sucrose-Regulatory Genes in High- and Low-Sucrose Sister Clones of Sugarcane.

Sugarcane is a significant primitive source of sugar and energy worldwide. The progress in enhancing the sugar content in sugarcane cultivars remains limited due to an insufficient understanding of specific genes related to sucrose production. The present investigation examined the enzyme activities, levels of reducing and non-reducing sugars, and transcript expression using RT-qPCR to assess the gene expression associated with sucrose metabolism in a high-sucrose sugarcane clone (GXB9) in comparison to a low-sucrose sister clone (B9). Sucrose phosphate synthase (SPS), sucrose phosphate phosphatase (SPP), sucrose synthase (SuSy), cell wall invertase (CWI), soluble acid invertase (SAI), and neutral invertase (NI) are essential enzymes involved in sucrose metabolism in sugarcane. The activities of these enzymes were comparatively quantified and analyzed in immature and maturing internodes of the high- and low-sucrose clones. The results showed that the higher-sucrose-accumulating clone had greater sucrose concentrations than the low-sucrose-accumulating clone; however, maturing internodes had higher sucrose levels than immature internodes in both clones. Hexose concentrations were higher in immature internodes than in maturing internodes for both clones. The SPS and SPP enzymes activities were higher in the high-sucrose-storing clone than in the low-sucrose clone. SuSy activity was higher in the low-sucrose clone than in the high-sucrose clone; further, the degree of SuSy activity was higher in immature internodes than in maturing internodes for both clones. The SPS gene expression was considerably higher in mature internodes of the high-sucrose clones than the low-sucrose clone. Conversely, the SuSy gene exhibited up-regulated expression in the low-sucrose clone. The enhanced expression of SPS in the high-sucrose clone compared to the low-sucrose clone suggests that SPS plays a major role in the increased accumulation of sucrose. These findings provide the opportunity to improve sugarcane cultivars by regulating the activity of genes related to sucrose metabolism using transgenic techniques.

POsHAK1:OsSUT1 Promotes Sugar Transport and Enhances Drought Tolerance in Rice.

Plant cells accumulate osmotic substances (e.g., sugar) to protect cell components and maintain osmotic balance under drought stress conditions. Previous studies found that pOsHAK1:OsFLN2 promotes sugar metabolism and improves the drought tolerance of rice plants under drought stress. This study further evaluated the effect of the ectopic expression of the OsSUT1 gene driven by the OsHAK1 promoter on the sugar transport and drought tolerance of rice. The results showed that the net photosynthetic rate and sucrose phosphate synthase activity of plants expressing the OsSUT1 gene were not significantly different from those of wild-type (WT) rice plants under drought conditions. However, the sucrose transport rate in the phloem increased in the transgenic plants, and the sucrose contents were significantly lower in the leaves but significantly higher in the roots of transgenic plants than those in WT plants. The pOsHAK1:OsSUT1 and pOsHAK1:OsFLN2 transgenic lines had similar rates of long-distance sucrose transport and drought tolerance, which were higher than those of the WT plants. The relative water content of the transgenic plants was higher, while their water loss rate, hydrogen peroxide (H2O2), and malondialdehyde (MDA) contents were lower than those of the WT plants. The stress-responsive gene OsbZIP23 and the antioxidant-related gene OsCATB were significantly upregulated in the drought-treated transgenic lines, while the senescence indicator gene SGR and the stress-responsive gene OsNAC2 were down-regulated compared to WT plants. These results showed that promoting the long-distance sugar transport through the expression of pOsHAK1:OsSUT1 could produce an improved drought tolerance effect similar to that of pOsHAK1:OsFLN2, providing an effective way to improve the drought tolerance of cereal crops at the seedling stage.

Drought stress reduces the photosynthetic source of subtending leaves and the transit sink function of podshells, leading to reduced seed weight in soybean plants.

Drought stress is the key factor limiting soybean yield potential. Soybean seed formation involves a coordinated "subtending leaf-podshell-seed" process, but little is known about the assimilation and transport of photoassimilates in subtending leaves, podshells and seeds or their relationships with soybean seed formation under drought stress. To address these research gaps, two-year experiments with two soybean cultivars, Wandou 37 (drought tolerant) and Zhonghuang 13 (drought sensitive), were conducted under three soil water content (SWC) conditions in 2020 and 2021 based on the responses of their yield to drought. We analyzed the photosynthetic assimilation and translocation of photoassimilates in subtending leaves, podshells and seeds by stable isotope labeling. Compared with those under 75% SWC, 60% SWC and 45% SWC significantly decreased the Wandou 37 seed weight by 19.4% and 37.5%, respectively, and that of Zhonghuang 13 by 26.9% and 48.6%, respectively. Compared with those under 75% SWC, drought stress decreased the net photosynthetic rate and the activities of sucrose phosphate synthase (SPS) and sucrose synthase (SuSy), which in turn decreased the photosynthetic capacity of the subtending leaves. The podshells ensure the input of photoassimilates by increasing the SuSy activity, but the weakened source-sink relationship between podshells and seeds under drought stress leads to a decrease in the translocation of assimilates from podshells to seeds. The lack of assimilates under drought stress is an important factor restricting the development of soybean seeds. We conclude that the decrease in seed weight was caused by the decrease in the photosynthetic capacity of the subtending leaves and the decrease in the overall availability of photoassimilates; moreover, by a decrease in the translocation of assimilates from podshells to seeds.

Transcriptome Profiles Reveals ScDREB10 from Syntrichia caninervis Regulated Phenylpropanoid Biosynthesis and Starch/Sucrose Metabolism to Enhance Plant Stress Tolerance.

Desiccation is a kind of extreme form of drought stress and desiccation tolerance (DT) is an ancient trait of plants that allows them to survive tissue water potentials reaching -100 MPa or lower. ScDREB10 is a DREB A-5 transcription factor gene from a DT moss named Syntrichia caninervis, which has strong comprehensive tolerance to osmotic and salt stresses. This study delves further into the molecular mechanism of ScDREB10 stress tolerance based on the transcriptome data of the overexpression of ScDREB10 in Arabidopsis under control, osmotic and salt treatments. The transcriptional analysis of weight gene co-expression network analysis (WGCNA) showed that "phenylpropanoid biosynthesis" and "starch and sucrose metabolism" were key pathways in the network of cyan and yellow modules. Meanwhile, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis of differentially expressed genes (DEGs) also showed that "phenylpropanoid biosynthesis" and "starch and sucrose metabolism" pathways demonstrate the highest enrichment in response to osmotic and salt stress, respectively. Quantitative real-time PCR (qRT-PCR) results confirmed that most genes related to phenylpropanoid biosynthesis" and "starch and sucrose metabolism" pathways in overexpressing ScDREB10 Arabidopsis were up-regulated in response to osmotic and salt stresses, respectively. In line with the results, the corresponding lignin, sucrose, and trehalose contents and sucrose phosphate synthase activities were also increased in overexpressing ScDREB10 Arabidopsis under osmotic and salt stress treatments. Additionally, cis-acting promoter element analyses and yeast one-hybrid experiments showed that ScDREB10 was not only able to bind with classical cis-elements, such as DRE and TATCCC (MYBST1), but also bind with unknown element CGTCCA. All of these findings suggest that ScDREB10 may regulate plant stress tolerance by effecting phenylpropanoid biosynthesis, and starch and sucrose metabolism pathways. This research provides insights into the molecular mechanisms underpinning ScDREB10-mediated stress tolerance and contributes to deeply understanding the A-5 DREB regulatory mechanism.

Physiological and biochemical processes underlying the differential sucrose yield and biomass production in sugarcane varieties

Summary Sucrose yield in sugarcane is a complex process regulated by both environmental and endogenous factors. However, the metabolic balance driving vegetative growth and sucrose accumulation remains poorly understood. Herein, we carried out a comprehensive assessment of carbohydrate dynamics throughout the crop cycle in two sugarcane varieties varying in biomass production, evaluating the carbon metabolism in both leaves and stalks. Our data revealed that the decline in photosynthetic rates during sugarcane maturation is associated not only to accumulation of sugars in leaves but also due to stomatal and non-stomatal limitations. We found that metabolic processes in leaves and stalks were intrinsically linked. While IACSP94-2094 had higher stalk sucrose concentration than IACSP95-5000, this latter produced more biomass. Compared to IACSP95-5000, IACSP94-2094 showed higher sucrose phosphate synthase (SPS) activity in leaves and stalks, along with lower soluble acid invertase (SAI) activity in leaves during the maximum growth stage. Interestingly, IACSP94-2094 also exhibited higher stalk SPS activity and lower stalk SAI activity than IACSP95-5000 during maturation. High biomass production by IACSP95-5000 was associated with higher sucrose synthase (SuSy) and SAI activity in leaves and higher SuSy and soluble neutral invertase (SNI) activity in stalks when compared to IACSP94-2094 during the maximum growth. Despite the contrasting strategies, both varieties displayed similar total sucrose yield, a balance between sucrose concentration and biomass production. This phenomenon implies the presence of a compensatory mechanism in sugarcane, with high biomass production compensating low sucrose accumulation and vice versa.

Calcium signal regulated carbohydrate metabolism in wheat seedlings under salinity stress.

This study aimed to explore the mechanism by which calcium (Ca) signal regulated carbohydrate metabolism and exogenous Ca alleviated salinity toxicity. Wheat seedlings were treated with sodium chloride (NaCl, 150mM) alone or combined with 500μM calcium chloride (CaCl2), lanthanum chloride (LaCl3) and/or ethylene glycol tetraacetic acid (EGTA) to primarily analyse carbohydrate starch and sucrose metabolism, as well as Ca signaling components. Treatment with NaCl, EGTA, or LaCl3 alone retarded wheat-seedling growth and decreased starch content accompanied by weakened ribulose-1,5-bisphosphate carboxylation/oxygenase (Rubisco) and Rubisco activase activities, as well as enhanced glyceraldehyde-3-phosphate dehydrogenase, phosphoglycerate kinase, alpha-amylase, and beta-amylase activities. However, it increased the sucrose level, up-regulated the sucrose phosphate synthase (SPS) and sucrose synthase (SuSy) activities and TaSPS and TaSuSy expression together, but down-regulated the acid invertase (SA-Inv) and alkaline/neutral invertase (A/N-Inv) activities and TaSA-Inv and TaA/N-Inv expression. Except for unchanged A/N-Inv activities and TaA/N-Inv expression, adding CaCl2 effectively blocked the sodium salt-induced changes of these parameters, which was partially eliminated by EGTA or LaCl3 presence. Furthermore, NaCl treatment also significantly inhibited Ca-dependent protein kinases and Ca2+-ATPase activities and their gene expression in wheat leaves, which was effectively relieved by adding CaCl2. Taken together, CaCl2 application effectively alleviated the sodium salt-induced retardation of wheat-seedling growth by enhancing starch anabolism and sucrose catabolism, and intracellular Ca signal regulated the enzyme activities and gene expression of starch and sucrose metabolism in the leaves of sodium salt-stressed wheat seedlings.

Source-sink coordinated peanut cultivar increases yield and kernel protein content through enhancing photosynthetic characteristics and regulating carbon and nitrogen metabolisms

The grain yield of crops is determined by the synergistic interaction between source activity and sink capacity. However, source-sink interactions are far from being fully understood of peanut. Therefore, a 2-year field study (2018–2019) was conducted to compare differences in photosynthetic characteristics, carbon and nitrogen metabolism, and yield and quality of different source–sink peanut varieties. Four representative source–sink types were examined: JH5 (source–sink coordination type), SH9 (sufficient source–large sink type), ZH24 (sufficient source–few sink type), and HY36 (large source–few sink type). The results showed that the photosynthetic potential of HY36 was higher than that of the other varieties after flowering because of a large source (leaves), whereas the chlorophyll content and net photosynthetic rate of HY36 were significantly lower than those of JH5 and ZH24. Proportions of dry matter transferred to pods were significantly different among four source–sink peanut varieties. From 50 days after flowering, the dry matter distribution ratio of pods exceeded that of stems and leaves in JH5, significantly earlier than other varieties, which prolong the duration of pod-filling period, followed by SH9 and ZH24. The activities of nitrate reductase, glutamine synthetase, glutamate dehydrogenase, and glutamate synthase in JH5 were the highest among the varieties, and thus, the highest protein content was also in JH5. The activities of sucrose synthase and sucrose phosphate synthase in ZH24 were significantly higher than those in HY36. The highest oil content was also in ZH24. Among pod sink characteristics and yield, SH9 had the longest flowering period and the highest gynophore formation rate but the lowest pod-bearing rate, and the effective proportion and pod fullness were also lower than those of other varieties. The highest pod rate was in ZH24. The effective proportion and pod fullness of JH5 were higher than those of the other varieties, and its yield was also the highest, followed by SH9 and ZH24, with the lowest yield in HY36. The obtained results indicate that the source–sink coordinated variety had high Pn and chlorophyll content in the late growth stage, a long functional period of leaves, and a high proportion of assimilates transported to pods, thus promoting effective proportions and pod fullness to improve peanut yield and protein content, suggesting that different cultivation and management measures should select for different peanut varieties to best coordinate the relationship between the source and sink.

Exogenous application of ALA enhanced sugar, acid and aroma qualities in tomato fruit.

The content and proportion of sugars and acids in tomato fruit directly affect its flavor quality. Previous studies have shown that 5-aminolevulinic acid (ALA) could promote fruit ripening and improve its aroma quality. In order to explore the effect of ALA on sugar and acid quality during tomato fruit development, 0, 100, and 200 mg L-1 ALA solutions were sprayed on the fruit surface 10 days after pollination of the fourth inflorescence, and the regulation of ALA on sugar, acid metabolism and flavor quality of tomato fruit was analyzed. The results showed that ALA treatment could enhance the activities of acid invertase (AI), neutral invertase (NI), and sucrose synthase (SS), reduce the activity of sucrose phosphate synthase (SPS), up-regulate the expression of SlAI, SlNI and SlSS, change the composition and content of sugar in tomato fruit at three stages, significantly increase the content of sugars in fruit, and promote the accumulation of sugars into flesh. Secondly, ALA treatments increased the activities of phosphoenolpyruvate carboxykinase (PEPC), malic enzyme (ME), and citrate synthase (CS), up-regulated the expression of SlPPC2, SlME1, and SlCS, and reduced the citric acid content at maturity stage, thereby reducing the total organic acid content. In addition, ALA could also increase the number and mass fraction of volatile components in mature tomato fruits. These results indicated that exogenous application of ALA during tomato fruit development could promote the formation of fruit aroma quality and were also conducive to the formation of fruit sugar and acid quality.

Phosphorus-induced improvement of photosynthate synthesis and transport in the leaf subtending to cotton boll provided sufficient sucrose for fiber thickening during the crucial period and improved fiber bundle strength

ContextCotton fiber is an important natural textile raw material, and its quality directly affects spinning performance. Phosphorus (P) deficiency reduces fiber quality, particularly fiber strength. Fiber strength formation relies on sucrose supplied by the leaf subtending to cotton boll (LSCB), and low P may affect it by regulating sucrose supply and utilization in LSCB and fiber. ObjectiveOur study aimed to investigate how P regulates the formation of fiber bundle strength through essential carbohydrate metabolic pathways. MethodsA three-year experiment on Lu 54 (low-P sensitive) and Yuzaomian 9110 (low-P tolerant) was carried out to study the influence of P on sucrose synthesis and export in LSCB, sucrose absorption and utilization in fiber under 0 (Deficient P), 100 (Critical P), and 200 (Excessive P) kg P2O5 ha−1 in soil with 16.9 mg kg−1 available P. ResultsP application significantly increased fiber bundle strength (8.9 %−14.6 %), primarily attributed to the enhanced sucrose supply and utilization in LSCB and fiber during the crucial period for fiber bundle strength formation (17–24 days post anthesis). During this period, P application promoted the sucrose synthesis in LSCB by improving the net photosynthetic rate (9.0 %−16.8 %), and sucrose phosphate synthase (6.3 %−12.4 %) and sucrose synthase (10.0 %−17.4 %) activities. Concomitantly, P application up-regulated the expressions of sucrose transporter genes in LSCB and fiber, promoting sucrose transport and increasing the sucrose content (28.1 %−54.0 %) in fiber. Furthermore, P application markedly enhanced the sucrose utilization in fiber by increasing sucrose synthase (9.7 %−19.2 %) and invertase (14.0 %−35.7 %) activities, improving cellulose content (6.7 %−11.5 %) and fiber bundle strength. Lu 54 exhibited a greater sensitivity to P in the carbohydrate metabolism of LSCB and fiber than Yuzaomian 9110, making its cellulose content and fiber bundle strength increase more. Incremental effects of unit P on the above measurements attenuated markedly with P rates. The critical P content in LSCB (LPC) required for effective fiber bundle strength formation was 0.40 % for Lu 54 versus 0.36 % for Yuzaomain 9110, on the basis of the content of sucrose (the key substrate) in LSCB and fiber. ConclusionP application increased cotton fiber bundle strength mainly by enhancing sucrose synthesis in LSCB, sucrose transport between LSCB and fiber, and sucrose utilization in fiber during the crucial period. ImplicationThis report revealed the crucial carbohydrate metabolic path by which P regulated the fiber bundle strength formation and quantified its LPC demand during the crucial period, providing theoretical support for P diagnosis and regulation in cotton cultivation.

MWCNTs Alleviated saline-alkali stress by optimizing photosynthesis and sucrose metabolism in rice seedling

ABSTRACTSaline and alkali stress affects the growth and development, survival rate, and final yield of rice, while new nano materials can have a positive effect on rice growth. In order to investing the effects of carboxymethyl multi walled carbon nanotubes (MWCNTs) on the growth and development of rice seedlings under salt alkali stress, rice seedlings were cultured using rice variety “Songjing 3” using nutrient solution water culture method. The effects of MWCNTs on water absorption capacity, leaf photosynthesis, and sucrose metabolism of rice seedlings under 50 mmol/L saline-alkali stress (1NaCl: 9Na2SO4: 9NaHCO3: 1Na2CO3) conditions were investigated. The results showed that MWCNTs can improve the water use ability of roots and leaves, especially the water absorption ability of roots, which provides a guarantee for the improvement of rice biomass and the enhancement of leaf photosynthetic capacity under adverse conditions. After treatment with MWCNTs, the photosynthetic rate (Pn), stomatal conductance (gs), and transpiration rate (Tr) of leaves increased significantly, and the photochemical quenching value (qP), photochemical quantum efficiency value (Fv/Fm), and electron transfer rate value (ETR) of chlorophyll fluorescence parameters increased significantly, which is beneficial to the improvement of the PSII photosynthetic system. MWCNTs treatment promoted the increase of photosynthetic pigment content in leaves under salt and alkali stress, improved the ratio of Chla and Chlb parameters, increased the activities of key photosynthetic enzymes (RUBPCase and PEPCase) in leaves, increased the value of total lutein cycle pool (VAZ), and significantly enhanced the deepoxidation effect of lutein cycle (DEPS), which can effectively alleviate the stomatal and non stomatal constraints on leaf photosynthesis caused by salt and alkali stress. MWCNTs treatment significantly enhanced the activities of sucrose phosphate synthase (SPS) and sucrose synthase (SS) under salt and alkali stress, and decreased the activities of soluble acid invertase (SAInv) and alkaline/neutral invertase (A/N-Inv), indicating that MWCNTs promoted sucrose synthesis while inhibiting sucrose decomposition, thereby promoting sucrose accumulation in rice leaves. This study can provide theoretical and experimental basis for the application of MWCNTs to the production of rice under salt and alkali stress, and can find a new way for rice production in saline and alkaline lands.

Response of maize stalk to plant density on cellulose accumulation by modulating enzymes activities

Hybrids and plant density are the main factors affecting maize lodging resistance, and cellulose plays an important role in lodging resistance. High plant density often decreases the stalk cellulose, but the cause is unclear. Therefore, we hypothesized that enzymatic activity might be the main factor affecting maize cellulose accumulation. For testing this hypothesis, three plant densities （4.5, 9.0 and 13.5 plants m−2） were set up and three maize hybrids （high lodging resistant cultivars: Xianyu 335, Zhengdan 958; low lodging resistant cultivar: Xinyu 41）with different lodging resistance were selected as materials. The results showed that high plant densities reduced the stalk cellulose contents and strength, and hence lodging risk increased. Through logistic curve fitting, we found that the period of rapid accumulation of cellulose in stalks was 10–26 DPV6 (days post 6-leaf stage). Analyse changes of stalk sucrose and related enzyme activity during 10–26 DPV6 showed that with the increase of plant density, the sucrose content of the stalks significantly decreased (P＜0.05), sucrose content began to differ at 16-DPV6. Both sucrose phosphate synthase(SPS) and sucrose synthase(SS) activity of stalks in this stage decreased with the increase of plant densities. At 20 DPV6, there has a great difference in SPS among different plant densities during this period. Compared with 4.5 plants m−2, the decreases for SPS in Xianyu 335 and Zhengdan 958 was lower than Xinyu 41 at 9.0 plants m−2 and 13.5 plants m−2. The SPS for Xianyu 335 decreased by 10.0%− 16.0% and 28.7%− 37.7% at 9.0 plants m−2 and 13.5 plants m−2, respectively. However, the SPS for Xinyu 41 decreased by 24.4%− 27.3% and 40.2%− 50.2%, respectively. At 16 DPV6, there was a large difference in SS among different plant densities, the decreases for SS in Xianyu 335 and Zhengdan 958 was lower than Xinyu 41. Xianyu 335 decreased by 3.6%− 15.9%, 18.8%− 32.7% at at 9.0 plants m−2 and 13.5 plants m−2, respectively. Similarly Xinyu 41 decreased by 18.2%− 26.4% and 31.8%− 50.6%, respectively. Thus, under high plant density, the activities of SS and SPS in maize stalks were decreased, the accumulation and conversion of sucrose into cellulose were inhibited, and the activity of cellulose synthesis-related enzymes in varieties with poor lodging resistance were more sensitive to the increase of density. Therefore, we concluded that higher activity of SS and SPS increased cellulose accumulation and stalk strength in maize varieties with better lodging resistance reduced the lodging percentage under close density conditions.

Melatonin mobilizes the metabolism of sugars, ascorbic acid and amino acids to cope with chilling injury in postharvest pear fruit

Chilling injury manifesting as internal browning is an irreversible physiological disorder in cold-stored ‘Yali’ pear fruit, which seriously restricts postharvest handling and commercialization of pear fruit. This study investigated the impact of melatonin (MN) treatment on chilling injury and the metabolism of sugars, ascorbic acid (AsA), proline, and γ-aminobutyric acid (GABA) during chilling injury development in ‘Yali’ pears. The findings revealed that MN-treated pear fruit exhibited considerably improved tolerance to chilling stress compared with untreated control fruit. MN application augmented proline accumulation, likely owing to increased activities and transcriptional concentrations of Δ1-pyrroline-5-carboxylate synthase and ornithine-δ-aminotransferase as well as diminished activity and transcriptional level of proline dehydrogenase. MN immersion increased enzyme activities and gene expression related to the GABA shunt pathway, leading to increased GABA concentrations in cold-stored pear fruit. Furthermore, MN treatment elevated AsA concentrations in the fruit, exerting protective effects against cell membrane damage and mitigating oxidative stress damage caused by chilling. Compared with the control group, MN-treated fruit exhibited reduced neutral invertase enzyme activity and gene expression. Additionally, MN treatment increased both sucrose-phosphate synthase and sucrose synthase activities and gene expression in pear fruit. These observations indicated that the enhanced chilling tolerance of ‘Yali’ pear fruit following MN treatment may be associated with the regulation of proline, GABA, AsA, and soluble sugars metabolism.

Sucrose phosphate synthase 8 is required for the remobilization of carbon reserves in rice stems during grain filling.

Carbon reserve remobilization in stems is closely related to rice grain filling. Sucrose phosphate synthase (SPS) is highly associated with carbon reserve remobilization. In this study, we investigated the expression pattern of SPS genes in various rice tissues, and found that SPS8 is the major SPS isoform in rice stems during the grain-filling stage. We then constructed sps8 mutants using the CRISPR/Cas9 system. The SPS activity of the sps8 mutants was markedly reduced in the stems. In addition, the sps8 mutants exhibited significant starch accumulation in stems. 14C-labelling experiments revealed that the remobilization of non-structural carbohydrates from rice stems to grains was impaired in the sps8 mutants. In the sps8 mutants, grain filling was delayed and yield decreased by 15% due to a reduced percentage of ripened grains. RNA sequencing and quantitative PCR analyses indicated that the genes involved in starch synthesis and degradation were up-regulated in the sps8 mutant stems. In addition, the activity of the enzymes involved in starch synthesis and degradation was increased in the sps8 stems. These results demonstrate that SPS8 is required for carbon reserve remobilization from rice stems to grains, and that its absence may enhance 'futile cycles' of starch synthesis and degradation in rice stems.

Proline interacts with Ca2+-dependent signaling to enhance chromium tolerance in rice by manipulating nitrate reductase and sucrose phosphate synthase

The entrance of chromium (Cr) into the agricultural system would exert a negative influence on the carbon/nitrogen metabolism (CNM) of plants. In this study, we investigated the role of exogenous proline-mediated Ca2+-dependent signaling in the regulation of CNM in rice subjected to Cr(VI) stress, with emphasis on the involvement of nitrate reductase (NR) and sucrose phosphate synthase (SPS). Results demonstrated that proline effectively mitigated the growth inhibition of rice imposed by Cr(VI) stress, which is achieved by a reduction in cytoplasmic Ca and Cr content and the activation of the downstream Ca2+-dependent signaling pathway. Additionally, proline displayed a positive effect in modulating the expression and activities of NR and SPS under Cr(VI) stress, which are attributed to the cross-regulation between calcium-dependent protein kinases (CDPKs) and 14-3-3 proteins (14-3-3s). Consequently, nitrogen use efficiency and sucrose content in rice under Cr(VI) + proline treatments were higher than Cr(VI) treatments. Gene expression variation factors underscored that the regulation of proline on NR is crucial to the Ca2+-dependent signaling pathway, initiated by the interaction between CDPKs and 14-3-3s in rice plants during Cr(VI) stress. These results reveal that proline interacts with Ca2+-dependent signaling pathways to enhance Cr tolerance in rice by regulating NR and SPS.

Phosphorus and naphthalene acetic acid increased the seed yield by regulating carbon and nitrogen assimilation of flax.

To evaluate the impact of phosphorus (P) combined with exogenous NAA on flax yield, enhance flax P utilization efficiency and productivity, minimize resource inputs and mitigate negative environmental and human effects. Therefore, it is crucial to comprehend the physiological and biochemical responses of flax to P and naphthylacetic acid (NAA) in order to guide future agronomic management strategies for increasing seed yield. A randomized complete block design trial was conducted under semi-arid conditions in Northwest China, using a factorial split-plot to investigate the effects of three P (0, 67.5, and 135.0kg P2O5 ha-1) and three exogenous spray NAA levels (0, 20, and 40 mg NAA L-1) on sucrose phosphate synthase (SPS) and diphosphoribulose carboxylase (Rubisco) activities as well as nitrogen (N) and P accumulation and translocation in flax. Results indicated that the SPS and Rubisco activities, N and P accumulation at flowering and maturity along with assimilation and translocation post-flowering, fruiting branches per plant, tillers per plant, capsules per plant, and seed yield were 95, 105, 14, 27, 55, 15, 13, 110, 103, 82, 16, 61, 8, and 13% greater in the P treatments compared to those in the zero P treatment, respectively. Moreover, those characteristics were observed to be greater with exogenous spray NAA treatments compared to that no spray NAA treatment. Additionally, the maximum SPS and Rubisco activities, N and P accumulation, assimilation post-flowering and translocation, capsules per plant, and seed yield were achieved with the application of 67.5kg P2O5 ha-1 with 20 mg NAA L-1. Therefore, these findings demonstrate that the appropriate combination of P fertilizer and spray NAA is an effective agronomic management strategy for regulating carbon and nitrogen assimilation by maintaining photosynthetic efficiency in plants to increase flax productivity.

An efficient triose phosphate synthesis and distribution in wheat provides tolerance to higher field temperatures.

High temperatures in the field hinder bread wheat high-yield production, mainly because of the adverse effects of heat over photosynthesis. The Yaqui Valley, the main wheat producer region in Mexico, is a zone prone to have temperatures over 30°C. The aim of this work was to test the flag leaf photosynthetic performance in 10 bread wheat genotypes grown under high temperatures in the field. The study took place during two seasons (2019-2020 and 2020-2021). In each season, control seeds were sown in December, while heat-stressed were sown in late January to subject wheat to heat stress (HS) during the grain-filling stage. HS reduced Grain yield from 20 to 58% in the first season. HS did not reduce chlorophyll content and light-dependent reactions were unaffected in any of the tested genotypes. Rubisco, chloroplast fructose 1,6-biphosphatase (FBPase), and sucrose phosphate synthase (SPS) activities were measured spectrophotometrically. Rubisco activity did not decrease under HS in any of the genotypes. FBPase activity was reduced by HS indicating that triose phosphate flux to starch synthesis was reduced, while SPS was not affected, and thus, sucrose synthesis was maintained. HS reduced aerial biomass in the 10 chosen genotypes. Genotypes SOKWB.1, SOKWB.3, and BORLAUG100 maintained their yield under HS, pointing to a potential success in their introduction in this region for breeding heat-tolerant bread wheat.
