Synthesis and application of some new heterocyclo‐s‐triazole derivatives as antimicrobial agents

Abstract3‐Aryloxymethyl‐4‐amino‐5‐mercapto‐s‐triazoles (1a–c) reacted with alkyl/aralkyl halides to give 5‐alkyl/aralkyl thio‐s‐triazole derivatives (2a–i) and with substituted phenacyl bromides afforded s‐triazolo‐thiadiazines (3a–l). Condensation of 1a–c with aromatic/heterocyclic aldehydes furnished the corresponding schiff bases 4a–r. The latter compounds (4b,h,n) were further reacted according to Mannich reaction affording 5a–f and 6a–c. Moreover, interaction of 4b,h,n with phenacyl bromides yielded the bicyclic compounds 7a–l. Some of these compounds were screened in vitro for their antimicrobial activity against seven strains of bacteria.

Carbon‐13 NMR spectra of some 4‐substituted phenacyl bromides

AbstractThe 13C NMR signals for some 4‐ substituted phenacyl bromides were assigned. The experimental chemical shifts of the aromatic ring carbons are in close agreement with those calculated using substituent chemical shifts. Both the carbonyl and the α‐methylene carbons exhibit upfield shifts compared with those of the corresponding 4‐substituted acetophenones.

Quaternisation at an Sp2 Nitrogen. II. An Analysis on the Substituent Effect and on the Nature of the Transition State

Abstract The kinetics of N-phenacylation of a number of substituted thiazoles and substituted phenacyl bromides have been investigated in nitrobenzene and in a number of other dipolar aprotic solvents. The rate constants of 2-amino-4-phenylthiazoles and 2-aminobenzothiazoles have been calculated with suitably developed equations. The deviation of the observed values from the calculated results has been ascribed to the steric effect. A seven membered hydrogen bonded transition state has been proposed on the basis of the results obtained from the substituent and medium effect.

Reaction of dienamine 4a with substituted phenacyl bromides gave steroidal[3,4- b] furans 5a–g. The same principle reaction was utilized for the total synthesis of (±) 2 - ( p - chlorophenyl) - 3 - oxa - A - nor - estra - 1,5(10), 9(11) - triene - 17 - acetate 12a. Treatment of 4a, b with benzenediazonium salts, in DMF, followed by a Fischer-indole cyclization yielded steroidal[6, 7- b] indoles 8a–k. Dienamine 4b could be annelated to benz[4, 5, 6] steroids 9a and 9b by reaction with methyl vinyl ketone and crotonaldehyde, respectively.

2-(Acylmethylmercapto)-3-amino- and 2-(acylmethylmercapto)-3-acetamidopyridines

The reaction of 2-mercapto-3-acetamido-5-(or 6-)chloropyridines with phenacyl bromide and substituted phenacyl bromides yielded 2-(phenacylmercapto)-3-acetamidopyridines, while the reaction of the former with γ-chloroacetoacetic ester yielded 2-(carbethoxyacyl-methylmercapto)-3-acetamidopyridlnes.
