Bicalutamide is an inhibitor of the androgen receptor. We examined whether anti-androgen drugs (goserelin and bicalutamide) and surgical castration decreased new cell formation and learning and memory function. Methods A total of 36 adult male S-D rats weighing 300 ¡¾ 50 g were were randomly assigned to the four groups (n = 9 in each group); Group 1: sham-operation, Group 2: orchiectomy group, Group 3: subcutaneous goserelin (goserelin acetate 3.6-mg housed in a 5-mm-long ¡¿ 1-mm-wide biodegradable cylinder) injected, and Group 4: bicalutamide (10 mg/kg daily for 4 wks) administered, respectively. We used immunohistochemistry to 5-bromo-2¡¯-deoxyuridine (BrdU), dobulecortin (DCX) in the hippocampal dentate gyrus, western blot forebrain-derived neurotrophic factor (BDNF), TrkB, Bax, Bcl-2 in the hippocampus. Results Complete suppression of total testosterone was observed with goserelin and orchiectomy. The rats in the orchiectomized group showed significantly suppressed sexual behavior compared to the rats in the control group (p< 0.05). Also, in the goserelin injected group and bicalutamide injection group exerted significant effect on the sexual behaviors. Goserelin and bicalutamide did not show side effects as memory and learning ability impairment although surgical castration did. Conclusions Goserelin and bicalutamide did not show side effects such as memory and learning ability impairment, cell proliferation impairment and neuronal apoptosis in contrast to surgical castration. Medical castration would be used without concern for decreasing in memory and learning function compared to surgical castration.
