ANTIBACTERIAL ACTIVITY Streptomyces spp. ENDHOPHYTIC TO Staphylococcus aureus AND Eschericia coli

The objective of this research was to known antibacterial activity of 4 endhophytic Streptomyces namely Streptomyces griseorubiginosus, Streptomyces vellosus, Streptomyces diastaticus and Streptomyces griseoruber against Staphylococcus aureus and Eschericia coli and measure the biggest antibacterial activity which is indicate by clear zone diameter. Antibacterial activity by using agar plug diffusion method showed that Streptomyces vellosus has capable to inhibits growth of Staphylococcus aureus but not with Eschericia coli and Streptomyces diastaticus has capable to inhibits growth of Eschericia coli but not with Staphylococcus aureus. The clear zone of Streptomyces vellosus and Streptomyces diastaticus are 5,18 mm and 7 mm respectively. Streptomyces griseoruber and Streptomyces griseorubiginosus can’t grow during isolation. Extracellular Synthesis of Selenium Nanoparticles from an Actinomycetes Streptomyces griseoruber and Evaluation of its Cytotoxicity on HT-29 Cell Line.

Selenium nanoparticles (SeNPs) have gained significant importance because of its bioavailability, least toxicity, its interaction with proteins and its biocompatibility. In the present study, the extracellular synthesis of SeNPs was carried out by using culture supernatant of Streptomyces griseoruber, an Actinomycetes member isolated from the soil and cytotoxicity was tested on HT-29 cell line. Culture supernatant was mixed with 1mM sodium selenite for the biosynthesis of SeNPs. Characterisation of the synthesised SeNPs was done by UV-Visible spectrophotometer, FTIR, XRD, DLS and HR-TEM. The cytotoxicity of nanoparticles on HT-29 cell line was studied by MTT assay and with different staining procedure. Bioreduction of SeNPs was confirmed by UV-Visible spectrophotometer that showed the peak at 575 nm. Size and distribution of the biosynthesised SeNPs were analysed by HR-TEM that showed the formation of particle size in the range of 100-250nm. The synthesised SeNPs showed good cytotoxic activity against HT-29 cell line with 40.5%, 33% and 23.7% of cell viability at 2μg/ ml, 4μg/ml and 30μg/ml concentration respectively. The present study reports the simple and eco-friendly synthesis of SeNPs that showed good cytotoxic activity against HT-29 cell line suggesting that biogenic SeNPs could be a potential chemotherapeutic agent.

Streptomyces griseoruber Y1B, a Novel Streptomyces for 1-Hydroxyphenazine Production

Strain Y1B was isolated from a soil sample collected from Wuxi in Jiangsu Province, China. The strain was identified as Streptomyces griseoruber based on phenotypic characteristics and 16S rRNA gene sequence analysis. phzE and phzF gene fragments were amplified by PCR from S. griseoruber Y1B, and showed 70–80% similarity to those of phenazine-producing Pseudomonas and Streptomyces species, indicating that this strain contained phenazine biosynthetic genes and had the potential to produce phenazine compounds. A crude extract, obtained from S. griseoruber Y1B fermentation broth by organic solvent extraction and evaporation under reduced pressure, showed significant antifungal activity against Rhizoctonia solani , Pythiumultimum , and Fusarium oxysporum , and had a broad spectrum of antifungal activity. A phenazine compound, 1-hydroxyphenazine, and a shikimic acid-derived metabolite, benzoic acid, were separated and purified from the crude extract by preparative high performance liquid chromatography. The effects of addition of intermediate metabolites on 1-hydroxyphenazine production implied that the phenazine biosynthesis pathway in S. griseoruber Y1B might branch off from the shikimate pathway, using phenazine-1-carboxylic acid as the phenazine precursor. Furthermore, this is the first demonstration that S. griseoruber can produce phenazine compounds. Therefore, as a novel Streptomyces strain, S. griseoruber Y1B might have potential applications for biocontrol in agricultural production.

A strain of Streptomyces griseoruber isolated from rhizospheric soil of Chinese cabbage as antagonist to Plasmodiophora brassicae

To find potential biocontrol agents for Plasmodiophora brassicae, a total of 63 actinobacteria were isolated from the rhizospheric soil of Chinese cabbage that were collected from Sichuan province in China. Isolates were screened by measuring the resting spores' germination inhibition. The suppressive effects of six antagonistic strains against clubroot on Chinese cabbage in the glasshouse and the field were assessed, respectively. Strain A316 was the most promising antagonist against clubroot, which showed the highest control values of 73.69% in the glasshouse experiment and 65.91% in the field trial. Based on the 16S rDNA sequence analysis, strain A316 was most closely related to Streptomyces griseoruber NBRC 12873 (99.9% similarity). Together with morphological, cultural and physio-biochemical characteristics, strain A316 was identified as Streptomyces griseoruber. The results indicate that the isolate A316 might have the potential possibility to be used as an antagonist against P. brassicae. This is the first report of S. griseoruber for controlling clubroot.

Streptomyces coacervatus sp. nov., isolated from the intestinal tract of Armadillidium vulgare

A Gram-staining-positive bacterium, designated AS-0823(T), which formed spiral spore chains on the aerial mycelium, was isolated from the intestinal tract of Armadillidium vulgare, a small terrestrial crustacean commonly found on the ground around houses in Japan. 16S rRNA gene sequence analysis showed that the isolate belonged to the genus Streptomyces and was most closely related to Streptomyces longisporus ISP 5166(T) (98.6 % 16S rRNA gene sequence similarity), Streptomyces curacoi NBRC 12761(T) (98.4 %) and Streptomyces griseoruber NBRC 12873(T) (98.4 %). The affiliation of strain AS-0823(T) to the genus Streptomyces was supported by chemotaxonomic data: iso-C(16 : 0), anteiso-C(15 : 0), C(16 : 0), iso-C(15 : 0) and anteiso-C(17 : 0) as the major cellular fatty acids, ll-diaminopimelic acid as the characteristic diamino acid in the peptidoglycan and the absence of mycolic acids. DNA-DNA hybridization and physiological and biochemical analysis supported the differentiation of strain AS-0823(T) from S. longisporus JCM 4395(T). Therefore, strain AS-0823(T) represents a novel species, for which the name Streptomyces coacervatus sp. nov. is proposed. The type strain is AS-0823(T) ( = IFM 11055(T) = DSM 41983(T) = JCM 17138(T)).

Gombapyrones, new α-pyrone metabolites produced by Streptomyces griseoruber Acta 3662

Gombapyrones A-D, new members of the alpha-pyrone family of secondary metabolites, were produced by Streptomyces griseoruber Acta 3662, which was isolated from bamboo tree rhizosphere. The strain was characterized by its morphological and chemotaxonomical features and by 16S rDNA sequencing as S. griseobuber. The gombapyrone structures were determined by mass spectrometry and by NMR experiments, and were found to have an inhibitory activity against protein tyrosine phosphatase 1B and glycogen synthase kinase 3beta.

The hedamycin locus implicates a novel aromatic PKS priming mechanism.

The biosynthetic gene cluster for the pluramycin-type antitumor antibiotic hedamycin has been cloned from Streptomyces griseoruber. Sequence analysis of the 45.6 kb region revealed a variety of unique features such as a fabH homolog (KSIII), an acyltransferase (AT) gene, a set of type I polyketide synthase (PKS) genes, and two putative C-glycosyltransferase genes. As the first report of the cloning of the biosynthetic gene cluster for the pluramycin antibiotics, this work suggests that the biosynthesis of pluramycins utilize an iterative type I PKS system for the generation of a novel starter unit that subsequently primes the type II PKS system. It also implicates the involvement of a second catalytic ketosynthase (KSIII) to regulate this unusual priming step. Gene disruption is used to confirm the importance of both type I and II PKS genes for the biosynthesis of hedamycin.

Production of rhodomycins in Streptomyces griseoruber 4620.

The strain Streptomyces griseoruber 4620 produces, besides the anthracycline antibiotics beromycins, some other anthracyclines of the rhodomycin type. Twelve isolates exhibiting a higher antibiotic activity (up to 2.5X), as compared to the parent strain, were obtained after a spontaneous selection. The following species were isolated from the hydrolysate of mycelial extract: beta-rhodomycinone, beta-isorhodomycinone, alpha 2-rhodomycinone, epsilon-rhodomycinone and 10-deoxy-beta-rhodomycinone, which has not yet been described.
